Abstract

Abstract

Chlorophyll is a green pigment in plants which is important for the process of photosynthesis. The photosynthesis process affects plant growth. Seaweed extract has been proven to be a source of biostimulants that can increase plant growth, one of which is Padina minor. Concentration and frequency are important factors that influence the work of biostimulants. One effort to increase the effectiveness of biostimulants is by changing the crude extract into the form of nanoparticles. This research aims to analyze the effect of concentration, frequency, and interaction between concentration and frequency of crude extract and Padina minor nano extract on chlorophyll levels in soybean plants. The method used was a completely randomized design (CRD) in factorial consisting of 2 factors and 4 replications. Factor A concentration of Padina minor extract, namely control, crude extract 0.4%, nano extract 0.2%, 0.3%, 0.4%, 0.5% and 0.6%. Factor B is the frequency of administration of Padina minor extract, namely 1, 2, and 3 times administration. Based on research that has been carried out, the results showed that administration of 0.4% crude extract increased the average chlorophyll a level by 6.6% and a total of 11%. Meanwhile, chlorophyll b gave the same results as 0.3% nano extract, namely an increase of 14% compared to the control. The total frequency of administration of Padina minor extract was 3 times, and the same as the other treatments, increasing chlorophyll a levels by 4.6%, b by 5.2%, and a total of 4.9% compared to the control. The interaction between concentration and frequency of administration of Padina minor nano extract gives the same results as the crude extract, with lower concentration and less frequency of application in increasing levels of chlorophyll a, chlorophyll b, and total chlorophyll in soybean plants.

Abstract

Dendrobium lasinthera is an orchid that has considerable potential as a raw material for anti-cancer drugs and for the production of cut flowers. Therefore, this orchid is a plant with high economic value. This orchid has beautiful and long-lasting flowers. The demand for this orchid is increasing along with its diverse uses, so the propagation of this orchid needs to be increased. In vitro propagation is the most appropriate step to reproduce this orchid. One of the stages that determines the success of orchid propagation through tissue culture is the root induction stage before acclimatization. Using the right type and concentration of Growth Regulatory Substances (ZPT) is very important. This research aims to determine the effect of several NAA Growth Regulatory Substance (ZPT) concentrations and determine the best NAA concentration for Dendrobium lasianthera root induction. This study used a Completely Randomized Design (CRD) with several NAA concentrations (Control (A); 0.5 mg/L (B), 1 mg/L (C), and 1.5 mg/L (D)), on Murashige & Skoog (MS) media. The results of this study show that NAA has a significant influence on the percentage of root formation, root emergence time, and root length in the Dendrobium lasianthera orchid. NAA 1.5 mg/L is the best concentration for induction of Dendrobium lasianthera orchid roots.

Abstract

The majority of Indonesian people consume rice as a staple food. According to data from the Central Statistics Agency (2022), rice production in West Sumatra reached 1,482,996.01 tons in 2019 and continued to decline until 2021, total rice production in West Sumatra was only 1,317,209.38 tons. This is inversely proportional to the population of West Sumatra which increases every year, so efforts need to be made to increase rice production. One of them is the biopriming technique to increase seed vigor. Priming is hydration carried out slowly which aims to ensure that the seed's water potential reaches balance and activates metabolism in the seed before the seed germinates. One of them is biopriming, which is a seed priming technique using biological agents in the form of beneficial microorganisms. Several factors influence the success of the priming technique, one of which is the soaking time. The right soaking time provides a deeper opportunity to absorb water to stimulate the seeds to germinate. This research aims to determine the effect of soaking time on biopriming of the local West Sumatran rice variety "Anak Daro" using Bacillus subtilis and Trichoderma harzianum on the germination of the Anak Daro rice variety. Completely Randomized Design (CRD) factorial pattern with 2 factors with 3 replications. Factor A is a biopriming agent, namely A1 (no microbes), A2 (Bacillus subtilis), and A3 (Trichoderma harzianum). Factor B is the length of soaking, namely B1 (24 hours), B2 (48 hours), and B3 (72 hours). The results showed that Bacillus subtilis and Trichoderma harzianum had a significant effect on maximum growth potential and germination, while soaking time had a significant effect on maximum growth potential and vigor index.

