Abstract

Endometriosis is a complex, chronic, and multifactorial gynecological disorder characterized by the presence of endometrial tissue outside the uterine cavity, leading to chronic inflammation, pelvic pain, and infertility. One of the major challenges in diagnosing endometriosis is the delay in detection due to nonspecific symptoms, along with the reliance on invasive laparoscopic procedures as the gold standard for confirmation. This condition highlights the urgent need for the development of safer, faster, and more accurate diagnostic methods through the identification of non-invasive biomarkers. This review aims to examine recent advances in various biomarker approaches, including hormonal, immunological, genetic, proteomic, metabolomic, and microbiome-based aspects, which contribute to the pathogenesis of endometriosis and hold potential as diagnostic tools as well as predictors of therapeutic response. The findings indicate that a multi-omics approach integrating genetic data, metabolite profiles, plasma proteins, and microbiota composition has strong potential to generate biomarker panels with higher sensitivity and specificity. Therefore, the development of multi omics-based biomarkers is expected to support early diagnosis and more personalized therapeutic strategies. This advancement represents a crucial step toward the implementation of precision medicine, ultimately improving patients’ quality of life through more effective and targeted management.

Abstract

Mesenchymal stem cells (MSCs) have gained significant interest for their potential application in repairing damaged tissues through immunomodulatory functions. Vitamin D has been shown to regulate both the innate and adaptive immune systems. Active forms of Vitamin D, such as 1,25-dihydroxyvitamin D3 (1,25(OH)2D3) are known to influence the action of mesenchymal stem cells (MSCs) in several mechanisms, such as stimulating their proliferation and osteogenic differentiation, as well as tissue regeneration. This study aimed to investigate the effects of vitamin D supplementation on the biological properties and expression of cytokines and growth factor genes in MSCs isolated from the human umbilical cord (UC). MSCs were isolated from Wharton’s jelly (WJ-MSC) of UC, cultured, and supplemented with various concentrations of vitamin D3. The Cell Counting Kit-8 (CCK-8) assay was used to measure cell viability, and a scratch wound healing assay was conducted to evaluate the migration capacity of MSCs. The mRNA expression levels of vascular endothelial growth factor (VEGF), tumor necrosis factor (TNF)-α, and interleukin (IL)-6 were quantified using reverse transcription-polymerase chain reaction (RT-PCR). This study showed that supplementation with 50 nM of vitamin D3 for 48 h significantly increased the viability and migratory capacity of MSCs. Furthermore, vitamin D supplementation significantly decreased the mRNA levels of TNF-α and VEGF but did not affect IL-6 gene expression compared to the control group. These findings suggest that vitamin D supplementation can enhance the biological characteristics and modulate the expression of key immunomodulatory factors in MSCs, potentially improving the effectiveness of MSC-based therapies.

Abstract

Malnutrition, especially among children under five, remains a significant public health concern in Indonesia, including Riau Province. One potential effort to address this issue is the utilization of traditional fermented foods from Kuantan Singingi Regency, such as pekasam, sarobuong, puduong, and cangkuok semaung. These foods are rich in nutrients, including protein, carbohydrates, amino acids, minerals, and probiotics beneficial to health; however, they are on the verge of extinction.This community service program aimed to revitalize traditional Kuansing fermented foods through a science and technology-based training approach. The program implementation involved video demonstrations of traditional food preparation, distribution of pocket books, and interactive discussions with health cadres, as well as pregnant and breastfeeding mothers, and mothers with toddlers.The results showed high enthusiasm among participants, demonstrated by their active involvement in discussions and question-and-answer sessions. The training not only improved participants’ knowledge but also fostered awareness and a sense of ownership of local food, while simultaneously opening opportunities for small-scale household businesses.
