Abstract

The purpose of this study was to examine the effect of adding various doses of virgin coconut oil to the quality of bali bull spermatozoa during preservation. Semen is accommodated twice a week using an artificial vaginal method from two 3-4 year old bali bull with normal body condition and reproductive organs. Semen with motility> 70%, concentration> 1000 × 106, and abnormality <15% was diluted with Tris-egg yolk (T-KT) diluent added with virgin coconut oil (VCO) at a concentration of: 0% (T0), 2 % (T1), 4% (T2), and 6% (T3). The diluted semen was stored at 3-5ᶱC. Evaluated every 24 hours for motility, viability, abnormalities and vitality of spermatozoa up to a motility of at least 40%. The results showed that semen diluted with T-EY with 6% VCO (T3) had a higher quality (P <0.05) compared to the other three treatments, with motility reaching (43.10 ± 2 , 83%), viability (48.68 ± 1.59%), abnormalities (5.92 ± 0.90%) and survival (6.00 ± 0.00 days). The conclusion of this study is the addition of 6% virgin coconut oil in Tris-egg yolk diluent is more effective in maintaining the quality of bali cattle liquid semen.

Abstract

This study aims to determine the effect of adding various doses of carrot juice into citrate-egg yolk diluents (C-EY) on bligon buck semen quality. Semen was collected twice a week using an artificial vaginal method of three buck with normal body condition and reproductive organs. Semen was 70% of motility, konsentration ≥1000x106mL and abnormality ≤ 15 diluted with C-EY added with carrot juice at a concentrations: 0% (CJ-0), 10% (CJ-10), 12.5% (CJ-12,5), 15% (CJ-15), 17.5% (CJ-17,5), and 20% (CJ-20), and stored in a refrigerator at 3-5o C. The quality of spermatozoa was evaluated every 24 hours. The Variables measured were spermatozoa motility, viability, and abnormalities. The results showed that spermatozoa preserved by C-EY supplemented by 17.5% carrot juice (CJ-17.5) had a higher quality (P<0.05) compared to the other four treatments, with motility (49.04 ± 1.0%), viability (55.70 ± 1.22%), dan abnormalities (4.57 ± 0.88 %) on the 4th day of storage. This study concludes that 17.5% carrot juice into citrate eggyolk extender improves the bligon buck semen

Abstract

This study aims to determine the effect of equilibration time on the quality of landrace spermatozoa both pre- and post-freezing and to find the best equilibration time. The materials used is fresh semen collected from a 2,5 years old landrace boar. Collecting was carried out twice per week using the glove hand method. Semen of good quality (sperm motility ≥70%, sperm concentration ≥200 x 106 cells/ml, and percentage of sperm abnormalities ≤15%) was diluted with durasperm that had been modified with lontar fruit juice, while the cryoprotectants used glycerol and sucrose. Holding time was carried out for 2 hour at 27-28°C, centrifuged at 2000 rpm for 15 minutes, and the sperm sediment was diluted with the same diluent. After that, the liquid semen was packed in a 0.5 mL straw and equilibrated at 3-5°C for 1 (P1), 2 (P2), or 3 hours (P3). The freezing of semen was carried out on the surface of liquid nitrogen at a distance of 5 cm for 10 minutes, and then stored in a liquid nitrogen container. Thawing is done by placing the frozen semen straws in warm water (37°C) for 30 seconds. The research data were analyzed with analysis of variance and continued with the Duncan test. The results showed that the equilibration time at 2 hours (P2) gave the best results (P<0,05) with the mean percentage of motility, viability, abnormality, intact plasma membrane and recovery rate respectively were 30,62%, 58,21%, 4,68%, 58,94% and 43,74%. It was concluded that the equilibration time did not affect the quality of pre-freezing spermatozoa, but it did affect the quality of post-freezing spermatozoa with the best equilibration time to maintain the quality of frozen semen of landrace boar was 2 hours.

