Abstract

Di Indonesia, kerang Anadara dikenal dengan nama umum kerang bulu dan kerang darah. Informasi mengenai variasi genetik spesies ini masih terbatas. Penelitian ini bertujuan untuk menganalisis sidik jari DNA kerang Anadara guna mengidentifikasi polimorfisme dan variasi genetik antar individu dan antarpopulasi. Penelitian ini menggunakan metode analisis keragaman genetik pada populasi kerang Anadara menggunakan aplikasi teknik sidik jadi DNA, jenis primer yang efektif untuk mengidentifikasi pola polimorfisme, dan menentukan kekerabatan yang terbentuk dari teknik sidik jari DNA menggunakan marker Random Amplified Polymorphic DNA (RAPD) menggunakan enam primer yaitu, OPD 20, OPC 02, OPA 2, OPA 13, UBC 101 dan UBC 456. Hasil penelitian menunjukkan bahwa teknik RAPD berhasil mengidentifikasi variasi genetik yang signifikan antar individu kerang Anadara. Total 27 pita DNA teramati dengan ukuran berkisar antara 200-3000 pasang basa. Tingkat polimorfisme mencapai 100% dengan rata-rata 5-8 pita polimorfik perprimer. Terdapat satu primer monomorfik dan satu primer tidak ter amplifikasi. Tingkat kemiripan genetik antar individu bervariasi dari 0,68 hingga 1.00, mengindikasikan keragaman genetik yang substantial. Dendogram hasil analisis menunjukkan pengelompokan yang jelas berdasarkan hasil yang di dapatkan dalam PCR-RAPD. Berdasarkan hasil penelitian yang telah dilakukan, dapat disimpulkan bahwa metode RAPD efektif dalam mengungkap variasi genetik kerang Anadara. Temuan ini memberikan kontribusi penting dalam memahami struktur genetik spesies.

Abstract

Ikan mas koki (Carrasius auratus) adalah ikan memiliki ciri khas yang tersendiri. Keunikan dan keragamannya dapat diukur dari warna yang cemerlang, bentuk dan kelengkapan fisik. Tujuan penelitian ini adalah untuk mengetahui primer yang efektif, polimorfik genetik DNA dan kekerabatan pada Carrasius auratus yang terdapat di pembudidayaan ikan hias, Binjai Utara dengan menggunakan metode RAPD dengar enam primer yaitu OPA 13, UBC 101, UBC 456, OPD 20, OPA 02 dan OPC 02. Tahapan yang dilakukan pada penelitian ini diantaranya isolasi DNA menggunakan Kit Favorgen, amplifikasi PCR, dan elektroforesis. Hasil penelitian menunjukkan primer yang teramplifikasi dan menghasilkan pita polimorfik adalah primer OPA 13, OPA 02 dan OPD 20. Jumlah pola pita dari hasil amplifikasi primer tersebut sebanyak 32 pita dengan panjang basa yang berkisar antara 100 bp – 1500 bp. Pita yang muncul kemudian diterjemahkan kedalam data biner dan dianalisis dengan program NTSYS versi 2.02. Menunjukkan bahwa kekerabatan terdekat adalah pada ikan mas koki jenis Ryukin dan Oranda dengan memiliki koefisien 0,74 atau sebesar 74%, sedangkan kekerabatan jauh pada Ikan Mas Koki Rachu dan Teleskop dengan koefisien sebesar 0,75 atau sebesar 75%. Dari data hasil panjang pita 900 bp, 600 bp, 400 bp, 300 bp 200 bp dan 100 bp dari komponen pertama memiliki peran penting dalam pengelompokkan Carrasius auratus dalam dendogram.

Abstract

Teaching English to children is crucial. English plays a significant role internationally, providing access to information and technology. Effective teaching for young learners requires good planning, suitable methods, and appropriate materials. The purposes of this study were to (1) identify the visual aids that English teachers at MIN 2 Langsa utilize to teach English to primary pupils, and (2) investigate the teacher's experiences with these aids. Data collection was done in December 2023 through a fifth-grade English teacher at MIN 2 Langsa. The phenomenological approach was combined with a qualitative method in this study. An interview and an observation served as the instruments. The purpose of the observation was to see how the fifth-grade pupils at MIN 2 Langsa's teachers using media, and the interview was given to the English teacher to learn more about the teacher's expertise with visual media. The results of this study demonstrated that the types of media that teachers used to teach English included whiteboards, English textbooks, dictionaries, posters, pictures, games, and other objects that were nearby. The interview with the teacher revealed that, depending on the subject, she employed visual aids to simplify the English lesson and help the pupils understand it better.

