Abstract

Uji secara molekuler dilakukan dengan menentukan satu penanda yang tepat untuk menganalisis beberapa kajian dalam bidang molekuler. Uji secara molekuler tumbuhan dilakukan menggunakan penanda gen rbcL (Ribulose-1,5-biphosphate carboxylase/oxygenase large subunit gene). Tumbuhan pegagan (Centella asatica (L.) Urban) merupakan herba yang pada umumnya hidup secara liar di tempat terbuka seperti padang rumput, kebun, tepi jalan, dan tepi parit. Tempat tumbuh pegagan seperti dataran tinggi, dataran rendah, dan pesisir pantai sangat mempengaruhi perubahan dari segi molekuler. Tujuan penelitian ini untuk mengetahui bagaimana karakteristik molekuler, keragaman molekuler (keragaman genetik) dan hubungan filogenetik tumbuhan pegagan (Centella asiatica (L.) Urban) dengan menggunakan gen rbcL. Penelitian ini memiliki tahapan yaitu koleksi sampel, isolasi DNA, amplifikasi DNA dengan gen rbcL, elektroforesis, sekuensing, dan analisis data dengan MEGA 11. Hasil penelitian dari amplifikasi menggunakan gen rbcL pada DNA genom Centella asiatica (L.) Urban yang diteliti menghasilkan panjang amplikon kurang lebih 600 bp. Pada penyatuan sekuens yang konsesus pada sampel mendapatkan sekuens dengan panjang D1 567 bp dan D2 562 bp. Pembentukan pohon filogenetik menggunakan metode Neighbor-Joining dan model perhitungan Kimura-2-Parameter menunjukkan Centella asatica (L.) Urban termasuk dalam kelompok monofiletik. Hasil analisis keragaman molekuler Centella asatica (L.) Urban menunjukkan bahwa pada seluruh sampel yang dikoleksi dari Desa Pantai Labu Pekan Kabupaten Deli Serdang dan Jl. Air Bersih Ujung Kota Medan memiliki keragaman molekuler. Maka dari itu, dapat disimpulkan bahwa gen rbcL menghasilkan barcode DNA dapat digunakan sebagai metode untuk mengidentifikasi Centella asatica (L.) Urban secara molekuler dan sangat efisien dalam menentukan hubungan kekerabatan dengan spesies yang lainnya.

Abstract

Indonesia recognizes several legal systems i.e., state law, customary law, Islamic law, and international law. Islamic sharia in Aceh is part of legal pluralism in Indonesia. This study aims to discuss the application of Islamic sharia from the perspective of legal pluralism. This normative legal study employed a legal pluralism theory analysis. Legal pluralism is a theory that views law not only as positive or written law made by the state but also as a recognition of the legal reality that exists in a pluralistic society. The study collected data by means of a literature review. Findings revealed that Islamic sharia in Aceh as part of the recognition of the concept of legal pluralism has been implemented well in the context of state law, custom, and Islamic law. The state has provided juridical legitimacy through laws in the context of legal substance and judicial institutions or sharia courts. Likewise, revitalization has also occurred in traditional institutions in terms of customary law and customary justice. The argument emphasized in this study is that Islamic sharia in the context of legal pluralism has succeeded in manifesting legal harmonization between the state, custom, and Islamic law

Abstract

Rajungan crab (Portunus sp.) is one type of crab that has a fairly high commodity value, this species has a fairly wide distribution covering the Indo-Pacific Ocean and also offshore, this crab has an important role in ecological stability. Portunus sp. has a more attractive appearance compared to other crabs. The purpose of this study is to look at the (dominant) crab species found in January (west wind season) in the waters of Percut Sei Tuan based on the genes CO1, and phylogenetics (kinship relationship) of Portunus sp. in the waters of Percut with Portunus sp. in NCBI or Genebank. The method used in this study is DNA isolation on specimens using the Favorgen kit locus of the CO1 gene in the genome of the Portunus sp. amplified by PCR technique with LCO1940 (forward) and HCO2198 (reverse) primers. The results of the research that have been carried out have found 3 species from the genus Portunus, namely P. sanguinolentus, P. trituberculatus and P. pelagicus. Of the three species found in the Percut Sei Tuan Waters, P. trituberculatus and P. pelagicus have a closer kinship relationship compared to P. sanguinolentus, based on the percentage of AT content and GC content, the AT content is higher at 61.03% which means that the three species have primitive properties. The genetic distance between P. sanguinolentus and P. trituberculatus is closer (19.32) compared to P. pelagicus. Based on the phylogenetic tree reconstruction, P. trituberculatus and P. sanguinolentus are closer to P. pelagicus with the Neighbor-Joining method and the Kimura-2-Parameter calculation model. Thus, it can be concluded that DNA barcoding using the CO1 gene locus can be used as a method in identifying Portunus sp. molecularly and kinship relationships with other species.

