Abstract

Tujuan penelitian ini adalah mengamati kemampuan yogurt sinbiotik yang mengandung probiotik lokal (Lactobacillus acidophilus 2B4) sebagai antidiare pada tikus yang terinfeksi Enteropathogenic Escherichia coli (EPEC), dan untuk mendeteksi dampak terhadap karakteristik imunomodulator (jumlah sel limfosit, kadar malonaldehid/MDA hati, dan aktivitas antioksidan superoksida dismutase/SOD hati). Infeksi EPEC (10 7 cfu/ml/hari) yang dilakukan secara oral ke tikus selama tujuh hari berturut-turut secara nyata menyebabkan diare ringan tanpa penurunan berat badan. Pemberian secara oral yogurt sinbiotik yang mengandung 10 9 cfu bakteri asam laktat/ml/hari selama 21 hari secara nyata meningkatkan respons imun tikus, yang ditunjukkan dengan meningkatnya sel limfosit di hari ke-14, penurunan MDA hati pada hari ke 14 dan 21, dan meningkatnya aktivitas SOD hati pada hari ke-14.

Abstract

Stress condition has been reported decreased the level of intracellular antioxidant, especially superoxide dismutase (SOD). The condition can lead to certain cell impairment and finally can reach to the degenerative processes, such as aging and carcinogenesis. We need several alternative treatments to solve the severe problem under antioxidant defiency status caused by stress condition. The aim of the study was to observe the effect of α-tocopherol on increasing the profile of superoxide dismutase (SOD) in the kidney of rats under stress condition. The stress condition was achieved by five days fasting together with smimming for 5 min/day and only drinking distiled water ad libitum. α-tocopherol was orally administrated at a dose of 60 mg/Kg/BW/day for seven days. By measuring SOD activity and Cu,Zn-SOD imunohistochemically, showed that α-tocopherol has effect on increasing the profile of intracellular antioxidant in the rats kidney tissue under stress condition.

Abstract

Intelligent is one of human quality factors that can be influenced by nutrition. Iodine is one of trace element nutrition that necessary for intelligent level. The purpose of this research was to learn the effect of seaweed as iodine source on rats learning ability. The rats were divided into four groups based on the ratio: A (without potassium iodide/KI-seaweed flour (SF) 0%), B (with KI-SF 0%), C (without KI-SF 5%) and D (without KI-SF 10%). The ratio had no significant effect on rats growth, but significant on iodine retention, neuron cell number and rats learning ability. Iodine retention value had a positive correlation with the neuron cell number. The neuron cell number of group A, B, C and D were 72.83, 89.33, 93.92 and 100.83 cells per view area, respectively. The number of neuron cells gave a significant influence on rats learning ability. Total time needed by the rats in food retrieval test for the group A, B, C and D was 57.84, 38.17, 33.39 and 20.55 seconds, respectively. The more neuron cell number, the more learning ability.

Abstract

This study was conducted to evaluate the activity of ginger oleoresin (Zingiber officinale) on the intracelluler antioxidant-superoxide dismutase (SOD) in the liver of rats under stress condition. A total of twuenty five Wistar rats were used for this study. They were divided into five groups ; (1)K (control), (2) S (stress), (3)O + S (oleoresin followed by stress), (4) S + O (stress followed by oleoresin and (5) O + S +O (oleoresin followed by stress then oleoresin). The dose of oleoresin is 60mg/Kg/BW/day for seven days. Stress condition was done by five days fasting and smimming for five minutes/day, while drinking water was provided ad libitum to all groups. The results showed that ginger oleoresin significantly decreased malonaldehyde (MDA) and elevated SOD activity. The immunohistochemical evaluation also showed that ginger oleoresin increased the content of copper,zinc-superoxide dismutase (Cu,Zn-SOD) in the liver of rats under stress condition. These effects were showed in the tissues of rats treated by ginger oleoresin before or after stress or combination of both.

