Abstract

This study investigates the activity of various compounds as inhibitors, enhancers, agonists, antagonists, and substrates in biochemical pathways. The results indicate that the highest activity is observed in chlordecone reductase inhibition (Pa: 0.984, Pi: 0.001), followed by HIF1A expression inhibition (Pa: 0.969, Pi: 0.002) and membrane integrity agonist activity (Pa: 0.968, Pi: 0.002). Notably, several compounds exhibit significant inhibitory effects on enzymes such as peroxidase (Pa: 0.966), kinase (Pa: 0.958), and NADPH oxidase (Pa: 0.939). Additionally, CYP1A substrates and inducers demonstrate relevant metabolic interactions, indicating potential roles in drug metabolism. These findings provide insights into the pharmacological significance of these compounds, which may contribute to the development of novel therapeutic agents.

Abstract

This study evaluates the ADMET (Absorption, Distribution, Metabolism, Excretion, and Toxicity) profile of nuciferine to assess its potential as a drug candidate. Based on in silico analysis, nuciferine demonstrates favorable oral absorption and extensive systemic distribution, including its capability to cross the blood-brain barrier into the central nervous system. Regarding metabolism, the compound acts as a substrate for CYP2D6 and CYP3A4 enzymes while serving as an inhibitor of CYP1A2 and CYP2D6, which may lead to drug-drug interactions. In terms of excretion, nuciferine displays moderate clearance values and utilizes renal transporter OCT2. However, toxicological evaluations suggest potential risks, including mutagenicity (positive AMES test), hepatotoxicity, and inhibition of hERG II potassium channels, which are associated with cardiotoxicity risks. Despite its favorable bioavailability and distribution properties, concerns about chronic toxic effects and metabolic interaction risks underscore the need for careful consideration in its development. Further research is necessary to validate its safety and efficacy for therapeutic applications.

Abstract

Sepsis is an irregular body response to severe infection, triggering uncontrolled inflammation that can lead to extensive tissue damage. It can progress to septic shock with multiple organ failure, resulting in death if left untreated. Laboratory examinations, such as leukocyte differential count in hematology, help understand the distribution pattern of white blood cells associated with health conditions. Laboratory research was conducted on mice injected with E. coli ESBL or K. pneumoniae carbapenemase. After 24 hours, observations were made on apoptosis in the spleen and liver of mice. Mouse blood was processed to count white blood cell types with a differential count. The results were analyzed to compare the control group with the bacterial infection groups of E. coli ESBL and K. pneumoniae carbapenemase. The research results indicate that the neutrophil count in the E. coli ESBL group is still within the normal range and lower (44.5±1.915%) compared to the K. pneumoniae carbapenemase group (55.75±8.342%). Similarly, the lymphocyte count in the E. coli ESBL group is within the normal range and lower (77.5±3.109%) compared to the K. pneumoniae carbapenemase group (91.25±7.588%). This highlights the crucial role of neutrophils and lymphocytes in responding to severe bacterial infections such as K. pneumoniae carbapenemase. Previous studies indicate neutrophilia and lymphocytopenia as markers of severe bacterial infections. Neutrophils are the primary defense against bacterial infections and can be rapidly recruited to the infection site, while specific infections can trigger prolonged neutrophil recruitment from hematopoietic tissues.

Abstract

Penelitian ini bertujuan untuk pengembangan suatu metode pengukuran kadar glukosa dalam darah menggunakan sensor kimia berbasis reagen Benedict. Sensor optik ini dikembangkan dengan mengimobilisasi reagen Benedict pada selulosa nata de pina. Membran selulose nata de pina dibuat dari kulit buah nanas kultivar pasir Kelud (PK1). Membran dari nata de pina in berperan sebagai material pendukung secara penjerapan/entrapment. Membran selulosa nata depina digunkaan untuk mengimmobilisasi reagent benedict.  Pengukuran menggunakan Spektrometer genesys 5 dan Spektrometer USB +2000. Penentuan glukosa memiliki kondisi optimum pengukuran pada panjang gelombang maksimum adalah 624,94 nm, konsentrasi Benedict sebesar 0,4480 M dan perbandingan massa selulosa nata de pina dengan volume Benedict yaitu 1: 3.  Hasil karakterisasi sensor optik selulosa nata de pina /Benedict yaitu pada range kerja 0-5000 ppm, limit deteksi sebesar 909,21 ppm, sensitivitas sebesar 0,0009  dan reprodusibilitas sebesar 0,2305%.

