Abstract

In advanced ovarian cancer, the main goal of surgery is complete debulking with no visible residual tumor, and predicting it before surgery has become a goal that still unresolved. The ability in advanced ovarian cancer surgery varied between healthcare facilities. Recent discovery put forward the role of fatty acid as one of the main source of energy for ovarian cancer aggresiveness and the role of fatty acid binding protein (FABP) escpecially FABP4 expression as an important biomarker for predicting residual disease in ovarian cancer. There are no data about FABP4 serum in ovarian cancer and it’s role in predicting the extensiveness of advanced stage ovarian cancer. To evaluate correlation between FABP4 serum level with PIV Fagotti Score and Peritoneal Carcinomatosis Index (PCI) in advanced stage ovarian cancer. Analytic observational study. Among 28 subjects, most age groups (46.4%) are 40–45 years of age, 46.4% with normal body mass index, 71.4% with ovarian cancer stage IIIC, 75% with high grade serous adenocarcinoma, average ascites volume 3232.1 ± 2006.9 mL, median CA125 serum 1094 u/mL (12–19425 u/mL). The rate of optimal and complete debulking is 53.5% from 28 subjects. Strong expression of FABP4 on 42.9% subjects. MeanFABP serum 69.6 ± 51.4 ng/mL. Mean PCI score is 14.5 (3–29), mean PIV score is 6 (2–12). There is a significant correlation between FABP4 expression and FABP4 serum level (p<0.05) with moderate power (r=0.55), and a significant correlation with moderate power (r=0.421) between FABP4 serum level with PCI and PIV score. There is a significant correlation with moderate power between rising FABP4 serum and the extent of peritoneal carcinomatosis evaluated by PCI and PIV Fagotti Score in advanced stage ovarian cancer.ABSTRAKPrediksi resektabilitas pada kanker ovarium epitel masih merupakan masalah penting yang belum terpecahkan. Penelitian-penelitian mutakhir mengemukakan peran dari asam lemak sebagai salah satu sumber energi penting bagi progresivitas kanker ovarium. FABP4 sebagai salah satu protein yang berperan dalam transportasi asam lemak merupakan salah satu biomarker yang dijumpai pada kanker ovarium dengan residu tumor. Akan tetapi, penelitian mengenai kadar serum FABP4 pada stadium lanjut belum ada. Selain itu, apakah PCI dan PIV skor Fagotti mempunyai hubungan dengan kadar serum FABP4 pada kanker ovarium stadium lanjut belum pernah diteliti. Penelitian ini bertujuan untuk menilai korelasi antara PIV skor Fagotti dan PCI dengan kadar serum FABP4 pada penderita kanker ovarium stadium lanjut. Observasional analitik digunakan dalam penelitian ini, di mana peneliti ingin melihat korelasi antara nilai skor PIV skor Fagotti dan PCI dengan kadar serum FABP4. Subjek penelitian berjumlah 28 orang dengan karakteristik: kelompok usia terbanyak 40–45 tahun (46,4%); indeks massa tubuh normal (46,4%); stadium FIGO IIIC (71,4%); high grade serous ovarian cancer (75%); jumlah rerata asites 3232,1 ± 2006,9 mL; median kadar serum CA125 1094 u/mL (12–19425 u/mL); 53,5% subjek dapat dilakukan sitoreduksi optimal dan komplit. Ekspresi FABP4 yang kuat terjadi pada 42,9% subjek. Rerata kadar serum FABP4 69,6 ± 51,4 ng/mL. Korelasi signifikan antara ekspresi FABP4 dengan kadar serum FABP4 pada penelitian ini (p<0,05), dengan tingkat korelasi sedang (r=0,55). Rerata PCI total 14,5. Rerata PIV adalah 6 dengan nilai minimum 2 dan maksimum 12.Didapatkan korelasi signifikan (p<0,05) antara kadar serum FABP4 dengan penilaian PCI dan PIV dengan tingkat korelasi sedang (r=0,421, r=0,458). Pada penelitian ini ditemukan korelasi signifikan dengan kekuatan korelasi sedang antara peningkatan skor PIC dan PIV skor Fagotti dengan peningkatan kadar serum FABP4 pada pasien kanker ovarium stadium lanjut.