Abstract

Sea cucumber (Holothuria scabra J.) is one of fishery commodities whichcontain high nutritional and economic value. This study aims were 1) thephysical characteristics of sea cucumber, 2) nutrient content (proximate) seacucumbers fresh meat, and 3) the yield and nutrient content (proximate) seacucumber powdery fresh meat. The study was conducted in four stages of theexperiment: 1) Preparation of meat of sea cucumber, 2) analysis of nutrientcontent (proximate) sea cucumber fresh meat, 3) The making of powder of seacucumber meat , and 4) analysis of yield and nutrient content (proximate) seacucumber meat powder. Results showed the proportion between the parts of thebody meat: offal and gonads: the skin: water and dirt were 4:3:2:1 (w / w). Freshsea cucumber meat has a high nutrient content that is 87.03% water, 9.94%protein, 0.54% fat, 1.86% ash, and 0.64%. carbohydrate. The average yieldproduced approximately 10.16%. While the nutrient content (proximate) powderysea cucumber meat that is 9.13% water, 61.31% protein, 3.68% fat, 12.52% ash,and 13.36% carbohydrate.

Abstract

Virgin Covonut Oil (VCO) used to  be produced with various methods and each methods claims ait advantege especially in its lauric acid content. The objective of this research was to study the effect of heat application in producing VCO and the efficacy of result VCO in reducing blood glucose of diabetes mellitus (DM) rat of sprague Dawley. Three types of samples were used in the study, i.e., (1) VCO produced without heat application and (2) VCO producd with controlled heat application and (3) Ordinary coconut cooking oil produced with the application of severe heating as well as typical chemical tretment. Each sample was introduced orally to each group consisting of 5 rats. Other two groups consisting of 5 rats were also used as control, i.e., (1) negative control was of healty rat and (2) positive control was of DM rats. The animal study was conduced for 28 days and the observation was carried out on the amount of feed consumed, body weight and blood glucose. At the end of observation, all the rats were terminated and analysis was carried out on blood chorestrol level. This study revealed no significant different between VCO and Coconut Cooking Oil in body weight gain effect. The content of bioactive conpound of Lauric Acid in two types of VCO and Coconut Cooking Oil were also not significantly different. However, the VCO produced without heating showed the most significant effect in reducing blood glucose (corellation value of-0,99 and significant value at a a=1%). All the VCO samples were also not significant in affecting the level of total cholestrol, HDL, LDL, and triglyceide of blood serum of DM rat.

Abstract

Virgin Coconut Oil (VCO) can be produced by different methods  related to health function. The objective was to study the efficacy of VCO processing method in reducing blood glucose level, cholesterol profile and pancreatic tissue profile of diabetic mellitus (DM) Sprague dawley rats. Four rat groups from five rat groups were injected using alloxan to induce the diabetic condition, while the rest was negative control. VCO-A produced without heat and VCO-B produced with controlled-heat treatment and Coconut Cooking Oil (RBD-CNO) produced with the application of severe heating and chemical (MG) were fed respectively to three rat groups, while the other two rat groups were fed by water and used as negative control (healthy rats) and positive control (DM rats). During 28 days observation, all of the rats blood glucose level were assessed, and at the end of the observation day the rats were terminated to assess the cholesterol and pancreatic tissues profile. VCO-A significantly positive reduced blood glucose level than the others.  VCO did not influence DM rats cholesterol profile and all VCO inhibits the rate of pancreatic β cells damage. On 28 days observation all VCO treatments suppressed blood glucose level of DM rats.

Abstract

Diabetes mellitus is a spectrum of inherited and acquired disorders characterized by elevating blood glucose levels. Diabetes is an abnormal carbohydrate metabolism, therefore, diet therapy for diabetics plays a key role in the management of the disease. Most Indonesian people consume rice as source of energy and protein. Generally, diabetics consume very limited rice because they believe that rice is the one of hyperglycemic food, even though different rice varieties have large range of glycemic index. The study aimed to evaluate hypoglycemic properties by using rat assay and chemical characteristics of 10 Indonesian rice varieties, i.e. Pandan Wangi, Rojolele, Bengawan Solo, Cenana Bali, Memberamo, Celebes, Ciherang, Batang Piaman, Cisokan, and Lusi. Taj Mahal, an herbal ponni imported rice, was used as a comparison. Male Sprague Dawley rats (150- 200 g body weight) were used for hypoglycemic assay. The rats were fasted overnight before the blood glucose was measured in the morning. The rats were then feed with 4.5 g rice per kg body weight by oral administration, followed by 1 ml of 10% glucose solution in the next 30 minutes. The blood glucose was measured for the next 30, 60, 90, and 120 minutes. Changes in blood glucose concentrations (mg dl-1) before and after the oral administrations were calculated for each rice variety tested. Results showed that Cisokan and Batang Piaman were categorized as low glycemic responses and Ciherang as high glycemic response, while the other varieties (Memberamo, Cenana Bali, Lusi, Bengawan Solo, Pandan Wangi, Celebes, and Rojo Lele) showed moderate glycemic responses. As the best hypoglycemic activity, Cisokan contained high amylose (27.6%), fat (0.87%), total dietary fiber (6.24%), resistant starch (2.02%), and lowest starch digestibility (52.2%), which are ideal for diabetic’s consumption. Ciherang as the worst hypoglycemic activity had low resistant starch (1.78%), low total dietary fiber (4.52%), and medium amylose (23.0%). This study implies that Cisokan variety is suitable for diabetic’s consumption.

