Abstract

The purpose of this study was to determine the quality of frozen semen from landrace boars in modified citrate diluents with different glycerol concentrations. The research material used was fresh cement from 2-3 year old landrace boar studs from 4 boars. This study used an experimental study with a completely randomized design method consisting of 4 treatments and 4 replications. The treatments in question are T1: S-KT + G 3%, T2: S-KT + G 5%, T3: S-KT + G 7% and T4: S-KT + G 9%. The diluted cement was stored at room temperature (27-28℃) for two hours (holding time), then centrifuged at 3000 RPM for 15 minutes. After centrifugation, the supernatant was discarded and the centrifuged pellet was re-diluted with basic diluent. The diluted semen was packed in 0.5 Ml straws, then packed, arranged in a freezing rack, and then equilibrated at 3-5℃ for 2 hours. Then the straws were frozen above the surface of liquid N2 with a distance of 10 cm at a temperature of ±-110℃ for 10 minutes. The results of this study indicate that the addition of different glycerols did not have a significant effect (P>0.05) on the motility, viability, abnormality and recovery rate of spermatozoa. It was concluded that the addition of different glycerols had the same impact but were not suitable for IB.

Abstract

Efforts to improve cattle productivity can be done by using artificial insemination (AI) technology. Many factors determine the success of AI, one of which is the diluent used in semen preservation. The purpose of this study was to determine.the effect.of zinc level in.tris-egg yolk.diluent (T-EY) on the.quality of liquid semen of Angus bull. Semen is collected once a week using.an artificial.vaginal method from a three-year-old Angus bull in an undisturbed or healthy body and reproductive organs. Semen was diluted with T-EY diluent which had been added with zinc at several levels, 0 mg control treatment (T0), 10.mg (T1), 20.mg (T2), 30.mg (T3), 40 mg (T4). The diluted semen was.stored at a temperature of 3-5oC, then observed every 24 hours for.motility, viability,.abnormalities and survival of.spermatozoa to a minimum motility of 40%. The.results showed.that the addition of 20 mg of zinc in the T-EY.diluent (T2) resulted in a higher.quality of liquid semen of Angus bull (P˂0.05) than other.treatments. The quality of the liquid semen (motility, viability, abnormalities and survival of spermatozoa) were motility 44±5.75%, viability 48.39±13.59%, abnormality 6.91±1.85% and durability.live 5.60±0.89 days. It was.concluded that the.addition.of 20 mg zinc in the.tris-egg yolk diluent produced liquid semen of Angus bull with higher quality than other treatments.

Abstract

This research aimed to determine the best level of sun-dried carrot extract (SDCE) in the spermax-egg yolk (EY-S) diluent for preserving the sperm of duroc boars. Semen was collected from two 2-year-old healthy duroc boars. The study employed an experimental method with a completely randomized design (CRD), consisting of five treatments and five replicates. The five treatments were T1 = EY-S + SDCE 10%, T2 = EY-S + SDCE 12.5%, T3 = EY-S + SDCE 15%, T4 = EY-S + SDCE 17.5%, T5 = EY-S + SDCE 20%. Semen, which was diluted with the treatment diluent, was stored in a cool box at a temperature of 18-20°C and observed every 12 hours until sperm motility reached 40%. The results indicated that treatment P5 was able to maintain the quality of liquid boar semen better than the other treatments (P<0.05) with sperm motility (46.00%), sperm viability (51.92%), sperm abnormality (4.90%), and sperm survival (66.33 hours). It was concluded that a 20% level of sun-dried carrot extract in spermax-egg yolk diluent can preserve motility for up to 60 hours of storage. This study demonstrates that dried carrot extract can enhance the quality and storage capacity of spermatozoa, making it an effective alternative in storage media. This is beneficial in animal reproduction, particularly in artificial insemination, where sperm motility is crucial for fertilization. Furthermore, this research opens up opportunities to explore other natural substances in sperm diluents, reducing reliance on synthetic chemicals and improving the sustainability of reproductive practices.

