Abstract

Phytoplankton plays an important role in the aquatic ecosystem for supporting the survival of organisms, such as zooplankton and fish. The aim of this research was to observe the phytoplankton in the common carp ponds and tilapia ponds with water quality parameters at IBAT Punten, Batu, East Java. The survey method was used in this research. The water quality assay was temperature, brightness, pH, DO, CO2, Nitrate, and Orthophosphate, and the phytoplankton analysis was phytoplankton abundance, diversity index, dominance index, relative abundance, similarity index, and evenness index. The water quality on the carp ponds and tilapia ponds was quite good for common carp and tilapia growth. The average of phytoplankton abundance in common carp ponds were 1538 cell/ml which classified oligotrophic and tilapia ponds were 4900 cell/ml which included mesotrophic respectively, diversity index was moderate in both ponds, no dominance specific phytoplankton in both ponds the relative abundance in both ponds showed different phytoplankton availability, the similarity, and evenness index showed the same in both ponds and evenly on tilapia ponds and equally on common carp pond respectively. The carbon dioxide (CO2), nitrate, and orthophosphate have a strong correlation to phytoplankton abundance with value 0.882; 0.949; and 0.967 respectively. In the future, pond management will be needed in IBAT Punten with growing natural food.

Abstract

Fish larvae are the early stage of the life cycle and the key to mass production of fish. Its nature which is susceptible to invade by pathogens, especially viruses, it is an important concern in increasing the innate immune system in virus invading. IFNa and Mx are inducer genes that have the most role in enhancing the innate immune system. The study was conducted in September 2017 with the experimental method. The sample used was 1-day-old medaka larvae in amount 30 fish in each tank with triplicate in each concentration. The cytotoxicity results showed that at 72 hours cinnamaldehyde immersion was 100% of medaka larvae survival rate at a concentration of 2.5 µg mL-1 and 0 µg mL-1 (control). One-Way ANOVA results, the genes expression of IFNa and Mx on 1-day-old medaka larvae showed decreasing at 2.5 µg mL-1 concentration of cinnamaldehyde immersion for 3 days with several observation periods (12h; 24h; 48h; and 72h) compared to control. The lowest expression of IFNa genes was 12h (2.54 x 10-5 ± 6.12 x 10-6) at a 2.5 µg mL-1concentration of cinnamaldehyde, and the highest expression of IFNa gene was 48h (3.95 x 10-4 ± 7.78 x 10-5)at 0 µg mL-1 (control)concentration of cinnamaldehyde. While the lowest Mx gene expression was 72h (8.96 x 10-3 ± 4.51 x 10-4) at a 2.5 µg mL-1concentration of cinnamaldehyde. And the highest Mx gene expression was 72h (0.88 ± 0.26) at 0 µg mL-1 (control)concentration of cinnamaldehyde. The immersion of cinnamaldehyde extract at a concentration of 2.5 µg mL-1 is not able to increase the innate immune system in medaka larvae. More assay is needed to determine the mechanism for the phenomenon of decreasing IFNa and Mx gene expression on fish larvae.

Abstract

Ekosistem lamun di wilayah Paciran, Lamongan, Jawa Timur, merupakan habitat bagi komunitas makrozoobentos. Tujuan dari penelitian ini adalah untuk mengetahui struktur komunitas makrozoobentos di wilayah pesisir Paciran dengan metode survei. Sampel makrozoobentos diamati dengan menggunakan transek saat air laut surut di tiga stasiun yang telah ditentukan. Stasiun 1 merupakan wilayah tempat bersandarnya kapal; stasiun 2 adalah daerah dekat pemukiman penduduk dan stasiun 3 di dekat daerah pembenihan ikan atau udang. Hasil penelitian ditemukan 3 filum organisme, yaitu Mollusca, Echinodermata, dan Annelida. Kelimpahan makrozoobentos di tiga stasiun berkisar 486-608 ind/m2, dengan indeks keanekaragaman (H’) sedang (1,68-1,71), indeks keseragaman (E) tinggi (0,70-0,73) dan indeks dominasi (C) rendah (0,23-0,25) dengan kelimpahan relatif tertinggi yaitu pada gastropoda Cerithium granosum dan tidak ditemukan makrozoobenthos yang mendominasi. Berdasarkan hasil penelitian dapat disimpulkan bahwa perlu adanya pemantauan dan penyuluhan bagi warga sekitar lingkungan pantai agar ketersediaan makrozoobentos tersebut tetap lestari.

Abstract

In this study, the effect of cinnamaldehyde (CA) towards Streptococcus agalactiae on zebrafish egg were investigated. To obtain the optimal concentration, CA and S. agalactiae were tested by short-term effects assay. Fresh zebrafish egg, CA and S. agalactiae were observed in different groups, single and mix treatment. The result showed that the larval survival rate (%) of single CA concentration (2.5 and 5 µg mL-1) were not significantly different compared to the control, with the value 94 ± 2.8% and 93 ± 3.3% respectively. Meanwhile, the single S. agalactiae concentration (109 and 1010 CFU mL-1) were significantly different in comparison to the control with the value 73 ± 2.1% and 6 ± 2.5%. Mix groups, larval survival rate (%) value were significantly different with the best combination value was 17 ± 4.7% at 5 µg mL-1 of CA and 109 CFU mL-1 of S. agalactiae. Even though CA may potential for zebrafish egg shelter against S. agalactiae infection. But a mixture (CA and S. agalactiae) treatments were not recommended for this study because the low value was shown compared to the single treatments. This possibility may occur because of the debris formation of two substances which leads to the poor environment and causes to low fish larval survival rate (%) value. For further study, the water quality assay is needed.