Abstract

Macro and microvascular disease are currently the principal causes of morbidity and mortality in diabetes mellitus patients. Loss of the modulatory role of  the  endothelium  may  be  a  critical and  initiating factor  in  the  development of diabetic  vascular  disease.  Polyphenol is known behave protective active compound to function endothelium cell.  Polyphenol that contained in brown algae was phlorotannin.  The  aim  of  the  present  study  was  to  investigate  the effects  of  S.  echinocarpum extract  on  endothelial  dysfunction  in  aorta  of streptozotocin  diabetic  rats.  This experiment was conducted by complete randomized design in 2 months old of male albino Sprague Dawley strain of rats, 125-150 grams body weight. The rats divided in fivegroups, each group consist of  6  rats.  First  group:  normal  which  is  given  1  mL  of  extract  vehicle  orally; second  group:  diabetic  rats  which  is  given  1  mL  of  extract  vehicle  orally;  third group:  diabetic  rats  which  is  treated  extract  150  mg/kg  BW  dose  orally;  fourth group: diabetic rats which is treated extract 300 mg/kg BW dose orally; and fifth group: diabetic rats which is treated extract 450 mg/kg BW orally. Diabetic rats prepared by injection of 45 mg/kg  streptozotocin  intraperitoneally.  Extract was given to diabetic rats during 90 days.  The  result  showed  that  brown  algae extract  rearrange  endothelium  cell  function  of  diabetic  rats  by  increasing relaxation,  releasing  of  EDRF  and  decreasing  of  MDA.  Based on these observations,  it  was  concluded  that  S.  echinocarpum extract  recovers  the endothelial  dysfunction  associated  with  diabetes.  This effect appears to be  due to its antioxidant properties.Key words: diabetes, EDRF, endothelial dysfunction, MDA, S. Abstract

High level of blood glucose is an indicator for diabetes mellitus (DM) condition. The condition iscaused by low level of insulin secretion or impairement of insulin receptor. The number of DM patientincreases every year. The World Health Organization reported that the number of DM patient in Indonesiawas the 4th highest in the world, after following China, India, and the United States of America, respectively.This study was conducted to analyze the effect of sea cucumber (Holothuria scabra J) on the profile ofantioxidant copper, zinc-superoxide dismutase (Cu, Zn-SOD) in the pancreatic tissues of diabetic rats. Atotal of 25 male white rats (Sprague Dawley) were used in this study. They were divided into five groups;(1) negative control (KN), (2) positive control, diabetic rats (KP), (3) diabetic rats treated with hydrolyzatedprotein of sea cucumber (HDL), (4) diabetic rats treated with concentrated protein of sea cucumber (KST),and (5) diabetic rats treated with isolated protein of sea cucumber (ISL), respectively. Diabetic conditionwas obtained by alloxan injection 110 mg/kg bw. The treatments were done for 28 days. At the end oftreatment period, the rats were sacrificed and pancreatic tissues were collected and fixed in Bouin solution and then processed to paraffin embedding standard method. The tissues were then stained withimmunohistochemical staining techniques using monoclonal antibody of Cu, Zn-SOD. The results showedthat treatment of HDL, KST, and ISL of sea cucumber (Holothuria scabra J) increased the content ofantioxidant Cu, Zn-SOD either in Langerhans islets and acinar cells of pancreatic tissues-diabetic rats.The HDL of sea cucumber treatment gave the best effect in increasing the antioxidant content of Cu, Zn-SOD in pancreatic tissue of diabetic rats.
