Abstract

This study was designed to examine motor and congnitive changes, infarct lesion and neurohistological changes, involving histologic staining and immunohistochemical expression of caspase-3 after induction by right unilateral common carotid artery occlusion (RUCCAO) for 90 minutes. The animals were divided into two groups: sham group and stroke model group. Cognitive impairment was evaluated by Y maze. Motor function was measured on days 0, 1, 3 and 7 using FUAT paradigm. Infarct area, histological and caspase-3 expressions were evaluated on day 14 after RUCCAO. The results showed that RUCCAO induced cognitive and motor impairment on day 3 and 7. Furthermore, stroke model group induced infarct lesion. Hispatology examination showed body damage of neuron cell in the ipsilateral hemisphere. Moreover, expression of caspase-3 on RUCCAO group was significantly higher than that in sham group. In conclusion, RUCCAO method caused significant cognitive and motor function impairment. Furthermore, RUCCAO also induced infarct lesions and cell death in the thalamus brain area. Thus, RUCCAO can be employed as a method for ischemic stroke model, especially in focal ischemia.

Abstract

Bone is an important organ for supports the body that stores reserve of calcium, phosphorus, and other minerals. In fracture conditions where bleeding, soft tissue edema, nerve damage, and blood vessels around the bone damage happen, they can cause the mobilization of these minerals in the surrounding tissue. One of the efforts made in the treatment of these fractures is reconnection, in which it works by filling of bone defect with a matrix and administration of anti-infection. Biomaterial filling in defective bone is thought to accelerate the healing process of bone fracture and prevent osteomyelitis. For this reason, this study evaluates the acceleration of bone fracture healing using natural hydroxyapatite (NHA) bone filler in rabbits with bone defect model. Fracture modeling was performed by surgical technique and drilling of bones with a 4.2 mm diameter to form a defect in the rabbit femur. Bone implant contained bovine hydroxyapatite-gelatin-glutaraldehyde (BHA implant) or bovine hydroxyapatite-gelatin-glutaraldehyde-gentamicin (BHA-GEN implant) that was inserted in bone defects. 27 rabbits were divided into 3 groups: the control group who had bone defect, the bone defect group was given BHA implant and the bone defect group was given BHA-GEN implant. Observation of osteoclast, osteoblast, osteocyte, BALP level, and bone morphological integrity was carried out on the 14th, 28th, and 42nd days after surgery. Histological observation of rabbit femur showed a significant difference on the number of osteoclast, osteoblast and osteocyte in all three groups. The BALP level also showed a significant difference in the group given the natural BHA bone implant compared to the control group on day 14 (p = 0.0361). Based on the result of the X-ray, there was also a better integration of rabbit femur bone in groups with the use of BHA or BHA-GEN bone implant. Thus, it can be concluded that the use of a natural BHA implant can accelerate the process of bone repair in the fracture of rabbit femur. In addition, BHA implants were compatible as a matrix for supporting the bone cell growth.

Abstract

Neuropathy pain is a pain that caused by nerves injury. Nowadays, treatment for neuropathic pain change to drugs that works as GABA agonist and cause reimbalance between excitatory and inhibitory neurotransmitter in central nervous system (CNS). The present study was designed to investigate the effectiveness of gabapentin and baclofen in decreasing N-Methyl-D-Aspartate (NMDA) receptor NR2B subunit activity in neuropathic pain. Fourty mice were divided into 8 groups i.e sham, negative control, gabapentin (10, 30, 100 nmol) and baclofen (1, 10, 30 nmol). Neuropathic pain was induced by ligation of sciatic nerve with Partial Sciatic Nerve Ligation (PSNL) method). Treatments were administrated intrathecally once a day for seven consecutive days, at a week after induction. On day 15th, mice were sacrified and the spinal cord were removed quickly. The expression of NMDA receptor NR2B subunit were examined with imunohistochemistry and data were analyzed by one way anova. The result from this research was gabapentin and baclofen administration significantly decrease expression of NMDA receptor NR2B subunit in mice compared to sham group. The higher the dose, the more effective to decrease the number of neuron that express NR2B. The conclusion of this research was gabapentin and baclofen treat neuropatic pain by decreased the number of NMDA receptor NR2B subunit.

