Abstract

The free-range system is a currentmodel of chicken production according to the concept of Animal Welfare. This article aims to discuss how the free-range system was implemented and its effect on health, performance and quality product as well as free-range development strategies as an alternative to chicken production systems. The free-range system must have access to outdoor to express their natural behavior. Based on regulation, the density of chickens in outdoor is a maximum of 10,000 birds/ha. Chickens reared using the free-range system show better welfare indicators, as evidenced by the Heterophil/Lymphocyte (H/L) value and the frequency of their natural behavior. Chicken eggs from the free-range system contain higher levels of protein and carotenoid. It also contains omega-3 & 6, DHA, vitamins A and E and have lower cholesterol level. Chicken meat from the free-range system contains higher protein and lower fat levels. However, the performance of chicken production using free-range system was still not consistent and was greatly influenced by the type of forage in the pasture. The introduction of grazing land with superior forage such as Indigofera zollingeriana and choosing kampung or local type of chickens may become development strategies of chicken production in the free range system.

Abstract

Broiler chicken has high fat content, especially abdominal and subcutan fat which reduced carcass quality and efficiency of feed energy. Genetic approach could be potentially applied to reduce high abdominal and intramuscular fat in broiler chicken through the selection program at gene level related to fat metabolism. This paper describes the metabolism and biosynthesis of body fat and the role of its controlled genes. Fat synthesis is controlled by feed quality and metabolism and biosynthesis process occurred in liver. These processes are controlled by many family genes, but certain genes have dominant role in the process; those are triglyceride lipase genes, fatty acid synthase genes and fatty acid binding protein genes. Expression of fatty acid synthase genes has positive correlation with fat content in liver and intramuscular. Expression of fatty acid binding protein genes was related to the increased abdominal fat deposit. These genes are polymorphic, so that they can be used as a genetic marker in selection to optimize feed efficiency, to minimize abdominal fat and to increase economic value of broiler chicken. Abstract

Penelitian ini bertujuan untuk mencari metode yang efektif dalam teknik ekstraksi DNA dari gen Mx (Myxovirus) ayam Tolaki. Gen Mx adalah gen yang bertanggungjawab terhadap kemampuan ayam untuk mertespon serangan penyakit viral seperti penyakit Flu Burung dan New Castle Disesase. Penelitian ini dilaksanakan mulai bulan April sampai dengan bulan Agustus 2013.  Sampel penelitan terdiri atas 25 ekor ayam Tolaki dewasa, yang diperoleh dari pemeliharaan secara ekstensif oleh masyarakat di Kabupaten Konawe. Penelitian ini menggunakan kedua metode ekstraksi DNA yakni metode ekstraksi DNA secara konvensional dan metode Kit ekstraksi DNA.  Selanjutnya hasil ekstraksi DNA diamplifikasi menggunakan mesin PCR. Produk DNA kedua metode tersebut kemudian dibandngkan dan dianalisis secara kualitatif. Secara umum kedua metode tersebut cukup efektif untuk menghasilkan kualitas DNA gen Mx yang baik. Ekstraksi DNA gen Mx dari kedua metode tersebut menghasilkan ukuran DNA dengan panjang 299 pb. Kata Kunci : Gen Mx, Ayam Tolaki, Ekstraksi DNA, PCR

Abstract

ABSTRAKPenelitian ini bertujuan untuk menganalisis gambaran sel darah putih dan sel darah merah sebagai indikator ketahanan tubuh pada anak ayam sentul. Penelitian ini menggunakan 20 ekor ayam betina dan 20 ekor ayam jantan yang dikawinkan, masing masing terdiri dari 10 ekor ayam dengan konsentrasi IgY tinggi dan 10 ekor ayam dengan konsentrasi IgY rendah. Berdasarkan hasil perkawinan tersebut diperolah 34 ekor anak ayam yang terdiri dari 17 ekor anak ayam yang berasaldari induk dengan konsentrasi IgY rendah. Parameter yang diamati meliputi gambaran sel darah merah (eritrosit, hematocrit dan indeks eritrosit) dan gambaran sel darah putih (leukosit dan diferensiasinya). Penelitian ini menggunakan rancangan acak lengkap (RAL) dengan 2 perlakuan, semua perlakuan diulang 10 kali. Data yang diperoleh dianalisis menggunakan t-test program SAS.  Hasil penelitian ini menunjukan bahwa perbedaan konsentrasi IgY pada induk tidak mempengaruhi sel darah merah dan sel darah putih. Namun terdapat kecendrungan nilai limfosit yang rendah dan H/L yang tinggi pada anak ayam yang berasal dari induk dengan konsentrasi IgY rendah. Hasil ini mengindikasi bahwa anak ayam yang berasal dari induk dengan konsentrasi IgY rendah lebih rentan terhadap cekaman panas. Kata Kunci: ayam sentul, IgY,  gambaran darah, ketahanan tubuhABSTRACTThis study was to aim the description of white blood cells and red blood cells as indicators of immunity in Sentul chicks. This study used 20 hen chickens and 20 cock-chicken mated, consisting of 10 chickens with high IgY concentration and 10 chickens with low IgY concentrations. Based on the results of mating, 34 chicks were obtained consisting of 17 chicks that were from the hen with low IgY concentration. The parameters observed included the description of red blood cells (erythrocytes, hematocrit and erythrocyte index) and the description of white blood cells (leukocytes and their differentiation). The experiment was conducted as a completely randomized design with 2 treatments, all treatments were repeated 10 times. The data obtained were analyzed using the SAS program t-test. The results of this study indicate that the difference in IgY concentration in the hen did not effect on red blood cells and white blood cells. However, there is a tendency for low lymphocyte values and high H/ L in chicks originating from hens with low IgY concentrations. These results indicate that chicks from hens with low IgY concentrations are more susceptible to heat stress.Keyword: blood picture, IgY, immunity, sentul chicken

