Abstract

Transformation two cry genes (cryIB-cryIAa) and transformation with a single the cry1B gene under the control of a wound-inducible maize proteinase inhibitor gene (mpi) promoter were two approaches that were used to get resistant rice to the rice stemborer which may be had a durable resistance. To obtain information on the effectiveness of seven transgenic rice lines to the rice yellow stemborer S. incertulas, a test was conducted in greenhouse. The seven lines were 1). line 4.2.3 and 2) line 4.2.4 both contain fusion of two cry genes (cryIB-cryIAa); 3) line 3R9 and 4) line 3R7 lines both contain of mpi-cryIB gene; and 5) line 6.11 contains of cryIAb gene by particle bombardment, 6) line DT-cry (Azygous) that do not contain cry gene (null), and 7) DT-cry line contains cryIAb gene by Agrobacterium, and as a negative control, we used three non transgenic rice varieties i.e., Rojolele, Cilosari, and Ciherang. The result showed that transgenic rice lines, except DT-cry and DT-cry (Azygous) lines were effective to suppress damage by the insect, and showed an inhibition effect on the growth of S. incertulas, and had a high level ofresistance than non transgenic rice varieties had. There were differences on resistance value/level among transgenic rice lines. Based on the resistance value, 6.11 line was the highest followed by 4.2.4 line and 3R7 line, these lines were categorized as high resistance. Transgenic rice-4.2.3 line and 3R9 line were categorized as moderat resistance. DT-cry and DT-cry (Azygous) lines were susceptible.

Abstract

Stalk eye flies (Diptera: Diopsidae) is an insect family which has unique eye morphology. Their eyes are in the distal end of a long stalk. The information of species and their habitats is still very limited in Indonesia. Therefore, the objectives of this research were studying species diversity, habitat, and distribution areas of stalk eye flies in soroundings in Bogor. The sample sites are classified into three areas; lowland, middle, and upland. The flies ware taken with two methods: net sweeping and direct capture, then parts of the body were measured. The observation of stalk eye flies activity and behavior was done in the morning, midday, and afternoon. The result of observation indicated that stalk eye flies were found almost in all areas, from lowland to upland. On 15 survey locations, stalk eye flies were found in 10 locations with the same habitat. The species are Cyrtodiopsis dalmanni and Teleopsis sp.. C. dalmanni was found in 5 locations, and Teleopsis sp. in 9 locations. The observation result of stalk eye flies activity and behavior indicated that the number of stalk eye flies which captured in three period of time were not obviously different. Result from measuring stalk eye flies bodyparts indicated that the average size of male and female flies was not obviously different, except the male stalk is longer than female. In addition, C. dalmanni stalks are longer than Teleopsis sp. stalks.

Abstract

Predatory arthropods inhabiting paddy rice ecosystems in Cianjur District, West Java. Predatory arthropods were surveyed in Cianjur District, West Java from July to November 1997, with objectives to observe the species of predatory arthropods inhabiting the ecosystems. Canopy-inhabiting and soil-dwelling arthropods were sampled using D-vac and pitfall traps, respectively. Results indicated that insecta and Arachnida were the most abundant predators in the ecosystem, with the total of 142 species. Predatory insects were dominated by Carabidae (27 species), and Staphylinidae (13 species) and spiders were dominated by Therididae (9 species), and Lycosidae (8 species). Thus, predatory-arthropods inhabiting the sawah ecosystems were very diverse.

Abstract

Whiteflies (Hemiptera: Aleyrodidae) can cause direct and indirect damages on plants, especially vegetables. There is only limited information regarding taxonomy and population dynamic of whiteflies attacking vegetables in Indonesia. This research is conducted to identify species of whitefly collected from chili pepper, tomato, and soybean, and to study their population dynamic. The information gathered from these studies will be useful to support whitefly management in the field. Based on morphology identification of the puparium collected directly from the host plants, there were four species of whitefly identified from chili pepper, tomato, and soybean in Bogor, Cianjur, and Sukabumi, i.e. Bemisia tabaci, Aleurodicus dispersus, Trialeurodes vaporariorum, and Dialeurodes sp. The presence of B. tabaci on chili pepper and tomato was associated with virus infection that causes yellowing and leaf curl disease. This population of B. tabaci tended to increase along with plant growth and generally reached the highest population when the plant was 60-70 days after planting.

Abstract

Soil samples were collected from each plot at the two field sites at 0, 1, 3, and 5 months after planting, and at one-month intervals after harvest for three months. Soil suspension from each sample was mixed with agar based diet (1 g soil/20 ml soil-diet mixture) then was presented to neonate larvae of H. armigera. Two reference standards were used, i.e. pure Cry1Ac protein (0.165 x 104 to 1.436 x 105 ng per ml of the proteindiet mixture) and non-Bt-soil spiked with Cry1Ac protein (0.11 x 104 to 9.573 x 104 ng per ml of the soil-diet-mixture). Larval mortality was recorded at 7 days of treatment. In both reference standards, mortality of H. armigera larvae occurred in a concentrationdependent fashion, i.e. larval mortality increased with the increase in Cry1Ac protein concentration, with LC50 of 0.95 x 104 and 1.65 x 104 ng/ml, respectively, for the pure protein and spiked non-Bt-soil standards. Result of the bioassay of soil samples from the field sites showed the residues from both transgenic cotton soils did not cause any mortality (0%) in the test insects. This indicates that the concentrations of Cry1Ac residue in soil at the study sites were very low (in the second standard, Cry1Ac at a concentration of 0.33 x 104 ng/ml caused about 10% larval mortality).

