Abstract

Diabetes melitus merupakan penyakit metabolic yang tiap tahun mengalami peningkatan tidak hanya diperkotaan, namun juga di pedesaan seperti desa Ngepung, Kecamatan Kedamean, Kabupaten Gresik. Tujuan dilaksanakan Program Pengabdian kepada Masyarakat (PkM) ini adalah untuk mengimplemetasikan terapi berjemur di pagi hari secara rutin guna menurunkan resiko diabetes melitus dan memberikan alternatif bagi masyarakat terutama lansia untuk menerapkan pola hidup sehat sebagai pencegahan terhadap penyakit diabetes melitus. Metode pelaksanaan yang dilakukan meliputi penyuluhan diet sehat dan manfaat terapi berjemur di pagi hari dan pemeriksaan kadar glukosa darah pada hari ke-1 sebelum terapi berjemur dan ke-7 setelah melakukan terapi berjemur. Kegiatan penyuluhan diikuti 29 warga dengan target usia usia 40 tahun keatas. Implementasi berjemur di pagi hari dilakukan selama 10 menit selama 7 hari. Berdasarkan hasil kegiatan menunjukkan bahwa penyuluhan mampu meningkatkan antusiasme masyarakat untuk hidup sehat. Namun, untuk implementasi kegiatan terapi berjemur hanya diikuti 16 peserta pada hari ke-1 namun hari ke-7 sejumlah 9 orang.

Abstract

Kolagen dan kitosan dapat digunakan sebagai bahan pembalut luka karena memiliki karakteristik yang baik. Namun, pembalut luka kolagen-kitosan perlu dilakukan uji sitotoksisitas sebelum diaplikasikan secara in vivo, seperti Brine Shrimp Lethally Test (BSLT). Pembalut luka kolagen-kitosan tidak dapat larut dalam Dimetil Sulfoksida (DMSO) dan aquadest dengan mudah, oleh karena itu perlu pertimbangan alternatif pelarut karena kolagen dan kitosan lebih mudah larut dalam pelarut asam seperti asam klorida (HCl) dan asam asetat ( CH3COOH). Penelitian ini bertujuan untuk mengetahui Lethal Concentration  50 (LC50) dari pembalut luka kolagen-kitosan yang dilarutkan dalam pelarut DMSO, HCl, CH3COOH dengan metode Brine Shrimp Lethality Test (BSLT). Pembalut luka kolagen-kitosan didapatkan dengan mencampurkan larutan kitosan 2% dan kolagen dengan perbandingan 1:1 w/w kemudian dihomogenkan, dicetak, dan dikeringkan.  Penelitian ini menggunakan uji sitotoksisitas dengan metode BSLT dan LC50 dihitung menggunakan Analisis Probit. Pembalut luka dilarutkan dalam pelarut DMSO 1%, CH3COOH 1%, dan HCl 1% hingga homogen, kemudian diencerkan dengan berbagai konsentrasi yaitu 100 ppm, 250 ppm, 250 ppm, 500 ppm, dan 1000 ppm dengan tiga kali ulangan untuk setiap perlakuan. Setelah itu uji BSLT dilakukan dengan menggunakan Artemia salina. Hasil penelitian menunjukkan bahwa pembalut luka yang dilarutkan dalam DMSO 1% memiliki LC50 > 1000 ppm, sedangkan pada pelarut CH3COOH dan pelarut  HCl menunjukkan  LC50< 30. Kesimpulan dari penelitian ini adalah pelarut DMSO bersifat non-toksik (LC50  > 1000 ppm), tetapi pelarut CH3COOH 1% dan HCl 1% bersifat sangat toksik (LC50 < 30 ppm) sebagai pelarut alternatif pembalut luka kolagen-kitosan pada uji BSLT.

Abstract

Setiap harinya, sampah organik dihasilkan dari limbah rumah tangga dengan jumlah yang tidak sedikit. Sampah organik merupakan bahan buangan yang berasal dari makhluk hidup, baik manusia, hewan, maupun tumbuhan. Berdasarkan hal tersebut, perlu adanya suatu usaha untuk mengolah sampah organik rumah tangga salah satunya dengan membuat pupuk kompos. Salah satu metode pengolahan pupuk kompos yang cocok dilakukan dalam skala rumah tangga adalah dengan keranjang Takakura. Takakura adalah metode pengomposan yang memiliki kelebihan dan dapat diaplikasikan pada lahan yang sangat sempit, ruang dapur keluarga, atau kamar kost. Metode pelaksanaan yang digunakan dalam kegiatan ini adalah penyuluhan secara langsung. Pada penyuluhan tersebut juga dilakukan pelatihan (demonstrasi) cara pembuatan keranjang Takakura oleh anggota tim. Hasil yang diperoleh setalah dilakukan demosntrasi persentase pengethuan meningkat sebnyak 5%. Kegiatan ini bermanfaat untuk meningkatkan kemampuan kader desa dalam memberikan pelayanan Kesehatan bagi warga secara umum, dan lingkup keluarga kader sendiri secara khusus.

