Abstract

Pada kucing, infeksi saluran pernapasan atas (URI) dapat dikaitkan dengan infeksi bakteri primer atau sekunder dan umumnya dilakukan pengobatan dengan antimikroba. Penggunaaan antimikroba yang tidak tepat, dan penggunaannya berlebihan. Tidak ada protokol rinci untuk pengobatan, seperti yang tersedia untuk pengobatan manusia. Tujuan penelitian ini adalah untuk mengetahui kepekaan antimikroba dari bakteri yang diidentifikasi pada kucing yang terinfeksi URI. Dilakukan isolasi dan identifikasi bakteri pada sampel kucing yang terinfeksi URI kemudian dilanjutkan dengan uji kepekaan antibiotik amoksisilin, doksisiklin, tetrasiklin, azitromisin, siprofloksasin, dan sefotaksim dengan metode difusi Kirbey Bauer Agar Dics. Berdasarkan hasil penelitian, bakteri yang dapat diidentifikasi adalah Enterobacter spp, Streptococcus spp, Staphylococcus spp, Pseudomonas spp, Seratia spp, Yersinia spp, Micrococcus spp, Klebsiella spp dan Hafnia spp. Ditemukan resistansi antibiotik amoksisilin pada empat isolat Staphylococcus spp dan dua isolat resistan terhadap sefotaksim. Resistansi antibiotik amoksisilin dan tetrasiklin ditemukan pada satu isolat Streptococcus spp, dua isolat resistan terhadap sefotaksim serta dua isolat intermediet terhadap doksisiklin. Satu isolat bakteri Enterobacter spp resistan terhadap amoksisilin, azitromisin dan dua isolat resistan terhadap sefotaksim. Semua isolat yang diuji sensitif terhadap siprofloksasin. Berdasarkan uji kepekaan antibiotik, sebagian besar isolat bersifat sensitif, namun terdapat ada isolat yang resistan terhadap antibiotik, terutama amoksisilin dan sefotaksim.

Abstract

The study aimed to evaluate the effect of the water application of probiotic Bacillus sp. DMP13 on the growth and stress response of giant freshwater prawn larvae Macrobrachium rosenbergii. The larvae used in this experiment were 1-day-old larvae post-hatching with an average body weight of 2.7±0.6 mg and an average length of 2.1±0.1 mm. The larvae were reared in 2 L plastic tanks with a density of 100 L-1 for 21 days. The study was conducted using 4 treatments with three replications of each, namely A (control without probiotic), B (Bacillus sp. DMP13 with cell concentration of 102 CFU (colony forming unit) mL-1), C (Bacillus sp. DMP13 with 104 CFU mL-1) and D (Bacillus sp. DMP13 with 106 CFU mL-1). Probiotic was applied in the rearing media on days 9th up to 18th with 3 days intervals. The results showed that application of Bacillus sp. DMP13 with a cell density of 102 CFU mL-1 was able significantly increased the absolute larvae weight (29.67±1.88 mg), increased larval survival in stress concentrations of 1,250 μL L-1 of formalin solution (the highest larval survival rate 98.3±2.9%). Treatment with 102 CFU mL-1 probiotic cell addition also increasing the total bacterial viable count in the media (the cell density reached 5.99±0.28 log CFU mL-1). However, other parameters such as the larvae stage index, survival rate, and absolute length of the larvae with probiotic treatments were not significantly different from those of control.