Abstract

Objective: To analyze the effect of curcumin in VEGF expression on Rattus norvegicus cervical cancer cell xenograft model.Methods: An experimental study with randomized post test only control group design. The subjects were Rattus norvegicus (Sprague Dawley), inoculated with He-la cervical cancer cells from American Type Culture Collection (ATCC) processed in stem cell laboratory Institute of Tropical Disease (ITD) Airlangga University. 5x106 of He-La cells were injected subcutaneously in dorsal flank area of Rattus norvegicus. After 30 days of observation we performed histopathological examination of xenograft tissue and randomized into 2 groups which were given curcumin orally 1000 mg/kg (curcumin group) vs. no therapy (control group). After another 30 days the xenograft tissue was dissected and underwent immunochemistry examination for VEGF expression.Results: 32 samples of Rattus norvegicus were divided into 2 groups, In curcumin group the VEGF median expression was 2,2 (0,3-7,6) and in control group the VEGF median expression was 6,6 (1,2-12). There was a statistically significant difference with p value =0,009 with Mann Whitney test (p<0,05).Conclusion: VEGF expression in Rattus norvegicus xenograft model of cervical cancer was suppressed by giving Curcumin 1000 mg/kgBB orally.

Abstract

Twin pregnancy with Hydatidiform Mole is a rare case, only about 1 in 22.000 to 100.000 pregnancies. Molar pregnancy with triplet pregnancy is even rarer, which is only 6 cases reported and mostly occurred in women who received therapy for infertility. Management in cases of twin pregnancy with hydatidiform mole is dilemmatic both for the patient and physician. We present a case of 29 years old woman with her first pregnancy of twin fetuses complicated with hydatidiform mole. Diagnostic approaches were made mainly by ultrasound examination, continued with laboratory and radiology examinations which some did not performed due to worsened vaginal bleeding. This case ended with premature labor with the result of delivery of two babies and placenta with vesicles which is a characteristic of hydatidiform mole.

Abstract

Obesitas  merupakan  suatu  penyakit  kelebihan  massa  lemak  tubuh  yang  mempunyai  efek  merugikan  bagi  kesehatan.  Pada saat  ini  proses  adipogenesis  menjadi salah  satu  target  terapi  obesitas.  Salah  satu  jalur  yang  diduga  teraktivasi  pada  proses adipogenesis  adalah  melalui aktivasi  ST A T3  yang  salah  satu  jalur  hulunya  melalui protein  mammalian target  of rapamycin complex 1 (mTOR). Penelitian ini bertujuan untuk membuktikan proses adipogenesis melalui jalur ST A T3 yang diaktivasi oleh  mTORC1.  Penelitian  ini  merupakan  penelitian  eksperimental  dengan  rancangan  post  test  only  control.  Untuk penghambatan  mTORC1  digunakan  rapamycin  dan  penghambatan  ST A T3  digunakan  inhibitor  ST A T3  peptide.  Subjek penelitian  adalah  kultur  primer  sel  preadiposit  yang  diambil  dari  lemak  viseral  tikus  putih  Rattus  norvegicus.  Setelah  kultur sel preadiposit dinilai konfluen minimal 70-80% dilakukan induksi diferensiasi dan dibagi menjadi  4 kelompok yakni (K) kontrol  (A):   diberi rapamycin  10  nM,  (B):  diberi  inhibitor  ST A T3  100  µM (C):  diberi  inhibitor  ST A T3  100  µM dan   rapamycin10  nM.  Parameter  yang  diukur  adalah  aktivasi  p70S6K1,  ST A T3,  ekspresi  C/EBPδ,  aktivitas  enzim  Glyserol-3-fosfodehidrogenase  (GPDH)  pada  hari  ke-2,  ke-4  dan  ke-6  serta  gambaran  morfologis  sel  adiposit.  Analisis  statistik menggunakan uji ANOVA, Duncan dan korelasi Pearson dengan tingkat kepercayaan 95%. Hasil penelitian membuktikan terjadi  penghambatan  proses  adipogenesis  karena  penghambatan  aktivasi  p70S6K1,  dan  ST A T3  oleh  rapamycin  dan inhibitor ST A T3.

