Abstract

Infections caused by Pseudomonas aeruginosa pose a considerable challenge in terms of treatment. The bacterium's key virulence factor, particularly LecB, plays a significant role in bacterial adherence, infections, biofilm formation, and suppression of the host immune response. This study aims to assess the affinity and interactions of hesperetin analogs against LecB in P. aeruginosa. The investigation utilized the molecular docking method, employing a combination of rigid and flexible docking. The docking results successfully identified hesperetin 7-O-glucoside and hesperetin 7-O-rhamnoside, demonstrating superior binding energies compared to MJO as the reference ligand. Rigid docking indicated binding energies of -7.0 and -7.2 kcal/mol for hesperetin 7-O-glucoside and hesperetin 7-O-rhamnoside, respectively. Subsequently, flexible docking revealed lower binding energies for both compounds, reaching -10.3 kcal/mol. These top-performing compounds exhibited interactions with critical residues, including Asn70, Arg72, Thr98, Asp99, Asp96, Asn21, and Ser23, strategically positioned in LecB's substrate-binding site in P. aeruginosa. Notably, hesperetin 7-O-rhamnoside displayed an excellent ADME profile and a favorable safety profile, as it was predicted not to inhibit CYP enzymes. The hesperetin analog also met Lipinski's rules, suggesting its suitability as an oral drug. In conclusion, this research unveils the potential antibacterial activity of hesperetin analogs and provides an opportunity for experimental verification of these compounds as candidates for antibacterial drugs against P. aeruginosa.

Abstract

Malaria is an infectious disease caused by Plasmodium protozoan parasites that transmit via the female Anopheles sp. mosquito. Plasmodium falciparum, Plasmodium vivax, Plasmodium ovale, Plasmodium malariae, and Plasmodium knowlesi are the five Plasmodium species known to infect humans. P. falciparum is the most virulent species, causing the most deaths worldwide. The decrease in efficacy of most antimalarials suggests drug resistance. Therefore, the development of new effective antimalarials, particularly against novel targets, was still required. Cysteine protease falcipain-2 (CPF-2) plays a role in hemoglobin degradation; therefore, inhibiting the activity of this enzyme could be a viable antimalarial target. This study aimed to identify a potential inhibitor for the CPF-2 with PDB ID 3BPF from the ZINC15 database for treating malaria. This study employs a virtual screening workflow with HTVS, SP, and XP docking methods on the Glide Maestro Schrodinger. Based on the glide XP docking score, 10 hit compounds were identified, and their conformational interactions with CPF-2 were compared to the natural ligand (E-64). The binding energy values of hit compounds vary from -7.131 kcal/mol to -8.074 kcal/mol, which is more negative than the E-64 (-6.011 kcal/mol). The three best compounds identified from the ZINC15 database are ZINC000025691540, ZINC000096436101, and ZINC000097797430. All the hit compounds discovered similar interaction with E-64, specifically on the CPF-2 binding pocket residues with Gln36, Cys42, Gly83, Asn173, and His174. All hit compounds exhibit suitable Lipinski rule profiles and are potentially evaluated experimentally as CPF-2 enzyme inhibitor candidates. Abstract

The main objective of this research is to analyze customer preferences of online motorcycle services in East Lombok using the Fishbein multi-attribute model. A deep understanding of the attributes most valued by customers is essential for service providers and local government to improve service quality of the community's needs. The research was conducted using purposive sampling with data collected over three months through interviews, questionnaires, observations, and literature reviews. The results indicate that all evaluation importance levels is categorized as important, with the highest score of 4.24 attributed to the safety and comfort attributes. On the other hand, the evaluation of belief levels (bi) falls into the good category, with the highest score of 3.83 attributed to driver experience. Based on the customers’ attitude analysis, several attributes receive positive scores: the safety and comfort attributes receive the highest attitude score of 15.94, followed by the driver’s experience attribute with a score of 15.87, and finally the ease of use of the online motorcycle application with a score of 15.61. In conclusion, deeper understanding of customers’ preferences could assist service providers and local government to improve the quality of online motorcycle services in East Lombok Regency.

