Abstract

The objectives of this experiment were to evaluate the effects of isoflavone-riched soybean flour with different treatment level of isoflavone on MDA level, SOD activity, and profile of Cu,Zn-superoxide dismutase (Cu,Zn-SOD) spermatogenic cells in the seminiferous tubules of male rat testes. Diet was given as isonitrogen and isocaloric with 10% of dietary protein. Twenty five males of Sprague Dawley rats were divided into five groups and treated with isoflavone-riched soybean flour by oral administration with different concentrations. The treatment was done for 2 months. The optimum concentration of isoflavone was 1.5 mg/day and resulted in : decreased level of MDA testes (2.29±0.05 nmol/g), mantained high SOD activity of rat testes (729.4±23.73 U/g), and increased content of Cu,Zn-SOD by immunohistochemical technique on spermatocytes and early spermatids. It was concluded that isoflavone-riched soybean flour diet (1.5 mg/day isoflavone) can prevent the formation of lipid peroxide and improve the antioxidant status of rat testes.

Abstract

The aim of the study was to observe the ability of indigenous probiotic lactic acid bacteria (LAB) Lactobacillus plantarum 2C12 and Lactobacillus fermentum 2B4 as antidiarrheal agent in rats infected by Enteropathogenic Escherichia coli (EPEC), and also to observe their effect as immunomodulator (malonaldehyde level and proliferation of lymphocyte cell). A total of 90 male Sprague Dawley rats were used for this study and divided into 6 groups i.e.: (1) Negative Control (not infected with of EPEC), (2) LAB L. plantarum 2C12, (3) LAB L. fermentum 2B4, (4) LAB L. plantarum 2C12 + EPEC, (5) LAB L. fermentum 2B4 + EPEC, and (6) Positive Control (infected with EPEC). The treatment of LAB was undertaken from 1st-21st day, while infection of EPEC using 106 cfu/ml per day was undertaken during 8th -14th day. Groups administered with LAB L. plantarum 2C12 + EPEC, LAB L. fermentum 2B4 + EPEC, and positive control, showed decreased body weight during 12th-21st day. At the 21st day, positive control group underwent acute diarrhea (fecal water content was 68.2 % b/b). Statistical analysis with Duncan Test showed that the treatment given to six groups of rats gave significant effect (p<0.05) toward PER value, number of lymphocyte cells and malonaldehyde level in liver and kidney of the rats.

Abstract

The objectives of this research were to evaluate the effect of Lablab protein extract in diet on blood glucose concentration and lipid profiles of alloxan-induced diabetic rats. Two month old male Sprague Dawley rats were divided into 4 groups, containing 5 rats in each group. Three groups were diabetic rats induced by alloxan injection (110 mg/kg of body weight by intra-pheritonial injection) while one group was a normal rat as a control. The experimental groups were normal group, diabetic group, diabetic+cholesterol 0.5% group, and diabetic+cholesterol 0.5% + Lablab protein group. The concentration of rat’s blood glucose were periodically measured during diet intervenion (day 0,14,27, and 42). The result demonstrated that after 42 day of intervention, diet containing Lablab protein extract decreased the blood glucose concentration sharply from 455,75 mg/dl to 104.50 mg/dl (77%), while glucose concentration of control group only descreased from 458 mg/dl to 455.33 mg/dl (0.6%). Lablab protein diet significantly (p<0.05) reduce the concentration of blood glucose as compared to the control group. Lablab protein diet had a significant effect (p<0.05) on the reduction of serum total cholesterol, LDL cholesterol and triglycerides. Lablab protein diet seem to be benefecial for treating hyperglikemia and preventing diabetic complications.

Abstract

Bawang dayak (Eleutherine palmifolia) is an indigenous plant in Borneo traditionally used by Dayak tribes to treat any kind of degenerative deseases including diabetes mellitus. The purpose of this research was to measure antioxidant and antidiabetic capacities of water and ethanolic extracts of bawang dayak bulb. Parameters evaluated in this research were phytochemical screening, total phenolics, flavonoid content, DPPH free-radical scavenging activity, and alpha glucosidase inhibiting (AGI) activity. The result showed that the total phenolics and flavonoid content in bawang dayak ethanolic extract (217.71 mg GAE/g and 65.35 mg QE/g) were higher than that of the water extract (139.93 mg GAE/g and 16.95 mg QE/g). The ethanolic extract also had higher antioxidant and AGI activities (IC50 112 and 241 ppm) than that of the water extract (IC50 526 and 505 ppm). In addition, the IC50 values for AGI in bawang dayak ethanolic extract was lower than acarbose which is known as a commercial antidiabetic agent.

