Biodiversitas 22: 5425- 5431. Research on the development of black rice has not received much attention. The anther culture technique can shorten the plant breeding process and produce doubled haploid (DH) lines. The target of this study was to evaluate DH black rice lines with early maturity, good agronomic performance, and high yields. Twenty-seven doubled haploid lines and three check varieties (Aek Sibundong, Inpari 24, and Jeliteng) were evaluated at two locations (Kediri, East Java, and Bogor, West Java) from January to July 2020. A randomized complete block design was applied to this study with three replicates. The combined analysis results showed that the genotype effect (G) was highly significant (p < 0.01) for all characters. In addition, all characters had a high broad-sense heritability. Characters that were positively correlated with yield based on genotypic and phenotypic correlation were the number of productive tillers, the number of vegetative tillers, panicle length, the number of filled grains, and the number of total grains. The 1000-grains weight and panicle length had a positive and the largest direct effect on yield. Principal component analysis showed that the number of productive tillers and the 1000-grains weight were related to yield. The selection index model that can be formulated is as follows I = (3*0.375*Yield) + (0.723* 1000-grains weight) + (0.280* Number of productive tillers). Based on the selection index, 14 genotypes were selected.

Breeding programs to obtain superior upland rice varieties with high productivity and adaptive in dryland must be prioritized to maximize the potential of dryland. This research aimed to obtain information on the agronomic performance of upland rice lines in advanced yield trials and select the best lines with high productivity and tolerance to drought stress and aluminum toxicity. Advanced yield trials were conducted from November 2016 until March 2017 in Bogor and Sukabumi using a randomized complete block design, where three replications were nested in the environments. Aluminum and drought stress evaluations were conducted at Muara Research Station, Bogor, in May-June 2018 and September-October 2018. Based on the selection index, twelve lines were selected with superior agronomic characters and high yield potential. These lines had characteristics as follows: days to harvesting (110.2-116.0 days), number of filled grains (70.6-101.3 grains), number of unfilled grains (27.9-58.4 grains), and productivity (2.2-2.9 tons ha-1). Evaluation of drought tolerance showed four lines with better drought tolerance than the drought-sensitive check IR20. The aluminum tolerance evaluation obtained two tolerant lines and nine lines with moderate responses to aluminum toxicity. The selected lines need to be further evaluated in multilocation trials.

Application of selection index for rice mutant screening under a drought stress condition imposed at reproductive growth phase. Biodiversitas 23: 5446-5452. Drought is a condition of limited availability of water that can reduce rice growth and productivity, and may threaten food security. Therefore, efforts are needed to produce drought-tolerant rice genotypes. This study aimed to select drought tolerant of the M3 mutant upland rice population at the reproductive growth phase. This research was carried out in dryland at BATAN, Indonesia, from September 2020 to February 2021, using an augmented design. The study employed 360 ??genotypes of mutant upland rice and 5 check varieties, namely Salumpikit, IR20, IR64, Limboto, and Situgintung. The selection index was constructed based on multivariate analyses such as correlation, path, and Principal Component Analysis (PCA). The coefficient of the selected main component can be used as a selection index. Before selecting the character values of each genotype, the values were standardized to obtain the same degree of value. The results showed that grain weight per hill (W.G.), panicle density (P.D.), panicle length (P.L.), and the number of filled grains per panicle (NFG) could be used as selection criteria for mutant upland rice tolerant to drought at reproductive growth phase. The selection index formula was I = (5*0.44*WG) + (2*0.41*PD) + (0.23*PL) + (0.42*NFG). Selection of genotypes of mutant upland rice based on weighted index selection resulted in 89 genotypes with high yields and good agronomic characteristics so that they could be tested in further research.

Rice is the staple food of most Asian, including Indonesian people. Plant breeding plays an important role in producing high-yielding varieties of rice to meet national carbohydrate sufficiency. Green Super Rice (GSR) is an alternative for rice variety development that has the advantages of high yield, good quality, pest resistance, and efficient fertilizer use to support sustainable agriculture. This study aimed to evaluate the agronomic character of GSR doubled haploid (DH) lines derived from anther culture and determine the best GSR rice lines based on index selection. The treatment was 65 genotypes consisting of 60 DH of GSR lines from anther culture of 5 combinations of crosses and 5 check varieties. The study used a randomized complete block design with three replications. The results showed that most of the tested lines had similar agronomic performances to the check variety Inpari 42 Agritan GSR. Almost all agronomic characters of the GSR DH lines had high genetic variability. Most characters had a high category of broad-sense heritability except panicle length. Line selection using index selection for economically important agronomic characters yielded 27 GSR DH lines having high productivity, number of productive tillers per hill, number of total grains per panicle, and percentage of filled grains per panicle.

