Abstract

Plant extract is one of the functional ingredients to support health and immune system. Black cumin (Nigella sativa) is known to have a strong effect on immunomodulatory function. Its bitter taste and characteristic smell will affect the palatability of a final product. The palatability is important for product development in the pet food industry. The palatability defines cat’s preference and acceptance of a product. This research was aimed to determine the palatability of functional foods containing black cumin’s extract in domestic shorthair cats (Felis catus). The modified two bowl test was used in this study. Domestic shorthair cats aged 1-6 years, 7 spayed females and 2 neutered males, unvaccinated, and clinically healthy were participated in the study. Three bowls of food containing 89,1 mg; 222,75 mg; and 445,5 mg concentration of black cumin extract were offered to cats simultaneously. The first choice consumed, the duration of food consumed, and the amount of food intake were evaluated. A significant difference (p<0,05) was observed in the first choice consumed. The duration of food consumed, and the amount of food consumed were not found a significant difference. Based on all observations, this study indicates the food containing 222,75 mg concentration of black cumin is more palatable compared with other foods. Abstract

Red Blood Cell (RBC) dan Red blood cell Distribution Width (RDW) merupakan variabel hematologi yang banyak dikaji sebagai biomarker sepsis dan prediktor mortalitas pasien sepsis yang dirawat secara intensif. Penelitian ini bertujuan untuk menanalisis potensi EKP sebagai terapi sepsis tikus model melalui variabel nilai RBC dan RDW. Ekstraksi kecoa P. americana menggunakan metode maserasi dengan etanol 70%. Induksi sepsis pada tikus model menggunakan metode bedah cecal ligation and puncture. Tikus model sepsis dibagi menjadi empat kelompok terapi yaitu NaCl 0,9%, deksametason 0,75 mg/kg, EKP 50 mg/kg, dan EKP 100 mg/kg. Terapi dilakukan secara per oral 24 dan 36 jam setelah bedah CLP. Rataan nilai RBC setelah induksi sepsis dan diterapi masing-masing sebesar 6,76; 7,82; 8,01; dan 6,80. Persentase nilai RDW setelah induksi sepsis dan diterapi masing-masing sebesar 14,7; 13,75; 13,88; dan 14,26. Hasil penelitian menunjukan tidak adanya perbedaan signifikan antara setiap kelompok (P > 0,05), namun nilai RBC dan RDW lebih baik pada kelompok yang diterapi dengan deksametason dan EKP 50 mg/kg. Kesimpulan dari penelitian ini adalah EKP 50 mg/kg berpotensi sebagai terapi sepsis berdasarkan nilai RBC dan RDW tikus model sepsis.

Abstract

Jintan putih (Cuminum cyminum) merupakan tanaman yang sudah sejak lama dikonsumsi sebagai bahan masakan dan obat, namun belum terdapat penelitian terkait manfaat jintan putih sebagai antidiare. Penelitian ini bertujuan mengetahui efektivitas infusa jintan putih sebagai antidiare dan menentukan konsentrasi yang memiliki efek antidiare paling efektif. Penelitian ini juga bertujuan mengetahui kandungan metabolit sekunder pada jintan putih melalui penapisan fitokimia. Pengujian antidiare dilakukan dengan menggunakan metode proteksi intestinal dengan pemberian sediaan secara peroral. Parameter yang diamati pada metode proteksi intestinal yaitu frekuensi defekasi, konsistensi feses, dan durasi diare. Penelitian ini menggunakan 30 ekor mencit yang dibagi menjadi 5 kelompok terdiri dari kelompok kontrol negatif (Tween-80 [1%]), kontrol positif (Loperamide HCl), dan tiga kelompok perlakuan infusa jintan putih dengan konsentrasi 25%, 50%, dan 100%. Konsentrasi 25% merupakan konsentrasi jintan putih yang paling efektif, karena menghasilkan durasi diare paling pendek dibandingkan kelompok perlakuan lainnya. Penapisan fitokimia infusa jintan putih menunjukkan bahwa terdapat senyawa metabolit sekunder meliputi alkaloid, saponin, flavonoid, tanin, dengan tanin sebagai senyawa yang paling berpotensi sebagai antidiare.

Abstract

Inflammation is the body’s response to harmful stimuli, and it plays a crucial role in boosting immunity. However, if inflammation is not managed properly, it can lead to degenerative diseases. This research aims to analyze the potential of Plectranthus scutellarioides (L.) R. Br. leaves as an anti-inflammatory agent. The study employs an in silico molecular docking method, using the active compound from the plant as the test ligand and the COX-2 receptor as the target. The results indicate that the test ligand, Antosianin, shows the best interaction as an inhibitor of the target receptor, as demonstrated by its superior affinity energy value compared to the other test ligands.

