Abstract

One of the medicinal Zingiberaceae plants which is regularly used by the society as a traditional remedy is turmeric rhizome. The pharmacological properties of turmeric rhizome include anti-inflammatory, anti-immunodeficiency, anti-viral. Anti-bacterial, anti-fungal, anti-oxidant, anti-carcinogenic, and anti-infectious. This study aimed to identify the immunomodulator activity of curcuminoid extract of turmeric rhizome against the increase of leucocyte and its components, lymphocyte and neutrophil in white Galur Wistar rats that had been injected with Staphylococcus aureus. These test animals were given curcuminoid extract in VCO 100 mg/ml on a dosage of 1 ml, 0,5 ml, 0,25 ml curcuminoid powder 200 mg/kg body weight. Curcuminoid was suspended in Na CMC0,5%, Levamisole 25 mg/kg of body weight suspended in Na CMC 0,5%, VCO 1 ml. For the negative control, merely Na CMC 0,5% was given. The extract was orally given daily for 14 days. On the 4th day, Staphylococcus aureus was injected intra-peritoneally. The rats’ blood was then drawn from the heart and was observed to calculate the volume of the leucocytes, lymphocyte, and neutrophil which was conducted using flow cytometry.The findings suggested that there was a significant difference between the immunomodulatory activity of the curcuminoid extract in VCO at the doses of 1 ml, 0,5 ml, 0,25 ml on the increase in leucocyte, lymphocyte, and neutrophil compared to that of the negative control.Therefore, the findings of this study indicated that the extracts of curcuminoid in VCO could increase the number of leucocytes, lymphocytes, and neutrophils. Thus, the curcuminoid extract of turmeric rhizome in VCO potentially possessed immunomodulatory activity.

Abstract

Non-steroidal anti-inflammatory drugs (NSAIDs) are among the most commonly prescribed drugs for pregnant women to treat fever, pain and inflammation. Specific NSAIDs can displace thyroid hormone from its protein binding sites, causing thyroid hormone measurement problems. {1,3 bis (p-Hydroxyphenyl)urea} is a modified p-aminophenol compound with potent analgesic and anti-inflammatory activity and less toxicity. This study is a follow-up to previous research to observe the toxic effect on triiodothyronine hormone levels after administration of {1,3 bis(p-Hydroxyphenyl)urea} compound in pregnant white rats. The toxic effect test was carried out by giving the test preparation to pregnant rats, which had been divided into five groups, namely the normal control group (CMC-Na 0.5%), the positive control (Gabapentin 50 mg/kg BW), Compound {1.3 bis (p- Hydroxyphenyl)urea} at a dose of 50 mg/kg BW, 500 mg/kg BW and 1000 mg/kg BW. Mice were given the test preparation every day from the 6th to the 15th day of pregnancy. Blood was taken on the 16th day, and T3 hormone levels were measured using the ELISA method. The results showed that the T3 hormone levels in the {1,3 bis(p-Hydroxyphenyl)urea group had no significant difference from the normal control group, so it was concluded that {1,3 bis(p-Hydroxyphenyl)urea) did not have a significant effect on the hormone T3.

Abstract

In Indonesia, shrimp underwent a "cold storage" process where the heads, tails, and shells were discarded as waste. This shrimp waste could pollute the environment around the factory so it needed to be utilized. So far, shrimp shells had only been used as ingredients for making crackers, shrimp paste, and animal feed supplements. The remaining shrimp shells that had been separated were made into chitosan which was then subjected to characterization tests. The three steps of the chitosan isolation process were deproteinization, demineralization, and deacetylation. The analytical tests carried out were testing the water content, ash content, and functional groups contained in chitosan which were identified using the Fourier Transform Infra Red (FT-IR) Spectrophotometer. Based on the test results, there was a moisture content of 9.27% ??and an ash content of 1.69% in the chitosan samples made from shrimp shells. Functional groups of shrimp shell chitosan were identified using FT-IR characteristics. The -NH- group, which was represented by the N-H and O-H stretching groups, could be seen in the absorption band 1456.26 in the wave range 3000–30500. The conclusion of this study was that chitosan could be extracted from shrimp shells and meet the characterization criteria.

Abstract

Dendrophthoe pentandra (L.) Miq is a parasitic plant with medicinal potential found in 3000 other plant species. The duku parasite is usually considered a parasite on the duku plant and is often discarded because it is thought to interfere with the growth of the duku plant. The purpose of this study was to see the effect of the duku parasite leaf extract on the reduction of malondialdehyde (MDA) in hyperglycemic rats. MDA levels were measured by thiobarbituric acid-reactive substance (TBARS) assay with doses of duku mistletoe leaf ethanol extract of 400, 200, and 100 mg/kg BW. MDA levels were analysed using rat blood plasma, which was measured using a spectrophotometer at a wavelength of 532 nm. Oral administration of EEDBD 400, 200, and 100 mg/kg BW reduced MDA levels in hyperglycemic rats. However, only EEDBD 400 mg/kg BW showed a significant difference between the negative control (CMC-Na), but with glibenclamide and the normal groups, there was no significant difference.

Abstract

Turmeric (Curcuma domestica Val.) is a traditional Indonesian plant that has many benefits. Curcuminoids are turmeric compounds known to have effects as immunomodulators and have been proven in several in vivo tests. Immunosuppressive drugs such as cyclophosphamide are widely given to improve clinical symptoms in autoimmune patients and cancer treatment which can cause susceptibility to infection.Therefore,this study aimed to determine the immunostimulating effect of curcuminoid extract from turmeric rhizome on cyclophosphamide-induced white rats. Proof of the immunostimulating effect of curcuminoid extracts was by testing the total leukocytes and leukocytesdifferential. The test results showed that curcuminoid extracts of 1 mL, 0.5 mL, and 0.25 mL increased total leukocytes and leukocyte differential (lymphocytes, neutrophils, monocytes, basophils, eosinophils) with a significant difference (p<0.05) to the Na group CMC 0.5% as a negative control. Therefore, curcuminoid extract had the potential to have an effect as an immunostimulant. The test results showed that curcuminoid extracts of 1 mL, 0.5 mL, and 0.25 mL increased total leukocytes and leukocyte differential (lymphocytes, neutrophils, monocytes, basophils, eosinophils) with a significant difference (p<0.05) to the Na group CMC 0.5% as a negative control. Abstract

Abstract. Multidrug resistance is still a major health problem worldwide. Biofilm formation was related to the resistance of bacteria mechanism. Acinetobacter baumannii, one of the bacteria that cause nosocomial infections, is given particular attention due to its high prevalence of resistance. The current study address observing the antibacterial activity and antibiofilm formation activity of Curcuma domestica rhizome extract against multidrug-resistant Acinetobacter baumannii (MDR A.baumannii). Curcuma domestica rhizome was macerated by ethanol 96%. Determination of Minimum Inhibitory Concentration (MIC) was carried out by the microdilution method. Antibiofilm formation analysis was determined by standard quantitative using crystal violet-stained assay. Ethanol extract of C. domestica possesses antibacterial activity against MDR A. baumannii with MIC value 250 µg/mL. Additionally, the ethanol extract of C. domestica could inhibit the biofilm formation of MDR A.baumannii, thus percentage of biofilm formed only 23.7% and significance different with control after analyzed using ANOVA post hoc test Tukey. According to these results, ethanol extract of C. domestica revealed antibacterial activity and had the potency in suppressing the biofilm formation of MDR A.baumannii, thus could be utilized in clinical application, especially in MDR A.baumannii infection.