Abstract

Anadara sp. clams are very popular clams among the community. Anadara sp. also usually live in waters with muddy and sandy surfaces. DNA barcoding is a barcode sequence to identify unknown species. CO1 is a molecular marker commonly used in molecular research on animals. The purpose of this study was to determine the species of Anadara sp. clams found in Percut Sei Tuan Waters using gene markers, the diversity of Anadara sp. clams using CO1 Gene Markers, and the kinship of Anadara sp. clams found in Percut Sei Tuan Waters. This study has 4 stages, namely DNA isolation using the Favorgen kit, PCR amplification, electrophoresis and sequencing. The DNA sequencing results were analyzed using the MEGA 11 application. The results showed that of the three samples that had been analyzed using the CO1 gene marker obtained using BLAST and NCBI, namely the Anadara granosa, Anadara antiquata, and Anadara gubernaculum samples. With the calculation results of higher AT content (28.9%) and lower GC content (20.12%) which states that the species is more primitive. In the pairwise distance matrix, the samples of Anadara antiquata (Percut Sei Tuan) and Anadara granosa (Percut Sei Tuan) which are in the same clade with different sub clades have a genetic distance ranging from 111.45 to 213.77. The phylogenetic tree also shows that the three samples have a close kinship relationship.

Abstract

This study aims to identify DNA barcodes with the rbcL gene locus and reconstruct the phylogenetic relationship of the Balakka plant with species belonging to the genus Phyllanthus. This research includes experimental research by isolating DNA, amplification DNA using rbcL gene as primer, electrophoresis and sequencing. DNA was isolated with a commercial kit, amplification was carried out using the Polymerase Chain Reaction method. Sequence results and phylogenetic tree reconstruction were analyzed by Bioinformatics (MEGA 11). The results showed that the number of amplicons from the consensus of the two primers on the balakka plant sample was 557 bp. The phylogenetic tree of the Balakka sequence formed a sister clade with Phyllantus emblica KY9888320.1 with 66% similarity, and P. emblica grouped together, but still had common ancestor branches with other species of the Phyllantus genus using the Neighbor-Joining method, and Kimura 2-parameter calculations.

Abstract

Healthy and clean environment is something that must be initiated for the survival of future generations so that it is not threatened by the consequences of irresponsible human behaviour. This community service is inspired by the lack of students’ awareness in maintaining environmental cleanliness and the lack of cleaning facilities at MTs MIM Langsa. The procedure of this assisting activity is carried out through 3 stages, namely: planning, implementing, and reflecting, which begins with field observation to the target school. This potential-based community service activity is in the form of counseling/socialization and mentoring on school environmental hygiene. Socialization activities targeting OSIM administrators of MTs MIM Langsa as role models are expected to motivate other students to always maintain school cleanliness. Meanwhile, other activities include painting garbage cans and providing posters with the theme of environmental cleanliness. The provision of cleaning facilities in the form of garbage cans in several places in the school environment has received positive responses and support from the school community, one of which is showed by students who responsibly do class pickets and do not litter.

Abstract

Fraction is one of the difficult topic for students. One way to reduce students’ difficulties is to implement Realistic Mathematics Education (RME) by integrating Islamic values such as Sacrifice Worship (Ibadah Kurban). In addition, the use of technology to integrate sacrificial worship in fraction learning is also expected to increase students' interest in learning in the 21st century and can improve students' understanding. Therefore, it is necessary to develop an e-module for learning fractions through RME with the context of Sacrifice worship. The developed e-module contains learning trajectories, live worksheets, videos, and post-tests. This type of research is research and development which follows the stages of Plomp, especially in the prototyping and assessment stages. The instruments in this study were validation sheets. The validators consisted of one mathematics education lecturer and two Banda Aceh City junior high school teachers. Based on the validity test it was concluded that the e-module meets the valid criteria. The implication of this research is the development of e-modules needs to be continued to test practicality and effectiveness.

