Abstract

Makrofag sebagai sel fagosit mononuklear berperan dalam respons imun innate dan adaptif. Ekstrak polisakarida temu putih (Curcuma zedoaria Rosc.) secara in vitro diketahui dapat meningkatkan aktivitas fagositosis. Tujuan penelitian ini adalah untuk mengetahui efek opsonisasi serum terhadap aktivitas fagositosis sel makrofag yang telah diinduksi minyak atsiri temu putih. Subjek penelitian adalah 10 ekor tikus Wistar jantan yang dibagi menjadi 2 kelompok (perlakuan dan kontrol), masing-masing kelompok terdiri atas 5 ekor. Kelompok perlakuan diberi minum minyak atsiri temu putih dosis 30,6 μl/ml dan kelompok kontrol diberi akuabides selama 14 hari pada hari ke-7. Gingiva anterior rahang bawah tikus diolesi A. actinomycetemcomitans sebanyak 100 μl dalam CMC 2% setelah pemberian minum. Pada hari ke-15, tikus pada masing-masing kelompok dianestesi lalu diambil darahnya dari plexus retroorbitalis untuk pembuatan serum. Latex beads diinkubasi dalam serum tikus selama 1 jam. Uji fagositosis makrofag peritoneal dilakukan terhadap latex beads teropsonisasi. Pewarnaan menggunakan Giemsa 20% lalu dilakukan pengamatan aktivitas fagositosis dengan menghitung indeks dan prosentase fagositosis di bawah mikroskop cahaya. Data dianalisis menggunakan uji t. Hasil penelitian menunjukkan terdapat perbedaan bermakna efek opsonisasi serum terhadap aktivitas fagositosis sel makrofag yang telah diinduksi minyak atsiri temu putih (p< 0,05). Sebagai kesimpulan, opsonisasi serum mampu meningkatkan aktivitas fagositosis sel makrofag yang telah diinduksi minyak atsiri temu putih.

Abstract

Getah batang pisang mengandung saponin, flavonoid, vitamin C, dan tanin yang berperan dalam proses penyembuhanluka. Penelitian ini bertujuan untuk mengetahui efek gel getah aplikasi topikal gel getah batang pisang raja terhadapkepadatan serabut kolagen pada proses penyembuhan luka pasca pencabutan pada soket gigi marmut (Cavia cobaya).Lima puluh empat marmut dibagi menjadi 3 kelompok, masing-masing terdiri atas 18 marmut. Pencabutan gigi marmutdilakukan pada gigi insisivus sentralis kiri rahang bawah. Kelompok kontrol positif diperlakukan dengan iod gliserin dankelompok kontrol negatif dengan CMC-Na. Subjek pada kelompok perlakuan diberi gel getah batang pisang 80% secaratopikal. Setelah perlakuan, pada 3 marmut dari masing-masing kelompok dilakukan dekapitasi pada 1, 3, 5, 7, 14,dan 24 hari pasca pencabutan gigi. Rahang bawah marmut dipotong lalu dibuat preparat histologis untuk pengecatanMallory. Kepadatan serabut kolagen diamati dengan mikroskop cahaya, kemudian data dianalisis dengan Uji KruskallWallis dan Mann-Whitney. Hasil penelitian menunjukkan ada perbedaan bermakna serabut kolagen (p < 0,05) antarakelompok perlakuan dan kontrol. Peningkatan serabut kolagen menunjukkan perbedaan yang signifikan pada 3, 5, 7, dan14 hari pasca pencabutan gigi antara kelompok perlakuan dengan kontrol negatif dan positif. Kesimpulannya, aplikasitopikal gel getah pisang 80% dapat mempercepat pembentukan serabut kolagen pada proses penyembuhan luka soketgigi marmut pasca pencabutan gigi marmut.

Abstract

Epigallocatechin gallate (EGCG) merupakan salah satu komponen polifenol dalam ekstrak teh (Camellia sinensis).Penelitian ini bertujuan untuk mengetahui aktivitas fagositosis sel mononuklear tikus Wistar setelah diinduksi denganEGCG. Subjek penelitian terdiri atas 20 ekor tikus Wistar jantan dibagi menjadi 2 kelompok (perlakuan dan kontrol),masing-masing kelompok terdiri atas 10 ekor. Kelompok perlakuan diberi minum EGCG konsentrasi 0,1% dankelompok kontrol diberi aquabides selama 14 hari. Sampel berupa sel mononuklear yang diisolasi dari darah tikuskemudian diuji aktivitas fagositosisnya menggunakan metode uji fagositosis sel mononuklear. Hasil penelitianmenggunakan uji-t menunjukkan terdapat perbedaan yang bermakna indeks fagositosis kelompok perlakuan dibandingkontrol (p<0,05). Disimpulkan bahwa EGCG konsentrasi dapat meningkatkan aktivitas fagositosis sel mononuklear.