Abstract

Sepsis is a severe medical condition characterized by a systemic immune response to infection, often leading to multi-organ failure and death if not treated effectively. This study aimed to investigate the antiseptic properties of Paederia foetida leaf extract in male mice induced with sepsis using Escherichia coli. A total of 24 mice were divided into six groups: a normal group (no treatment), a negative control group (distilled water), a positive control group (ciprofloxacin), and three treatment groups receiving P. foetida leaf extract at doses of 100, 300, and 500 mg/kg BW. After 15 days of oral administration, liver tissue samples were analyzed for IL-6 expression, a pro-inflammatory cytokine associated with sepsis. The results revealed a significant reduction in IL-6 expression in the treatment groups, particularly at the 500 mg/kg BW dose. This dose showed the most effective anti-inflammatory response, with IL-6 expression levels comparable to those of the positive control group treated with ciprofloxacin. However, the 100 mg/kg BW dose demonstrated minimal effects, similar to the negative control. These findings suggest that P. foetida leaf extract, especially at higher doses, has potential as an anti-inflammatory agent in sepsis management. The bioactive compounds in the extract, including flavonoids, alkaloids, and terpenoids, likely contribute to its efficacy. This study provides preliminary evidence supporting the use of P. foetida as a potential herbal alternative for managing sepsis, but further clinical research is necessary to confirm its therapeutic potential.

Abstract

This community service program aimed to increase adolescents' awareness and knowledge regarding the role of Vitamin A (Retinol) in preventing myopia, particularly in rural areas such as Gembleb Village, Trenggalek. The activity was conducted on Saturday, May 3, 2025, in collaboration with the Government of Gembleb Village and supported by the Institute for Research, Community Service, and Publication (LP3M) of Universitas Kadiri. A total of 15 adolescents participated in the educational intervention, which included a pre-test, a lecture session, and a post-test evaluation. The results showed a significant improvement in participants’ knowledge about myopia and the importance of Vitamin A for eye health. Pre-test scores averaged lower compared to post-test results, with an average increase of 20–25 points. This finding suggests that structured education can effectively enhance health literacy among youth, particularly regarding nutritional prevention of visual disorders. These outcomes underscore the importance of continued community-based health education programs, especially in underserved areas, as a strategic effort to reduce the prevalence of preventable eye diseases

Abstract

This community service program aimed to educate adolescent girls in Dusun Gembleb, Trenggalek, on the benefits of red ginger (Zingiber officinale) as a natural remedy for alleviating menstrual pain. Menstrual pain, or dysmenorrhea, is a common health issue affecting a significant number of adolescent girls, often leading to disruption in daily activities. While conventional pain relief medications are commonly used, their long-term side effects can be detrimental. Red ginger, known for its anti-inflammatory and analgesic properties, offers a safer alternative. The program, held on May 3, 2025, at the Gembleb Village Office, included educational sessions on the medicinal properties of red ginger, practical demonstrations on how to prepare red ginger tea, and discussions on its health benefits. The results showed a significant improvement in participants' understanding of red ginger's therapeutic effects, as evidenced by pre-test and post-test comparisons. This initiative successfully raised awareness about alternative treatments and empowered the community to utilize locally available resources for health improvement.

Abstract

Minat masyarakat dalam penggunaan obat tradisional di Indonesia dapat dikatakan tinggi. Salah satunya adalah masyarakat di Desa Pulungdowo, Kecamatan Tumpang, Kabupaten Malang. Jika dilihat dari segi keamanan dan manfaatnya, penggunaan obat tradisional dapat menunjang kualitas hidup masyarakat. Pengukuran kualitas hidup dapat dilakukan dengan menggunakan instrumen kualitas hidup generik yaitu 36-item Short Form and Health Survey (SF-36). Tujuan penelitian ini untuk mengetahui hubungan antara persepsi penggunaan obat tradisional terhadap kualitas hidup masyarakat di Desa Pulungdowo, Kecamatan Tumpang, Kabupaten Malang. Penelitian ini menggunakan metode deskriptif analitik dengan pendekatan kuantitatif dengan menggunakan metode pengambilan sampel purposive sampling. Pengambilan data penelitian menggunakan kuesioner yang disebar kepada 100 responden di Desa Pulungdowo. Hasil penelitian menunjukkan bahwa persepsi responden tentang penggunaan obat tradisional baik artinya responden dapat menerima dan mengolah dengan baik informasi mengenai penggunaan obat tradisional. Selain itu, kualitas hidup masyarakat setelah menggunakan obat tradisional juga dikatakan baik. Hasil uji analisis hubungan antara persepsi penggunaan obat tradisional dengan kualitas hidup masyarakat Desa Pulungdowo, didapatkan hasil yaitu terdapat hubungan antara persepsi penggunaan obat tradisional dengan kualitas hidup masyarakat. Perlunya dilakukan peningkatan program promosi kesehatan seperti penyuluhan tentang pengenalan jenis obat tradisional mengingat masih besarnya masyarakat yang hanya mengenal jamu. Bagi peneliti selanjutnya diharapkan dapat lebih mendalami penelitian seperti penambahan variabel penelitian yang diklasifikasikan berdasarkan jenis obat tradisional.