Abstract

This study aimed to examine the quality of frozen sperm of duroc boar in modified Tris diluent with different equilibration time. Semen was collected from two duroc males and evaluated macroscopically and microscopically. Semen with motility 75%, concentration 200x106 cell/mL and abnormality <20% was diluted in Tris modified diluent, then held for two hours at room temperature (27-28ºC). The semen that has been in the holding time is then centrifuged at 2000 rpm for 15 minutes. The pellet from centrifuge were diluted again with modified Tris diluent + 3% glycerol. The diluted semen was packed in 0,50 mL mini straws and equilibrated according to the treatment equilibration time, namely P1: 1 hour equilibration time, P2: 2 hour equilibration time, P3: 3 hour equilibration time. After equilibration , it was frozen over liquid nitrogen vapor (-110ºC) at a distance of 5 cm for 10 minutes, then stored in liquid nitrogen (-196ºC) and left for 24 hours. Then thawing with warm water at 37ºC for 30 seconds in a water bath. The result showed that motility, viability, abnormality, intact plasma membrane and recovery rate were significantly higher (P<0,05) at two hour equilibration , namely 23.12%, 45.44%, 4.65%, 50,15%, 33.03% respectively compared to one and two hour equilibration time, namely 20%, 42.17%, 8.19%, 42.67%, 28.57% and 16,87%, 36.39%, 7.64%, 37.73%, 24.99% respectively. This study conclude that the equilibration time of two hours resulted in higher quality of frozen sperm of duroc boar than one and three hours.

Abstract

One of the diluent containing lipoproteins and lecithins that function in maintaining and protecting the integrity of the plasma membrane of spermatozoa from cold shock is soy milk. The purpose of this study was to determine the effect of the combination of soy bean milk and egg yolk citrate liqud on the quality of liquid semen of bali cattle. The material used in this study was fresh semen of 3 year old bali cattle. Semen was diluted with egg yolk citrate diluent with a combination of soy bean milk, namely T0: 100% citrate egg yolk (CEY); T1: CEY 75%+soy bean milk (SBM) 25%; T2: CEY 50%+SBM 50%; T3: CEY 25%+SBM 75%; and T4: 100% SBM. Liqud semen was stored at a temperature of 3-5oC. The parameter studied were motility, viability, abnormality and liveability of spermatozoa. The research data ware analyzed using an analysis of variance (ANOVA) and followed by the multiple range test. The results showed that T1 significantly difference (P<0.05) from other treatments, with motility of 42.40±2.50%, viability 52.15±6.31%, abnormality 6.44±0.84%, and liveability 6.00±0.00 days. From these result it could be concluded that the combination of 25% soy bean milk and 75% egg yolk citrate liqud was the best treatment, and spermatozoa can be stored until six days.

Abstract

The research was conducted to find out the effect of Aloe vera gel administrated orally onreproductive performance of female rats. Fifteen two months old female rats were alloted to fivetreatments of per oral Aloe vera gel administration: P0 (control), P1(1 mg /g body weight), P2 (2 mg/gbody weight, P3 (3 mg/g body weight), and P4 ( 4 mg/g body weight). The gel were administrated everyday for four weeks. The body weight were measured on the same day as last treatment. The animal thenmated to proven male on 1:1 rasio. Positively mated animals were sacrificed on D-10 after mating tocollect uterus and ovary so the number of fetus, corpus luteum, and ovary weight can be determined. Thetreatments significantly decreased (P0.05) the average body weight gain. The P0 (4.1 g/head/day) wasdifferent to P1 (2.69 g/head/day), P2 (2.87 g/head/day, and P4 (2.68 g/head/day), but was not different toP3 (3.39 g/head/day). The ovarium weight were not different (P0.05) among the treatments, so did thenumber of corpus luteum (CL), the number of fetuses and the CL: fetus rasio.Keywords: Aloe vera, rats, reproductive performance .

Abstract

This study aims to find the best level of sun-dried Moringa leaf extract (SDMLE) in Beltsville Thawing Solution (BTS) diluent to preserve the spermatozoa of Landrace boars. Semen from a two-year-old Landrace boar was collected using the massage method. Semen with spermatozoa motility ≥70% and spermatozoa abnormalities <20% was diluted with BTS (P0, control), BTS + 3% SDMLE (P1), BTS + 6% SDMLE (P2), BTS + 9% SDMLE (P3), and BTS + 12% SDMLE (P4). Storage of liquid semen using a cool box with a temperature of 15-20oC, while evaluating the quality of semen every 8 hours for spermatozoa motility, viability, abnormality, and longevity. The results showed that spermatozoa stored for 40 hours in BTS + 3% SDMLE (P1) diluent had higher quality (P<0.05) than the other four diluents, namely motility (54.00%), viability (61.16%), abnormality (3.62%), and longevity (45.77%). The results of this study concluded that 3% SDMLE added to BTS diluent was the best level for the preservation of spermatozoa in landrace boars.