Abstract

Catfish Clarias sp. is one of the leading commodities in demand and has economic value. Low survival due to cannibalism and disease is a serious problem. To overcome these problems is application of bioflocs and probiotics Bacillus NP5. This study aimed to analyze the appropriate dose of Bacillus NP5 probiotic addition to the biofloc system to improve the health status, growth performance, water quality, and resistance to A. hydrophila. The catfish fry (Clarias sp.) with an average weight of 1.79±0.05 g was used in the experiment for 40 days, reared in tanks with the dimension of 60×30×35 cm3. The experiment applied a completely randomized design (CRD) of five treatments with three replicates, namely KN (negative control), KP (positive control), BFT (biofloc application without probiotic bacteria), BFT4 (biofloc application with Bacillus NP5 104 CFU mL-1), BFT6 (biofloc application with Bacillus NP5 106 CFU mL-1). Treatments tested were challenged with A. hydrophila density 104 CFU mL-1 by immersion, except the negative control. After 40 days of rearing, biofloc and Bacillus NP5 treatments had a significant effect (P < 0.05) on growth performance, immune response, water quality, total bacteria in water and the gut compared to the control treatment (P < 0.05). In addition, total A. hydrophila in liver, kidney and water were lower (P < 0.05) in BFT4 and BFT6 treatments than the control. The conclusion of this study is that the bioflocs supplemented with Bacillus NP5 improved the growth performance, immune response and resistance of catfish to A. hydrophila infection.

Abstract

Latar Belakang: Penggunaan antibiotik yang tidak tepat pada ternak menyebabkan munculnya resistansi bakteri terhadap antibiotik. Salah satu bakteri yang telah dilaporkan resistan adalah Escherichia coli sehingga pengobatan untuk infeksi bakteri ini menjadi kurang efektif. Tujuan: Penelitian ini bertujuan untuk mendeskripsikan resistansi E. coli patogen yang berasal dari usap kloaka ayam petelur terhadap antibiotik. Metode: Sebanyak 20 sampel usap kloaka diidentifikasi keberadaan E. coli melalui pengamatan makroskopis, mikroskopis, uji biokimia, dan fermentasi karbohidrat uji patogenisitas dilakukan dengan menggunakan media Congo red. Isolat yang patogen selanjutnya diuji kepekaannya terhadap 6 jenis antibiotik dengan uji kepekaan difusi cakram Kirby-Bauer. Hasil: Penelitian memperoleh 15 isolat E. coli dan uji Congo red mendapatkan 4 isolat E. coli patogen. Hasil penelitian menunjukkan bahwa 75% isolat resistan terhadap azitromisin, 100% isolat resistan intermediet terhadap doksisiklin dan siprofloksasin. Sementara itu, 75% isolat masih sensitif terhadap gentamisin dan 100% isolat sensitif terhadap kloramfenikol dan amoksisilin. Kesimpulan: Penelitian ini berhasil mengisolasi E. coli yang bersifat patogen dari ayam petelur yang telah resistan terhadap beberapa antibiotik yang diujikan.

Abstract

Bakteri merupakan satu di antara penyebab terjadinya beberapa penyakit infeksi. Jenis bakteri yang dapat menginfeksi tubuh berbeda-beda tergantung organ atau lokasi target. Organ yang sering diinfeksi oleh bakteri adalah saluran pencernaan. Penelitian ini bertujuan mengidentifikasi jenis bakteri saluran pencernaan dan pola resistansi bakteri terhadap beberapa jenis antibiotik. Feses primata diperoleh dari Kebun Binatang Bukittinggi yang diisolasi pada media Eosin Methylene Blue Agar (EMBA), MacConkey agar (MCA), dan agar darah. Isolat bakteri yang didapat kemudian diuji dengan pewarnaan Gram, dan uji biokimia untuk diidentifikasi. Bakteri Escherichia coli dan Staphylococcus aureus yang didapat dilakukan uji resistansi dengan metode disk Kirby Bauer. Jenis bakteri Gram negatif yang dapat diidentifikasi yaitu Shigella sp., Proteus sp., Escherichia coli, Klebsiella oxytoca, dan Yersinia sp., serta bakteri Gram positif yaitu Staphylococcus aureus, Staphylococcus epidermidis, Streptococcus sp., Stomatococcus mucilaginosus, dan Bacillus sp.. Bakteri E. coli mengalami resistan terhadap antibiotik ampisilin, streptomisin, eritromisin (2 isolat dari 2 isolat), amoksisilin tetrasiklin, oksitetrasiklin, doksisiklin, gentamisin, kloramfenikol dan asam nalidiksat (1 isolat dari 2 isolat), sedangkan bakteri S. aureus hanya mengalami resistansi terhadap antibiotik ampisilin dan amoksisilin (1 isolat dari 1 isolat). Resistansi tersebut dapat terjadi karena penggunaan antibiotik yang tidak tepat, perpindahan gen antarmikroorganisme.