Abstract

Tempe is a potential source of protein for 250 million people in Indonesia. Indonesian soybean demand reached 2.2 million tons per year, and 1.3 million tons of which are used for tempe production. Approximately 70% of the soybeans are imported from different countries. To improve national food security, it is important to use local soybean as a raw material. Therefore, this study used Grobogan local soybean as raw material. The purpose of this study was to evaluate the impact of the consumption of tempe (from local soybean Grobogan) in the long term, ie over a 90-day subchronic test using rats as a model. Parameters measured were serum biochemical profile, hematology, malonaldehyde (MDA), and superoxide dismutase (SOD) in liver and kidney organs of rats. This research was conducted using three groups of rats were fed tempe, boiled soybean, and casein as a source of protein in the ration. Data feed convertion efficiency showed tempe of Grobogan soybean had a better quality protein than that of boiled soybean and casein. As compared to the groups of rats fed boiled soybean and casein, the tempe group had higher HDL cholesterol, but not significantly different in hematological parameters, MDA and SOD in liver and kidney organs. This suggests consumption of tempe made from Grobogan soybean in the long term does not have a negative impact on health.

Abstract

The aim of this study was to evaluate acute toxicity of methanol extract of Sargassum echinocarpum on male and female of mice (Mus musculus) strain BALB/c two month age. S. echinocarpum was obtained from Talango Island in April 2008. The powder of S. echinocarpum was macerated by methanol (1:3) at 4 °C for 3 x 24 hours, then concentrated, dried, and extracted. The extract was screened to observe the phytochemical compounds. For acute toxicity extracts was given orally and once on one hour before feeding. The dose treatments were 0; 625; 1250; 2500; and 5000 mg/kg body weight (BW). Parameters measured were phytochemical contain, body weight during the study, 50 percent of lethal doses, and histology score of liver and kidney. The phytochemical study showed that methanol extract of S. echinocarpum contained tannins, polyphenols, saponins, glycosides, and steroids. On 1250 mg/kg BW or more, methanol extract of S. echinocarpum inhibited the increasing of body weight on mice, but, until 5000 mg/kg BW, it did not cause mortality on mice. On 1250 mg/kg BW or more, methanol extracts caused necrosis on hepatocyt and tubulus.Key words: acute toxicity, methanol extract, Sargassum echinocarpum

Abstract

Fish oil production in Indonesia is not only directly sourced from fish raw materials, but also could beprocessed from by-products of canning and fish meal through purification process. The result of purificationwith one process was not satisfy yet for the parameter oxidation quality. Therefore, fish oil from by-productsof fish meal needs to be purified more intensively, such as stratified purification. The aim of this study wasto improve the quality of fish oil from by-product through stratified purification. The multilevel purificationof once purification (P1), twice purification (P2), and three times purification (P3) gives varying results onphysical properties, oxidation parameters, and fatty acid profiles. The color of the brightest fish oil was foundin sample P2, the lowest density was found in sample P3 (0.63 g /cm3), the highest yield was in P1 sample(44.42%), FFA was lowest in P3 sample (0.37%), The lowest PV was in the P1 sample (25.59 meq / kg),the lowest TOTOX was in the P1 sample (48.11 meq / kg), and the highest PUFA content in the fatty acidcomposition was found in sample P2 (37.02%). Purified sardine oil from by-products of fish meal conductedby three-stage purification, which parameters have met the criteria of IFOS 2014 standards, while for PV,AnV, and TOTOX parameters have met IFOMA standards.