Abstract

This study aims to identify the substitution of native honey as an energy source to replace fructose in Tris-KT diluent to improve the quality of spermatozoa of Etawah crossbreed goats (PE). The study was conducted at the Nekamese Vocational School and the Wiliams and Laura Foundation Biological and Reproductive Laboratory for 2 months. In this study, four PE male goats were used as a source of semen. Cement is accommodated twice a week. The semen obtained was evaluated macroscopically and microscopically then diluted with T-KT diluent with the addition of honey divided into P0 (control), P1 (T-KT + 0.25 w/v honey), P2 (T-KT + 0.50 w/v honey) and P3 (T-KT + 0.75 w/v honey). After dilution the cement is stored at a temperature of 3-5°C.  Semen is evaluated post-dilution and post-storage every 24 hours, until the movement of spermatozoa is at least 40%. The variables tested were motility and viability of spermatozoa. P3 provides the best results in maintaining cement quality. The results showed that honey substitution in Tris-KT diluent exerted a marked effect on the motility and viability of spermatozoa (P<0.05) during storage.

Abstract

This study was conducted with the aim of evaluating the effect of adding aloe vera extract (ave) in egg yolktris diluent on the quality of spermatozoa of landrace boar during storage. The material used was good qualitysemen of landrace boar, diluted with treatment diluents viz: TO: (100%), T-EY, T1: 98% T- EY + 2% AVE, T2: 96% T-EY + 4% AVE, T3: 94% T- EY + 6% AVE.Storage of diluted semen at 180 C, and evaluationmotility, viability, abnormality and longerly of spermatozoa every 8 hours. If the data obtained, there aresignificant differences, continue with the Duncan Test. The results showed that the addition of aloveraextract in the Tris-Egg Yolk diluent has no significant effect (P>0.05) on the spermatozoa quality of landraceboar, but the storage time for 8-48 hours P0, P1 and P2 were significant effect (P < 0.05) with P3 on themotility and viability. The conclusion of this study is that the addition of aloevera extract in Tris-Egg Yolkdiluent did not effective in maintaining the quality of spermatozoa landrace boars. Penelitian ini dilaksanakan dengan tujuan untuk mengevaluasi pengaruh penambahan ekstrak lidah buaya(ELB) dalam pengencer tris kuning telur (T-KT) terhadap kualitas spermatozoa babi landrace selamapenyimpanan. Materi yang digunakan adalah semen segar babi landrace yang berkualitas baik, masingmasing diencerkan dengan: PO: (100%) tris-kuning telur, P1: 98% tris kuning telur + 2% ekstrak lidahbuaya, P2: 96% tris kuning telur + 4% ekstrak lidah buaya, P3: 94% tris kuning telur+ 6% ekstrak lidahbuaya. Penyimpanan semen hasil pengenceran pada temperature 180C, dan evaluasi motilitas, viabilitas,abnormalitas dan daya tahan hidup spermatozoa setiap 8 jam. Data dianalisis menggunakan analysis ofvariance (ANOVA) dan jika terdapat pengaruh perlakuan dilanjutkan dengan uji Duncan menggunakanprogram softwere SPSS 29.0 for windows. Hasil penelitian menunjukkan bahwa penambahan ekstrak lidahbuaya dalam pengencer Tris-Kuning telur tidak berpengaruh nyata (P>0,05) pada kualitas spermatozoa babilandrace, tetapi pada lama waktu penyimpanan selama 8-48 jam P0: (100%) tris-kuning telur, P1: 98% triskuning telur + 2% ekstrak lidah buaya, dan P2: 96% tris kuning telur + 4% ekstrak lidah buaya berbeda nyata(P<0,05) dengan P3: 94% tris kuning telur+ 6% ekstrak lidah buaya pada motilitas dan viabilitas.Kesimpulan dari penelitian ini adalah penambahan ekstrak lidah buaya dalam pengencer Tris-Kuning Telurtidak efektif dalam mempertahankan kualitas spermatozoa babi landrace.
