Abstract

Kappaphycus alvarezii is a globally significant tropical red seaweed renowned for its carrageenan content. While tissue culture is a valuable technique for enhancing seedling quality and stress resilience in vegetative propagation, it is time-consuming and costly. Biostimulants have demonstrated the potential to enhance plant growth. This study aimed to evaluate the potential of biostimulants in enhancing the growth of K. alvarezii propagules. Three different biostimulant products were tested: biostimulant A (amino acid-based), biostimulant B (bacteria-based), and biostimulant C (seaweed extract-based). Each biostimulant was applied at two different concentrations and compared to a negative control and a positive control. Artificial seawater enriched with PES and supplemented with biostimulants according to the treatment was used as the growth medium. Growth of K. alvarezii propagules was monitored weekly, with the primary parameters being propagule weight and specific growth rate. Growth medium quality was assessed by in situ pH measurements and ex situ nitrate and phosphate analyses. Fermented biostimulants negatively impacted growth by reducing the pH of the culture medium, while biostimulant B (photosynthetic bacteria) maintained a near-neutral pH and showed the most promising results, with treatment B2 displaying stable growth and treatment B1 achieving the highest specific growth rate at week 6. Though treatments A1 and A2 showed higher nitrate and phosphate concentrations, these did not correlate with improved growth, likely due to the low pH. While these findings suggest the potential of photosynthetic bacteria for K. alvarezii growth, further research is necessary to fully understand the underlying mechanisms and to develop strategies to overcome the limitations associated with the acidic nature of fermented biostimulants.

Abstract

Indonesia is a leading global producer of seaweed, with kotoni seaweed highly valued for its high-quality carrageenan, an essential ingredient in various industries. This study aimed to evaluate the growth, carrageenan quality, and genetic diversity of kotoni seaweed cultivated at three distinct sites in eastern Indonesia: Banggai, West Halmahera, and Biak. The research involved cultivating kotoni seaweed over a 45-day period using the long-line technique, with growth monitoring conducted every five days and water quality parameters measured concurrently. Additionally, ex situ analyses of nitrate, phosphate, and ammonia were performed every 15 days. Carrageenan quality was assessed by measuring carrageenan content, gel strength, and viscosity, in addition to proximate composition analysis. Genetic diversity was evaluated using the cytochrome oxidase subunit 1 (COI) gene, involving DNA extraction, PCR amplification, and sequencing to determine genetic similarity across the cultivation sites. Significant differences in growth rates and carrageenan quality were observed across the three sites. West Halmahera exhibited the highest growth rate, making it the most favorable site for large-scale seaweed cultivation. Although Biak had a lower growth rate, it produced carrageenan with superior gel strength and viscosity, indicating higher product quality. Genetic analysis confirmed 100% similarity among the samples across sites. These findings underscore the importance of site-specific cultivation practices to optimize both yield and carrageenan quality, supporting the sustainability and economic viability of kotoni seaweed cultivation in Indonesia.

Abstract

The commercially important red seaweed Kappaphycus alvarezii is extensively cultivated for carrageenan production. Despite its economic value, large-scale reproduction and genetic enhancement remain limited due to its low regeneration potential. This study aimed to optimize plant growth regulator (PGR) concentrations for efficient callus induction in K. alvarezii. A completely randomized design was employed, comprising five treatments with varying concentrations of indole-3-acetic acid (IAA) and 6-benzylaminopurine (BAP), along with a control lacking PGRs. A total of 180 explants from meristematic tissues of acclimatized thalli were cultured (30 explants per treatment). The highest callus induction rate (88%) was achieved with 1.50 mg/L IAA and 5 mg/L BAP (Treatment F), with visible callus formation beginning around day 38. A progressive color change from brown to white was observed, indicating active cellular proliferation. Other treatments exhibited lower induction rates, ranging from 0% (control) to 61% (Treatment D). These findings underscore the critical influence of auxin–cytokinin interactions on callogenesis and offer an optimized hormonal regime for improving in vitro culture efficiency. The established protocol provides a valuable platform for future large-scale propagation and genetic improvement strategies in K. alvarezii, contributing to the advancement of seaweed biotechnology.
