Abstract

This study aims to analyze the effect of titanium dioxide (TiO₂) nanoparticle concentrations on the characteristics of Talinum paniculatum (Java ginseng) callus. The experiment was conducted using a completely randomized design with six TiO₂ concentration treatments (0, 1, 2, 3, 4, and 5 ppm) over a 14-day incubation period. The observed parameters included callus morphology (color and texture), fresh weight, and dry weight. Fresh and dry weight data were analyzed using one-way analysis of variance (ANOVA) followed by Duncan’s New Multiple Range Test (DNMRT), while callus color and texture were analyzed descriptively. The results showed that the addition of TiO₂ nanoparticles induced changes in callus color ranging from yellow to yellow-reddish, and the resulting callus exhibited a compact texture. TiO₂ nanoparticles up to a concentration of 5 ppm did not show negative effects on either fresh or dry callus weight. These findings indicate that TiO₂ nanoparticles at the tested concentrations play a role in modifying the visual characteristics of callus without affecting biomass accumulation. Therefore, TiO₂ nanoparticles have potential application in enhancing secondary metabolite production.

Abstract

In vitro culture provides an effective strategy for conserving and mass propagating the endangered orchid Dendrobium mussauense. This study aimed to evaluate the effects of different concentrations of 2.4-D on somatic embryogenesis (SE) induction using transverse thin cell layer (TCL) and non-TCL techniques. A factorial Completely Randomized Design (CRD) was employed, involving two factors: culture technique (TCL and non-TCL) and 2.4-D concentrations (1, 2, 3, and 4 mg L-¹), with four replications for each treatment. Explants were stem segments from 12-month-old D. mussauense plantlets. The highest survival and SE formation rates were observed in non-TCL explants treated with 4 mg L⁻¹ 2.4-D, reaching 75% and 50%, respectively. The earliest shoot and root emergence occurred at 13–14 and 23–25 days, respectively, under the same treatment. SE development in non-TCL explants treated with 2 mg L⁻¹ 2.4-D progressed through globular, scutellar, and coleoptilar stages within two weeks. Overall, 2.4-D concentrations of 2 and 4 mg L-¹ were most effective for SE induction in D. mussauense using both TCL and non-TCL techniques. Abstract

Maize (Zea mays L.) is one of the major cereal commodities with a strategic role in various sectors of societal needs. The application of plant-derived biostimulants has emerged as an alternative approach for enhancing plant physiological performance, including the use of leaf extract from Cnidoscolus aconitifolius (chaya). This study aimed to evaluate the effects of different solvent types and extract concentrations of C. aconitifolius leaves on the chlorophyll content of maize plants. A two-factor Completely Randomized Design (CRD) was used, consisting of: (a) solvent types (aqueous, ethanol, methanol, and butanol) and (b) extract concentrations (control, 25 mg/L, 50 mg/L, and 100 mg/L). The results indicated that neither solvent type nor extract concentration had a significant effect on any chlorophyll parameter. However, the aqueous-based extract at 25 mg/L tended to produce the highest chlorophyll a and total chlorophyll content, while the highest chlorophyll b content was observed in the 25 mg/L butanol extract treatment. In contrast, the 100 mg/L ethanol extract treatment showed a tendency toward the lowest chlorophyll content. These findings suggest that the effectiveness of C. aconitifolius extract as a photosynthetic biostimulant in Z. mays remains limited and is likely influenced by the solubility of its bioactive compounds in the solvents used.

Abstract

Lamiaceae family are rich in natural bioactive compounds, particularly essential oils, with antioxidant, antimicrobial, antifungal, and anti-inflammatory activities, making them a potential source of therapeutic agents for managing inflammation, which is a biological response to tissue damage. This review article aims to provide an overview and analyze the potential of terpenoid compounds from the Lamiaceae family as anti-inflammatory agents. A systematic literature search was conducted using ScienceDirect, PubMed, SciFinder Scholar, EBSCO, and Google Books, covering publications from 2014 to 2025. Using relevant keywords, a total of 100 publications were initially identified. After title and abstract screening, 72 articles were selected for eligibility assessment, of which 20 peer-reviewed research articles reporting in vitro and in vivo anti-inflammatory activities of Lamiaceae terpenoids were analyzed in detail. This study showed that terpene compounds in Lamiaceae essential oils have significant potential to inhibit the production of inflammatory mediators, including cytokines (IL-6, IL-8, IL-1β, IL-2, IL-4, IL-17A, and TNF-α), and nitric oxide (NO). These findings strongly support the Lamiaceae family as a promising source of natural anti-inflammatory agents, warranting further investigation through mechanistic studies, in vivo validation, toxicity assessment, and formulation development to advance their potential therapeutic application.