Abstract

This study aimed to evaluate the effect of BAP concentrations on explant response and callus formation in Massoia. The experiment was arranged in a Completely Randomized Design (CRD) with three BAP concentrations: 0.5 mg/L, 1.0 mg/L, and 1.5 mg/L, each with 24 replications. The culture medium used was Murashige and Skoog (MS), and observations were carried out for 7 days after planting. Parameters observed included the percentage of viable explants, browning, contamination, average callus formation, callus color, and texture. Data were analyzed using analysis of variance (ANOVA) at a 5% significance level. The results showed that 0.5 mg/L BAP yielded the highest explant survival rate (54%), the lowest browning rate (13%), and 35% contamination. Meanwhile, 1.5 mg/L BAP produced the highest average callus formation (0.21) with compact texture and brown coloration. Although not statistically significant, BAP concentrations affected explant viability and callus formation. A concentration of 0.5 mg/L tended to be optimal for maintaining explant viability, while 1.5 mg/L was more effective in inducing callus. These findings provide a preliminary basis for optimizing Massoia tissue culture techniques.

Abstract

Dendrobium mussauense has attractive flowers and potential as a parent of a hybrid orchid. The color of this species is cream with violaceous purple venations on the labellum. D. mussauense is an orchid species listed as vulnerable in IUCN. This species is not located in protected areas nor known to be conserved ex-situ. Clonal propagation is an important biotechnological tool for the conservation of orchids. Thin cell layer (TCL) is a micropropagation method using thin-sized explants that facilitate the diffusion of nutrients to tissues. This study examined induction shoot through two explant cutting techniques, namely TCL and non-TCL explant, at various concentrations of BAP. The non-TCL explant was more effective than the TCL explant in inducing the shoot of D. mussauense. The half-strength Murashige and Skoog (MS) medium supplemented with 0.9 mg.L-1 BAP was a suitable concentration to induce the shoot of D. mussauense through non-TCL explant. The average shoot length was 1,8 cm, and the average number of shoots was 5/explant.

Abstract

Cryptocarya massoy (Oken) Kosterm is a plant known for producing essential oils with many applications. However, overexploitation has threatened the sustainability of this plant. This study aims to identify the effect of adding methyl jasmonate to the culture medium on producing secondary metabolites through the callus culture of C. massoy. The methods in this study include callus induction, callus elicitation in liquid culture, callus extraction, analysis of secondary metabolite compounds in C. massoy callus culture, and data analysis. The results show a significant decrease in fresh weight of callus exposed to MeJA stress, depending on the concentration. Regarding secondary metabolites, massoia lactone, the primary compound in C. massoy, was not detected. It is possible that the duration of elicitation was insufficient to induce the biosynthesis and accumulation of detectable levels of massoia lactone. Secondary metabolite production often follows a specific temporal pattern, and certain compounds may require longer exposure times to reach detectable concentrations. The concentration of MeJA influenced the level of other target compound production, where at 0.5 mM MeJA increased the percentage of humulene by 24.64%. MeJA at 1.5 mM enhanced the percentage of ß-copaene (413.11%), ß-ylangene (100%). MeJA at 2 mM increased benzyl benzoate (9.61%). MeJA at 2.5 mM increased α-ylangene (31.25%) and caryophyllene (22.83%). Each MeJA concentration activated specific biosynthesis pathways, influencing the types of secondary metabolites produced. The metabolite screening revealed a predominance of terpenoids, especially sesquiterpenes, which play a role in plant defense.

Abstract

The community service activity aimed to enhance public understanding and skills in utilizing ginger (Zingiber officinale) as a natural ingredient for functional food through the production of jelly candy. This initiative was motivated by the low level of public knowledge, as indicated by pre-test results showing limited understanding of the health benefits and processing techniques of ginger. The activity consisted of educational sessions, demonstrations, and hands-on practice on jelly candy production using natural ingredients, including ginger extract, sugar, jelly powder, vanilla, salt, and cinnamon. Evaluation through post-tests, interactive quizzes, and observation of practical sessions demonstrated a significant improvement in participants’ knowledge and skills. Participants showed enthusiasm and were able to independently produce ginger-based jelly candy. This program not only increased public literacy on the use of herbal ingredients but also encouraged the development of small-scale functional snack enterprises. Future implementations are expected to expand to other communities with continuous mentoring to strengthen economic and health impacts.