Abstract

Ficus septica is a plant that has demonstrated significant potential as an anti-inflammatory agent. This study evaluates the anti-inflammatory activity and molecular mechanisms of compounds from Ficus septica, focusing on their interactions with Bovine Serum Albumin (BSA) and Cyclooxygenase-2 (COX-2). The results show that the purified extracts from Kadia and Batalaiworu exhibited better anti-inflammatory activity, with IC50 values of 32.04 µg/mL and 32.06 µg/mL, respectively, compared to the crude extracts, which had IC50 values of 42.07 µg/mL and 42.44 µg/mL. Despite this, Diclofenac Sodium exhibited a significantly lower IC50 value of 7.99 µg/mL. Computational studies revealed that Genistein had the strongest binding affinity to BSA with a binding energy of -7.52 kcal/mol, comparable to Diclofenac Sodium (-7.51 kcal/mol). Furthermore, Ficuseptine B and Septicine demonstrated strong binding to COX-2, with binding energies of -9.51 kcal/mol and -9.45 kcal/mol, respectively, outperforming Diclofenac Sodium (-8.49 kcal/mol). These findings suggest that compounds from Ficus septica, particularly Genistein, Ficuseptine B, and Septicine, may serve as potential anti-inflammatory agents through their ability to bind to BSA and COX-2.

Abstract

Speaking is the special moment when the student can be improve their speaking in daily life or their classroom. With speaking the students can practice their knowledge of English and they can get new vocabularies. The objective of the research is to improve speaking skill for the eight grade students of SMP Negeri 1 Pedongga in the academic year of 2019/2020 with the use of role play method. Based on the observation, there were some problems in the teaching and learning of speaking. To solve those problems, the researcher implemented role play as a speaking activity.The research was carried out through action research that consisted of two cycles. Each cycle consisted of two meetings. The results of the research showed that there were some improvements in the students speaking skill. In cycle I, there were some unsuccessful actions. Meanwhile, in cycle II, the implementation of role play techniques was successful in improving the students’ speaking skill. The conclusion of this research were the students low in speaking was caused by lack in vocabulary, self-confident, and they were confused how to pronounced the word. The success in teaching doesn’t depend on the lesson program only, but more important is how to the teacher present the lesson and uses various method to manage the class.

Abstract

TOEFL has played a very important role in predicting students’ academic performance. However, very limited research has specifically addressed the relationship between the TOEFL reading section and students’ critical reading comprehension, as most existing studies predominantly focused on the correlation between overall TOEFL scores and students’ academic performance in general, such as GPA. This research examined the correlation between TOEFL reading score and critical reading comprehension of TBI students at UIN Datokarama Palu. A quantitative approach through correlational design was employed with 30 students as participants. Data were collected by using TOEFL reading test and GPA of critical reading comprehension course of students. Pearson product moment correlation was used to analyze the data. The results revealed a significant positive moderate correlation (r = 0.587, p = 0.01) between two variables. Reflecting that as one variable increases, the other variables tend to increase as well with a meaningful but not perfectly linear relationship. This indicates that students’ proficiency in the TOEFL reading section not only reflects their reading ability in English generally, but also correlates with their capacity to understand, analyze, interpret, and evaluate information in the reading text critically. By integrating TOEFL reading test into students’ critical reading exercises, educators can strengthen students’ critical reading comprehension that emphasizes determining main idea, identifying arguments, and evaluating evidences.

Abstract

Mathematics learning is often considered abstract and less relevant to real life, making it difficult for students to understand the concept. Realistic Mathematics Education (RME) offers an approach that helps students build understanding through real situations. The Peresean tradition with its competition structure and cultural values ​​has the potential to be a concrete and meaningful combination learning context. This study aims to design a realistic mathematics learning design that integrates the Peresean tradition into a combination of materials at the Senior High School level. The main problem behind this study is the low understanding of students towards the combination concept due to the delivery of material that is too abstract and has minimal relevance to real life. The RME approach offers a solution through the use of the real context of the Peresean Tradition which was chosen because it has a competition structure that has the potential to be mathematically modeled in the combination context, such as selecting opponents or compiling match schemes. This study uses a qualitative approach with the type of design research in the development studies category. The research process is limited to the preliminary design stage, which includes context analysis, literature studies, and the development of learning designs. The research results are in the form of a theoretical and contextual culture-based learning design, which is validated reflectively based on RME principles and suitability to the curriculum.