Development of superior rice varieties requires selection of the best lines having good agronomic characters and high yields. This study aimed to select high-yielding doubled haploid rice lines obtained from anther culture through index selection and agronomic characters. The study was conducted using a single-factor randomized complete block design (RCBD) with genotype as treatment. Sixty doubled haploid lines and 5 check varieties were evaluated along with three replications, hence, the total experimental unit was 195. Analysis of variance showed that the genotypes had a significant effect on all observed characters, except for the number of productive tillers. The agronomic characters observed had a high broad sense of heritability, except for the number of productive tillers which was in the medium category. Furthermore, index selection was carried out based on productivity, the number of productive tillers, the total number of grains, as well as the percentage of filled grains. Based on the index selection, 29 lines were selected among the 60 doubled haploid rice lines tested. These lines had the range in productivity from 2.95 to 6.77 ton ha-1, the number of productive tillers from 12.3 to 20.8 tillers, the total number of grains from 135 to 275.4 seeds, the filled grain percentage from 53.23 to 87.61%, and the selection index from -1.48 to 7.88. The selected lines will be further tested at the next experimental stage in several locations to produce high-yielding, brown planthopper-resistant, and drought-tolerant rice varieties.

Landraces play an important role in the local food security and sustainable development of agriculture as a source of genes that control important traits adaptive to environmental conditions. The study aimed to characterize the agro-morphological and molecular characteristics of 22 rice landraces of East Timor (Timor Leste). The research was conducted in April-August 2018 in Caibada-Baucau-East Timor. Soil analysis was conducted at the Soil Laboratory of the Agronomy and Horticulture Department, IPB University. Materials used were 22 rice landraces seeds and one check variety. The seeds were germinated in seedbed seedlings with a basin (30 cm x 25 cm). The seedlings were planted in the field 16 days after sowing in a plot of 1.5 m x 7.0 m and a spacing of 25 cm x 25 cm, with an isolation distance between plots of ±3 m. Data collection was based on nine growth phases of rice. The cluster analysis based on agro-morphological characters resulted in 6 groups with a similarity coefficient of 0.17 or 17%, showing a high level of diversity. The lowest yield was Ale Kukefuhu (290 g/m2), and the highest was Hare Nona Portu (979 g/m2). Using 10 RAPD markers, the 22 rice landraces samples produced 77 DNA fragment bands with sizes between 300 bp-1475 bp, averaging 3.35 alleles per landrace; polymorphic rates of 8.1. OPA8, OPH1, and OPH7 markers were the highest (100.00%), and OPF5 was the lowest (66.67%) polymorphic percentage. Based on the molecular marker, the coefficient of similarity of the landraces ranged from 0.12 to 0.51, and they were grouped into five clusters.

Abstract

Plant cell walls as sources of lignocellulosic biomass are abundant and used in diverse applications. Lignocellulosic contents manipulation can be applied to develop biomass plants. This study aimed to determine the lignocellulosic components, expressions patterns of OsCAD2 and OsMYB55/61 related to lignin biosynthesis, and deposition patterns of lignin in previously determined three low and three high total lignin content rice accessions based on TGA method. The total Klason and acid-soluble lignin showed variations between 18.03-24.53%, consistent with the TGA method. The ash content of the low lignin content (12.50-21.68%) were significantly differed from the high lignin content (24.84-27.91%) accessions. The extractive, α-cellulose, and hemicellulose contents in each accession showed almost similar proportion. Py-GC-MS proved that the S/G ratio and the syringyl contents were directly proportional to the total lignin contents. OsCAD2 expression levels at the heading stages were consistent with their total lignin contents and their lignin histochemical assay results in the vascular bundle and cortical fiber, suggesting its important roles in rice lignin biosynthesis. However, OsMYB55/61 showed inconsistent expression patterns to total lignin contents, which may also upregulate not only lignin biosynthesis but also cellulose biosynthesis.