Abstract

Objective:  To compare the successful of the identification of the ureteral orifice at cervical cancer patients who received phenazopyridine orally and to analyze the correlation between hydronephrosis and successful rate for retrograde stenting. Material & Method: This was a comparative experimental study, using phenazopyridine hcl orally prior to cystoscopy to identify the ureteral orifice on cervical cancer patients and to perform retrograde stenting on spotted ureteral orifice. Forty samples provided consent to enroll in this study. Mean age was 49.2 ± 5.16 years. Thirty-four out of 40 samples was confirmed as squamous cell carcinoma type. Results: Chi-square test demonstrated no significant differentiation in finding ureteral orifice between treatment and control group on stage 3B (p = 0.408). However, result shown conversely on stage 4A (p = 0.046). There was no significant disparity in conducting retrograde stenting between treatment and control group on stage 2B and 3B (p = 0.221 and p = 0.197). There was no significant correlation between hydronephrosis gradation and retrograde stenting on control group (p = 0.144). Conclusion: Administration of phenazopyridine HCL orally in cervical cancer patients increase success for ureteral orifice identification and retrograde stenting, but not statistically significant. There is no correlation between hydronephrosis with a success retrograde stenting.Keywords: Cervical cancer, hydronephrosis, phenazopyridine hcl, ureteral orifice, retrograde stenting.

Abstract

Background: Adult stem cells are currently reliable sources of mesenchymal stem cells for regenerativetherapy, include diabetes mellitus. The aim of this study is to develop endocrine pancreatic progenitor cellscharacterized by Pdx1 and insulin expression from rat adipose-derived mesenchymal stem cells using twosteps in-vitro differentiation.Methods: In this experimental study, ADMSCs were isolated from rat adipose tissue and exposed toinsulinogenic differentiation medium containing nicotinamide, activin A and glucagon-like peptide-1 (GLP1). After induction, the existence of pancreatic progenitor cells (PPCs) was confirmed by immune-stainingassay of Pdx1 and insulin.Results: After three weeks of in-vitro differentiation, expression of Pdx1 and insulin proteins showed upas green in the immunofluorescence assay. Immunofluorescence intensity of Pdx1 was higher in PPCs thanin ADMSCs control (p<0.05). Immunofluorescence intensity of insulin was also higher in PPCs than inADMSCs control (p<0.05). Therefore, in-vitro differentiation was successful to develop PPCs from ratADMSCsConclusion: This study has demonstrated the in-vitro differentiation of ADMSCs into PPCs that expressedPdx1 and insulin

Abstract

Tujuan: mencari peningkatan ekspresi cleaved caspase-3 pada pasien kanker serviks IIB tipe sel skuamosa sesudah pemberian kemoterapi neoadjuvan cisplatin dan mencari kapan waktu yang tepat untuk mendeteksi apoptosis menggunakan cleaved caspase-3. Bahan dan Metode: Jenis penelitian analitik observasional berpasangan. Dilakukan pada penderita kanker serviks IIB tipe sel skuamosa berdasarkan kriteria FIGO yang berobat di POSA RSUD Dr. Soetomo Surabaya. Penelitian dilakukan mulai bulan Mei 2014. Sebelum dilakukan terapi semua penderita diambil biopsi serviks. Kemudian penderita diberikan cisplatin 50mg/m2/minggu Diambil biopsi serviks setelah diberikan kemoterapi ke 1 dan ke 4 (maksimal 8 jam setelah kemoterapi, sampel kedua). Sampel diperiksakan imunohistokimia cleaved caspase 3. Dilakukan penghitungan ekspresi cleaved caspase 3 di bawah mikroskop dengan perbesaran 400x pada 10 lapang pandang. Pengujian statistik dilakukan dengan nilai kemaknaan p<0,05.Hasil: Rata-rata ekspresi cleaved caspase 3 sebelum kemoterapi adalah 1,46± 1,854. Setelah kemoterapi ke 1 didapatkan rata- rata 10,77± 3,655.Setelah kemoterapi ke 4 didapatkan rata- rata 12,77± 5,703. Pada pemeriksaan didapatkan ekspresi cleaved caspase 3 setelah pemberian kemoterapi lebih meningkat dibandingkan sebelum pemberian kemoterapi (p<0,01), sedangkan ekspresi cleaved caspase 3 setelah kemoterapi ke 1 dan ke 4 tidak didapat-kan perbedaan bermakna (p=0,882).Simpulan: Pemberian kemoterapi terbukti memberikan efek peningkatan ekspresi cleaved caspase-3. Sehingga cleaved caaspase-3 dapat dijadikan sebagai salah satu uji apoptosis pada efek kemoterapi terhadap sel kanker.