Abstract

Plankton merupakan salah satu jenis pakan alami yang berperan penting untuk pertumbuhan organisme akuatik terutama ikan. Tujuan dari penelitian ini adalah untuk mengetahui jenis-jenis plankton beserta kelimpahannya di perairan dan di lambung ikan nila yang tertangkap di Waduk Wonorejo, beserta aktivitas enzimatis lambung ikan nila tersebut. Penelitian ini bertempat di Waduk Wonorejo, Tulungagung, Jawa Timur dengan metode survei. Sampel diambil dari 2 stasiun, yaitu stasiun pertama di daerah wisata Waduk Wonorejo, sedangkan stasiun 2 berada di wilayah pemancingan umum Waduk Wonorejo. Plankton di perairan Waduk Wonorejo ditemukan 17 genus fitoplankton, dan 5 genus zooplankton, sedangkan di lambung ikan nila ditemukan 30 genus fitoplankton dan 2 genus zooplankton. Genus Spirogyra sp. merupakan genus yang paling tinggi kehadirannya baik di lambung maupun perairan Waduk Wonorejo. Kelimpahan plankton di perairan waduk dapat digolongkan oligotrofik dengan nilai kelimpahan plankton sebesar 1487 ind/ml di stasiun 1 dengan keanekaragaman tinggi dan 746 ind/ml di stasiun 2 dengan keanekaragaman sedang, serta tidak ada jenis plankton tertentu yang mendominasi di kedua stasiun tersebut. Analisis aktivitas enzim lambung ikan nila menunjukkan aktivitas enzim protease sebesar 0,84 ± 0,02 µmol tirosin/g enzim menit; enzim amilase 14,59 ± 1,07 µmol glukosa/g enzim menit; enzim lipase 17,83 ± 0,14 µmol asam lemak/g enzim menit. Kualitas air di Waduk Wonorejo tergolong baik dengan suhu berkisar 26,3-27,3 oC, pH 7, DO 7,1-8,4 mg/L, Kecerahan 110-154 cm, TOM 19-30,3 mg/L. Maka dari itu upaya untuk mempertahankan kondisi Waduk Wonorejo agar tetap oligotrofik perlu dilakukan.

Abstract

Warna pada ikan cupang memiliki daya tarik tersendiri bagi para pecinta ikan hias. Selain sebagai antibakteri, daun ketapang diketahui memiliki manfaat dalam mempercepat kemunculan warna pada ikan cupang. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh mutasi warna pada ikan cupang yang direndam dalam ekstrak daun ketapang. Dalam penelitian ini terdapat dua kelompok perlakuan, yaitu ikan cupang dengan perendaman air tawar (kontrol), dan ikan cupang dengan perendaman ekstrak daun ketapang, dengan pengulangan masing-masing kelompok sebanyak tiga kali, ikan cupang yang digunakan dalam penelitian ini adalah ikan cupang berusia 4 bulan yang berasal dari indukan dan kelompok breeding yang sama dan belum mengeluarkan corak warna pada tubuhnya. Penelitian ini dilakukan mulai bulan 4 Mei hingga 25 Mei 2021 di Laboratorium Nutrisi/Pakan Alami Politeknik Kelautan dan Perikanan Sorong. Pada kelompok kontrol, tidak ditemukan adanya perubahan atau mutasi warna pada ikan cupang; sedangkan pada perlakuan cahaya terang menunjukkan adanya mutasi warna dan pola yang terdapat pada tubuh ikan cupang. Daun ketapang terbukti mampu membantu pengeluaran warna pada ikan cupang.

Abstract

The aim of this study was to determine the effect of differences addition of culture medium concentrations on the growth of microalgae type Chlorella sp. Samples were obtained from the Center for Brackish Water Cultivation Fisheries (BBPBAP), Jepara. This research was conducted at the Laboratory of Nutrition and Natural Feed Sorong Marine and Fisheries Polytechnic with an experimental method using four treatments with addition of walne medium, there were A (adding only at the beginning of cultivation); treatment B (adding every day); C (adding every two days), and treatment; D (adding every three days). The cell growth performance was significantly found in treatment C with the addition of nutrients every two days with a peak cell density of Chlorella sp was 2,628,450 cells / ml. There is a need for mass culture assay to determine the level of effectiveness and efficiency.

Abstract

The low grade of mud crab catched causing market crashes. Aim of this actvities was to improve the skills of local partner in processing low grade mud crab to be more valuable. This activity was collaboration between Polytechnic KP Sorong and Conservation International using demonstration and direct instruction model through training on processing of nugget and sumpia based mud crab with 10 participants from Komplek Rimba Pala, Jalan Mandiri RT. 06, RW. 06, District Remu Selatan, Sorong City, Papua Barat Province. Post-test which part of the evaluation, it was showen an increasing in the knowledge and skill possess by local partner.

Abstract

STICOM (Smart Tilapia Community) is a program initiated by the study program. Sorong Marine and Fisheries Polytechnic Fisheries Cultivation Techniques collaboration with the Majaran Fish Seed Center. This activity expected to increase public awareness about the use of land or narrow areas to cultivate fish such as vegetables that are effective in buckets. That this will also increase the income of the residents of the Mayamuk district. Then the activity was carried out using the STICOM socialization method and the practice of making BUDIKDAMBER (fish in buckets). The socialization activity invited the residents of the Mayamuk district. After that a satisfaction survey was carried out. The results obtained are that the Mayamuk community is satisfied with the activities that have been carried out by the team and hopes that these activities will be repeated to evaluate activities.