Abstract

The integration of technology into Islamic Education (PAI) in secondary schools encounters challenges such as limited facilities and low digital competence. This study analyzes implementation strategies, supporting and inhibiting factors, and the impact of digital-based PAI learning on student quality in Central Sulawesi. A descriptive qualitative approach was used with data collected through participatory observation, in-depth interviews, and document analysis. Informants included three principals, three PAI teachers, and three students from three schools. Data analysis involved reduction, visualization with N-Vivo software, and validation with informants. The findings show that strategies include principals’ roles in policymaking and supervision, the use of educational videos, the Canva application, and the TPACK (Technological Pedagogical Content Knowledge) model. Supporting factors are leadership support, teacher readiness, and student enthusiasm. Key obstacles are limited technological devices, low teacher digital literacy, and restricted internet access. Digital-based PAI learning improves students’ digital skills, enhances learning outcomes, and strengthens critical thinking. The study concludes that with appropriate strategies and adequate resources, digital-based PAI learning can enhance student competencies. It recommends providing digital literacy training for teachers, ensuring adequate technological infrastructure, and reinforcing digital learning concepts to optimize Islamic Education in secondary schools.

Abstract

Upaya peningkatan mutu pelayanan kesehatan, Puskesmas Lepo-Lepo senantiasa menampung semua saran, keluhan dan pengaduan dari masyarakat yang menyoroti tentang kualitas pelayanan yang telah diterima. Sehingga perlu dilakukan evaluasi kualitas pelayanan. Pengukuran kualitas pelayanani di Puskesmas Lepo-Lepo dapat i diukuri dengani menggunakani metodei servicei qualityi (servqual).i Metodei servquali pertamai kalii dikenalkani olehi Parasuramani meliputii 5i i dimensii yaitui Tangible,ii Reliability,ii Responsiveness,ii Assurance,ii Emphaty.i Metodei inii seringi digunakani untuki melihati sejauhi manai kesenjangani antarai harapani padai suatui layanani dani kenyataani layanani yangi diterima. Tujuan penelitian ini untuk mengevaluasi kualitas pelayanan di Puskesmas Lepo-Lepo apakah sudah  sesuai dengan harapan pasien atau sebaliknya sehingga Puskesmas Lepo-Lepo dapat melakukan perbaikan yang lebih baik. Pada penelitian ini telahi dilakukani wawancarai dengani menggunakani kuisoner kepada 100 responden jumlah pasien Puskesmas Lepo-Lepo tahun 2023.  Hasil dari penelitian ini diperoleh nilai GAP untuk dimensi tangible nilai GAP terkecil -0,57  dan terbesar -0,46, untuk dimensi reliability nilai GAP terkecil -0,49  dan terbesar -0,46, untuk dimensi responsiveness  nilai GAP terkecil -0,51  dan terbesar -0,31, untuk dimensi assurance  nilai GAP terkecil -0,51  dan terbesar -0,32, dan untuk dimensi emphaty  nilai GAP terkecil -0,39  dan terbesar -0,28. Nilai GAP pada tiap dimensi masih negatif menunjukkan bahwa ada beberapa pasien merasa tidak puas terhadap pelayanan Puskesmas Lepo-Lepo. Nilai GAP kemudian dianalisis dengan metode Zone of  Tolerance (ZoT) untuk melihat apakah ketidakpuasan pasien masih bisa diterima atau tidak.  Hasil analisis dengan ZoT  positif  yaitu pada  0,36 – 0,58, dapat disimpulkan bahwa meskipun pasien tidak sepenuhnya puas, kualitas pelayanan yang diberikan masih dalam batas yang dapat diterima.

Abstract

Perkembangan apotek retail modern di Kota Kendari semakin cepat, mempengaruhi dinamika pasar produk farmasi dan perilaku konsumen. Penelitian ini bertujuan untuk menganalisis pengaruh tampilan produk dan persepsi harga terhadap keputusan pembelian di apotek retail modern. Menggunakan pendekatan kuantitatif, penelitian ini melibatkan 100 responden yang merupakan pelanggan di salah satu Apotek retail modern. Data dikumpulkan melalui kuesioner berbasis skala Likert dan dianalisis menggunakan metode deskriptif serta SEM PLS 4.0 dengan software Smart PLS 4.0. Hasil penelitian menunjukkan bahwa tampilan produk dan persepsi harga memiliki pengaruh signifikan terhadap keputusan pembelian. Model yang dibangun memiliki nilai R Square sebesar 0,710, yang menunjukkan bahwa 71% variasi dalam keputusan pembelian dapat dijelaskan oleh faktor-faktor yang diteliti. Nilai Q Square sebesar 0,692 mengindikasikan bahwa model ini memiliki relevansi prediktif yang tinggi. Temuan ini memberikan wawasan penting bagi manajer apotek dalam merancang strategi pemasaran yang lebih efektif dan meningkatkan pengalaman pelanggan.