Abstract

Cytokinins are growth regulators that play a crucial role in cell division and shoot growth. Thidiazuron, a potent cytokinin, was the focus of our research. The research aimed to determine the effect of several concentrations of Thidiazuron on the protocorm growth of Grammatophyllum stapeliiflorum and to identify the best concentration of Thidiazuron (mg/L) for the protocorm growth of G. stapeliflorum orchids in vitro. The research was conducted from January until March 2022 at the Plant Physiology Laboratory, Universitas Andalas, Padang. The research used a Completely Randomized Design (CRD) with 5 treatments and 5 replications. Several concentrations of Thidiazuron were added to MS media with various concentrations: 0 mg L-1 (A), 0.50 mg L-1 (B), 1 mg L-1  (C), 1.50 mg L-1(D), and 2 mg L-1  (E). The parameters in this research were the percentage of life explants, day of shoot appearance, number of shoots, shoot length, number of leaves, number of roots, and root length. The results showed that Thidiazuron significantly affected the protocorm growth of the G. stapeliiflorum orchid compared to the control treatment. Notably, 2 mg L-1  Thidiazuron emerged as the best concentration for all growth parameters of protocorm G. stapeliiflorum, a finding that could significantly impact our understanding of orchid cultivation and plant physiology.

Abstract

This study aimed to evaluate the effects of nanopriming using Padina minor nanoparticles on the germination parameters and early seedling growth of Kopay chili pepper (Capsicum annuum L.) seeds, as well as to assess the physiological responses and enzyme activities associated with nanopriming treatment. The experiment was arranged in a completely randomized design (CRD) with five treatments: no priming, hydropriming, and nanopriming at concentrations of 100, 150, and 200 ppm, each with five replications. The observed parameters included time to germination emergence, germination potential, vigor index, root length, shoot length, and the activities of amylase and catalase enzymes. Data were analyzed using analysis of variance (ANOVA) at the 5% significance level, followed by Duncan’s Multiple Range Test (DMRT). The results showed that nanopriming significantly accelerated germination emergence and increased root length, with the best response observed at 100 ppm. However, the treatment did not have a significant effect on germination potential, vigor index, shoot length, or the activities of amylase and catalase during the early germination phase. Thus, Padina minor nanopriming contributes to the improvement of specific early growth parameters, particularly those related to germination speed and root development.

Abstract

This study aimed to evaluate the effects of the dosage and application frequency of chicken bone meal on the growth of water spinach (Ipomoea reptans L.). The experiment employed a factorial design within a completely randomized design (CRD) consisting of two factors, namely chicken bone meal dosage and application frequency. The observed parameters included plant height, number of leaves, and plant dry weight. The results showed that both the dosage and application frequency of chicken bone meal significantly affected the growth of water spinach. A dosage of 60 g produced the highest dry weight, reaching 20.76 g, compared with 6.10 g in the control treatment, while an application frequency of once per week resulted in better plant growth. The interaction effect indicated that the A3B1 combination produced the best growth performance, with dry weight reaching 24.97 g. Therefore, the application of chicken bone meal at an appropriate dosage and frequency can optimize the growth of water spinach.

Abstract

Dendrobium lasianthera (Orchidaceae) is one of the potential plants as ornamental plants because it has beautiful and long-lasting flowers. Propagation of orchid plants through in vitro culture technique has been widely done to meet market demand. One of the supporting factors in plant propagation through in vitro culture techniques is growth regulators. Cytokinin is one of the growth regulators commonly used in plant propagation, including orchids. The study aims to determine the effect of several cytokinins and find the best type for shoot induction of Dendrobium lasianthera. The method used a Completely Randomized Design (CRD) with 4 treatments and 6 replications with several types of cytokinis (control; BAP 1.2 mg/L, Kinetin 2 mg/L, and Thidiazuron 1 mg/L) in Murashige & Skoog (MS) media. The results showed that several cytokinins significantly affected the number of shoots, time of shoot appearance, and shoot length of D. lasianthera. Abstract

Drought stress severely reduces plant productivity by disrupting photosynthesis and cellular homeostasis. Plants respond through regulatory networks involving stress-responsive genes, including heat shock proteins (HSPs), regulated by heat shock transcription factors (HSFs). Among these, HsfA2 plays an important role in abiotic stress tolerance, including drought. Although CaHsfA2 expression has been reported in Capsicum annuum, validated primers for quantitative PCR (qPCR) analysis remain limited. This study aimed to design and validate specific primers for CaHsfA2 expression analysis. Primer candidates were designed from sequence NM_001324561.1 using Primer-BLAST and evaluated based on length, GC content, melting temperature (Tm), and self-complementarity. Total RNA was extracted from chili roots subjected to drought stress (50% field capacity) and nanopriming with Padina minor, followed by cDNA synthesis, gradient PCR and agarose gel electrophoresis.. Secondary structures were analyzed using OligoAnalyzer. Of ten primer pairs, four met the criteria, and primer set 1 (1_CaHsfA2) showed optimal characteristics (%GC 55%, Tm 60°C, product length 190 bp) and minimal secondary structure potential (ΔG > −9 kcal/mol). Gradient PCR (55.3–59.2°C) produced a single band (~193 bp), consistent with the predicted size (190 bp), with an optimal annealing temperature of 57.3°C, confirming its specificity and suitability for qPCR-based gene expression analysis under drought conditions.
