Abstract

The study aimed to isolate the Salmonella sp on quail eggs that failed to hatch in Garot village, Darul Imarah Subdistrict, Aceh Besar. A total of thirty quail eggs that failed to hatch were examined in microbiology laboratory of Veterinary Medicine Faculty, Syiah Kuala University. Modification of Carter method was used in this study to isolated samples. The eggs were opened for taking the embryo and then it was swabbed with sterile swabs.  The swab was put into Selenite Cystine Broth (SCB) and incubated at 37o C for 24 hours. The breed was then transferred onto Salmonella Shigella Agar (SSA). The growing Bacteria were observed for their morphology and Gram staining to make sure the bacteria belonged to the negative Gram. The result showed that 10 of the 30 of quail eggs that failed to hatch were positively infected with Salmonella sp. Based on this study, it can be concluded that the salmonella sp.can be isolated from quail eggs which were not hatched in the farm of Garot, Darul Imarah Subdistrict, Aceh Besar

Abstract

The purpose of this study is to determine the effect of the red dragon fruit peels (Hylocereus polyrhizus) extract on the growth of Salmonella pullorum. This research was conducted at the Laboratory of Microbiology, Veterinary Faculty, University of Syiah Kuala, Banda Aceh. Red dragon fruit peels that used in this study is obtained from  ripe fruit and Salmonella pullorum cultures is  obtained from  Microbiology Laboratory of Veterinary Faculty Unsyiah. Salmonella pullorum culture added evenly on the surface of Mueller Hinton Agar, the number of the bacteria have been standarized using Mc Farland'1 method. Discs soaked in the red dragon fruit peels extracts at different concentrations (20 mg/ml, 40 mg/mL, and 60 mg/ml). Ampicillin is used as positive controls and DMSO (dimethyl sulfoxide) was placed on the surface of Mueller Hinton Agar. The parameters in this study is the inhibition zone diameter. Data were analyzed descriptively. The results showed that the red dragon fruit peel extract is able to inhibit the growth of Salmonella  pullorum and the average of  inhibition diameter zone at a concentration 60 mg / ml is 9.6 mm, 40 mg / ml is 9.4 mm and 20 mg / ml is 9.3 mm

Abstract

The purpose of this study was to detect the existence of the Streptococcus sp bacteria in the horses’ dental caries in Aceh Tengah Regency. The caries samples used in this study were taken from 8 horses in Aceh Tengah Regency. The samples were collected using swab on the teeth and then put into Nutrient Broth (NB) media and further were cultured on selective media TSY20B. The bacteria identification was conducted by observing the bacteria’s morphology, Gram staining and biochemical test. The eight samples of the horses’ dental caries showed that Streptococcus sp bacteria could be isolated from all of the collected samples. From this research it could be concluded that Streptococcus sp bacteria was found in the horses’ dental caries and it was one of the causes of the dental caries.

Abstract

The aim of this study was to count the total sum of cellulolitic bacteria colony in  aceh cattle rumen. The sample used was aceh cattle rumen liquid which taken from salughtered house in Banda Aceh. Aceh cattle rumen liquid were diluted from 10-2 to 10-5, then inoculated to agar BHM-CMC media with pour plate method. Petri dish was entered to candle jar to be incubated in an incubator with temperature 37oC about 96 hours. Colony of cellulolitic bacteria which were grown then were observed by using colony bacteria criteria (colour, position, edge, and diameter of colony), counting total of bacteria which was grown, and Gram staining. The results showed that there is a white translucent colony morphology, growth inside agar, smooth periphery, Gram-positive bacil. The average number bacteria of total bacteria in the cattle rumen liquid was 4,81 x 105 cfu/ml

Abstract

This study aimed to isolate and identify the bacteria Salmonella sp in spleen of male layer chicken that died in farms Sibreh, Aceh Besar. A sample of the research is the spleen of male layer chickens. Isolation of Salmonella sp performed by inoculating spleen that were pierced using a swab on the media Selenite Cystine Broth (SCB) then planted on selective media Salmonella Shigella Agar (SSA) and colony morphology was observed, continued with observation of Gram stain under microscopic. Furthermore, colonies of suspected Salmonella sp planted in the media MR-VP (Oxoid), Indol (Difco), Simmons's Citrate Agar (Oxoid), Triple Sugar Iron Agar (TSIA), Sulfite Indol Motiliti (SIM), and media sugars (sucrose, manitol, glucose, lactose , maltose). The results of this study were analyzed descriptively. The research shows a results from a total of 33 samples, we found that 15 samples are positive that can be isolated and identified as Salmonella sp. Therefore it can be concluded that Salmonella sp is possible the death cause of male layer chicken in farms Sibreh, Aceh Besar

Abstract

This research has purposes to isolating and identifying the lactic acid bacteria genus Pediococcus in sumatran orangutan (Pongo abelii) feaces. Feaces are isolated from one orangutan at Kandi Zoo and two orangutans at Kinantan Zoo. The isolating of the lactic acid bacteria was done by using streak plate method with the MRSA (de Man Rogosa Sharpe Agar) specific media. Identification was done by the morphological observing, Gram stain, catalase test, oxidase test and biochemical test. The result showed that one isolate feaces from Kandi Zoo has cocci shaped with the isolate prediction approaches genus Pediococcus sp. The characteristic features of isolate are cocci shaped, Gram positive, tetrad cell structure, nonmotile, catalase negative, and oxidase negative. Based on these characteristics, it can be concluded that there is a lactic acid bacteria coloni which grouped into genus Pediococcus in male orangutan feaces from Kandi Zoo

Abstract

The research aimed to isolate Pseudomonas sp from fail to hatch quail eggs in Garot village, Darul Imarah subdistrict, Aceh Besar. Thirty failed to hatch quail eggs were examined in microbiology laboratory of Veternery Medical, Faculty Syiah Kuala University. Modification of examined Carter method was used in this research. The result of this research showed 12 of 30 samples examine we were positive infected by pseudomonas sp. It can concluded that bacteria Pseudomonas sp is one of the causes of failure hatching quail eggs in Garot village, Darul Imarah subdistrict, Aceh Besar