Abstract

Kalanchoe pinnata (Lmk) Pers is a medicinal plant that has many activities, such as antioxidant, anti-infl ammation and immunosuppressant activities. Based on our previous study, aqueous extract of this plant leaves has a repairing eff ect on lupus nephritis model. The expected active compounds are flavonoids. Those compounds are separated in the ethyl acetate fraction. Therefore, this study observed the activity of the ethyl acetate fraction of Kalanchoe pinnata (Lmk) Pers on reducing the level of Anti- Smith autoantibody and the regulation function of CD4+CD25+ T reg in lupus mice. The experimental groups were a negative control group that received placebo, ethyl acetate fraction of Kalanchoe pinnata (Lmk) Pers (EF-KP) group and a positive control group that received cyclophosphamide. The anti-Sm level was measured by using indirect ELISA. The spleen cells were prepared for CD4+CD25+ T reg assay using fl ow cytometry. The result of this experiment was a reduction of anti-Sm antibody level. The decrease was suffi cient to stabilize lupus condition. The fl ow cytometry assay results showed the increase in the relative percentage of CD4+CD25+ T reg. This function can inhibit the reactivity of autoantigens, so it prevents tissue and organ damage from lupus. However, further research is needed to observe the complete mechanism.

Abstract

Endothelial dysfunction has been considered as one of the key initial events in the pathogenesis of atherosclerosis and other cardiovascular diseases (CVDs). Several studies imply that chronic inflammation and oxidative stress play a critical role in endothelial dysfunction. The present study was designed to evaluate the preventive effect of alpha-lipoic acid (ALA) on lipopolysaccharide (LPS) induced endothelial dysfunction in rat based on malondialdehyde (MDA) level and vascular cell adhesion molecules-1 (VCAM-1) expression. Thirty Wistar rats were administered ALA for 2 weeks in different doses (30, 60, 120 mg.kg-1 BW) 1 hour before LPS 0.5 mg.kg-1 BW i.p challenge. The LPS was injected on 1st, 4th, and 9th day. MDA plasma level was analyzed with spectrophotometer λ 529 nm, and VCAM-1 expression was determined by immunohistochemistry. Pretreatment with ALA for 14 days could decrease plasma MDA level on LPS-induced endothelial dysfunction in rats. However, only one group of ALA doses, 120 mg.kg-1 BW, showed significant difference with LPS untreated group statistically. It was also found that ALA, in all treatment groups, could attenuate VCAM-1 expression. These findings suggest that ALA had a protective effect against endothelial dysfunction and may potentially prevent vascular inflammatory disease.

Abstract

Background: In developing a pharmaceutical product, it is necessary to conduct pre-clinical and clinical trials to ensure its safety and effectiveness. The toxicity test is conducted to assess the safety of a substance to determine its toxic effect of the substance. Objective: This study aims to determine the acute and subchronic toxicity of administering IHDM allergenic extract using experimental animal models. Methods: Female BALB/c mice and female and male Wistar rats were used as experimental animal models. While the IHDM allergenic extract was used with the level of Der p1 is 11.3-26.6 ng/mL and was administered by intravenous route. The acute toxicity test was carried out for 14 days on four different dose groups of experimental animals. The subchronic toxicity test was carried out for 28 days using three other dose groups of experimental animals. Results: The administration of a single dose of IHDM allergenic extract at various doses did not cause mice behaviour changes, and no death was shown in each group. Likewise, there was no change in the principal organs by macroscopic observations. Meanwhile, administering IHDM allergenic extract at repeated doses for 28 days could show signs of toxicity. The symptoms were shown in the histopathological structure of the liver, kidney, and heart organs. Conclusion: It can be concluded that the IHDM allergenic extract is safe for single-dose administration but shows toxic signs when given in repeated doses. Further tests are needed for 90 days of subchronic toxicity and satellite testing.

Abstract

This study aims to study the process of fabricating and characterization nanofibers based on cellulose acetate polymer filled with ceftriaxone and ibuprofen using the electrospinning method as a model for a targeted drug delivery system. Cellulose acetate has been successfully fabricated into nanofibers using the electrospinning method and filled with ceftriaxone and ibuprofen. The resulting nanofibers have an average diameter in the range of 412.5 – 558.5 nm under the conditions of the electrospinning process, namely 22 kV voltage, 15 cm distance, and flow rate. 0.005 mL/min using acetone/DMAc (2:1). Based on the data generated from the FTIR test, homogeneity, and drug release test, it is known that the resulting nanofibers, namely cellulose acetate, cellulose acetate-ceftriaxone, and cellulose acetate-ibuprofen, have good morphological characteristics, homogeneity, and release rate so that they have the potential to be used as a targeted drug delivery system

