Abstract

Pre-growing weed control by glyphosate herbicides is effective for increasing yield, but glyphosate residues in the soil might reduce soil quality and can accumulate in agricultural products. Naturally, microbes are able to breakdown glyphosate into nontoxic substances orthophosphate and glycine. Glyphosate degradation in soil by single soil microbes are reported elsewhere, but the information about glyphosate removal by soil bacterial consortia was limited. The objective of this research was to determine the effect of carbon (C), nitrogen (N), and phosphorus (P) composition in liquid media to increase glyphosate degradation and its degradation product by soil bacterial consortia and 2) verify the effect of bacterial consortia on maize seedlings growth, their N and P uptake, as well as total and soluble P in soil. Glyphosate degradation test was set up by incubating bacterial consortia in a different composition of C-N-P liquid basal media. Greenhouse experiment has been performed in a randomized block design to treat maize grown in Inceptisols with bacterial and glyphosate application. The results showed that C-N-P composition of liquid media affected the concentration of glyphosate, as well as orthophosphate and glycine as by-products. In-planta experiment verified that inoculation of glyphosate-degrading bacterial to maize seedling grown in glyphosate-contaminated soil enabled to enhance shoot dry weight of maize seedling and N and P uptake at 4 weeks after inoculation.

Abstract

Nitrogen-fixing Azotobacter synthesizes exopolysaccharide, which is important among other to improve aggregate stability and hence nutrients uptake. A pot experiment has been conducted to determine the effect of exopolysaccharide-producing Azotobacter and organic matter on nitrogen, phosphor, and potassium uptake by the shoot of sorghum (Sorghum bicolor (L.) Moench), and plant growth. The pot experiment was setup in randomized block design which test eight combination treatments of Azotobacter isolates (AS5, AS6, and AS5 + AS6) and organic matter application (with and without 20 t ha-1 of cow manure). The result showed dual inoculation of Azotobacter AS5 and AS6 inoculation combined with cow manure application increased N and P uptake. The dual inoculation treatment did not affect root length; but increased the shoot height and dry weight when accompanied by the application of cow manure. The ratio of root and shoot dry weight was not influenced by single or dual Azotobacter inoculation with or without organic matter.

Abstract

Indonesia has a diverse of new and renewable energy sources including biofuels. Biofuels are promising eco-friendly energy sources generated from biological raw materials. Biofuels can also be derived from the seeds of  Capilong trees. However, the occurrence of Capilong trees in North Maluku has not been tapped to support a local economic income due to the lack of knowledge  of the local community. The present study was undertaken with the aim at investigating the distribution and number of Capilong trees across Ternate Island. A field investigation using a census method was held to collect data on the number and distribution of Capilong trees while observing the following parameters: stage of growth, topography, rock materials and landforms. The findings revealed that Capilong trees are mostly occurred in a tree group, while the seedling population number was low. Interestingly, Capilong trees have demonstrated a natural characteristic that are tolerance to various topography, rock materials and landforms condition. It is therefore recommended to cultivate the plant through a community development program and classify the plant as a potential species that can be used for land rehabilitation programs.

Abstract

Agricultural productivity in Buru Island, Maluku is threatened by tailings which are generated from formerly gold mine in Botak Mountain in Wamsait Village. Gold that extracted by using mercury was carried out in mining area as well agricultural field.  High content of mercury in tailings and agricultural field pose a serious problem of food production and quality; and further endangers human health. The purpose of this research was to determine the contaminant level of mercury in tailing, soil and its accumulation in edible part of some food crops. Soil, tailing and plant samples for Hg testing were taken by purposive method based on mining activities in Waelata, Waeapo and Namlea sub district. Six soil samples had been analyzed for their chemical properties. Total mercury levels in tailings and plants were measured by Atomic Adsorption Spectrophotometer. This study showed that agricultural field where tailings were deposited contained Hg above the threshold but agricultural area which is far from hot spot did not. Most edible parts of food crops accumulated mercury more than Indonesian threshold for mercury content in food. This evidence explained that tailings deposited on the surface of agricultural field had an impact on soil quality and crop quality. Tailing accumulated on soil will decreased soil quality since naturally soil fertility in agricultural field in Buru is low.