Abstract

Pseudomonas aeruginosa is one of the bacteria that causes nosocomial infections. The incidence of nosocomial infections caused by Pseudomonas aeruginosa is around 10-15%. Nosocomial infections caused by these bacteria are increasingly difficult to treat. This happens because more and more strains are resistant to several antibiotics (Multidrug Resistance), so the selection of antibiotics against Pseudomonas aeruginosa is very difficult. This study aims to determine and analyze the effect of using polar and nonpolar solvents on Dayak onion bulb extract on the inhibitory ability of Pseudomonas aeruginosa. The method used in this study was Kirby-Bauer disk diffusion using 8 sample and 5 replication with assay solvent 96% ethanol and n-hexane extract of Dayak onion bulbs (Eleutherine palmifolia L. Merr) with concentration variations of 75%, 80%, and 85%. Positive control used ciprofloxacin 5 µg and positive control DMSO 10%. The results showed that on phytochemical screening of solvent ethanol 96% contained flavonoids and n-hexane contained triterpenoids. Ethanol 96% solvent obtained a higher presentase yield than n-heksan solvents and the highest inhibition zone diameter was 85% concentration in 96% ethanol solvent. The results of statistical analysis with Mann Whitney test showed a significant difference between the treatment groups (p<0.05).

Abstract

Adolescence is a unique and vital developmental phase to build the foundation of health. During this period, maturation is about the reproductive system, so adolescents must understand reproductive health. According to WHO, to grow and develop with good health, adolescents need some information, including sex education, the opportunity to improve life skills, equality in health services, effective and supportive environment. This community service aims to provide health education to students at PP Wahid Hasyim Bangil. This community service was implemented at PP Wahid Hasyim Bangil on May 21 and 22 May 2022. The target of this community service is students at PP Wahid Hasyim, totaling 12 students. The method used is presentation and discussion in an online conference and a questionnaire to measure students' knowledge about reproductive health in pre and post-webinar. The results of data analysis using Wilcoxon showed that the p-value was 0.04, indicating a difference before and after the webinar on reproductive health. Community service effectively increases students' knowledge about reproductive health, so this event can do routine and make the adolescents community in Islamic boarding schools.

Abstract

Background: The concentration of NaOH in the alkaline lysis method needs to be optimized, especially when isolating DNA for its application in PCR which has not been maximized using the Candida albicans fungus which has a complex cell wall and is difficult to break. Thus, this optimization is expected to increase the efficiency of the alkaline lysis DNA isolation method in fungal species. Objective: The purpose of this studied was to determine the effect of optimizing the concentration of NaOH in the alkaline lysis method on the quality and quantity of Candida albicans DNA. Materials and Methods: This researched is experimental by performed DNA isolation used alkaline lysis method used NaOH concentrations of 1.5 N, 1.75 N, and 2.0 N and controlled (NaOH 0.2 N). Calculation of DNA quality and quantity using a nanodrop spectrophotometer. Results: The results showed the ordered of obtained the high to low DNA quality (A260/280) was NaOH concentration 2.0 N>1.75 N>1.5 N>0.2 N (1.86±0.44-0.95±0.18) while for DNA quantity was NaOH concentration 2.0 N>1.75 N>1.5 N>0.2 N (187.7±58.3-9.6±3.5 ng/?l). Conclusion: There is an effect of optimizing NaOH concentration on the lysis of fungal cell walls in the alkaline lysis method, namely increased the purity valued and concentration of sample DNA isolation results gradually. The use of 2.0 N NaOH concentration produced the best quality and quantity of DNA, namely 1.86±0.44 for DNA quality and 187.7±58.3 for DNA quantity.