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian fraksi air lingzi (G. lucidum) terhadap penurunan kadar glukosa darah dan gambaran histopatologi di jaringan hepar pada tikus yang mengalami diabetes akibat induksi aloksan monohidrat. Sejumlah 36 ekor tikus dibagi dalam 6 kelompok, yaitu kelompok normal, kelompok diabetes, kelompok pembanding glibenklamid, dan kelompok fraksi air G. lucidum dengan dosis 225 mg/KgBB. Pada hari ke-0 diberikan injeksi aloksan monohidrat 150 mg/KgBB secara i.p. Pemberian glibenklamid dan fraksi air G. lucidum selama 9 hari diberikan setelah timbul hiperglikemia yang persisten. Hari ke-10 setelah treatment tikus dikorbankan dan diambil jaringan heparnya untuk dilakukan pengujian histokimia menggunakan hematoxylin eosin. Hasil penelitian menunjukkan bahwa pemberian fraksi air G. lucidum dosis 225 mg/kgBB Lingzhi segar mampu menurunkan kadar glukosa darah yang hampir sama dengan kelompok tikus DM yang diberikan glibenklamid 0,45 mg/KgBB serta mampu melindungi jaringan hepar dari kerusakan karena kondisi DM. Ditunjukkan oleh perubahan morfologi sel hepatosit mengalami perbaikan. Kata kunci  : Fraksi air Lingzi, G. lucidum, Diabetes Melitus, Resistensi insulin dan Hepatoprotektif

Abstract

The antioxidant properties from medicinal plants have protective effects against oxidative stress-linked diseases. Their antioxidant capacities are related to phenolic compounds, including phenols and flavonoids. Clinacanthus nutans, a native to Asia, including Malaysia, Thailand, and Indonesia, are used for treating diabetes and other diseases, but the exploration of the plant in Indonesia is still limited. Therefore, this study aimed to reveal the phytochemical constituents, metabolic compounds, and antioxidant activity of C. nutans extracts. Dried leaf powder was extracted with 70% ethanol and aqueous, each extract was subjected to qualitative phytochemical screening. Subsequently, total phenolic and flavonoid content was calculated using Folin-Ciocalteu and AlCl3 colorimetric assay. The antioxidant activity was determined using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging method. The profiles of phenolic and flavonoid compounds of 70% ethanolic extract were investigated by LC MS/MS. Phytochemical screening showed that the extracts contain phenolic, flavonoid, tannin, and saponin. The total phenolic content of the 70% ethanol and aqueous extract was 4.14±1.9% mgGAE/g and 3.89±3.2% mgGAE/g, respectively. Meanwhile, the total flavonoid content obtained was 0.19±1.0% mgQE/g and 0.03±0.5% mgQE/g for 70% ethanol and aqueous extracts, respectively. Both extracts showed high antioxidant activity, with DPPH IC50 30.57 ????g/mL for 70% ethanol and 37.24 ????g/mL for aqueous extract. LC-MS/MS analysis of 70% ethanol extract revealed 5 phenolic and 3 flavonoid compounds, where DL-glutamic monohydrate and Boesenbergin B were the most dominant based on the percentage of area under the curve. C. nutans originating from Indonesia can be a good source of natural antioxidants because 70% ethanol and aqueous extract show high phytochemical content and antioxidant activity.

Abstract

The Covid-19 Pandemic outbreak has changed various forms of life in society, including restrictions on activities in an open-source environment. The pandemic has also created various kinds of unresolved mental health problems, both in the national dan global area, both those who are directly exposed to the virus and those who are not. Today, the community is still struggling to control the spread of Covid-19, but on the other hand, it has caused the spread of feelings of anxiety, fear and mental pressure due to isolation, physical distancing restrictions, social relations, and other uncertainties. Therefore, these webinar activities were held as one of the online learning communication media, in terms of providing mattery regarding the molecular mechanism of antidepressant therapy, which during the post-Covid-19 pandemic its use increased due to social reasons. This webinar media was very effective without having to gather a crowd and aims to increase knowledge, especially among participants in terms of handling mental health such as depression in terms of its pharmacology therapy. Then, knowing the mechanism of molecular action can improve the optimization of antidepressant therapy according to the patient’s condition. The target participants in this webinar are health workers, students of health study programs, and common people. The webinar activity was conducted by the online system, using the Zoom meeting platform, and was held on April 15th, 2023, then was attended by 241 people, who came from all over Indonesia. From the implementation of the webinar activities, it is known that there has been an increase in understanding of the molecular matter that has been given, this can be seen from the percentage of the correct answers in the post-test evaluation compared to the answers in the pre-test.