Abstract

Diseases caused by oxidative stress and inflammation are global health problems that encourage the search for alternative treatments from natural materials. Grapevine leaves (Gynura procumbens (Lour.) Merr.) have long been empirically used, but scientific validation of their potential is still limited. The objective of this research is to identify and describe the active substances in sambung nyawa leaves and assess their effectiveness as natural antioxidants and anti-inflammatory agents. The study involves conducting laboratory experiments, starting with the extraction of sambung nyawa leaves using 96% ethanol solvent through a maceration process. The extract obtained is then analysed for its phytochemical composition. The antioxidant activity is evaluated using the DPPH method to determine the IC₅₀ value. On the other hand, the anti-inflammatory properties are tested in male Wistar rats following the induction of rat hind paw edema with 1% carrageenan, using extract doses of 75, 150, and 300 mg/kgBW. The findings indicate a 13.4% yield from the extraction process. The phytochemical analysis reveals the presence of flavonoids, alkaloids, saponins, tannins, phenols, and steroids/triterpenoids. The antioxidant test results show a moderate level of activity with an IC₅₀ value of 89.26 µg/mL. In the anti-inflammatory test, the 150 mg/kgBW dose demonstrates the highest efficacy with a 52.00% reduction in edema, comparable to the positive control triamcinolone (55.00%). In summary, the ethanol extract of sambung nyawa leaves exhibits promising potential as an antioxidant and anti-inflammatory agent, indicating its suitability for further development as a botanical pharmaceutical ingredient.

Abstract

Background: Anemia merupakan masalah kesehatan masyarakat yang masih tinggi, terutama pada ibu hamil dan balita. Penelitian ini bertujuan untuk memberdayakan kader melalui pelatihan budidaya tanaman burdock (Arctium lappa L.) sebagai intervensi berbasis apotek hidup dalam upaya pencegahan anemia. Jumlah kasus anemia di Indonesia meningkat dari 37,01% pada 2013 menjadi 48,9% pada 2018, dengan kasus signifikan terjadi pada remaja putri dan anak-anak di Kabupaten Cilacap. Faktor penyebab utama meliputi pola makan yang tidak seimbang, kekurangan zat besi, serta akses layanan kesehatan yang terbatas. Metode: Kegiatan ini dilakukan di Desa Sidanegara dengan pendekatan sosialisasi materi ilmiah, pemberdayaan ibu balita dalam pencegahan stunting, serta pelatihan teknik budidaya burdock. Kader dilatih untuk menanam burdock di pekarangan rumah sebagai sumber pangan fungsional yang kaya zat besi, yang berpotensi meningkatkan kadar hemoglobin dan hematokrit. Evaluasi dilaksanakan melalui observasi praktik, wawancara, dan pendampingan pekarangan secara berkala untuk menjamin keberlanjutan program. Hasil: Hasil menunjukkan bahwa kader memahami manfaat tanaman burdock menunjukkan bahwa pengetahuan kader telah meningkat, dengan nilai rata-rata post-test 93, dan mampu menerapkan teknik budidaya secara mandiri. Selain itu, tercipta lahan apotek hidup yang meningkatkan ketahanan gizi dan kesadaran masyarakat terhadap pemanfaatan herbal sebagai solusi kesehatan jangka panjang. Kesimpulan: Program ini berkontribusi pada pemberdayaan kader, penguatan ketahanan pangan lokal, serta pencegahan anemia berbasis komunitas. Pendekatan terpadu ini efektif diterapkan di Desa Sidanegara sebagai model intervensi kesehatan berkelanjutan.