Abstract

This study elaborates the perspectives of teachers and primary school students learning English as a Foreign Language (EFL) regarding the application of the shadowing technique for pronunciation. Scholars have acknowledged shadowing as an effective technique for improving learners' pronunciation, where pupils replicate speech as carefully and promptly as possible following a speaker. This qualitative study was carried out at MIN 2 Langsa throughout the academic year of 2022-2023. Subject of the research was an English teacher and 30 fifth-grade pupils selected purposively. Data were gathered through semi-structured interviews with an EFL teacher and focus group discussions with the primary school students, followed by classroom observations to provide a deeper comprehension of the actual use of the method. Thematic analysis was employed to identify key patterns and themes in participants' responses. Results revealed that teacher considered the technique effective for enhancing students' speech accuracy, rhythm, and intonation. It offers students an immediate opportunity to correct their pronunciation and actively involves them in learning English, particularly in terms of pronunciation. The learners primarily viewed shadowing activity as an enjoyable exercise, where they practiced English chorally. This technique fosters students' confidence in the English classroom by alleviating individual pressure, while all participants learn and practice English together. Thus, it can be a powerful tool for enhancing young learners' pronunciation in an engaging way. English teachers might employ the technique as an alternative strategy to engage young learners in foreign language acquisition, particularly in pronunciation.

Abstract

: The armyworm (Spodoptera exigua) is one of the most common plant pests that lower plant yields.  The purpose of this study is to ascertain how armyworm (Spodoptera exigua) attacks on long bean plants are impacted by botanical pesticides and planting distance.  This study employs experiments and is quantitative in nature.  In this investigation, papaya leaf trials and garlic peels were used as plant insecticides in covert studies.  The one-way ANOVA test was the statistical method employed in this investigation. The findings demonstrated that while the botanical concentration had no effect on plant height or attack severity, it significantly affected the percentage of attacks and the leaf area index.  Plant height, attack intensity, attack %, and leaf area index were not significantly affected by planting distance in long bean plants.  The study's conclusions include the need for more research on plant insecticides that use plant leaves to fend against attacks, particularly in lowering the incidence of armyworm attacks.

Abstract

Gourami is a type of freshwater fish that is widely distributed in Indonesia, including in the waters of Kualuh Leidong District, North Sumatra. However, the kinship relationship between gourami species in the region has not been widely studied. Therefore, this study aims to analyze the kinship relationship of several gourami species found in the waters of Kualuh Leidong using the DNA fingerprint method. This study uses a molecular characterization method with several stages, namely collecting fish samples, isolating and extracting DNA, amplifying DNA using Polymerase Chain Reaction (PCR) with several primers, electrophoresis, and analyzing data using NTSYS software. Sampling was carried out by collecting specimens from the waters of Kualuh Leidong, including four types of gouramis, namely Trichopodus trichopterus, Trichopodus pectoralis, and Helostoma temmincki. The electrophoresis results showed that the DNA of the gourami fish could be amplified with primers OPA 02, OPC 02, and UBC 456, but not amplified with primer OPA 20. The DNA banding pattern obtained consisted of polymorphic and monomorphic bands with varying locus sizes. Jaccard coefficient analysis showed a high level of kinship (>70%), with the species Trichopodus pectoralis having a similarity of 0.78, Trichopodus trichopterus 0.76, and Helostoma temmincki 0.71. The results of this study can be used as a basis for the conservation and management of gourami fish resources in the waters of North Sumatera.