Abstract

Pengenceran semen memiliki dua fungsi utama yaitu untuk memperpanjang masa simpan dan memperbesar volume semen. Penelitian ini bertujuan untuk mengetahui perbandingan penambahan ekstrak daun kelor (EDK) 5% dalam pengencer tris- kuning telur (T-KT), sitrat- kuning telur (S-KT) dan beltsville thawing solution®-kuning telur (BTS® -KT) terhadap kualitas semen babi landrace selama penyimpanan. Semen babi diambil seminggu sekali menggunakan metode masase dari satu ekor babi jantan berumur 2 tahun dengan kodisi sehat dan organ reproduksi yang normal. Semen yang berkualitas baik dibagi dan diencerkan dengan pengencer perlakuan yaitu : P1: T-KT – 5% EDK , P2: S-KT – 5% EDK dan P3: BTS® -KT – 5% EDK. Kemudian pengenceran semen disimpan dalam syrofoam pada suhu 18-22ᶱ C dan dievaluasi setiap 8 jam meliputi motilitas, viabilitas, abnormalitas serta membran plasma utuh (MPU). Rancangan yang digunakan adalah rancangan acak lengkap yang terdiri atas tiga perlakuan dan lima ulangan. Data yang telah diperoleh dianalisis dengan uji anova, apabila terdapat perbedaan yang nyata antar perlakuan maka dilanjutkan dengan uji duncan. Hasil penelitian menunjukkan bahwa pengencer S-KT modifkasi (P2) mempunyai kualitas yang lebih baik (P<0,05) dibandingkan dengan perlakuan lainnya yaitu selama 40 jam dengan motilitas (45,40±4,15 %), viabilitas (56,66±1,32) dan MPU (55,44±5,18%). Kesimpulan dari hasil penelitian ini yaitu pengenceran modifikasi terbaik adalah pengencer S- KT dengan penambahan EDK 5 % (P2).

Abstract

The research was conducted to find out the effect of Aloe vera gel administrated orally onreproductive performance of female rats. Fifteen two months old female rats were alloted to fivetreatments of per oral Aloe vera gel administration: P0 (control), P1(1 mg /g body weight), P2 (2 mg/gbody weight, P3 (3 mg/g body weight), and P4 ( 4 mg/g body weight). The gel were administrated everyday for four weeks. The body weight were measured on the same day as last treatment. The animal thenmated to proven male on 1:1 rasio. Positively mated animals were sacrificed on D-10 after mating tocollect uterus and ovary so the number of fetus, corpus luteum, and ovary weight can be determined. Thetreatments significantly decreased (P0.05) the average body weight gain. The P0 (4.1 g/head/day) wasdifferent to P1 (2.69 g/head/day), P2 (2.87 g/head/day, and P4 (2.68 g/head/day), but was not different toP3 (3.39 g/head/day). The ovarium weight were not different (P0.05) among the treatments, so did thenumber of corpus luteum (CL), the number of fetuses and the CL: fetus rasio.Keywords: Aloe vera, rats, reproductive performance .

Abstract

The aim of this research was to determine the effect of adding dry moringa seed extract (DMSE) added to the Beltsville thawing solution (BTS) diluent on the quality of landrace boar semen during storage. This research used experimental methods and a completely randomized design  consisting of 5 treatments and 5 replications to form 25 experimental units. The treatment in question was : T0 = BTS EY + 0% DMSE, T1 = BTS EY + 1% DMSE, T2 = BTS EY + 2% DMSE, T3 = BTS EY + 3% DMSE, T4 = BTS EY + 4% DMSE. This diluted semen will be stored at a temperature of 18-20ºC and evaluation of motility, viability, abnormalities and survival of spermatozoa was carried out every 8 hours of storage. Data was analysis of the average, standard deviation, variance and continued with the Duncan test. The results showed that T2 treatment with a 2% DMSE level at 32 hours of storage gave the best results (P<0.05) compared to other treatments, with motility values (51, 80±4.71), viability (65.54±5.00), abnormality (5.81±0.81 and survival value of (38.89±1.21). It was concluded that the addition of EBKK at a level of 2% to the BTS diluent provided a good response in maintaining motility, viability and abnormalities.

Abstract

This study aims to identify the substitution of native honey as an energy source to replace fructose in Tris-KT diluent to improve the quality of spermatozoa of Etawah crossbreed goats (PE). The study was conducted at the Nekamese Vocational School and the Wiliams and Laura Foundation Biological and Reproductive Laboratory for 2 months. In this study, four PE male goats were used as a source of semen. Cement is accommodated twice a week. The semen obtained was evaluated macroscopically and microscopically then diluted with T-KT diluent with the addition of honey divided into P0 (control), P1 (T-KT + 0.25 w/v honey), P2 (T-KT + 0.50 w/v honey) and P3 (T-KT + 0.75 w/v honey). After dilution the cement is stored at a temperature of 3-5°C.  Semen is evaluated post-dilution and post-storage every 24 hours, until the movement of spermatozoa is at least 40%. The variables tested were motility and viability of spermatozoa. P3 provides the best results in maintaining cement quality. The results showed that honey substitution in Tris-KT diluent exerted a marked effect on the motility and viability of spermatozoa (P<0.05) during storage.

