Abstract

The present study was to investigated the effects of cow serum supplementation on meiotic resumption of canine oocytes from various estms stage. Selected cummulus oocyte complexes (COCs) were matured in TCM-199 as control and TCM-199 + 10% cow serum as treatment, on incubator with 38C, 5% CO2, and 5% O2. After 72 h incubation and Hoechst 33342 staining, the result shows that cow serum supplement have higher meiotic resumption significantly (P<0.05) in MI achievement han in control and oocyte from proestrusi rave higher meiotic competent(46.5%) comparing oocyte from anestrus(29,4%)s ignificantly(P<0.05) but not significantly(P>0.05) with diestrus stages(34.8%).Keywords: meiosis, oocyte, ovarium, cows serum

Abstract

Hormon tiroid mengontrol pertumbuhan, diferensiasi dan metabolisme semua sel somatik jaringan. Level abnormal hormon tiroid akan menyebabkan infertilitas atau berkurangnya fungsi reproduksi. Hormon tiroid ini kemungkinanm empunyai peranan penting dalam pengaturan steroidogenesis dalam folikel ovarium. Penelitian ini bertujuan untuk mendeteksi level T3 dan T4 sapi bali pada saat estrus dan ovulasi. Sapi bali yang digunakan tiga atau empat ekor dengan umur yang seragam (dua tahun) dan dalam keadaan sehat. Pengamatan estrusdilakukan setiap hari sedangkan penentuan waktu ovulasi menggunakan USG. Sampel darah diambil dari vena jugularis pada saat estrus dan ovulasi. Hasil penelitian menunjuk level T3 1,46ng/dl±0,173 saat estrus dan 1,98ng/ml±0,41 saat ovulasi, level T4 10,43 ±0,442 saat estus dan 10,48±0,896 saat ovulasi sedangkan ratio T3/T4. Level T3, T4 dan ratioT3/T4 tidak berbeda signifikan antara waktu estrus dan ovulasi. Kesimpulan penelitian ini adalah bahwa hormon T3 dan T4 kemungkinan berpengaruh terhadap fungsi ovarium.Kata kunci: triiodothyrosin, estrus, sapi Bali, waktu ovulasi

Abstract

The aim of this research was to know the heart activity of rats (Rattus norvegicus) at some environmental temperature. Ten rats (Rattus norvegicus), male, 2 months old, the average weight of 200 gr purchased at experiment Animal Care Unit Gadjah Mada University, Yogyakarta. Allf rats were adapted in a cage with food and water ad libitum. Furthermore, each tail rat was measured heart activity is heart rate frequences (beats per minute), mean blood pressure (mmHg), systolic blood pressure (mmHg) and diastolic blood pressure (mmHg) at the environmental temperature of 30o C, 32o C, 34o C, 36o C, 37o C, 38o C, and 40o C  measured with a blood pressure cuff BP-98A at coccygea vein for 3 minutes at each temperature. The measurement was repeated 3 times. The data of the  heart activities obtained were analyzed by using one way analysis of variance. It was concluded that the environmental temperature of 30o C was optimal to the heart activities of the rats, including  heart rate frequences , blood pressure, systolic and diastolic blood pressures.Keywords:    heart rate frequences, blood pressure, systolic and diastolic, environment temperature, Rattus norvegicus

Abstract

Digestive dysfunction is a common symptom of diabetes. Diabetes can lead to the loss of enteric neurons as well as nerve dysfunction and gastrointestinal complications. There has been no complete report of mienteric nitrergic of small intestines on hyperglycaemia condition, so the purpose of this study was to assess changes of nitrergic neural profile of small intestines on hyperglycemic Wistar strain rats (Rattus norvegicus). Twelve rats were adapted for 3 days, then they were randomly divided into 3 groups namely group I (K-1, as control), group II (K-2), and group III (K-3). Before treatment all rats were fasted for 12 hours, rats K-1 were induced with 1.0 ml of buffer citrate single peritoneal dose; rats K-2 were induced with 40 mg streptozotocin (STZ)/ kg body weight of single peritoneal dose; and rats K-3 were induced with 45 mg STZ /kg body weight of single peritoneal dose. On the 1st and 5th days after STZ induction, blood was collected through the coccigea vein to measure blood glucose levels and weigh the weight. On the day 5th the rats were uthanated with a high dose of ketamine, dissected and taken small intestines (duodenum, jejunum, and ileum) for staining with Nicotinamide Adenine Dinucleotide Phosphate-diaphorase (NADPH-d). Morphometric changes of nitrergic neurons of the small intestine segments were observed. The results showed that on the 5th day increased blood glucose levels of the K-2 and K-3 groups were significantly higher than K-1 (P <0.01). The weight of three groups did not show any significant improvement difference (P> 0.05). The number of neurons in the duodenum, jejunum and ileum of K-2 group increased higher than that of K-1 and K-3 groups. These results indicate that in hyperglycaemic states, the number of nitrergic neurons increases (K-2 group), and there is a significant decrease in the number of neurons (group K-3) compared with K-1 and K-2 groups (P <0.01). The results of this study concluded, that the STZ induction treatment causes hyperglycemia. Hyperglycemia does not affect to the body weight and the number of  nitrergic neurons of small intestines in Wistar strain rats (Rattus norvegicus).

