Abstract

Paradoxical sleep deprivation (PSD) or sleep disorders can affect sexual function and potentially reduce fertility rates. The active substance of Purwoceng (Pimpinella pruatjan Molk.), namely flavonoid and tannin compounds can improve the quality of function. Drug nanoemulsion is expected to increase the therapeutic effect and reduce toxicity. This study aims to determine the effect of purwoceng nanoemulsion on serum testosterone levels, testicular volume and testicular histopathology of male Wistar rats after stress induction of paradoxical sleep deprivation for 96 hours. This research is an experimental study with a post-test only design with a control group. A total of 24 rats were divided into 6 treatment groups. Group A was given PSD, group B was given PSD and sleep recovery, group C was given PSD and purwoceng extract 25 mg/300 gBW/day, group D was given PSD and purwoceng nanoemulsion 25 mg/300 gBW/day, group E was given PSD and purwoceng nanoemulsion. 50 mg/300 gBW/day, and group F was given PSD and purwoceng nanoemulsion 75 mg/300 gBW/day. Serum testosterone levels were measured by the ELISA method, testicular volume and histopathology determined using Image J software and data were analyzed by One Way ANOVA test and followed by Post Hoc Bonferroni test. Statistical analysis showed that there were significant differences in serum testosterone levels, testicular volume and testicular histopathology (p<0.005). Abstract

Obesity is considered a global health problem that has reached epidemic proportions and significantly affects almost all physiological functions of the body. Swimming can reduce fat levels so that it can reduce obesity conditions which can trigger other cases. However, training that exceeds capacity can cause skeletal muscle damage. This study aims to determine the effect of swimming of various intensities (light, moderate and heavy) on creatine kinase (CK) activity and the histopathological appearance of the musculus vastus lateralis in obese rats (Rattus norvegicus). This study used a true experimental using a Completely Randomized Design (CRD) posttest-only control group design approach. The sample in this research; white rat (Rattus norvegicus) male Wistar strain, body weight 150-200 grams, and aged 6-8 weeks. Obesity induction; Pokphand 551, BOLT, cassavas and carrots. The rats were divided into 5 groups and each group consisted of 4 rats. Swimming is given once per day every morning for 14 days. CK activity was measured using UV-Vis spectrophotometer and histopathological preparations of the musculus vastus lateralis using Hematoxyline Eosine (HE) staining. Swimming significantly (p 0.05) reduced CK activity. The best results were found in group 4 with a reduction of 37.29% from obese controls, and an increase of 11.53% from healthy controls. The best CK activity values were obtained by the group that had values closest to those of healthy controls. The histopathology results of group 4 showed histopathological improvement based on reduced inflammatory infiltration, fiber profile abnormalities and the presence of myofiber hypertrophy as a form of cell adaptation. Group 4 (obesity + moderate intensity swimming) is the best treatment to reduce CK activity and improve the histopathology of the musculus vatsus lateralis.

Abstract

Purwoceng merupakan tanaman obat asli Indonesia yang dipercaya berkhasiat sebagai Afrodisiak (meningkatkan vitalitas seksualitas pria). Purwoceng sebagai afrodisiak mengandung beberapa zat kimia seperti steroid, minyak atsiri, furanocoumarin,flavonoid dan vitamin, yang terdapat pada bagian daun dan akarnya. Tujuan studi pustaka ini adalah   menyusun pemahaman yang lebih komprehensif tentang zat aktif dalam tanaman purwoceng dan manfaatnya bagi kesehatan manusia. Beberapa penelitian dengan hewan coba tikus, menunjukan bahwa zat aktif didalam Purwoceng memiliki efek positif androgenik karena dapat meningkatkan LH, FSH, dan testosteron. Selain itu ekstrak purwoceng memiliki aktivitas antioksidan Bioflavonoidyang memiliki kemampuan menangkap radikal bebas, menurunkan ROS, dan mengurangi stres oksidatif.  Purwoceng memiliki kandungan zat aktif antioksidan yang dapat mencegah stres oksidatif dan bermanfaat meningkatkan kadar hormone LH, FSH, dan testosteron.

Abstract

Background: Vitamin D is thought to have a role in the pathogenesis of acne vulgaris through anti-inflammatory effects. Research regarding the relationship between vitamin D and the severity of acne vulgaris still shows mixed results. Objective: To determine the correlation between vitamin D levels and clinical severity in acne vulgaris patients. Method: This systematic review uses Pubmed, Google Scholar, and Science Direct databases. Of the 362 articles, 13 articles that met the inclusion criteria were analyzed narratively. Results: Of 13 studies, 4 showed significant differences in serum vitamin D levels between degrees of acne severity. Meanwhile, the other 9 did not show any significant differences. One study showed significant differences in tissue vitamin D levels with severity. Conclusion: Overall, results are still inconsistent. There is evidence of a higher risk of vitamin D deficiency and decreased tissue vitamin D levels in people with acne vulgaris as severity increases. It is suspected that this is related to the role of vitamin D in suppressing inflammation. Further studies considering confounding factors are needed to clarify this association.