Abstract

Sago starch production leaves solid and liquid wastes that has not been used optimally. Both organic substances still contains a lot of nutrients, and could be processed into raw material for bio-fertilizers media. Solid waste, known as ela sago, has been developed into compost while the liquid waste did not. The objective of this study was to verify volume ratio of liquid inoculant of Azotobacter chroococcum in solid inoculant produced from ela sago; and determine the concentration of sago waste water as a growth medium of biofertilizer A. chroococcum. This study confirms population of A.chroocccum and T. harzianum in ela sago compost reached 106 cfu/g and 108 cfu/g consecutively following enrichment with either 2% or 4% of A. chroococcum liquid inoculant. Sago wastewater can used support A.chroococcum growth. This study proved that waste from sago starch production could be used as a natural medium for biofertilizer.

Abstract

Low-land rice (Oryza sativa L.) cultivation in saline soils face some constraints include nitrogen availability. Saline-resistant nitrogen fixing bacteria Azotobacter are expected to increase supply nitrogen in saline soils. The objectives of the study were to determine the effect of liquid inoculant concentration of two Azotobacter isolates on early vegetative growth of lowland rice grown in potted saline soil.  The greenhouse trial design was a randomized block design with seven treatments and four replications. The treatments were combination of isolates and Azotobacter liquid inoculant concentrations in single and mixed inoculation. The results showed that all plants experienced chlorosis and stunt due to high Electrical Conductivity. Inoculation of different isolates and concentrations did not influence the growth of lowland rice in soil with high EC at the end of experiment. Therefore, neither isolates nor concentration of Azotobacter could improve retarded-growth of lowland rice in saline soil.

Abstract

Fertilizer is one of the important limited factor in plant cultivation system. Biofertilizer is a kind of fertilizer which improve nutrient availability in soil and subsequently plant yield. A research was carried out in village Waiheru, District Baguala, Ambon to determine the effect of different level of mixed biofertilizer on the growth and yield of caisim that grown on Alluvial. The research used Randomized Completely Block Design (RCBD) with three concentration of liquid biofertilizer, namely: 0.1%, 0.5% and 1%. Treatment control was without biofertilizer. The results showed that inoculation of liquid biological fertilizer at concentration of 1% improved growth and yield of caisim higher than the concentration of 0.1% and 0.5%. Inoculation of 1% biofertilizer increased the fresh weight of caisim shoot up to 37.36% compared to that of controls. While, fresh weight of caisim with biofertilizer of 0.1% and 0.5% was 24.52% and 28.94% respectively.Keyword: Ambon, Mixed Biofertilizer, Caisim

Abstract

Kadar merkuri (Hg) yang tinggi pada tailing tambang emas adalah sumber pencemaran lingkungan termasuk lahan pertanian. Penurunan kadar Hg di tailing dengan metode bioremediasi adalah strategi yang efektif, murah dan mudah. Azotobacter adalah rizobakteri pemfiksasi nitrogen dan penghasil eksopolisakarida yang mengubah mobilitas logam berat serta memicu pertumbuhan tanaman dengan optimal jika terdapat bahan organik. Penelitian ini dilakukan untuk mengevaluasi pengaruh dosis bahan organik dan Azotobacter strain resisten Hg terhadap perubahan kadar Hg di tailing dan tanaman jagung. Percobaan rumah kaca dirancang dalam Rancangan Petak Terbagi dengan tiga ulangan. Petak utama adalah dosis bahan organik yang terdiri atas 22,5; 30 dan 37,5 g/polibeg. Anak petak adalah strain bakteri terdiri atas tanpa inokulan, Azotobacter indigen, A. chroococcum, dan konsorsium kedua bakteri. Hasil penelitian menunjukkan bahwa inokulasi Azotobacter meningkatkan tinggi dan bobot kering tanaman jagung umur 3 minggu. Aplikasi kompos kotoran sapi 30 g/polibeg disertai inokulasi A. chroococcum maupun konsorsium Azotobacter menurunkan kadar Hg tanah dan meningkatkan serapan Hg di tanaman. Penelitian ini menjelaskan bahwa inokulasi Azotobacter menginduksi serapan Hg oleh tanaman sehingga berpotensi digunakan untuk bioremediasi tailing terkontaminasi Hg dengan tanaman fitoakumulator.