Abstract

Smoking may increase dental caries prevalence, meanwhile dental caries is primarly caused by Streptococcus mutans. The aim of this research was to investigate the effect of clove cigarette smoking on the growth of salivary Streptococcus mutans. In this cross-sectional study, twenty healthy male subjects aged 17 to 25 years was volunteered to join this study. Those subjects were divided into smoking and non-smoking group. Subjects in the smoking group had smoked regularly 6 to 9 clove cigarettes per day for 25 to 35 months. Both groups were asked not to brush their teeth at night before sample was taken. Before having breakfast in the early morning, unstimulated saliva was collected into a tube. The collected saliva was diluted using potassium phosphate buffer and then cultured on spesific S. mutans media agar (TYS20B agar). Streptococcus mutans colonies were identified as milky white colonies. After three days of anaerobic incubation at 37oC, the S. mutans colonies were counted. Data was analyzed using unpaired student’s t test. The result showed that S. mutanscolonies in the smoking group was significantly higher than the non-smoking group (p<0,05). This study indicated that smoking could increase the growth of S. mutans in saliva. In conclusion, smoking increase the growth of salivary streptococcus mutans.

Abstract

Dental caries is the most dominant tooth disease that is caused predominantly by Streptococcus mutans. S. mutans is Gram positive facultative anaerobic bacteria and can be found in saliva. Cashew leaves extract (Anacardium occidentale L.) contains of antibacterial substance namely flavonoid, tanin and triterpenoid. The objective of this study was to examine the effect of gargling with 10% cashew leaves extract in S. mutans growth inhibition in saliva. In this study, 15 subjects were divided randomly into 3 groups (treatment, positive and negative control). Each group consists of 5 persons. Treatment group gargled with 10% cashew leaves extract, while positive control group gargled with 0.2% Chlorhexidine gluconate and negative control group gargled with aquades for 5 consecutive days in the morning and night after they brushed their teeth. S. mutans in saliva was cultivated anaerobicly on Trypticase soy extract of Yeast Sucrose Bacitracin agar for 24 hours. The results showed that 10% cashew leaves extract inhibited S. mutans growth in saliva (p<0.05). There was no significant difference between the number of S. mutans in samples who gargled with 10% cashew leaves extract and 0.2% Chlorhexidine gluconate (p>0.05). In conclusion, gargling with 10% cashew leaves extract may inhibit the growth of S. mutans in saliva. Ten percent cashew leaves extract may has similar potency as 0.2 % Chlorhexidine gluconate in S. mutans growth inhibition.

Abstract

The nicotine in the cigarette affects the sympathetic and parasympathetic nerves. Smoking causes heat in oral mucose.This study aimed to evaluate the effects the smoking to salivary pH and volume among the young adults and elderlymales. Forty males were as subjects with approval of ethical clearance from Ethic Committee Medical Faculty ofYogyakarta Muhammadiyah University, Yogyakarta Indonesia. The subjects were divided into four groups, i.e. 10smoker adults (18 – 40 years) and 10 elderly (>60 years). The controlled subjects consisted of 10 non smoker adults (18 –40 years) and 10 elderly non smokers (>60 years). The subjects were instructed to gargle mineral water for one minutebefore collecting sample. The saliva samples were collected in the morning, around 60 minutes after smoking usingunstimulated method. The pH meter was used to measure salivary pH and the volume by using injection. Analysis datafor the salivary volume and pH was compared using unpaired t-test (p<0.05). The results showed that the salivary pH andvolume in elderly smokers were lower than adult smokers. In conclusion, smoking could not affect on the salivary pH andvolume in adults, and the salivary volume in elderly, but it can decrease the salivary pH in elderly.

Abstract

Green tea (Camellia sinensis) has been widely known as a healthy beverage for years and the most polyphenolcomponents (catechin) of tea is Epigallocatechin gallate (EGCG). The aim of this study was to evaluate the effectivenessof tea leaf ethanolic extract toothpaste and EGCG toothpaste on dental plaque index. The subjects were 45 girls, 10-12yeras old, under ethical clearance from Ethic Comittee Medical Faculty of Gadjah Mada University, YogyakartaIndonesia. The subjecs were divided into 3 groups based on caries status (Group I), gingivitis status (Group II) andhealthy status (Group III). Each group consisted of 15 subjects, and divided into 3 sub groups based on toothpaste (tea leafethanolic extract toothpaste, EGCG toothpaste, and base material toothpaste as control). The subjects were instructed tobrush their teeth during 21 consecutive days in the morning and at night. Clinical evaluation was done by PHP Index.Data were taken before treatment in the 7th, 14th and 21st day. Then, they were analyzed using ANOVA and LSD tests.The results showed that the index of PHP was significant difference in each group after using leaf ethanolic extracttoothpaste, EGCG toothpaste, and base material toothpaste. This study suggested that the leaf ethanolic extract toothpasteand EGCG toothpaste could have effected to decrease the PHP Index. As conclusion, the leaf ethanolic extract toothpasteis more effective than the EGCG toothpaste to decrease the PHP Index.