Abstract

Semen babi pada dasarnya bersifat produktif dengan kosentrasi yang rendah, hal ini berpengaruh terhadap kelebihan semen setelah penampungan. Penelitian ini bertujuan untuk menguji efektivitas sari buah melon-kuning telur (SBM-KT) dalam pengencer sitrat-kuning telur (S-KT) terhadap kualitas spermatozoa babi landrace. Bahan yang digunakan adalah semen segar babi landrace kualitas baik yang diencerkan dengan pengencer perlakuan sebagai berikut: P0: S-KT 100%, P1: S-KT 90% + SBM-KT 10%, P2: S-KT 80% + SBM-KT 20%, P3: S-KT 70% + SBM-KT 30%, P4: S-KT 60% + SBM-KT 40%. Semen yang telah diencerkan disimpan pada suhu 18-20o C dan dievaluasi setiap 8 jam terhadap motilitas, viabilitas, abnormalitas dan daya tahan hidup spermatozoa. Hasil penelitian menunjukkan bahwa kualitas spermatozoa dalam pengencer sitrat-kuning telur tanpa penambahan sari buah melon (P0) memperlihatkan hasil lebih tinggi dibandingkan dengan perlakuan lainnya dengan motilitas (45,00±1,76), viabilitas (55,86±1,11), abnormalitas (4,55±0,54) dan daya tahan hidup (45,30 jam). Dari data diatas maka dapat disimpulkan bahwa substitusi sari buah melon dalam pengencer sitrat-kuning telur tidak efektif untuk mempertahankan kualitas spermatozoa babi landrace. Kesimpulan penelitian ini menunjukkan bahwa pengencer dengan komposisi yang berbeda dapat memengaruhi kualitas spermatozoa selama penyimpanan, dan perlakuan tanpa penambahan sari buah melon memiliki efek yang lebih baik dalam mempertahankan motilitas, viabilitas, abnormalitas, dan daya tahan hidup spermatozoa babi Landrace.

Abstract

ABSTRACT Background: The research material used was fresh semen of 2-year-old landrace boars of good quality (spermatozoa motility ≥ 70% and spermatozoa abnormality < 20%) Purpose: This study aimed to find the best combination of young coconut water (YCW) and Tris to improve the quality of landrace boars spermatozoa. Method: %). This study used an experimental method according to a complete randomized design procedure with five treatments and five replicates. The treatments were: T0= YCW, T1= TCW 75% + tris 25%, T2= YCW 50% + tris 50%, T3= YCW 25% + tris 75%, T4= Tris. All treatment diluents were added with 20% egg yolk, 3% moringa leaf extract, 1,000 IU/mL penicillin, and 1,000 µg/mL streptomycin. The variables tested were motility, viability, abnormality, and survival. Results: The observation results at the 40th hour of storage showed that treatment T3 was significantly different (P<0.05) on motility, viability, abnormality and survival but on the variable viability treatment T3 was insignificantly different from T1 (p>0.05), but significantly different from treatment T0, T2, and T4; and for spermatozoa abnormality variables in all treatments were the same (p>0.05) Conclusion: It was concluded that the combination of YCW 75% and tris 25% and 100% tris effectively maintained the quality of landrace boars spermatozoa

Abstract

Background: The benefits of frozen semen can improve the genetic quality of the offspring, the wide distribution of superior genetics to various locations even in distant areas. Purpose: The purpose of this study was to determine the optimal level of egg yolk in spermax diluent for the preservation of frozen semen of landrace boars. Methods: The design used was a completely randomized design consisting of four treatments with four replications each, namely: P1 = Spermax + 5% egg yolk, P2 = Spermax + 10% egg yolk, P3 = Spermax + 15% egg yolk, and P4 = Spermax + 20% egg yolk. Each treatment diluent was supplemented with 6% glycerol. Semen was diluted in the base diluent and stored for 2 hours at room temperature (27–28 °C). Next, the semen was centrifuged at 2000 rpm for 10 minutes, the supernatant was discarded, and the sediment was re-diluted. The semen was then evaluated for sperm motility, viability, and abnormalities, packaged in 0.5 mL straws, and equilibrated for 2 hours at a temperature of 3–5 °C. The semen was re-evaluated and frozen on the surface of liquid nitrogen at a distance of 10 cm for 10 minutes, then stored in a container filled with liquid nitrogen for 24 hours. The frozen semen was subsequently evaluated for motility, viability, abnormalities, and recovery rate. Results: The results of the study did not have a significant effect (T> 0.05) with the highest values in each treatment after thawing being motility (T3 = 28.75 ± 11.08%), viability (T4 = 47.52 ± 18.14%), abnormality (T1 = 9.84 ± 2.05%) and recovery rate (T3 = 35.93%). Conclusion: Addition of egg yolk with levels of 5%, 10%, 15% and 20% in the spermax diluent produces the same quality of frozen landrace boar semen.
