Abstract

Hyperlipidemia is a major risk factor for cardiovascular disease, with a continuously increasing prevalence worldwide. Turmeric leaves (Curcuma longa L.) contain various bioactive compounds with potential antihyperlipidemic properties through modulation of metabolic biomarkers such as acyl ghrelin, growth hormone (GH), and AMP-activated protein kinase (AMPK). To analyze the effects of ethanolic turmeric leaf extract on acyl ghrelin, GH, and AMPK levels in hyperlipidemic rats induced by a high-fat diet (HFD) and propylthiouracil (PTU) using combined in silico and in vivo approaches. This laboratory experimental study used Federer's formula to determine the sample size, resulting in 36 male Wistar rats randomly divided into six groups: normal control (K1), hyperlipidemia control receiving HFD+PTU (K2), positive control receiving HFD+PTU+simvastatin 0.9 mg/kg body weight (BW) (K3), and three treatment groups receiving HFD+PTU plus ethanolic turmeric leaf extract at doses of 300 mg/kg BW (K4), 600 mg/kg BW (K5), and 1200 mg/kg BW (K6). In silico molecular docking was conducted to evaluate interactions between active compounds and target proteins. Measured parameters included lipid profiles, aortic histopathology, and levels of acyl ghrelin, GH, and AMPK assessed using ELISA.  Phytochemical screening confirmed the presence of five classes of bioactive compounds in the extract. In silico analysis identified rutin as the principal compound, exhibiting strong binding affinities to acyl ghrelin (−10.6 kcal/mol), growth hormone (−8.0 kcal/mol), and AMPK (−9.3 kcal/mol). The 300 mg/kg BW dose demonstrated the most pronounced lipid-lowering effects, with significant reductions in total cholesterol (210.50±2.74 to 181.67±6.98 mg/dL), triglycerides (246.83±8.47 to 179.17±8.80 mg/dL), and LDL (96.33±5.68 to 70.17±8.11 mg/dL), alongside an increase in HDL (65.00±3.74 to 75.67±1.97 mg/dL) (p<0.001). Histopathological analysis revealed reduced aortic wall thickness (192.59±6.99 μm) and foam cell count (24.17±4.31) in this group. Acyl ghrelin levels differed significantly among groups (p<0.001), with the hyperlipidemic control showing the lowest levels. The 600 mg/kg BW dose optimally increased AMPK (10.33±0.95 ng/mL) and GH (1.96±0.15 ng/mL) compared with the hyperlipidemic control (p<0.05). Ethanolic turmeric leaf extract exerts significant antihyperlipidemic effects by improving lipid profiles, attenuating atherosclerosis, and modulating metabolic biomarkers including acyl ghrelin, GH, and AMPK. A dose of 300 mg/kg BW was most effective for lipid profile improvement and atherosclerosis reduction, while 600 mg/kg BW was optimal for hormonal biomarker modulation.

Abstract

Hyperlipidemia is a major cardiovascular risk factor with increasing global prevalence. Turmeric leaves (Curcuma longa L.) contain bioactive compounds with potential antihyperlipidemic properties through modulation of leptin and HMG-CoA reductase. To analyze the effects of ethanolic turmeric leaf extract on leptin, HMG-CoA levels, and blood vessel histopathology in hyperlipidemic rats using in silico and in vivo approaches. Thirty-six male Wistar rats were divided into six groups: normal control (K1), hyperlipidemia control (K2), simvastatin 0.9 mg/kg BW (K3), and turmeric extract at 300 (K4), 600 (K5), and 1200 mg/kg BW (K6). Hyperlipidemia was induced with high-fat diet and propylthiouracil. In silico molecular docking evaluated compound-protein interactions. Phytochemical screening confirmed five bioactive compound classes. In silico analysis showed rutin with strong binding affinities to leptin (−10.2 kcal/mol) and HMG-CoA reductase (−9.8 kcal/mol). The 300 mg/kg dose significantly improved lipid profiles, reduced aortic wall thickness (192.59±6.99 μm), foam cells (24.17±4.31), and HMG-CoA levels (3.45±0.42 ng/mL) (p<0.001). The 600 mg/kg dose optimally restored leptin (485.67±62.34 pg/mL). Ethanolic turmeric leaf extract exerts significant antihyperlipidemic effects by improving lipid profiles, attenuating atherosclerosis, and modulating leptin and HMG-CoA reductase.

Abstract

Background Recurrent Bell’s palsy is a rare condition that can affect the ipsilateral or contralateral to the affected side of the primary attack. There is still no consensus on treatment guidelines for recurrent Bell’s palsy. Therefore, we report a case of idiopathic recurrent contralateral facial palsy treated with autologous peripheral blood stem cell (PBSC) transplantation. Case Presentation A 72-year-old man presented with the sudden onset of right facial weakness, which was contralateral to the affected side of the first episode of facial palsy. The weakness had progressed with difficulty in closing the right eye, drooling the water from the right side of the mouth, and chewing impairment which was consistent with House Brackmann grade IV or moderately severe facial nerve grading system. He had exposure to cold stimulation, and a history of hypertension. He was given G-CSF 10 ug/kg/day for 5 days to mobilize stem cells, followed by apheresis on the 5th day. He did not receive acupuncture treatment. He had a complete recovery in 3 weeks with House Brackmann grade I or normal function of facial nerve. Conclusion Peripheral blood stem cell transplantation had a beneficial effect on the complete recovery of this recurrent Bell’s palsy and provides a treatment strategy in patients with unilateral facial palsy.

Abstract

Background: Diabetes mellitus (DM) sufferers in the world and even inIndonesia are expected to continue to increase, especially type 2 DM(DMT2). DMT2 often occurs in people who are obese, the cause of obesityis often consuming foods that contain a lot of fat. Hyperlipidemia plays arole in the pathogenesis of pancreatic β-cell dysfunction.Research method: This study is an experimental laboratory study with apretest-posttest only controlled group design, using male white rats(Rattus norvegicus) with a sample size of 30 divided into 6 groups, namelythe normal group, the negative control group, the positive control group,the group given a high fatty diet (HFD). The independent variable is HFD,while the dependent variable is blood sugar levels. Data were analyzedusing the Anova test.Results: Giving HFD of 4 ml/day for 4 weeks has not been able to increaseKGD to hyperglycemic, this is thought to be because the HFD was notgiven for long enough so that insulin resistance had not occurred in thetest animals.Conclusion: Based on the results of the study, it was concluded that givingHFD for 4 weeks still could not increase KGD in male white mice tobecome hyperglycemic
