Abstract

Insuline-like growth factor binding protein 2 (IGFBP2) is one of the principal binding proteins that has biological functions involved in growth, development, and differentiation. Selection for rapid growth based on molecular Marker Assisted Selection is required to increase production performance. The present study was designed to analyze the associations of IGFBP2 gene polymorphisms with chicken growth and body composition traits. Kampong chicken, a native chicken in Indonesia, is slow-growing chicken. A total of 59 males were used in the current study. Growth and body composition were measured in 24 wk of age. Primers for intron 2 region were designed from genomic chicken sequence. A c.1032C>T SNP of the IGFBP2 gene intron 2 region was detected and PCR-RFLP method was then used to genotype Kampong chicken population. The result showed that IGFBP2 polymorphism was significantly associated with body, carcass, breast, breast muscle, pectoralis minor, leg, and wings weight in Kampong chicken population (P < 0.05). The research suggests that the IGFBP2 gene could be a candidate gene that affects growth and body composition traits in chicken.

Abstract

Effect of zeolites spreading on litters to broiler performances under different floor spaces had been performed in five weeks. As many as 324 chicks of a day-old Hubbard strain were fed rations contained 3,000 "“ 3,100 Kcal/Kg ME and 21% crude protein during starter period and fed the ration contained 20% crude protein with similar energy during finisher period. The chicks were randomly allocated to three different floor spaces (10, 12, and 14 heads/m2) and three different zeolites spread-litters (0.0, 2.5, and 5.0 kg zeolites/m2) under completely randomized design with factor 3 x 3 and 3 replications. Parameters concerned of present research including feed intake, live weight gain, and mortality rate were analyzed with analysis of variance (ANOVA) and least significant different (LSD). Abstract

Toll-like Receptor 4 (TLR4) gene is one of the genes that control resistance of chickens againstSalmonella sp. through non-specific immune response. These gene can be used as genetic markers inIndonesian local chickens, if known its diversity. Therefore this research was aimed at evaluating thegenetic polymorphism of TLR4 gene in several types of Indonesian lokal chickens (kampung, kampungpelungcrossbreed, sentul, and tolaki chicken) using PCR-RFLP. The total of samples were 136. There werethree stages of identification : extraction of DNA genom, PCR amplification of TLR4 gene (with size 220 bpon exon 2), and RFLP method using restriction enzym (MscI). The data were analyzed include frequency ofallele, frequency of genotype, heterozygosity values , and value of Polymorphic Informative Content (PIC).The result showed that TLR4|MscI was found polymorphic in all kind of chickens that was genotyped. Itwere found two alleles (A and G), and three genotypes (AA, AG, and GG). The value of x2 showed thatTLR4|MscI was in Hardy-Weinberg equilibrium. The value of Ho and He were 0,11-0,41 and 0,11-0,35. Thevalue of PIC (0,10-0,29) included in the category of low to moderately high. These results declared thatTLR4|MscI gave hope for used as genetic markers in resistance to Salmonella sp. infection in Indonesianlocal chickens.

Abstract

Stunning method before slaughtering is considered more animal welfare compared to a non-stunning method and also reduces stress levels in broilers. As for the bleed-out efficiency, testing was done indirectly by blood volume measurement which consists of erythrocyte count, leucocyte count, hemoglobin levels, hematocrit values, and erythrocyte index in broilers.   Brain histopathology was also carried out to see the effect of electrical stunning towards the brain tissues.  The stunning method used is electrical stunning via water bath with the capacity of 146 mAh.  The results showed that the stunning method before slaughtering caused smaller number of erythrocytes and hematocrit values, and the brain histopathology showed that stunning causes encephalopathy.

Abstract

This study aims to study the impact on Maintenance broilers of temperature differences on ammonia, carbon dioxide, and dust. The completely randomized design (CRD) was used for the experiment design, with temperature of 20oC (KS20) and 30oC (KS30) as treatments. The experiment was repeated four times and each replication contained of 10 chickens. The ammonia, carbon dioxide, and dust sample was taken three times, at the 3rd, 4th, and 5th week of the experiment. Ammonia concentration during the 4th week was 0.08 ppm and increased to 2.022 -2.027 ppm at the 5th week; the carbon dioxide concentration at low temperature and high temperature was increased 17.76-22.13 ppm at the 5th week. The total dust increased at the 5th on the low temperature condition. The conclusion of this study, broilers were maintained at high temperature (30oC) produced lower air quality, in conditions this indicate that the broilers environment with conditions reared at high temperatures is still in a good condition.