Abstract

Caplak menjadi faktor penyebab gangguan kulit yang banyak terjadi pada anjing. Resiko akibat caplak membuat kulit anjing mengalami kerusakan jaringan dan reaksi inflamasi sehingga penampilan terlihat jelek. Berbagai metode penaggulangan infeksi caplak telah berkembang. Namun, perlu adanya penyediaan obat baru yang lebih efektif. Penelitian ini bertujuan menguji waktu kematian caplak terhadap kombinasi rosemari, eucalyptus, dan minyak peppermin. Caplak Rhipicephalus sanguineus diperoleh melalui teleskopik dari anjing yang positif terinfeksi caplak di Asrama Mahasiswa Papua Dramaga Bogor. Perlakukan yang digunakan dengan komposisi rosemari 2 ml, eucalypytus 2 ml, dan minyak peppermin 1 ml; rosemari 2 ml, eucalypytus 2 ml, dan minyak peppermin 1,5 ml; serta rosemari 2 ml, eucalypytus 2 ml, dan minyak peppermin 2 ml dengan berlipat ganda sebanyak tiga kali. Analisis data dilakukan dengan melihat jumlah mortalitas akibat pengaruh paparan konsentrasi yang berbeda pada kombinasi rosemari, eucalyptus, dan minyak peppermin terhadap mortalitas caplak. Komposisi rosemari 2ml, eucalypytus 2 ml, dan minyak peppermin 2 ml pada hasil penelitian merupakan kombinasi paling efektif membunuh caplak dibandingkan 2 kombinasi lainnya.

Abstract

Physalis angulata was known for its pharmacological potential, including antioxidant activity, yet the specific bioactive compounds responsible for enhancing endogenous antioxidant enzymes, such as superoxide dismutase 1 (SOD 1), remained unclear. This study aimed to identify and evaluate the active compounds of P. angulata as natural SOD 1 activators using an In silico molecular docking approach. Forty-nine ligands derived from the active constituents of P. angulata were screened based on Lipinski’s Rule of Five and ADMET properties. Molecular docking was performed using the SOD 1 protein (PDB ID: 5YTO), and docking validation yielded an RMSD value of 0.005 Å. Among the test ligands, withanolide exhibited the most favorable binding energy (−7.011 kcal/mol) and the lowest inhibition constant (7.0 pM), forming strong interactions with key catalytic residues of the enzyme. These findings indicated that withanolide had promising potential as a natural SOD 1 activator, providing a basis for future antioxidant drug discovery.

Abstract

The prevalence of burns ranks fourth in trauma after traffic accidents, falls, and violence. The prevalence of burns ranks fourth in trauma after traffic accidents, falls, and violence. The healing of burn wounds results from a complex inflammatory response where pro-inflammatory cytokines, for instance, TNF-α and MMP-9, play a pivotal role. Flavonoids with strong anti-inflammatory activity are apigenin, this compound comes from plants. In this study, we evaluated apigenin and its derivatives to determine their effectiveness as burn wound healing agents using molecular docking modeling techniques. To evaluate the molecular docking of these compounds with their target proteins, we used computer-aided drug design tools such as AutoDock Vina, PyMOL, and Discovery Studio. The data showed that apigenin compounds have better affinity than natural ligands to inhibit MMP-9 which can increase the rate of the inflammatory phase of burn wound healing. These findings indicate that apigenin and its derivatives have good potential to be candidates in the development of new therapeutic interventions for burn wound healing.

Abstract

Gastrointestinal neoplasia is an increasingly recognized condition in dogs, particularly in predisposed breeds, such as Rottweilers. A 4-year-old Rottweiler was presented by its owner to the Veterinary Teaching Hospital, School of Veterinary Medicine and Biomedical Sciences, IPB University, with clinical signs including coughing, dyspnea, lethargy, and intermittent yellow-brown, malodorous feces. Diagnostic evaluations revealed significant abnormalities in the respiratory and gastrointestinal systems. This report describes the clinical presentation, diagnostic approach, and management of a dog with suspected gastrointestinal neoplasia. The patient was provisionally diagnosed with gastrointestinal neoplasia with a poor prognosis. Although surgical intervention is generally recommended, the compromised pulmonary function of the dog precluded general anesthesia; therefore, management was limited to symptomatic and supportive therapy.

Abstract

Surgery can trigger systemic inflammatory response syndrome. This study aims to determine the effectiveness of Periplaneta americana cockroach extract as an oral systemic anti-inflammatory agent using molecular docking. The molecular docking method consisted of five steps. The first step was preparing the TNF-α converting enzyme (TACE) receptor as a native ligand, dexamethasone as a control ligand, and three test ligands of Periplaneta americana cockroach extract. The second step was docking preparation. The third step was analyzing Gibbs free energy (∆G) and root mean square deviation (RMSD). The fourth step was docking the test and control ligands with TACE. The fifth step was analyzing ∆G, inhibition constant (Ki), visualization of two- and three-dimensional interactions, percentage of binding site similarity (%BSS), and the rule of five (Ro5) on test ligands. The ∆G results for the native, control, and test ligands 1, 2, and 3 were -12.8, -7.1, -7.7, -6.9, and -9.0 kcal/mol, respectively. The Ki values for the native, control, and test ligands 1, 2, and 3 were 4.091, 6.205, 2.253, 9.521, and 2.507 µM, respectively. The results of the Ro5 analysis suggested that the three test ligands satisfied Lipinski's rule of five. This study concluded that Periplaneta americana cockroach extract is an effective oral systemic anti-inflammatory agent.