Abstract

Stress induced by sleep deprivation can increase inflammation and oxidative stress, destroying the pyramidal and neuroglia cells in the prefrontal cerebral cortex and interrupting cognitive and behavioral functions. This study aims to observe the difference in the number of pyramidal and neuroglia cells in the prefrontal cortex of male white rats (Rattus norvegicus) after stress induction by paradoxical sleep deprivation (PSD) and total sleep deprivation (TSD). This study was conducted in the Anatomy Laboratory of the Faculty of Medicine, Universitas Jenderal Soedirman, from November 2019 to February 2020. The method of this study was a posttest-only design with a control group approach using ten rats for each group; that was control (K.I.), PSD (KII), and TSD (K.I.). PSD and TSD groups received sleep deprivation treatment for eight days for 20 hours/day and 24 hours/day, respectively. The mean pyramidal cell number decreased in the PSD (66.67±24.55) and TSD (65.90±34.91) compared to the control (77.10±26.11) group, but no significant differences were found between all groups (p>0.05). The mean neuroglial cell number was lower in the PSD (97.78±28.17) and TSD (75.80±22.39) compared to the control (126.00±48.81). Post-hoc Bonferroni test showed a significant difference between control and TSD (p<0.05) but not between control and PSD or PSD and TSD (p>0.05). In conclusion, there was a significant difference in the number of neuroglial cells but not pyramidal cells in the prefrontal cortex of male white rats (Rattus norvegicus) after stress induction with total sleep deprivation (TSD).

Abstract

Background: Salivary pH can rise or fall influenced by intrinsic and extrinsic factors. Sleep deprivation is one example of intrinsic factors. Sleep deprivation causes a reduction in sleep time at a certain time. Purpose: Analyze the effect of different types of sleep deprivations and sleep recovery on salivary pH. Method: This study was experimental research with a post-test only with a control group design. Thirty white Wistar strain rats were randomly divided into 5 groups: healthy control group (KI), partial sleep deprivation (PSD/KII), total sleep deprivation (TSD/KIII), partial sleep deprivation, and continued sleep recovery (PSD+SR/KIV) and total sleep deprivation and continued sleep recovery (TSD+SR/KV). The treatment is carried out on a single platform method. Salivary pH was measured with the help of color-coded pH strips that were given grading after the completion of sleep deprivation induction. Result: The mean decrease in salivary pH was highest in the TSD group. One Way ANOVA test showed significant differences (p <0.05) in the control group with PSD and TSD, the PSD group with PSD+SR, TSD group with PSD+SR and TSD+SR. Conclusion: Sleep deprivation is proven to reduce the pH of Saliva. Total sleep deprivation is a chronic condition that has the most influence on decreasing salivary pH. The effect of decreasing salivary pH due to sleep deprivation is proven to be overcome by sleep recovery.

Abstract

Cadavers retain significant value within the realm of medicine, particularly in the context of anatomy laboratories, where they serve as crucial educational tools. Cadavers are of significant importance in facilitating the educational experience within the anatomy laboratory. Numerous research investigations have indicated that the decrease in anatomical knowledge is purportedly a contributing factor to the rise in “surgical errors” and subsequent surge in medicolegal litigation. Utilisation of cadavers for medical education and research should be conducted in a discerning, proficient, and optimal manner. At the fundamental level, individuals can access educational materials from sources such as prosection, plastination, or anatomy museums. The primary focus in higher education is placed on cadaveric dissection, particularly in relation to the development of clinical skills relevant to surgical practise.

Abstract

The nasal is a fundamental anatomical feature of humans and serves as a benchmark for beauty ideals. Rhinoplasty is a popular procedure worldwide. References to anatomical elements are crucial when discussing the distinct nasal shapes of each race, particularly in the context of Indonesia. Research is scarce on the nasal morphology of the Javanese ethnic group, resulting in a lack of precise quantitative measurements for their specific nasal form. The study conducted was a cross-sectional investigation with a sample of 50 individuals, consisting of 20 males and 30 females of Javanese ethnicity. The analysis was performed to determine the precise quantities of the three variables under investigation, specifically intercanthal distance, alar width, and base of nasal width. The mean figure at the intercanthal was 2.38 cm, alar base 2.92 cm, and base of nasal width 3.46 cm. The T-test results are significant for the base of nasal width variable at a p-value of 0.002. The acquired data were subsequently juxtaposed with data from various races in Indonesia and other regions, serving as a benchmark for rhinoplasty operations specifically for individuals of Javanese descent in Indonesia. The data reveal a significant gender gap in the Javanese base of nasal width. A study reveals that Javanese nasals are shorter and wider than those of other ethnicities, suggesting that rhinoplasty may be recommended over non-surgical methods.

Abstract

The growing global interest in herbal medicines underscores the importance of safety assessments for traditional plants. Physalis angulata L. (Ciplukan) demonstrates therapeutic potential, yet data on Indonesian extracts remain limited. This study evaluated the acute oral toxicity and determined the median lethal dose (LD50) of Indonesian Ciplukan extract. Twenty-five female Sprague-Dawley rats were divided into five groups (n=5): control (aqueous), and four groups that received extract at 100, 400, 800, or 1200 mg/kg body weight. Over 14 days, researchers monitored clinical signs, body weight, food intake, and organ weights in accordance with Acute Oral Toxicity (OECD Test Guideline 425). Major organs were examined histopathological using the modified International Harmonization of Nomenclature and Diagnostic Criteria. No mortality or significant behavioral changes were observed at any dose. Rats maintained normal body weight gain and food intake. The LD50 was determined to be exceeded 1200 mg/kg, indicating favourable acute safety. However, histopathological analysis revealed significant dose-dependent necrotic changes (p < 0.05) in the lungs, kidneys, liver, and spleen at doses of 400 mg/kg and above. Liver and spleen damage were detected first at this threshold. These findings indicate an LD50 above 1200 mg/kg body weight for Indonesian Ciplukan extract, supporting its acute safety. Nonetheless, subclinical organ toxicity occurred at doses of 400 mg/kg or higher, emphasizing the need for dose optimization in phytomedicine. The study provides regulatory-grade toxicological data to support evidence-based standardization of Indonesian herbal medicines and highlights the necessity for further research on sub chronic and chronic toxicity to establish safe therapeutic doses.
