Abstract

Single Nucleotide Polymorphisms (SNPs) on the Leptin gene may alter the quality and quantity of Leptin hormone, which plays an active role in energy metabolism and it can contribute to fat deposition in mammals. This study proposed to determine the effect of SNPs R25C and R25H on Cholesterol, and Triglycerides levels in the blood of Ongole Crossbreed cattle. The research was carried out experimentally with a Completely Randomized Design using the sixteen heads of Ongole Crossbreed. There were three different genotypes based on SNPs R25C and R25H: CC, TT, and  AA. The cattle were reared intensively for two months. The feed consists of corn kernels and concentrates, and It is calculated based on the Dry Matter (DM) intake, which is 3% of body weight. The result showed SNPs R25C and R25H on the Leptin gene did not have a significant effect on blood Cholesterol, and Triglyceride levels of Ongole Crossbreed cattle. Those SNPs in the Leptin gene cannot be used as selection markers in Ongole Crossbreed cattle. Further research is needed to include SNPs in other genes contributing to Cholesterol and Triglycerides on blood cattle

Abstract

Kesehatan ternak merupakan salah satu kunci penentu keberhasilan suatu usaha peternakan. Melalui penerapan manajemen kesehatan ternak yang baik dan dilakukan secara berkelanjutan, diharapkan dapat meningkatkan produktivitas ternak. Guna mencapai hal tersebut, diperlukan penyampaian informasi kepada peternak, khususnya bagi peternak yang masih menerapkan pola pemeliharaan tradisional. Salah satu metode penyampaian informasi yang sederhana dan dinilai efektif adalah melalui sosialisasi. Kegiatan Pengabdian Kepada Masyarakat (PKM) inidilaksanakandiKelompok Peternak Domba Lentera Kirei, Kecamatan Cijulang, Kabupaten Pangandaran. Kegiatan dilakukan dengan melakukan kunjungan secara langsung ke kandang peternak, dan selanjutnya dilakukan sosialisasi dan diskusi mengenai kesehatan hewan, serta observasi dan pemeriksaan fisik terhadap domba. Berdasarkan hasil pengamatan di lapangan terungkap bahwa, sebelum dilakukan sosialisasi ternyata mayoritas peternak telah memiliki pengetahuan dan kesadaran yang baik mengenai pentingnya kondisi kesehatan ternak, selain itu peternak juga telah memiliki pemahaman yang baik mengenai bagaimana cara memberikan pertolongan pertama pada domba yang sakit serta kondisi gawat darurat. Sebaliknya, pengetahuan tentang prinsip dasar pencegahan penyakit, pengetahuan tentang jenis penyakit pada domba masih sangat rendah, yaitu antara 30-40 %. Setelah dilakukan sosialisasi, terjadi peningkatan pengetahuan peternak terhadap semua parameter, terutama pengetahuan peternak terhadap prinsip dasar pencegahan penyakit, pengetahuan tentang jenis penyakit menular, penyakit tidak menular, dan penyakit reproduksi pada domba.

Abstract

Sapi Pasundan merupakan sapi lokal yang memiliki potensi dalam memenuhi kebutuhan daging nasional khususnya di Jawa Barat. Kualitas daging salah satunya dipengaruhi oleh komposisi asam lemaknya. Sintesis asam lemak pada daging sapi dipengaruhi oleh protein yang diproduksi oleh gen SREBP-1 . Penelitian ini bertujuan untuk mengidentifikasi gen SREBP-1 sebagai kandidat gen untuk kualitas daging pada Sapi Pasundan. Metode penelitian yang digunakan adalah eksperimental eksploratif. Penelitian ini dilakukan dengan mengamplifikasi DNA dari 25 ekor Sapi Pasundan. Amplifikasi target DNA untuk mendeteksi polimorfisme 84-bp indel pada intron 5 gen SREBP-1 , dilakukan dengan teknik PCR, dengan primer spesifik. Visualisasi hasil PCR dilakukan dengan elektroforesis gel agarose. Sekuensing DNA target hasil PCR dilakukan untuk mengkonfirmasi mutasi pada gen SREBP-1 . Hasil penelitian menunjukkan adanya dua genotip, yaitu LL (96%) dan LS (4%), dengan frekuensi alel L sebesar 0,98 dan alel S sebesar 0,02. Selain itu, ditemukan dua mutasi baru, yaitu SNP g.9969T>C dan SNP g.10265A>G. Penelitian ini menunjukkan bahwa gen SREBP-1 pada Sapi Pasundan bersifat polimorfik dan berpotensi menjadi kandidat gen untuk kualitas daging yang berkaitan dengan komposisi asam lemak.

Abstract

Abortus pada sapi merupakan kondisi ketidakmampuan fetus untuk bertahan hidup di dalam rahim hingga saat akan dilahirkan. Tulisan ini merupakan laporan pertamakali kasus abortus idiopatik superfetasi dari perkawinan alami yang terjadi pada induk sapi Pasundan primipara. Sapi-sapi awalnya dipelihara dengan sistem kandang koloni dan bercampur antara jantan (n=10) dan betina dara (n=18) selama 2 minggu pascapemindahan sapi dari kandang ternak di Kabupaten Ciamis. Terjadi kebuntingan oleh perkawinan alami pada sapi dara tanpa diketahui usia dan riwayat kebuntingan. Seekor induk sapi teramati menunjukkan gejala perdarahan pervagina, mengejan dan disertai pengeluaran fetus berukuran kecil (panjang ~25 cm, usia fetus ~120 hari) beberapa jam kemudian, sebelumnya sapi telah mengalami abortus dengan ukuran fetus yang besar (panjang ~36 cm, usia fetus ~150 hari). Induk sapi Pasundan primipara yang mengalami abortus kemudian dipisahkan ke kandang individu untuk mengurangi stress dan untuk observasi lebih lanjut. Terapi yang diberikan adalah dengan pemberian antibiotik dan multivitamin.