Abstract

The Thy1 gene is a member of the immunoglobulin superfamily, which is found in many different animals. This gene encodes a Thy1 protein (CD90) which measures 25–37 kDa. Although the function of the Thy1 gene is unknown, it is thought to play a crucial role in the immunological response, particularly in the activation of T cells. The goal of this study was to find a variation in the Thy1 gene in IPB D2  chickens that might be utilized as candidates for immunoresistance genetic markers. A total of 123 samples from IPB D2 chickens consisting of 38 samples from IPB D2 G0 (basic generation) chickens and 85 samples from IPB D2 G2 (second generation) chickens were used in this study. PCR methods and direct-DNA sequencing were used in this study. Data analysis in this study consisted of genotype frequency, allele frequency, observed heterozigosity, expected heterozygosity, and Hardy-Weinberg equilibrium. The results of the study  discovered two SNPs on the intron (g.2139 CT and g.2542 AG). Four haplotypes were created by combining two SNPs. The g.2139 CTand g.2542 AG were polymorphic, but not in the Hardy-Weinberg equilibrium.

Abstract

Quails are classified according to their immunity to different IgY concentrations. Quail IgY concentrations range from 0 to 1200 µg ml-1, and these IgY concentrations may affect production and reproductive performance. This study aimed to analyze IgY concentrations in male quail and to compare production and reproductive performance at different IgY concentrations. Forty-two male quail at five weeks of age participated in the study, including 29 quail with low IgY concentration (210-393 µg ml-1)   and, 13 with intermediate IgY concentration (403-564 µg ml-1). The observed productive performances were feed intake, initial body weight, final body weight, weight gain, feed conversion, morbidity, and mortality. In addition, this study observed testicular weight and macroscopic and microscopic semen quality for reproductive traits. Differences in production and reproductive performance of each group were analyzed using an independent-sample t-test. The result showed that male quail with different IgY concentrations were equal in all productive traits (P0.05). This means that male quail with low IgY and medium IgY concentrations are equally good. Testicular weight, semen color and pH were identical at different IgY concentrations. Quails with low IgY concentrations have better sperm consistency, which is related to the weight of the testes, which are heavier in quails with low IgY concentrations than in those with moderate IgY concentrations. This consistency is related to the concentration of sperm in the semen. The result concludes IgY concentrations in male quail did not affect production performance. Quails with low IgY concentration had thicker sperm consistency and higher sperm concentration.

Abstract

Pentingnya pemahaman yang baik tentang karakteristik SDGT sangat diperlukan untuk dijadikan sebagai referensi pengambilan keputusan dalam pengembangan dan program pemuliaan ternak. Penelitian ini bertujuan mengidentifikasi keragaman fenotipe sifat kuantitatif ayam burgo, ayam hutan merah, dan ayam kampung dalam upaya pelestarian dan pemanfaatan SDG ayam lokal khususnya di Bengkulu, dan menganalisis faktor-faktor peubah penentu ukuran dan bentuk ayam burgo, ayam hutan merah, dan ayam kampung. Bahan penelitian yang digunakan adalah 47 ekor ayam burgo, 9 ekor ayam hutan merah (Gallus gallus spadiceus), dan 18 ekor ayam kampung (Gallus gallus domesticus). Sampel diambil menggunakan teknik purposive sampling dan ayam yang diambil merupakan ayam yang sudah dewasa di Kabupaten Mukomuko, Kabupaten Bengkulu Utara, Kabupaten Bengkulu Tengah, Kota Bengkulu, dan Kabupaten Bengkulu Selatan, Provinsi Bengkulu. Data-data sifat kuantitatif berupa berat badan, panjang tubuh, panjang dada, lingkar dada, rentang sayap, panjang paha atas, panjang paha bawah, panjang shank, panjang jari ketiga, dan lingkar shank ayam burgo, ayam hutan merah, dan ayam kampung. Hasil uji-t menunjukkan bahwa karakteristik sifat kuantitatif ayam burgo jantan lebih mendekati karakteristik sifat kuantitatif ayam kampung jantan, sedangkan karakteristik sifat kuantitatif ayam burgo betina lebih mendekati karakteristik sifat kuantitatif ayam hutan merah betina. Berdasarkan hasil penelitian dapat disimpulkan bahwa: (1) ayam burgo dan ayam hutan merah memiliki sifat kuantitatif dengan tingkat keragaman yang rendah (monomorfik), sebaliknya dengan ayam kampung memiliki sifat kuantitatif dengan tingkat keragaman yang tinggi (polimorfik), dan (2) peubah penentu ukuran ayam burgo adalah panjang paha bawah (jantan) dan panjang jari kaki ketiga (betina), ayam hutan merah adalah panjang paha bawah (jantan) dan panjang paha atas (betina), dan ayam kampung adalah panjang shank, sedangkan peubah penentu bentuk ayam burgo adalah panjang paha bawah, ayam hutan merah adalah panjang jari kaki ketiga, dan ayam kampung adalah panjang shank.