Abstract

Objectives: This study aimed to determine the resistance of cisplatin (chemotherapy drugs) at three point areas including expression of CTR1, ERCC1, and HSP70 in cervical cancer patients.Materials and Methods: This research used a cross sectional approach. The population in this study were patients with stage IIB of cervical carcinoma. The study sample were patients with stage IIB of cervical carcinoma according to the FIGO classification, patients received cisplatin chemotherapy treatment (50 mg/m2 4 times), had good kidney and liver function. The independent variables in this study were the expression of CTR1, ERCC1, and HSP70 and the dependent variable of this study was the response to cisplatin therapy in cervical cancer. Primary data was obtained through MRI examination after the administration of cisplatin 50 mg/m2 every week. The parameters of this study included the expression of CTR1, ERCC1 and HSP70. Data obtained were analyzed using Wilcoxon rank test, Spearman test and categorical regression with a significance level of p <0.05.Results: The treatment of cisplatin therapy in cervical cancer patients had no significant correlation between the expression of CTR1 and ERCC1, but in the expression of HSP70 there was a significant negative correlation which means that the higher the expression of HSP70, the worse the response of therapy.Conclusion: This study showed that HSP70 expression can be used as an indicator in treatment of cisplatin therapy in cancer patients through MRI examination.

Abstract

At present, ten years have passed since the human papillomavirus (HPV) vaccine was first approved for use in humans. Research related to HPV vaccine, both in terms of effectiveness and immugenicity in its development, has been widely carried out, such as in terms of the indications of the HPV vaccine use that is not only for preventing cervical cancer, the guidelines for administering 2-dose HPV vaccines for those under 15 years of age, and the discovery of the latest HPV vaccine types: nonavalent HPV vaccine. This review literature discusses all aspects of the development of HPV vaccine since it was first approved for use in humans in 2006.

Abstract

HIGHLIGHTS1. This retrospective cohort study found an increase in the diagnosis of malnutrition between the IMT method and NRI.2. A significant correlation was found between preoperative malnutrition and the incidence of anaemia after adjuvant chemotherapy for ovarian cancer patients.3. There were no significant correlations in this study in the incidence of leukopenia and thrombocytopenia.ABSTRACTObjectives: To observe correlation of nutritional status using Nutritional Risk Index with the side effects of adjunctive haematological chemotherapy.Materials and Methods: This study was a retrospective cohort study observing whether or not hematologic side effects occurred during chemotherapy based on medical records of postoperative ovarian cancer patients receiving adjuvant chemotherapy.Results: Sixty-eight subjects with age range of 31-50 years (44.1%) multipara (68.8%), and advanced stage (52.1%) were observed. An increase was found in the diagnosis of malnutrition between the IMT method and NRI, which was 18.7% compared to 43.7%. A significant correlation was found between preoperative malnutrition and the incidence of anaemia after adjuvant chemotherapy for ovarian cancer patients (p=0.002). Whereas, in the event of leukopenia and thrombocytopenia, there were no significant correlations with p=0.675 and p=0.415, respectively.Conclusion: There was an increase in malnutrition rate with the use of NRI compared with BMI and there was a significant correlation between malnutrition and side effects of anaemia in patients with ovarian cancer who underwent surgery and continued with adjuvant chemotherapy.
