Abstract

Bioagumentation by used of nonsimbiotic N2 fixing Azotobacter  is a way to enhance soil N availability in sustainable Agricultural land.  A green house experiment has been done to verify effect of Azotobacter sp. inoculation in two soil order, Inceptisols and Entisols, on NH4+danNO3- content , N uptake as well as Azotobacter population in rhizosphere soil of sorghum (Sorghum bicolor L.). Experiment was set up in Randomized Block Design with eight treatments and three replicates, sorghum was maintained in green house until maximum vegetative phase. Resuls showed that Azotobacter sp. AS4 was more enable to increase the availability of soil N rather than isolate AS3.  Bacterial  bioaugmentation with Azotobacter sp. AS4 on Inceptisols increased more NO3-, Azotobacter sp. population in soil rhizosphere and shoot height of sorghum genotype 2.24.

Abstract

Characteristics of mineral-mine tailings deposited in agricultural areas do not meet the requirements for food crop cultivation. The objective of the experiment was to obtain the information on the improvement of plant growth planted in mercury-contaminated tailings after adding organic matter and nitrogen fixing bacteria, Azotobacter. The greenhouse trial was set up in randomized factorial design which tested the Azotobacter inoculation consentrations and organic matter doses. There was no interaction effect between the two treatments on the growth of four-week old corn. Both organic matter and Azotobacter had a positive impact on vegetative growth of corn.

Abstract

Substituting a part of soil on tea Camellia Sinensis (L.) O. Kuntze) nurseries by using organic matter could minimize the remove fertile soil from nature. Biofertilizing are a way to improve the biological quality of soil and subsequently tea seedling quality. The aim of the research was to determine the effect of planting media composition and biofertilizer doses on total fungal and bacterial population on planting media; as well as tea seedlings growth. The experiment was conducted using a randomized block design consisting of 9 treatments and 4 replications, each replication consisted of three plants. The treatment consisted of soil without biofertilizer (control), soil + 5% biofertilizer, soil + 10% biofertilizer, cocopeat without biofertilizer, cocopeat + 5% biofertilizer, cocopeat + 10% biofertilizer,  filter cake without biofertilizer, filter cake + 5% biofertilizer, and filter cake + 10% biofertilizer. Biofertilizer used in this experiment were Azotobacter vinelandii and Azotobacter chroococcum that was mixed with growing media before planting. Result of the experiment showed that planting media affect microbial populations differently. The population of total bacteria, total fungi and Azotobacter sp. in the filter cake media with 10% biofertilizer was significantly higher than those of other growing media. However, Azotobacter biofertilizer had no effect on seedling growth of tea (Camellia Sinensis (L.) O. Kuntze) clones Gambung-7

Abstract

Contaminant levels of cadmium (Cd) in tin mine tailings often exceeds the upper limit.  Relatively high Cd content might  decrease crop yields and threatened food chain. Bioremediation by using  indigenous microbes to lower soil Cd shoud be done prior to use tailings for cultivation. The objective of this experiment was to obtain information about the capacity of  indigenous bacterias to induce bacterial proliferation  in the rhizosphere, corn growth during the early vegetative stage and change the concentration of Cd in soil and plants. Pot experiment was set up in  a Randomized Block Design consist of several combination of  two bacterial isolates and their doses.  Maize seeds were sown in  tailings from the mine site on the island of Bangka and inoculated with indigenous bacteria. The results showed that the bacterial inoculation increased number of bacteria in the rhizosphere  and plant height, but only isolates BKH1 with a dose of 5 mL/plants that increased plant dry weight. Soil Cd content decreased when inoculated by bacteria although not statistically significant, while the effect of inoculation on plant Cd uptake was inconsistent.

Abstract

Azotobacter chroococcum based biofertilizers have been widely used in the production of environmentally agriculture. For commercial scale, cheap growth media for Azotobcter inoculant  will affect the quality and price of liquid biofertilizer. Growth medium should support cell proliferation as well as its  nitrogen fixation and phytohormone production.  The objective of this study was to determine whether  liquid organic fertilizer (LOF) containing low N can be used as a medium for scaling up  A. chroococcum inoculant without lowering the survival of bacteria and its activity in N2 fixation and phytohormones production. LOF generally contains a complete nutrient although in a small quantities. Experiment was set up in completely randomized design which tested two types of commercial LOF. The results showed that both of  LOF were only able to support cell growth up to 72 hours and at 96 hours cell viability began to decline compared to the control medium,  free-N Ashby. At 72 hour total N and cytokinin  concentration in LOF was lower than those in Ashby media. This study revealed  that LOF  could be used as A. chroococcum growth media, but without optimization of its composition, cell density and cytokinin content would be lower than those of inoculant  in Ashby Media.