Abstract

ABSTRAK Gen Toll-like Receptor 4 (TLR4) adalah salah satu gen yang mengontrol ketahanan ayam terhadap serangan Salmonella sp, melalui respon immun non-specific. Gen ini dapat digunakan sebagai marka genetik pada ayam.  Oleh karena itu penelitian ini bertujuan mengevaluasi polimorfisme genetik gen TLR4 pada beberapa jenis ayam (Ayam Tolaki, Ayam Ras pedaging dan petelur). Total sampel sebanyak 126. Penelitian ini melalui 3 tahapan yakni ekstraksi DNA, amplifikasi PCR (dengan ukuran DNA 220 pb pada ekson 2), dan metode RFLP menggunakan  enzim MscI.  Hasil penelitian menunjukkan gen TLR4 | MscI bersifat polimorfik pada semua jenis ayam. Diperoleh 2 alel (A dan G). Frekuensi alel G (membawa sfat resisten) pada ayam Tolaki, Ras pedaging, dan petelur masing-masing 0.77, 0.87 dan 0.20. Nilaix2menunjukkan genTLR4|MscI berada dalan keseimbangan Hardy-Weinberg dengan Ho dan He 0,31 dan 0,29. Nilai PIC 0,25 termasuk kategori medium. Gen TLR4| MscI dapat digunakan sebagai marka genetik sifat ketahanan ayam Tolaki terhadap infeksi Salmonellasp. Keywords : Polimorfik, Gen TLR4, Salmonella sp., Non-Specific Immune Response

Abstract

The aim of this study was to evaluate the effect of different IgY concentrations on the production, reproduction and immunity of Sentul chicken. This research used 20 month-old Sentul chickens which consisting of 10 chickens with high IgY concentration and 10 chickens with low IgY concentration Parameters observed included prelayer performance (feed consumption, weight gain, feed conversion, morbidity and mortality), layer performance (feed consumption, egg production, egg weight, feed conversion, hatchability, morbidity and mortality), and immunity at chicks (IgY concentration of eggs and chicks serum). The experiment was conducted as a completely randomized design. The treatments were high and low IgY concentration. All treatments were repeated 10 times. The data obtained were analyzed using the SAS program t-test. Morbidity and mortality were analyzed descriptively. The results of this research indicated that the prelayer period, chickens with high IgY concentration had a higher performance than chicken with low IgY concentration. This is caused by about 30% chickens with low IgY were in sick condition for approximately seven days. Further, the layer period showed that the chickens with high IgY concentration had lower production performance than the chicken with low IgY concentration. The study concluded that chickens at low IgY concentrations are susceptible to exposure to antigens that lead to decreased productivity. In normal conditions hens, Sentul with high IgY concentration resulted in low production. The observation of chicken immunity showed that concentration of IgY from chicks and eggs did not differ between hens with high IgY concentrations and hens with low IgY concentrations.

Abstract

The aim of this study was to evaluate the effect of different IgY concentrations on the quality of semen (macroscopic and microscopic) and testicular morphometry (weight, length and width of the testes) of Sentul cock testis and its ability to fertilize hens. This study used 4 months of 20 Sentul cocks consisting 10 high IgY concentration cocks and low IgY concentration cocks. This study used a complete randomized design (RAL) with 2 treatments namely low and high IgY concentration and10 times repetitions. Data were analyzed using t-test minitab program versions 16. Based on the result, different IgY concentrations did not affect the quality of semen and testicular morphometrics, but high IgY concentration cocks were able to fertilize more hens than the low IgY concentrations one by using dilution methods. This proves that high IgY concentration cocks are more effective and efficient cocks or stud.

Abstract

Heat stress in the poor transportation systems will impact the oxidative stress, affecting the quality of chicken meat. The decrease in heat stress levels can be trigerred through antioxidants, one of which is ZnSO4 antioxidants, before cutting. This study aimed to examine the reduction of post-transport stress on the performance of chickens, carcasses, and the physical and chemical qualities of broiler meat treated with ZnSO4. The total broiler chickens transported were 324 roosters aged 4 weeks (with 3 replicates each). All of the transported chickens were treated with ZnSO4 at doses of 0, 80, and 160 ppm for 7 days. Next, the chickens were transported with a travel time of 1, 2, and 3 hours. The breast meats were taken (filet) to be used as research samples. A factorial randomized block design was used as the research design and the data were analyzed using ANOVA (analysis of variance). The results showed that there was no correlation between travel time and the ZnSO4 levels on broiler chicken performance (weight and carcass loss) and physicochemical quality of broiler meat (pH value, percentage of water lost, aw, cooking loss, glycogen content and MDA). Longer travel time significantly reduced live weight, carcass percentage, pH value, glycogen content, while increasing the MDA levels. The addition of ZnSO4 can significantly increase the pH value of breast meat, maintain high glycogen levels, increase carcass percentage, and inhibit MDA formation.

Abstract

Effect of zeolites spreading on litters to broiler performances under different floor spaces had been performed in five weeks. As many as 324 chicks of a day-old Hubbard strain were fed rations contained 3,000 "“ 3,100 Kcal/Kg ME and 21% crude protein during starter period and fed the ration contained 20% crude protein with similar energy during finisher period. The chicks were randomly allocated to three different floor spaces (10, 12, and 14 heads/m2) and three different zeolites spread-litters (0.0, 2.5, and 5.0 kg zeolites/m2) under completely randomized design with factor 3 x 3 and 3 replications. Parameters concerned of present research including feed intake, live weight gain, and mortality rate were analyzed with analysis of variance (ANOVA) and least significant different (LSD).
