Abstract

Angiogenesis is a neovascularization process which its function is to supply nutrient and oxygen for cell survival. Pasak bumi is one of the Indonesian original plants having anti cancer activity. Extract of methanol, buthanol, kloroform, and water akar pasak bumi was proved to have cytotoxic effect on cancer cell culture. This research aim to know activity of antiangiogenic extract of metanol akar pasak bumi at chorioallantoic membrane of chicken embryo. Antiangiogenic activity test conducted by divide 8-9 days old egg embrio into 8 groups. Group I as paper disc control, group II as bFGF control, group III as bFGF control + DMSO 0.8% solvent, group IV, V, VI, VII, and VIII as  groups of treatment. Each groups of treatment were gave by bFGF 1ng/µL extract of metanol akar pasak bumi respectively with concentration 20, 40, 60, 80 and 100 µg/mL concentration. After 3 days incubation, then the chorioallantoic membrane macroscopic and microcospically analized. The antiangiogenic activity extract of metanol akar pasak bumi at chorioallantoic membrane bFGF induce at used concentration, that is start from 40 µg/mL concentration. The increases of the concentration also increase the prohibition activity to angiogenesis.

Abstract

Keloid is a skin lesion caused by an abnormal proliferation of fibroblasts and the deposition of collagen in the wound healing process. Therapy for keloids is relatively limited and mostly has side effects. Recent studies show that 5α-oleandrin from N. indicum Mill. (N. indicum) has anti-keloid effect by inhibiting (proliferation, migration activity and TGF-β1 expression) keloid fibroblast. Further studies with N. indicum extract standardized 5α-oleandrin should be conducted to develop it as an anti-keloid agent by performing at proliferative effects, collagen accumulation, and keloid fibroblast migration. The antifibrotic activity of N. indicum extract on the proliferation of keloid fibroblasts was measured by MTT assay, whereas collagen deposits were observed by Sirius Red method. Observation of keloid fibroblast migration by scratch assay is according to Liang et al. (2009). Ethanol extract of N. indicum standardized 5α-oleandrin can inhibit the proliferation of keloid fibroblas with IC50 0.458 μg/mL, also inhibited collagen deposits with IC50 0.055 μg/mL at incubation for 72 hours. In incubation 48 hours after treatment with ethanol extract of 1.0 μg/mL occurred inhibition of migration significantly compared with control.

Abstract

Previous research revealed that the extracts and fractions of Tithonia diversifolia (Hemsley) A.Gray leaves had antiplasmodial activity in vitro. For further development as an antiplasmodial agent, the mechanisms of action and safety of compounds are important to disclose. Heme polymerization inhibition is one of the main targets of antiplasmodial action. The aim of the study was to investigate the activity of T. diversifolia fractions in inhibiting heme polymerization and its cytotoxic effect on Vero cells. Heme polymerization inhibition assay from Bassilico and cytotoxic test on Vero cell using MTT method were conducted for three fractions (F5, F6, and F7) of T. diversifolia leaves. The inhibitory activity of heme polymerization expressed as IC50 and cytotoxicity effect expressed as CC50 were determined by probit analysis. The best heme polymerization inhibition activity was F5 with IC50 = 162.20 ± 57.81 μg/mL followed by F6 and F7 with IC50 216.30 ± 26.56 and 231.54 ± 44.26 μg/mL respectively. All the fractions had a low cytotoxic effect with CC50 for F5, F6, and F7 were over than 100, 34.81 ± 9.94 and 56.26 ± 6.73 μg/mL, respectively and the toxicity index fraction is below 10 or categorized as low selectivity. Conclusion: The fraction of T. diversifolia inhibited heme polymerization in vitro and had low cytotoxic effect on Vero cells but no selective toxicity. Further research using pure compounds may improve its selectivity.

Abstract

Pegagan (Centella asiatica L) has been traditionally used to treat wounds, disordered nerve system, blood circulation, and prevent muscle disorders. Extract of Pegagan was considered safe and non-toxic as the acuut and chronic toxicity tests on male SD rats were not significant. It meant that further study was potential to be proceeded. Study of C.asiatica on muscle function of the elderly has not been done, therefore this study is aimed to observe  effects of C.asiatica extract on muscle masses, holding hands, walking speed (4.57 meter), walking test (6 minutes), and Time Up and Go test of the elderly. This study is designed for clinical double blind study with 2 groups of people, these are C. asiatica (18 people) treated with capsul (550 mg C. asiatica extract/day for 90 days) and placebo (17 people). This study is performed in the RSUP Dr. Sardjito hospital at the geriatric division. Subjects for this study were healthy elderly people and fit with the criteria of subjects for experiment. Result of the study, muscle masses do not decrease and all the measurements are not different with that of placebo. Except that the walking speed (6 minutes) of the treated subjects is better than that of the placebo. The conclusion of the study, the capsul containing C. asiatica extract (550 mg, once a day for 90 days) increases the strengh of hands holding, and the walking test (6 minutes) almost twice better than that of the placebo although the muscle masses does not increase significantly.

Abstract

Lempuyang gajah rhizome (Zingiber zerumbet (L) Smith is considered potential as larvicidal. A previous study has shown that the purified extract of Z. zerumbet rhizome was toxic against Adese aegypti larvae.  The aim of the study it to formulate a purified extract of Z. zerumbet (L) Smith in granule preparations by combining Sodium starch glycolate, PVP K-30 and tween 80.  The granule formulation was optimized by the Simplex Lattice Design (SLD) method with using Design expert program 7.1.5. The results showed that the interaction of the three components can increase the flow rate index, the angle of repose, and reduce absorption. The optimum formula obtained was Sodium starch glycolate 2%, PVP K-30 2%, and Tween 80 5%. The analysis of one sample t-test shows that there is no significant difference between the predicted parameter values and the experimental results of the flow rate index and angle of repose, while the absorption response is significantly different. The optimum formula for granules has larvicidal activity with 100% larval death during 12-hour treatment.