Abstract

Teaching and learning process is inseparable from method and technique applied by educators, especially teachers as an attempt to improve the quality of output learning. Place mat technique is one of the cooperative learning methods in which students could work by themselves first and then team up with other group’s members in solving an issue or topic given. They can write down their idea as well as group’s opinion in a big place mat paper. Through the initial observation by interviewing some teachers of SMAN 1 Kuta Baro, Aceh Besar (Great aceh), it was found out that most of them never use this kind of technique in the classroom setting; they use some other kinds of techniques such as role play, jigsaw, numbered head together, and conventional technique. Therefore, the writers intend to carry on a community service to the students in the school as an effort to vary and increase the quality of teaching and learning process. This community service is hold on October 5-7, 2022 in SMAN 1 Kuta Baro, Great Aceh. This service is meant to train those teachers to be able to apply the technique during the teaching. Then, it is suggested to further community service activists to do the same activity in the rural areas as they basically need more input and insight from other educators in upgrading the way they teach.

Abstract

This study investigates how gender influences suprasegmental features—specifically pitch, duration, and intensity—in the pronunciation of Indonesian-origin words. The research addresses the problem of limited phonetic studies examining gender-based acoustic variation in Indonesian, particularly in devoiced stop consonants. The objective is to analyze whether and how gender differences manifest in these prosodic elements. Using a descriptive qualitative method, data were collected from ten participants—five males and five females—from diverse ethnic backgrounds (Bataknese, Javanese, Karonese, and Melayunese), all residing in Medan, North Sumatra. Nine commonly used Indonesian-origin words were selected, and acoustic features were analyzed using PRAAT software. The results show that female speakers consistently produced higher pitch values across all words, with peaks reaching up to 499 Hz, while male speakers demonstrated lower and narrower pitch ranges, as low as 94 Hz. Duration values also varied more among female speakers, ranging from 0.097 ms to 0.469 ms, indicating more dynamic articulation patterns, although some male speakers also showed extended durations in specific contexts. Intensity levels ranged from 23 dB to 54 dB, with female speakers exhibiting greater variability in loudness—from soft to very loud—whereas male speakers maintained a more stable and moderate intensity. These findings suggest that pitch differences are primarily influenced by physiological factors such as vocal fold structure, while duration and intensity are more reflective of individual articulation style, emotional expressiveness, and speech clarity. Despite limitations in audio editing features in PRAAT, the study provides new insights into how gender and speaker identity shape suprasegmental variation in Indonesian phonetics.

Abstract

Background: Meatballs are a popular processed meat product in Indonesia. High consumption of meatballs is accompanied by increasing public concern, especially among Muslims, regarding the halal aspect of the product. The potential contamination of pork DNA (Sus scrofa domesticus) in beef and chicken meatballs is a critical issue, whether due to unintentional cross-contamination or adulteration. Tuntungan area, North Sumatra, with its diverse demographic and trading characteristics, is an important location to assess the level of contamination. Objective: This study aimed to detect the presence of pork DNA (Sus scrofa domesticus) contamination in processed beef and chicken meatball products traded in the Tuntungan area, North Sumatra. Methods: Four meatball samples (three chicken meatballs and one beef meatball) were collected from street vendors and meatball stalls at different locations. DNA detection was performed molecularly using the Real-Time Polymerase Chain Reaction (qPCR) method with the *Genechecker UF-300* instrument. The procedure began with sample preparation, DNA extraction using the Genolution tool, master mix preparation, and analysis using the PCR Gen Checker chip. Result validation was based on the Cycle threshold (Ct) value in the target channel (FAM for pork DNA) and the internal control channel (ROX). Results: The analysis results of all four samples showed a FAM Ct value of 0 (not detected), indicating no amplification of specific pork target DNA. Meanwhile, the ROX Ct value (internal control) in all samples was detected in the range of 19.41–20.50, proving that the DNA extraction and amplification process ran optimally without inhibition. The positive control showed valid amplification signals, and the negative control showed no contamination. Conclusion: Based on the molecular detection results, it can be concluded that all tested beef and chicken meatball samples from the Tuntungan area were not contaminated with pork DNA (Sus scrofa domesticus). This finding indicates that traders in the area have applied good processing practices and separated raw materials, so the meatballs sold meet the halal aspect in terms of ingredient authenticity.