Abstract

Mastitis pada ruminansia kecil menimbulkan tantangan besar dalam kedokteran hewan karena berdampak signifikan pada kesejahteraan dan produktivitas hewan. Studi kasus ini menggambarkan mastitis klinis pada domba Etawah betina berumur empat tahun setelah melahirkan. Pemeriksaan fisik mengungkapkan adanya peradangan di kelenjar susu kanan. Kondisi domba tersebut memburuk akibat luka terbuka yang muncul dari gesekan kulit kelenjar susu dengan lantai saat berbaring. Lesi tersebut meluas, menghasilkan eksudat purulen yang berubah menjadi koreng. Penghentian pemerahan pada kelenjar susu yang terinfeksi berakibat buruk pada kesehatannya karena akumulasi susu dalam alveoli mendukung pertumbuhan dan reproduksi bakteri. Regimen pengobatan yang diterapkan meliputi penggunaan penisilin dan sulpidon; namun, kondisi domba tersebut terus memburuk, menunjukkan bahwa pengobatan tersebut tidak efektif.

Abstract

Reproductive technologies have the potential to enhance livestock productivity, with superovulation being one such method aimed at increasing the number of ovulated oocytes per estrous cycle through hormonal stimulation. This study aimed to evaluate the reproductive performance of three cattle breedsOngole Grade (OG) (n=20), Simmental (n=18), and Limousin (n=20)subjected to superovulation treatment. The cattle, aged 2.5 to 4 years with a body condition score of 3.5 to 4, were intensively managed. Superovulation was induced using CIDR (Controlled Internal Drug Release) for synchronization, followed by follicle-stimulating hormone (FSH) administration. Reproductive performance data, including age at first estrus, number of corpora lutea (CL), and embryo production, were collected over three estrous cycles. A quasi-experimental design was employed with breed as the primary treatment factor, and data were analyzed using analysis of variance (ANOVA). The results indicated that breed had a significant effect (P0.05) on the age of first estrus, with OG cattle reaching first estrus at 621.393.3 days, Simmental at 522.475.5 days, and Limousin at 570.488.1 days. However, no significant differences (P0.05) were observed among the breeds in terms of the number of CL, collected embryos, or transferable embryos across the superovulation cycles.

Abstract

Pasundan cattle are one of Indonesia’s local cattle thathave the potential to be developed, especially in terms of meatquality. Meat quality is strongly influenced by fatty acidcomposition, which is closely related to gene regulation. This studyaims to identify the diversity of the FASN (fatty acid synthase) geneas a candidate gene for meat quality in female Pasundan cattle. Thisstudy used an experimental descriptive method with 25 DNAsamples of female Pasundan cattle with an age range of 1,5-2 years.DNA amplification using PCR was perfomed with specific primersdesigned with NCBI Primer Blast, flanking the target gene along560 bp. The analysis was followed by PCR-RFLP (RestrictionFragment Length Polymorphism) method using MscI enzyme todetect single nucleotide polymorphism (SNP) g.17924 A>G in exon39 and confirm the target mutation using direct sequencingmethod. The results showed only one genotype, the GG genotype(100%), with a G allele frequency of 1.00. Based on these results, itcan be concluded that the FASN gene mutation g.17924 A>Gmutation in Pasundan cattle is monomorphic. SNP g.17924 A>G inthe FASN gene has the potential to affect the fatty acid compositionof Pasundan female beef

Abstract

Meat quality is influenced by genetic factors,environmental conditions, and their interactions. Variations in thenucleotide sequence of the leptin gene may affect the quality andquantity of leptin hormone, thereby influencing meat quality. Thisstudy aimed to identify the genetic diversity of the leptin genebased on nucleotide sequence mutations in Pasundan cattle raisedat the Ciparanje Beef Cattle Teaching Farm, Faculty of AnimalScience, Universitas Padjadjaran. This Study used 16 DNA samplesfrom female Pasundan cattle were isolated from blood andanalyzed. PCR amplification was performed to amplify a 620 bpfragment of the leptin gene using specific primers designed via theNCBI Primer-BLAST program, with an annealing temperature of59°C. PCR products were evaluated through electrophoresis, andsequencing was carried out to determine the nucleotide sequence.Sequence analysis and genotyping were conducted using BIOEDITand MEGA 11 software. The results revealed a Single NucleotidePolymorphism (SNP) at position g.1047C>T/R25C in the leptingene, with the CT genotype (56.35%) being more frequent than CC(18.8%) and TT (25%). Additionally, several point mutations inintron 1 were identified, including g.806G>A, g.845C>T,g.856G>A, g.868G>C, g.885C>T, g.911C>T, and g.913A>G,consisting of both transition and transversion mutations. Thesefindings indicate that the leptin gene in Pasundan cattle ispolymorphic. Leptin gene polymorphisms in Pasundan cattle arepotentially associated with meat quality traitrs and fat depositionvariation, thus providing a genetic basics for selection to improvethe quality of local cattle