Abstract

This study was aimed to determine the secure level of the n-hexane extract of kebar grass on hypercholesterolemia rats based the acute oral toxicity test. Based the literature, no report on the toxic dosage before, initial dose was started at 300 mg/kg body weight (BW) and to be increased 2000 to 5000 mg/kg BW. Toxicity data were set for 24 hours and continued until 14 days after treatment for several parameters, i.e : toxicity symptoms, death of animals, changes in body weight and the manifestation of toxicity effects. At the end of the test, the rats were sacrificed and organs were taken for abnormalities (macroscopy). The results of this study showed that the administration of the n-hexane extract of grass kebar at all doses did not show the toxicity symptoms, mortality and body weight change. The absolute and relative weights and gross pathology observation of the internal organ were normal. Conclusions: the n-hexane extract of grass kebar is safe and the LD was determined on category 5, 50 unclassified or minimum practically non toxic

Abstract

Gonadotropine Releasing Hormone (GnRH) is one of the recommended hormones to overcome ovulation problems and it can increase pregnancy rate so that it is used in government programs to increase cattle population in Indonesia, although the results are not yet optimal. The purpose of this study is to compare the composition of bovine GnRH receptor nucleotides with the GenBank reference so that the level of genetic diversity of bovine receptors in Indonesia is known. PCR product sequencing using Promoter F and Exon 1 R primers were further aligned with the reference sequences of Bos Taurus GnRHR mRNA GenBank using the MEGA X program. The results of the analysis found the presence of Single Nucleotide Polymorphism (SNP) in the coding region  of 1st cow  position 38(A> T), 261(C > T), 342(C >T), 411(C >T) and 495(C > T) and 2nd cow positions 261(C >T). Changes in amino acids were also detected in 1st cow position 13 (H> L). The presence of SNP was found to indicate genomic variation between individuals at cattle receptors in Indonesia.

Abstract

The effects of pregnancy and lactation on hematochemical and mineral parameters were evaluated in Ettawa cross-bred goats in Disctrict Kulonprogo, Yogyakarta, Indonesia. Blood plasma samples were collected from 14 th pregnant goats (2 month n=4; 3 month n=6; 4 month n=4) and 16 lactation goats (1 month, n= 4; 2 month, n=5; 3 month, n=3 and 4 month, n=4). Blood serum samples were collected from 18 pregnanct goats (1 month, n=3; 2 month, n=2; 3 month, n=6; 4 month, n=4) and 16 lactation goats (1 month, n=4; 2 month, n=5; 3 month, n=3; 4 month, n=4).  The does averaged 2.5-4 years old and their mean body weights were 30-40 kg. The current result showed that pregnant does recorded higher (p<0.05) values of RBCs and hematocrites compared to lactating does. The higher RBCs during the first and third month of pregnantion and the higher hematocrites values during the first of pregnantion. There were not significant differences in MCV, MCH, MCHC, neutrophyls,  limphocytes, monocytes, WBCs, eosinophyls, and minerals Mg2+, in organic  , Na+, K+ and Cl-during pregnantion and lactation.  The results indicated that pregnantion and lactation has significant effect on RBCs and hematocrites of Ettawa crossbred goats in district Disctrict Kulonprogo, Yogyakarta, Indonesia

Abstract

High fat diet can result in the loss of nitrergic neurons in the myenteric plexus. The study aimed at finding out the effect of high fat diet on the adaptation of nitrergic nerve of rat intestine. It used 15 male rats (Rattus norvegicus) of a month of age with mean body weight of 53.73 gr. The rats were adapted for 7 days to individual cages with ad libitum feeding. After random adaptation, all of the rats were assigned to 3 groups of five rats, namely K-7, K-10, and K-13 groups. Feed and drinking water were given ad libitum. The treatment of the high fat diet lasted for 7 weeks. After the treatment, all of the rats were fasted for 12 hours and then killed. Subsequently, small intestine segments (duodenum, jejunum, and ileum) were taken. The intestinal segments were prepared by using NADPH-d histopathological technique to determine the morphometric changes of nitrergic myenteric neurons. During the treatment the rats were weighed every week and at the end of the study orbitalis vein blood measurement was carried out to see its glucose, cholesterol and cholecystokinin (CCK) plasma levels. The data of body weight, glucose, cholesterol, CCK levels, the total number of the nippergenic myenteric segments of the small intestine were statistically analyzed using Anova. The results of the study showed that the treatment of 7% to 13% fat diets for 7 weeks did not indicate any weight gain and any increase in cholescystokinin level, and any decrease in glucose level. However, it indicated significant increase in cholesterol level. The treatment of 10% and13% fat diets increased the total number of neurons in the jejunum and the ileum. Thus, it was concluded that the treatments of the high-fat diet of the rats (K-7, K-10, and K-13) for 7 weeks had significant effect on the adaptation of the neurons of the jejunum and the ileum.