Abstract

The objectives of the study were to evaluate the effect of ethanolic extract of P. indica Less leaf (EPI) on serum growth hormone (GH), milk yield, body weight gain of dams, and dam’s organ weight in lactating rats. A total of 25 lactating rats with six pups were randomized and distributed to one of five treatments (control (RO water), standard (domperidone 2,5 mg/kg BW/day), EPI 250 (250 mg/kg BW of EPI), EPI 500 (500 mg/kg BW of EPI), and EPI 750 (750 mg/kg BW of EPI)). The treatment was administered daily starting from the 2nd until the 15th day of lactation. On the 16th day serum growth hormone level, body and organ weight of rats were measured. Serum GH levels in the EPI 750 group (1963.25 ± 360.91 pg/µL) increased significantly compared to the domperidone (409.46 ± 28.80 pg/µL) and the control group (723.40 ± 95.78 pg/µL, p0.05). There was significant body weight gain in the EPI 750 group compared with the domperidone group. There was no significant difference in organ weight gain in each group. The study revealed that ethanolic extract of P. indica Less leaf can increase serum growth hormone in lactating rats.

Abstract

Cases of dengue infection are still high in various parts of the world and no antiviral has been found to treat dengue infection. Jatropha multifida is one of the herbs used by the community to treat dengue infection, but pre-clinical and clinical scientific evidence has not been carried out. Phytochemical screening of ethanol extract of Jatropha multifida leaves was carried out using thin layer chromatography. Cytotoxic assay was performed on Vero cells using the MTT (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide). The cytotoxic concentration 50 (CC50) was determined using probit analysis. Anti-dengue test on dengue  virus serotype 2 (DENV-2) was performed on Vero cells and the RNA (ribonucleic acid) copy number was quantified using quantitative polymerase chain reactions ((qPCR). Phytochemical screening results showed that the ethanol extract of Jatropha multifida leaves contains flavonoid and terpenoid compounds. CC50 of the extract was found to be 651.8 µg/mL. The RNA copy number of the treated group was lower than the control group and the difference was significant. The ethanol extract of Jatropha multifida leaves has anti-dengue activity against DENV-2.

Abstract

Oleandrin is one of cardenolida compound isolated from an active fraction of Nerium indicum Mill leaves. (fam. Apocynaceae), which have cytotoxic effect on several human cancer cells, and also to normal cells in vitro. Another compound which was potential as renal anticancer has also been isolated from N. indicum. However, its chemical structure has not been discovered. The aim of this study was to identify the potential compound as selective to renal cancer present in the leaves of N. indicum.The potential compound was isolated from the active fraction using Preparative TLC and structure elucidation was done by using spectroscopic methods (UV, IR, MS and NMR).Base on this spectra and by comparison with oleandrin data, it was indicated that the potential compound as Renal anticancer was as 16,17-dehydrodeacetyl-5a-oleandrin.Key word: N.indicum, oleandrin, cytotoxic, renal anticancer, spectroscopic

Abstract

Pasak bumi is one of the Indonesian original plants having anti cancer activity. One compound of akar pasak bumi, i.e eurycomanone was proven to have cytotoxic effect on cancer cell culture by apoptosis induction on HeLa cells. Therefore, this study was aimed to isolate and identify euycomanone from akar pasak bumi and to look at the anti angiogenic activity of the compounds on corio alantois membrane (CAM) of chicken embryo induced by bFGF.The akar pasak bumi was ground and was macerated with methanol. The macerate was fractioned by mean of VLC. Six fractions was obtained. The second fraction exhibited two spots on a TLC system, one spot was cochromatographed with eurycomanone as the reference standart and another spot exhibited blue fluorecence at 254 nm. This second fraction, then, was chromatographed on a preparative scale TLC to separate isolate A. Anti angiogenic test was performed for Isolate A at concentration series of 5, 10, 15, 20, 40 μg/mL respectively. Egg embrio (8-9 days) were divided into 8 groups. The first group was implanted paper disc only, the second group were given bFGF on Tris HCl buffer and the third group were given bFGF (on Tris HCl buffer) + 0,8 % DMSO. The fourth to eighth group were induced by 10 ng bFGF and the test preparation at the respective concentration. After incubation at 39 °C for 3 days, each egg was opened, the content of each was aspired and discarded, the CAM sticked to its shell was examined macroscopically and microscopically for the formation of new blood vessels.Based on spectral data of UV, IR, 1H NMR, 13C NMR and MS of isolate A and comparison with the spectral data of the standart, the isolate A was eurycomanone. Eurycomanone inhibited angiogenesis at concentration of 10 μg/mL.Key words: antiangiogenic, angiogenesis, E.longifolia, eurycomanone,CAM.

Abstract

Finding anticancer drugs from natural resources still proceeds. Oleandrin isolated from Nerium indicum Mill. inhibited the growth of mieloma cell line in vitro better than that of vincristine sulphate. This study was aimed to determine the cytotoxic effect of oleandrin on various human cancer cell lines. Cytotoxic test of oleandrin on seven human cancer cell lines was done by SRB-method. The analysis was conducted by comparing the ID50 of oleandrin with that of doxorubicin and cisplatin as positive controls. This result indicated that oleandrin possessed the best cytotoxic effect on breast cancer (MCF7) with ID50 at 8.85 nM. Keywords : Oleandrin, cytotoxicity, human cancer cells, ID50