Abstract

This paper examines the phonological differences between Bahasa Indonesia and Singkil language in linguistic variation and identity among Singkils. Language variation includes one of human language attributes which demonstrated by differences in terms of pronunciation and structure. The objectives of this research are to find out phonological similarities between Bahasa Indonesia and Singkil language, to figure out the differences in the phonological systems of both languages, and to elaborate the attitudes of Singkils students toward their language and Bahasa Indonesia. This research was conducted on the third class of SDN 1 students speaking Singkil in KutaBahari district. Ten students engaged in reading an Indonesian passage. The evidence gathered via observation, analyses of secondary data, structured interviews, and questionnaires show that the sound ᵐb, ᵐp, ⁿc, ⁿd, ⁿg, and ⁿj are not dialects at all. All speakers of Singkil pronounce words containing these sounds with the same pronunciation. It is only possible because of the influence of the language outside (other than the language of Singkil). In this case, some students pronounce [ᵐb] became[b], [ᵐp] into [p], and this is only a very small speaker of Singkil. From this condition, we can conclude that language shift occurs in bilinguals. Bilingualism began when the people move into contact with other people and languages, and also from the other needs. It can be seen from The change [ᶬp] to [p],[ⁿb] to [b],[ⁿɟ] to [ɟ] is one of the language shifts. In the end, the identity of Singkil speakers display in their language attitude towards their language based on 12 questions is mostly positive. The average value obtained was 2, 7. Abstract

This article explores the political dynamics behind the legislative process of the Qanun on Ahwal al-Syakhshiyah in Aceh, a province with special autonomy to implement Islamic law in Indonesia. Rather than focusing solely on the substance of the qanun, this study investigates the complex interplay of power, ideology, and institutional interests during its formulation. The main research question addressed is: How do political, religious, and institutional actors shape the legislative process of Islamic family law in Aceh? Using a socio-legal approach and political legal theory, this article analyzes the roles of key actors such as the Aceh House of Representatives (DPRA), the Ulama Consultative Council (MPU), the provincial executive, civil society, and the central government. The study reveals that the legislative process is not merely normative or religious in character, but is deeply embedded in political contestation, ideological negotiation, and regional–national power dynamics. This article contributes to the discourse on Islamic legal politics by demonstrating how sharia-based legislation in autonomous regions is shaped by broader negotiations of authority and legitimacy.

Abstract

Work fatigue is one of the occupational health and safety problems. This study was to evaluate the potential of active compounds from pomegranate plants (Punica granatum L.) as heart disease inhibitor agents through a Molecular docking approach. The method used involved 3D structural analysis of four active compounds – Quercetin, Caffeic acid, Ellagic acid, and Phenol – in this plant which were downloaded from PubChem and processed using Swiss Target Prediction, Super Prediction, PyMol, Pyrx software. The interaction between compounds and biological targets in this case Clopidogrel, was studied to determine binding energy, binding affinity and stability. The results showed the Ellagic acid and Quercetin had significant interaction with the lowest binding energy of -7.8 kcal/mol and -8.7 kcal/mol and the lowest RMSD value of 0 (Å) so that this can be said to have good interaction stability with the protein target. The 3D molecular structure of these two compounds also shows the presence of hydrogen bonds and effective interactions with amino acids as well as low toxicity and favorable affinity energy. Based on these results, Ellagic acid and Quercetin were identified as potential candidates in the development of drugs to treat heart disease (cardiovascular). This study is expected to review the potential of active compounds in pomegranates through the insilico method, which can support the development of natural alternatives, in accordance with herbal-based prevention efforts that are increasingly in demand by the community.

Abstract

This study examined the emergence of the phenomenon of hijra among millennials in Aceh, Indonesia, and how this phenomenon has changed the Islamic landscape. This study applied qualitative research methods while the data were collected through observation, in-depth interviews, and documentation. The research subjects consisted of 4 hijra community founders and 10 teenage members of 3 hijra communities in Langsa. These communities include Komunitas Generasi Rabbani (KGR), the Akhwat Ilmu Agama (AKHIA) and the Muslimah Aceh Fillah (MAF). This research integrated Michael Foucault's theory of power and knowledge and Jurgen Habermas’s theory of public space. The findings of this study indicated that the phenomenon of hijra among them has implications for changes in the Islamic landscape. It was triggered by the rapid development of information technology, such as the use of the internet through social media and content production on various digital platforms such as Instagram, Facebook, Twitter, and YouTube. The impact of this phenomenon fosters the autonomy of the millennials to find their interpretations and choose new religious patrons that can be accessed easily through various existing digital platforms. The emergence of these new religious patrons does not come from traditional religious institutions like dayah. Such Islamic expression will expose young Muslims to pop culture and information technology in the future.
