Abstract

Penelitian ini bertujuan untuk mengembangkan dan menghasilkan media pembelajaran e-book dan praktikum berbasis analisis DNA lalat buah isolat lokal pada materi genetika di SMA yang layak digunakan. Penelitian ini merupakan penelitian pengembangan (Research and Development) dengan model ADDIE yang terdiri 5 tahapan, yaitu Analysis, Design, Development, Implementation, dan Evaluation. Instrumen tes pada penelitian ini adalah angket, wawancara dan dokumentasi. Lembar angket penilaian diberikan kepada subjek penelitian yaitu ahli materi, ahli media, guru mata pelajaran biologi dan siswa kelas XII di SMA Negeri 1 Pagimana. Hasil penelitian ahli materi dengan persentase 93% kriteria sangat valid, ahli media dengan persentase 96% kriteria sangat valid, guru biologi dengan persentase 97% kriteria sangat valid dan respons siswa dengan persentase 88% kriteria sangat baik. Dapat disimpulkan bahwa media e-book dan praktikum berbasis analisis DNA lalat buah isolat lokal pada materi genetika di SMA produk yang dihasilkan sangat layak untuk digunakan sebagai media pembelajaran.

Abstract

Cancer is the leading cause of death in the world, with a high prevalence in Indonesia, especially breast cancer. Conventional therapies such as chemotherapy and radiotherapy have various limitations, including severe side effects and cancer cell resistance. This study aims to explore the anticancer potential of the ethanol extract of Apis dorsata Binghami nests originating from Uluway Barat Village, Tana Toraja Regency. Extraction was carried out using the maceration method with 96% ethanol solvent, with a yield of 51.42%, and bioactive compounds were identified through GC-MS analysis. Anticancer activity tests were carried out in vitro on MCF-7 breast cancer cells using the resazurin assay method . The results showed that the extract contained various bioactive compounds such as flavonoids, furanones, and aromatic aldehydes which are known to have antioxidant and anticancer activities. The IC50 value obtained was 434.30 µg/mL, indicating weak cytotoxic activity. Nevertheless, this extract showed significant ability to kill cancer cells at certain concentrations, especially 15.63 µg/mL which reduced cell viability by 44.39%. These results indicate that A. dorsata Binghami nests contain compounds that have the potential as natural anticancer agents, but require further research for fractionation and identification of more specific active compounds and further testing in vivo

Abstract

The existence of insects cannot be separated from the condition of their habitat. The mangrove area of ​​Tatengesan Village is one of the ecotourism site in Minahasa, North Sulawesi, that can support the survival of insects. This study aimed to determine the distribution and diversity of insects and the factors that influence the insect diversity in the mangrove area. The type of research used is descriptive quantitative research by conducting data collection activities, analyzing data and interpreting data with the aim of making a description of the events that occurred. The animals were exploratory collected by using insect nets (sweep net), beating cloth (beating), and bare hands (hand collecting). The results showed that there were 1225 individual insects consisting of 14 insect species, which were divided into 14 genera, 11 families and 6 orders of insects. The insect diversity index value was 0.27 which was classified as low. The dominance index value was 0.90 which was classified as high. The evenness index value was 0.10 which was classified as low.

Abstract

Sulawesi Island in the Wallaceae line area has diverse endemic animals, including the white-tailed forest rats. White-tailed forest rat (Maxomys hellwandii) is one of the endemic animals in North Sulawesi, so this study aimed to determine the type of bacterial genus in the small intestine of Maxomys hellwandii. This research used a laboratory-based qualitative descriptive method using a pour plate isolation method and bacterial identification based on morphological, and physiological characters with biochemical tests. The small intestines of Maxomys hellwandii taken from the South Minahasa forest area, North Sulawesi were used as samples of this research. The results of the study obtained 22 isolates of bacteria that were successfully isolated and have been identified based on morphological characteristics, and physiology with biochemical tests based on the book Bergey's Manual of Systematic Bacteriology suspected 14 isolates as genus Bacillus and 8 isolates as genus Lactobacillus.

Abstract

Various types of eggs are one of the foodstuffs that are in great demand by the community, especially the people of North Sulawesi, because they are cheap and easy to obtain. Furthermore, eggs have the nutritional content of animal protein, which is good for humans. Numerous studies on eggs have revealed that microorganisms, including Salmonella sp., can contaminate them. This study aims to detect Salmonella sp. bacteria in various types of eggs in the Tondano Market, Minahasa, North Sulawesi. This research uses a descriptive method (qualitative method) to describe an event objectively. We collected up to three samples, the first of which is located outside the market. Three samples of purebred chicken eggs, duck eggs, and quail eggs were taken. Then, the second point inside the market was taken: 3 samples of purebred chicken eggs, duck eggs, and quail eggs from the same trader. To prepare the samples, the eggshells were separated from the insides, then 2 grams of the eggshells were crushed and mixed with 5 ml of Buffered Peptone Water (BPW) 0.1%. This mixture was then added to 10 ml of Rappaport Vassiliadis (RV) media, followed by scraping onto Salmonella Shigella Agar (SSA) media, and then inoculating onto Xylose Lysine Deoxycholate agar (XLD) media. Four eggshell samples tested positive for Salmonella sp. bacteria: duck eggs (TB 1), quail eggs (TP 1), broiler eggs (TA 2), and quail eggs (TP 2), while two samples were negative: broiler eggs (TA 1) and duck eggs (TB 2). In contrast, two samples were negative for Salmonella sp., namely broiler eggs (TA 1) and duck eggs (TB 2).

Abstract

This research is motivated by chilies, which are one of the plant products that become raw materials for daily needs and are therefore classified as a secondary food source. Insect pests are a factor affecting chilli productivity, use of fungi as biological controllers, to determine the use of the fungus Beauveria bassiana as a biological control against the insect pest of chilli aphids Myzus persicae Sulz, the research method used in this study is an experimental method using the complete randomized design (CRD) method with five treatments and three replications, where each plot has five samples, the treatment doses were Control, P1 (6 g/500 ml), P2 (7 g/500 ml), P3 (8 g/500 ml), P4 (9 g/500 ml) and Lethal Dose 50, the results of this study showed that the application of B. bassiana had a significant effect on the mortality of M. persicae Sulz with the mortality obtained was 0.75% of the total number of samples 120 outside the control treatment, so the higher the dose, the higher the mortality, the LD50 result was 6.025 g/ml, showing that the toxicity of B. bassiana falls into the category of mild toxins. Therefore, we can conclude that the fungus B. bassiana serves as an effective biological control agent against the insect pest M. persicae Sulz.

Abstract

Dua puluh tujuh ekor tikus putih (Rattus norvegicus L.) jantan dibagi menjadi 9 kelompok perlakuan yaitu kontrol normal (K1), kontrol negatif (K2), kontrol positif (K3), serbuk daun kersen 800 mg/kgB (K4), serbuk daun kelor 800 mg/kgBB (K5), serbuk daun kersen 1600 mg/kg BB (K6), serbuk daun kelor 1600 mg/kgBB (K7), kombinasi serbuk daun kersen 400 mg/kgBB dan daun kelor 400 mg/kg BB (K8), kombinasi serbuk daun kersen 800 mg/kg BB dan daun kelor 800 mg/kgBB (K9). Pengukuran kadar glukosa darah tikus dilakukan pada hari ke-3, 7, dan 10. Hasil analisis menunjukan bahwa pemberian dosis kombinasi serbuk daun kersen 800 mg/kgBB dan daun kelor 800 mg/kgBB merupakan dosis yang paling efektif dalam menurunkan kadar glukosa darah pada tikus putih jantan