Abstract

Resistensi Escherichia coli terhadap antibiotik telah menjadi permasalahan dalam dunia kesehatan, meskipun E. coli berperan sebagai flora normal, akan tetapi ada beberapa strain bersifat patogen. Tujuan dari penelitian ini adalah untuk mengetahui pola resistensi isolat E. coli dari swab kloaka buffy fish owl (Ketupa ketupu) dari penangkaran burung terhadap streptomisin, tetrasiklin, gentamisin, asam nalidiksat, ampisilin, dan doksisiklin dengan uji difusi cakram Kirby-Bauer. Zona hambat yang terbentuk, dibandingkan dengan Clinical Laboratory Standards Institute (CLSI) 2020. Hasil penelitian menunjukkan bahwa semua isolat telah mengalami resistensi terhadap antibiotik ampisilin, namun masih sensitif terhadap gentamisin. Dua isolat masih sensitif terhadap tetrasiklin, asam nalidiksat, doksisiklin, dan streptomisin. Satu isolat menunjukkan intermediet terhadap streptomisin dan menunjukkan resistensi terhadap tetrasiklin, asam nalidiksat dan doksisiklin.

Abstract

Kegiatan pengenalan dan pelatihan pembuatan eco enzyme pada peternak ayam dan ikan di Kabupaten Tapanuli Selatan, Sumatera Utara, bertujuan untuk meningkatkan pemahaman dan kemampuan para peternak dalam memanfaatkan limbah organik. Eco enzyme, yang dihasilkan dari fermentasi limbah organik seperti sisa buah dan sayuran, digunakan sebagai pupuk organik yang ramah lingkungan, desinfektan rumah tangga, maupun solusi sanitasi untuk peternakan. Program ini melibatkan penyuluhan dan pelatihan langsung kepada peternak ayam dan petani ikan, yang terbukti meningkatkan pengetahuan peserta tentang pembuatan dan aplikasi eco enzyme berdasarkan hasil pengetahuan pre test dan post test. Hasil dari kegiatan ini diharapkan dapat mendukung praktik peternakan yang berkelanjutan dan meningkatkan kesejahteraan ekonomi para peternak.

Abstract

Objective This study aimed to quantify the total bacterial count in salted eggs and identify bacterial species that may affect their quality. Methods Fifty samples consisted of 10 fresh salted eggs that passed the candling test, 10 portions of pasta dough at 0, 3, and 6 hours, and 10 fresh salted eggs that had failed the candling test (black egg yolk). Each sample was tested in triplicate. The total bacterial count was determined using the plate count agar method, and bacterial identification was based on phenotypic analysis, which included Gram staining and biochemical tests. Results The total bacterial load in fresh salted eggs was below the maximum limit set by SNI 7388:2009 (1×105 CFU/g), whereas the pasta dough and black egg yolk exceeded this limit. The identified bacteria included Escherichia spp., Enterobacter spp., Proteus spp., Staphylococcus spp., Pseudomonas spp., and Bacillus spp. Conclusion Although the bacterial count in salted eggs meets SNI standards, the presence of potentially harmful bacteria highlights the need for enhanced hygiene and sanitation measures to be implemented during the production of salt eggs.