Abstract

Eucalyptus spp. are commonly cultivated in the forest industry sector as the raw material for the pulp and paper industry. One of the problems in Eucalyptus plantations is leaf galls caused by insects. Leaf galls can cause leaf damage and inhibit plant growth. The objective of this study was to identify insects associated with leaf galls on eucalyptus plants. Leaves with gall symptoms were collected in the Tele sector located between Humbang Hasundutan and Samosir Regencies, North Sumatra Province. Identification was based on morphological characters of the antenna, dorsal mesosoma, fore wings, and hind legs. Results showed that there were seven insect species associated with leaf galls on Eucalyptus, i.e. Ophelimus eucalypti (Gahan) (Eulophidae) pest gall on eucalyptus leaf, Eurytoma sp. Illiger (Eurytomidae), Aprostocetus sp. Westwood, Quadrastichus mendeli Kim & La Salle, Chrysonotomyia germanica (Erdös), and Closterocerus chamaeleon (Girault) (Eulophidae) acts as a parasitoid and Diplesiostigma bisetosum Boucek (Tetracampidae).

Abstract

The whitefly Bemisia tabaci (Gennadius) biotype B also known as Bemisia argentifolii (Gennadius) is a more malignant whitefly biotype in damaging plants compared to non-B biotype. Currently the whitefly B. tabaci biotype B has been reported to exist in Indonesia. Basic information such as life cycle, length of life, fecundity, and breeding ability of a whitefly is important information as a basis in preparing the whitefly control strategy. The aim of this research was to study the life cycle and demographic statistic of the B. tabaci biotype B and the non-B biotype on chili pepper. The study was conducted by observing the development of the whiteflies from eggs to adult in a growth chamber that the temperature and lighting were controlled. Observations were made on several aspects of biology and some parameters of demographic statistics. The results showed that the biotype B of B. tabaci has several different biological aspects with the non-B whitefly in chili pepper. The life cycle of the biotype B of B. tabaci and the non-B were different, 33.27 and 30.86 days respectively. The biotype B of B. tabaci had a net reproduction rate (R0) which was similar to that of the non-B biotype as well as the average of its generation. However, the intrinsic increase rate (r) of the biotype B B. tabaci was 2.5 times shorter than the non-B biotype. The biotype B of B. tabaci doubled its population (DT) 2 times faster than the non-B biotype. It is clear that the biotype B of B. tabaci potentially more dangerous than the non-B.

Abstract

The African oil palm weevil, Elaeidobius kamerunicus is an effective pollinator of oil palm. Each individual palm produces exclusively male or female inflorescence so that the success of pollination depends on the ability of the pollinator to transfer pollen from male to female flowers. The objective of this research was to study the amount of pollen carried by E. kamerunicus between male and female inflorescences (pollen load) and the amount of pollen carried on each part of the weevil’s body (pollen distribution). Fifty each of male and female  E. kamerunicus individuals were collected from male and female flowers on trees in 3 locations: Siantar (North Sumatra), Dramaga (West Java), and Morowali (Central Sulawesi). Data on pollen load and pollen distribution on the weevil’s body were analyzed using ImageJ software. Results show that E. kamerunicus individuals collected more pollen from male flowers than from female flowers. In addition, male insects carried more pollen on their bodies than female insects. Pollen distribution on weevil body parts was highest on the elytra, followed by the thorax, abdomen, legs, and head respectively.

Abstract

The sweet potato whitefly, Bemisia tabaci (Gennadius) is a cosmopolitan pest on various types of agricultural crops. The whitefly is able to cause damage to plants by directly consuming plant parts using stylet and as a vector of plant viruses. Red chili (Capsicum annuum) and tomato (Solanum lycopersicum) plants are important horticultural plants that are often infested by B. tabaci. Whiteflies are known to reproduce with copulation that produce male and female offspring and without copulation that produce male offspring. The purpose of this study was to determine the reproduction ability of B. tabaci with and without copulation in red chili pepper and tomato. Adult of B. tabaci was taken from a greenhouse in Cikabayan, IPB Dramaga. One female who just emerged from the pupa or final instar nymph was invested in red chilli and tomato plants in polybags to determine the B. tabaci offspring produced without copulation. The same method was also done for whitefly with copulation, but using a pair of adult whitefly (male and female). Each treatment was repeated 5 times. Observations were made every day on the number of eggs produced by one female adult per day, the egg incubation period, the length of the nymph period for each instar, pupa, and adult. The results of this study showed that the fertility of B. tabaci which reproduces with and without copulation in tomato plants was higher than in chili pepper plants, but there was no difference in life cycle and sex ratio.

Abstract

The whitefly, Bemisia tabaci (Gennadius) and the aphid, Myzus persicae (Sulzer) are important pests in tomato and chilli pepper. These pests directly attack and as a viral vector can decrease the production of tomato and chili pepper. Menochilus sexmaculatus (Fabricius) and Micraspis lineata (Thunberg) are predator beetles of B. tabaci and M. persicae. This study aimed to know the potential predation of M. sexmaculatus and M. lineata as a predator of B. tabaci and M. persicae. Completely randomized factorial design using 2 factors consist of type and density of prey with 3 replicates used in this study. The potency of predatory coccinellidae as predator of B. tabaci and M. persicae was evaluated in 2 observations, i.e. predation and functional response assay. Based on logistic regression analysis, both of M. sexmaculatus and M. lineata showed characteristic of type I functional, this means that the higher density of prey, the ability and predation rate of M. sexmaculatus and M. lineata were also increased. The predator M. sexmaculatus and M. lineata were more effective to control M. persicae than B. tabaci.  M. sexmaculatus was the effective predator to control adult of M. persicae.