Abstract

ABSTRACT. The objetive of this study was to evaluate the effect offeeding with multimicronutrients fortified foods on anemia status and weight gain ofpregnan womrql. This study was carried out in three sub-districts in Bogor, i. e.: Leuwiliang, Leuwisadeng and Ciampea. A double blind randomized control trial was assigned on this study. About 210 pregnant women from these sub-districts were selected for this study and devided into 2groups, (1) 70 pregnant women received fortified foods with multi-micronutrients i.e. iron, iodine, zinc, folic acid, vitamin C, and vitamin A, and (2) 70 pregnant mothers received nonfortifiedfoods, each for 6 mounths. Product chosen as intervention carrier consisted of vermicelli, milk, and biscuit. Distribution ofproducts were carried out every week to thetarget pregnant women. The results of the study showed: (a) intervention could reduce prevalence ofanemia as much as 28% compared with placebo (b) Intervention affected the micronutrients consumption andfulfilled: iron 55,5% RDA, vitamin A 65% RDA, vitamin C 126% RDA, folat 15,3% RDA, zinc 26,7% RDA, iodium 26,3%, (c) intervention didn'tinfluence weight gain during pregnancy.

Abstract

Indonesia placed the fourth biggest diabetics in the world after India, China, and the USA with prevalence amounting to 8.6% of the population. Diabetes is an abnormal carbohydrate metabolism. Therefore, nutrition plays a key role in the management of the disease. This study aimed to find hypoglycemic sweet potatoes and appropriate processing to create low glycemic foods. Eight Indonesian sweet potato varieties/clones were used in this experiment, i.e. Kidal, Sukuh, Sari, Ungu, Jago, BB00105.10, B0464, and BB00106.18. Samples were firstly analyzed for their physicochemical and nutritional properties, which then followed by evaluation of their hypoglycemic responses. The selected variety was processed into three different basic processing methods, i.e. boiling, baking, and frying, and then their glycemic indexes (GI) were evaluated. Result showed that among eight sweet potato varieties/clones studied, BB00105.10 clone indicated the best hypoglycemic response. The highest hypoglycemic activity was supported with the highest resistant starch content (3.8%), protein content (5.47%), and low starch digestibility (51.4%). The sweet potato tubers contained medium to high amylose (24.94%). Processing methods influenced the GI value of foods. Fried sweet potatoes had the lowest GI (47), followed by the boiled one (GI = 62) and the baked one (GI = 80).

Abstract

Prevalensi penyakit degeneratif seperti diabetes melitus (DM) terjadi akibat perubahan gaya hidup masyarakat. Saat ini, jumlah penderita DM di Indonesia sekitar 14 juta jiwa. Diet pangan ber-indeks glikemik (IG) rendah akan membantu dalam  pencegahan primer dan pengendalian DM. Penelitian ini bertujuan untuk mengevaluasi penurunan IG beras dan perubahan mutu gizinya akibat proses pratanak. Prinsip proses pratanak yaitu perendaman gabah di dalam air (60ºC, 4 jam), pengukusan (20 menit), pengeringan I (100ºC, k.a. 18-20%) dan pengeringan II (60ºC, k.a. ≤ 12%). Hasil penelitian menunjukkan proses pratanak dapat meningkatkan kadar amilosa (15,44-26,32% menjadi 19,35-27,23%) dan serat pangan (4,67-7,57% menjadi 8,19-10,27%), tetapi menurunkan daya cerna pati in vitro (62,21-78,63% menjadi 36,40-49,74%). Proses pratanak menunjukkan hasil positif untuk memproduksi beras IG rendah, karena kemampuannya dalam menurunkan IG (54,43-97,29 menjadi 44,22- 76,32). Reducing glycemic index on some rice varieties using parboiled processing.Degenerative diseases such as diabetes mellitus (DM) become prevalence to the people' health due to the changing of lifestyles. Nowadays, the number of Indonesian diabetics around 14 million people. Diet of low glycemic index (GI) foods will assist in primarly production and controlling of DM. This research was aimed to evaluate reducing of GI rice and its nutritional quality changing due to parboiled processing. The basic process of parboiled rice i.e. rough rice steeping in waters (60·e, 4 h), steaming (20 minutes). drying I (100 °C. m.c. 18-20%) and drying II (60·e. m.c. d" 12%). Result showed that_parboiled processing increased rice amylose (15.44-26.32% to 19.35-27.23%) and dietary fiber content (4.67-7.57% to 8.19-10.27%), but reduced in in vitro starch digestibility (62.21-78.63% to 36.40-49.74%). Parboiled processing shared positive result in providing low GI rice, because of its capability in reducing GI (54.43-97.29 to 44.22-76.32).