Abstract

This study aimed to evaluate the effectivity of microencapsulated synbiotic (MS), Bacillus sp. NP5 and mannan oligosaccharide (MOS) dietary in different feeding frequencies in Pacific white shrimp culture field experiment. The MS was administered as a feed supplementation to enhance the immunity for prevention against co-infection with WSSV (White Spot Syndrome Virus) and Vibrio harveyi. The synbiotic was microencapsulated by the spray dryer method. Shrimps were reared in the floating net cages in the pond. Treatments included the administration of MS at different frequencies i.e, daily (A), twice a week (B), once a week (C), and without MS supplementation (consisted of negative and positive controls) with a feeding rate of 6% of shrimp biomass (5 times a day). During the challenge trial, shrimps were removed and further reared in plastic tanks, for 7 days. The shrimps (except negative control treatment) were intramuscularly injected by WSSV filtrate at the infective dosage of 10-4 copies.ml-1. Twenty four hours after WSSV injection the shrimps were immersed in the water-containing cells suspension of V. harveyi at the cell’s population dosage of 106 CFU.ml-1. Immune responses were observed for 7 days after experimental infection. The shrimps that have been treated with daily MS supplementation (A) showed better immune responses i.e., total haemocyte counts, phenoloxidase, respiratory burst, and the lower pathogenic cells abundance in the intestine compared to other treatments groups.

Abstract

ABSTRACT This study aimed to conduct the characterization and molecular detection of the pathogenicity and antibiotic-resistance genes in Vibrio parahaemolyticus, as the causative agent of vibriosis in Pacific white shrimp. The V. parahaemolyticus isolates were collected from the shrimp’s hepatopancreas, before biochemical test and polymerase chain reaction (PCR) sequencing of the 16S rRNA gene confirmation. The hemolysis test and PCR were applied to detect the presence of virulence genes, namely toxR, thermostable direct haemolysin (tdh), and tdh-related haemolysin (trh). The Kirby-Bauer method was used for characterizing the resistance patterns against ampicillin (AMP), tetracycline (TET), cyprofloxacin (CIP), enrofloxacin (ENR), and chloramphenicol (CHL). The biochemical tests and PCR-16SrRNA gene sequencing confirmed that 12 isolates belonged to V. parahaemolyticus that were further verified by amplification of the toxR gene in 382 bp (100% of the isolates). The alpha hemolysis activity was also confirmed by the amplicon of 199 bp in all isolates. All V. parahaemolyticus isolates showed their resistance to AMP and 42% of the isolates were TET-resistant. However, no resistance was shown to CIP, ENR, and CHL. The PCR-based analysis resulted a detectable resistance gene of ampC (42% of the isolates) and tetB (83% of the isolates). Isolat V. parahaemolyticus dikoleksi dari hepatopankreas, diuji secara biokimiawi dan selanjutnya dikonfirmasi dengan polymerase chain reaction (PCR)-sekuensing dari gen 16S rRNA. Tes hemolisis dan metode PCR diterapkan untuk mendeteksi keberadaan gen virulensi toxR, thermostable direct haemolysin (tdh) and tdh-related haemolysin (trh). Metode Kirby Bauer digunakan untuk karakterisasi pola resistansi terhadap ampisilin (AMP), tetrasiklin (TET), kloramfenikol (CHL), siprofloksasin (CIP) dan enrofloksasin (ENR). Uji biokimia dan sekuensing gen PCR-16SrRNA memastikan bahwa 12 isolat adalah V. parahaemolyticus yang selanjutnya diverifikasi dengan amplifikasi gen toxR berukuran 382 bp (100% isolat). Aktivitas alfa hemolisis juga dikonfirmasi dengan amplikon PCR (199 bp) di semua isolat. Seluruh isolat V. parahaemolyticus menunjukkan resistansinya terhadap AMP, 42% resistan TET, tidak ada resistansi yang ditunjukkan pada CIP, ENR dan CHL. Analisis berbasis PCR menghasilkan gen resistan yang terdeteksi dari gen ampC (42% isolat) dan gen tetB (83% isolat). Kata kunci: Antibiotik, udang, resistansi, virulensi, Vibrio parahaemolyticus