Abstract

ABSTRACTGingival epithelium has an important role in the protection of the gingival tissue from mechanical, physical, and chemical trauma also microbial invasion. Re-epithelialization is an important phase of post extraction socket healing. Previous study showed that banana (Musa sapientum L) latex containing tanin, saponin, flavanoid, vitamin C, and minerals i.e: kalium, magnesium, calcium induced re-epithelialization in skin wounds. The objective of the present research was to study the effectivity of banana latex gel in the re-epithelialization of post extraction socket in guinea pigs (Cavia cobaya).Fifty four guinea pigs were divided into 3 groups, there were negative control group, positive control group and banana latex gel group. Each group consisted of 18 guinea pigs. Mandibular left central incisive were extracted and CMC-Na 1% (negative control), iod-glycerin (positive control) and banana latex gel were applied into the wound socket. Guinea pigs were then sacrified at 1,3,5,7,14,24 day post extraction and processed for histological examination. The specimens were stained with hematoxilin eosin. Epithelial thickness was measured with optilab (µm). The result of Anova showed significant differences in epithelial thickness among groups (p<0,05). Least Significant of Difference test showed significant differences (p<0,05) at 3,5,7,14,24 day post extraction between banana latex gel group compared to negative control group and also at 7 and 24 day post extraction between banana latex gel group compared to positive control group. In conclusion, banana latex gel may induce re-epithelialization of post extraction socket. The effect of banana latex gel is similar to iod-glycerin.Key words: re-epithelialization, banana  latex  gel, tooth socket

Abstract

Aggregatibacter actinomycetemcomitans (A. actinomycetemcomitans)merupakan bakteri Gram-negatif yang dikaitkan dengan penyakit periodontal. Ekspresi positif CD4+ tampak pada keadaan gingivitis(inflamasi gingiva) maupun periodontitis. Tujuan dari penelitian ini adalah untuk mengetahui efek minyak atsiri temu putih (Curcuma zedoaria Rosc.) terhadap ekspresi CD4+ pada gingiva terpapar A.actinomycetemcomitans. Penelitian dilakukan pada tikus Wistar jantan umur 3 bulan yang diberi paparan bakteri A.actinomycetemcomitanspada gingiva anterior mandibula. Kelompok perlakuan sebanyak 5 ekor tikus diberikan minum minyak atsiri temu putih dosis 30,6μl/ml selama 14 hari dan kontrol diberikan aquades. Bakteri diaplikasikan sebanyak100 μl hari ke-7 sampai ke-14 lalu hari ke-15 dilakukan pengorbanantikus teranestesi. Gingiva anterior mandibula tikus diambil untuk pembuatan preparat histologis selanjutnya dilakukan pewarnaan Hematoxylin-Eosin (HE) dan imunohistokimia. Antibodi CD4+ (Santa Cruz Biotechnology, USA) digunakan pada pewarnaan imunohistokimia.Pengamatan preparat histologis menggunakan mikroskop cahaya. Hasil penelitian menunjukkan ekspresi CD4+ positif kuat di lamina propria dan stratum basale pada kelompok kontrol sedangkan ekspresi positif lemah di area lamina propria tampak pada kelompok perlakuan. Disimpulkanbahwa induksi minyak atsiri temu putih dapat menurunkan inflamasi gingiva ditandai dengan penurunan ekspresi CD4+. Reducing CD4+ Expression at Gingiva Exposed A.Actinomycetemcomitans using Curcuma Zedoaria Rosc., Zingiberaceae. Aggregatibacter actinomycetemcomitans (A. actinomycetemcomitans) is a gram-negative bacteria associated withperiodontal disease. Positive expression of CD4+ is shown in gingivitis and periodontitis. The aim of this study was to determine the effect of curcuma zedoaria Rosc., volatile oil against CD4+ expression on gingiva exposed A. actinomycetemcomitans. The study was conducted on male Wistar rats aged 3 months that were exposed bacteria A. Actinomycetemcomitans. Treatment group as many as 5 rats were administered orally with curcuma zedoaria Rosc. volatile oil at dose of 30.6 μl/ml for 14 days and distilled water in control. A 100 μl bacteria were applied on the 7th to 14th day on the gingiva anterior mandibular. At the 15th day, rat was anesthetized then sacrificed. The mandibular gingiva anterior of the rat was taken as histological preparations for HE and immuno-histo chemical staining. CD4+ antibody (Santa Cruz Biotechnology, USA) was used for immune staining. Histological preparations were observed under a light microscope.The results showed a strong positive CD4+ expression in the lamina propria and stratum basale of control group while a weak positive expression in lamina propria area of treatment group. From the experiment, it is concluded that theinduction of curcuma zedoaria Rosc. volatile oil may reduce gingivitis, indicated by the decrease in CD4+ expression.