Abstract

Background: DNA was an essential component in organisms that regulates biological activity. DNA analysis began with isolating pure, highly concentrated DNA, often used the CTAB method to isolate DNA from bacteria. DNA quality affected by incubation timed and temperature as well as its purity. The right incubation timed could optimize the degradation of proteins in the cell wall and maximize the released of DNA from the cell. DNA purity is good if the 260/280 absorbance ratio valued is 1.8-2.0. Objectives: This studied aimed to obtained the highest concentration and quality of DNA by modifying the incubation time of CTAB used Staphylococcus aureus DNA. Materials and Methods: This studied uses quantitative methods with the typed in experimental researched. The isolation stage included cell lysis with CTAB buffer and proteinase-K, purification stage with chloroform: isoamyl alcohol (24:1), DNA precipitation stage with isopropanol. Results: The average result of DNA purity obtained from the incubation treatment for 10 minutes was 1.89, for 20 minutes incubation was 1.46, and for 30 minutes incubation was 1.23. The average DNA concentration obtained from the incubation treatment for 10 minutes was 153 ng/µL, for 20 minutes incubation was 78.2 ng/µL, and for 30 minutes incubation was 26 ng/µL. Electrophoresis results at 10 minutes incubation time clearly visible DNA bands, 20 minutes incubation time DNA bands looked faint, and 30 minutes incubation time is not visible DNA bands. Conclusions: Based on the results of the studied, modification of CTAB incubation time with an incubation time of 10 minutes is the result of the highest concentration and quality of DNA using Staphylococcus aureus DNA.

Abstract

The Snakehead fish contains high protein that was mostly used as a treatment during healing process in either invasive or passive surgery. Therefore, it could be developed as well as a mixture of wounds dressing for skin healing. The aim of this study was to know the effect of combination of concentrations between chitosan and collagen of Snakehead fish (Channa striata) skin and scales on fungal total number recovered from the composite. Snakehead fish skin and scales was treated by soaking in 2% HCl solvent for 48 hours to obtain collagen from its filtrate. Filtrate obtained continued to neutralize with 1 M NaOH until soluble collagen appeared. Collagen obtained in this study then mixed with 2% chitosan liquid to make wound dressing in various concentrations group. Combination of colagen mixture in this study were chitosan liquid only as control, 25% collagen-75% chitosan (C1), 50% collagen-50% chitosan (C2) and 75% collagen-25% chitosan (C3). The study results showed that on Control, C1 and C2 group there was no fungal contamination, but on C3 group there was contamination of fungal with total counting about 2,43 x 103 CFU. Based on statistically test showed that there was discrepancy for each group with p-value was 0,02 (p<0,05).

Abstract

The program "Empowering the Community of Simo Angin-Angin Village, Sidoarjo through the Use of Natural Materials to Achieve Health Independence" was implemented at the Simo Angin-Angin Village Hall, Krian District, Sidoarjo Regency in May-June 2023. This Community Empowerment Program (Community Service) carries a preventive and consists of a series of activities, including counseling about the use of natural materials based on the recommendations of the Koran, considering that the majority of Simo Angin-Angin village residents are Muslim. This activity also includes training in farming using the hydroponic method. The aim of this program is to strengthen health independence in Simo Angin-Angin Village in order to improve collective welfare. The Community Service program in Simo Angin-Angin Village was attended by 22 participants, consisting of health cadres and the general public. The implementation is divided into 2 stages, namely counseling regarding the Black Cumin herbal plant and training regarding vertical hydroponic procedures, as well as the use of plastic bottles with the Wick system. The evaluation results showed that participants' understanding was very good, with 95% of them achieving a score of 100, while 5% obtained a score of 80. The evaluation results of participant satisfaction with the implementation of the extension showed very good scores for the topic material and delivery of the material by resource persons on herbal plant education and hydroponic training. This Community Service activity provides skills for the community to grow crops on narrow land using a hydroponic system

Abstract

Background: Multiplex PCR techniques are used to detect multiple pathogens at the same time simultaneously. In order to achieve the specificity and sensitivity of detection, it is important to optimize the multiplex PCR method properly. Developing suitable primers and optimizing PCR temperature to boost particular genes from pathogens are key factors for this optimization. Methods: For the simultaneous detection of Escherichia coli and Salmonella enterica in food samples, this research aims to develop a set of primers for multiplex polymerase chain reaction (PCR). Using Primer-BLAST software, the study utilizes specific primer designs for the phoA gene (Eschericia coli) and invA gene (Salmonella enterica). DNA isolation has confirmed successful extraction from the two bacterial samples. PCR was performed under different conditions, including Singleplex and Multiplex PCR, using two annealing temperatures of 53oC and 50oC. Results: The results showed that this method can effectively amplify target genes and indicate their specificity and reliability at both temperature levels. Given these results, it has successfully conducted multiplex PCR using the built primer pairs. Both annealing temperatures of 50oC and 53oC can be used to perform multiplex PCR to detect E. coli and Salmonella enterica in one PCR reaction. Conclusion: Through this research, we have created a new set of Multiplex PCR Primers and an optimized multiplex PCR technique for the simultaneous detection of E coli and Salmonella enterica in food samples. The rapid and simultaneous screening of E. coli and S. enterica, which contributes to improved food safety measures and pathogen detection in the food industry, is promising with this optimized PCR approach.