Abstract

Organic matter and inorganic fertilizer application are important to maintain the existence of potential microbes in the rhizosphere and maintain the soil quality. A greenhouse experiment has been carried out to determine the effect of  vermicompost and NPK fertilizer on total bacterial and Azotobacter  population in rhizosphere and yield of lettuce (Lactuca sativa L.) grown in soil order of  Inceptisols. The experimental design was a factorial randomized complete block design with three replicates were tested three doses of vermicompost (7.5, 15 and 22.5 t ha-1) and four doses of inorganic fertilizers; NPK (300; 450; 600 and 750 kg ha-1). The results showed that there was no significant interaction between vermicompost and NPK fertilizer on either total bacterial or Azotobacter population, as well as on fresh weight of lettuce. Both vermicompost and NPK fertilizer did not affect the biological parameters. However, the application of 750 kg/ha NPK fertilizer can increased lettuce weight up to 24% compared to that of control. This study suggested that in the relatively poor soil nutrients, lettuce growth was depend on inorganic fertilizers.

Abstract

The use of biofertilizers is important to prevent the negative impact of the synthetic fertilizers in vegetable production.  This study aimed to determine the effect of single biofertilizer  Azotobacter, and consortial biofertilizer which consist of nitrogen fixer bacteria and pohosphate solubilizing bacteria on the growth and yield of green cabbage (Brassica juncea L). The experimental designed was  randomized block design with five replications. Green cabbage seedling were inoculated with biofertilizer Azotobacter and consortial biofertilizer  on the growth and yield of green cabbage. Plants with consortial biofertilizer showed better growth and yield than that received single biofertilizer. Consortial biofertilizer  can increase plant height and shoot fresh weight up to 15.68 % and 17.05 %, while those with  Azotobacter  were 7.77 % and 9.58 % consecutively, compared with plants control.

Abstract

Illegal artisanal gold mining in Kebunlado, Riau Province has damaged the smallholders estate. The use of mercury for extracting the gold is potentially pollute the soil and syrrounding water as well as threats plantation productivity and it’s food chain. The purpose of this research is to study the mining impact towards the physical and chemical properties of the soil around the plantation, and measure the mercury contamination within the tailings, soil and river to be compared with the soil sample near the mine area. Sampling was carried out in 10 locations to study the properties of the land surrounding the artisanal gold mining. Physical and chemical properties of the soil were studied. The samples came from the mine sites (7 samples) and its surrounding area (3 samples). Sampling was conducted using a toposequence transect method, based on its topographical conditions and soil parent material with the purposive method. Mercury level was measured from eight soil samples that were sampled from surrounding artisanal mine area and six water samples from the surrounding rivers. The research showed that the soil characteristics had changed after mining activities. The soil properties were higher in sand, with bulk density and permeability compared to intact soil. However, there was no distinct change in pH, total nitrogen, organic carbon as well as available P, except C/N in post mine area was relatively low. Mercury content in the gold mine area was higher than that of the average mercury within the soil around the world. The Mercury in the river stream exceeds the threshold based on Indonesia Regulation. The study suggested that land reclamation is needed to develop oil palm plantation in terms of improving the soil physics, increasing C/N and remediation as well as reducing the mercury in soil and water.

Abstract

Gold mine tailing in Buru island, Maluku Province, Indonesia contains high level of mercury but low in carbon and plant nutrients. Revegetation in such condition needs certain soil treatment which is suitable for plant growth. The objective of pot trials was to study the effect of indigenous Plant Growth Promoting Rhizobacteria Azotobacter and organic matter on bacterial survival as well as growth of groundnut grown in mine tailing. The experimental design was a Split Plot Design which tested three types of Azotobacter liquid inoculant and three soil total organic carbon (TOC) contents. Results showed that Azotobacter inoculation increased Azotobacter population in tailing at the end of vegetative growth of groundnut. Total nitrogen content in soil decreased when TOC level increased. However, nitrogen uptake and growth of groundnut did not change after Azotobacter inoculation or manure amendment. These experiments provided information that Azotobacter inoculation on groundnut maintain its proliferation in Hg-contaminated mine tailing.

Abstract

Plant Growth Promoting Rhizobacteria Pseudomonas and Bacillus are well known biofertilizer due to their ability for solubilizing inorganic phosphate and producing phytohormones in soil. However, their potential to decreased diseases incidence caused by Pratylenchus coffeae has been documented. The objective of this laboratory experiment was to formulate solid biofertilizer contained Pseudomonas diminuta and Bacillus subtilis. The laboratory experiments consisted of three-step experiment: 1) selection of the molasses-based liquid media for P. diminuta and B. subtilis growth, 2) organic liquid inoculant formulation and 3) organic carrier-based inoculant formulation. The population of both bacteria in all steps was count by serial dilution plate method with specific medium. The result verifeid that cell density of P. diminuta and B. subtilis in liquid culture with 2% molasses were higher than cell count in the culture contained 4% molasses. Bagasse-based carrier inoculated with liquid culture of mixed bacteria has higher bacterial count compared to manure-based carrier. After 90-day incubation, bagasse-based solid inoculant maintained P. diminuta and B. subtilis density up to 108 CFU/mL and the acidity of solid inculaant was about neutral. The bacterial count and acidity of solid inoculant agreed with Indonesian standard for solid biofertilizer