Abstract

Abstrak. Inokulan bakteri pemfiksasi nitrogen (N2) Azotobacter umumnya diproduksi pada  media bebas N untuk mengoptimalkan fiksasi N2, padahal Azotobacter juga mensintesis eksopolisakarida (EPS) yang berperan sebagai amelioran tanah. Fiksasi nitrogen dihambat oleh N anorganik sedangkan produksi EPS memerlukan N anorganik. Dengan demikian diperlukan komposisi media yang mendukung produksi EPS tetapi tetap menjaga aktivitas fiksasi N2. Tujuan penelitian ini adalah mendapatkan komposisi media cair Azotobacter berbasis molase yang diperkaya dengan asam amino sisteina dan serina untuk menghasilkan sel, EPS, dan N tersedia dengan konsentrasi yang sama dengan media anorganik. Optimasi media dilakukan pada fermentor 2 L pada suhu kamar dengan pengadukan 115 rpm selama 7 hari, kemudian inokulan cair disimpan selama dua bulan pada suhu kamar. Hasil penelitian menjelaskan bahwa pada hari ke-7, media molase 1% + NH4Cl 0,05% dengan maupun tanpa asam amino menurunkan populasi bakteri dibandingkan dengan media anorganik. Namun produksi EPS di media molase dengan asam amino menyamai kadar EPS di media anorganik. Setelah dua bulan penyimpanan media cair berbasis molase dengan penambahan sisteina 0,25% dan serina 0,1%  meningkatkan populasi sel, serta kadar EPS dan N dibandingkan media anorganik, serta mempertahankan pH netral. Percobaan ini menjelaskan bahwa molase dengan NH4Cl yang diperkaya sisteina dan serina dapat digunakan untuk mengganti media anorganik untuk produksi  EPS oleh Azotobacter.Abstract. Inculants of nitrogen-fixing Azotobacter are generally produced on N-free media to optimize N fixation. Nonetheless Azotobacter synthesizes exopolysaccharide (EPS) that has a function as soil ameliorant. Nitrogen fixation was carried out in the absence of inorganic N while EPS production was induced by N. Therefore, another media composition is required to produce EPS while maintaining N fixation. The purpose of this study was to obtain the composition of molasses-based Azotobacter liquid media with the amino acid cysteine and serine additives to produce cells, EPS, and N with the same concentration as that in inorganic media. The media optimization was carried out on a 2 L fermentor at room temperature with stirring at 115 rpm for 7 days, then the liquid inoculant was stored for two months at room temperature. The results showed that on the 7th day, 1% molasses + 0,05% NH4Cl media with or without amino acids decreased the bacterial population compared to inorganic media. However, the production of EPS in molasses-based media with amino acids was comparable to that in inorganic media. After two-month storage, molasses-based liquid media with 0.25% cysteine and 0.1% serine increased the cell population, and levels of EPS and N compared to inorganic media; and maintain neutral acidity. This experiment suggested that molasses with NH4Clenriched with cysteine and serine can  replace  inorganic media for EPS production by Azotobacter.

Abstract

Low nitrogen content in soil is usually overcome by chemical fertilization. After long application period, high-dose and intensive use of N fertilizers can cause ammonia volatilization and nitrates accumulation in soil. In sustainable agriculture, the use of bacterial inoculant integrated with nutrient management system has a role in soil health and productivity. Azotobacter-based biofertilizer is suggested as a chemical nitrogen fertilizer substitute or addition in crop production to improve available nutrients in the soil, provide some metabolites during plant growth, and minimize fertilizer doses. The objective of this literature reviewed paper is to discuss the role of Azotobacter in agriculture; and the prospective of Azotobacter to increase yield and substitute the chemical fertilizer in food crops production. The results revealed that mechanisms by Azotobacter in plant growth enhancement are as biofertilizer, biostimulant, and bioprotectant. Nitrogen fixation by Azotobacter is the mechanism to provide available nitrogen for uptake by roots. Azotobacter stimulates plant growth through phytohormones synthesis; indole acetic acid, cytokinins, and gibberellins are detected in the liquid culture of Azotobacter. An indirect effect of Azotobacter is exopolysaccharide production and plant protection. Inoculation of Azotobacter in the field integrated with organic matter and reduced chemical fertilizer are reported to improve plant growth and yield.
