Abstract

Overexpression of Human Epidermal Receptor-2 (HER-2) is the cause of the development of breast cancer, so HER-2 can be a target for anticancer therapy. Trastuzumab is the drug of choice in inhibiting the HER-2 receptor and tyrosine kinase, but the weakness of this drug is that drug resistance can occur. Naringin has potential as a breast cancer therapy. The research aims to determine the inhibiting overexpression of the HER-2 receptor by naringin using in silico method. Molecular docking has been carried out by optimizing naringin and trastuzumab, preparing the HER-2 protein target (PDB ID 3PP0), validating the molecular docking protocol, and docking naringin and trastuzumab on HER-2. The research produced a binding energy of -8,66 kcal/mol, while the binding energies of the original ligand and trastuzumab to HER-2 were -9,94 kcal/mol and -5,87 kcal/mol, respectively. The binding energy shows that naringin has potential as an anti-breast cancer agent as indicated by its stronger affinity for HER2.

Abstract

Unpleasant sensory and emotional experiences arising from injury or damage to organs are often referred to as pain. Synthetic drugs are often used to treat the pain but often cause harmful side effects. Cat whisker leaves (Orthosiphon stamineus Benth) and celery leaves (Apium graveolens) are often an alternative choice of treatment for pain because they have potential as analgesics. The purpose of this study was to determine the analgesic effect of a combination of ethanol extracts of cat's whiskers and celery leaves on male mice (Mus musculus) using acetic acid as an inducer with a combination dose of 1:1, 1:5 and 5:1. The research is experimental using the writhing test method followed by data analysis including normality, homogeneity, one-way anova and LSD post hoc tests. The results obtained from the study showed that the analgesic effectiveness of the three doses of the combination was greater than the positive control of paracetamol, with a ratio of 2.9 times for combination I, 2.7 times for combination II and the largest comparison was in combination III which was 3 times with the highest percent protection of 74.64%. So it can be concluded that the combination of ethanol extracts of cat whisker leaves and celery leaves has an effect as an analgesic and can inhibit pain better than paracetamol.

Abstract

Belimbing wuluh leaves (Averrhoa bilimbi L.) contain flavonoids, saponins, and tannins that can be used as antibacterials in acne treatments. Acne treatment requires good delivery so that it can penetrate into the epidermis. A preparation that can deliver active substances directly into the epidermis is a microneedle patch (MN) preparation. This study aimed to formulate a microneedle acne patch of ethanol extract of A. bilimbi leaves. A. bilimbi leaves were extracted using maceration method. MN preparation base was optimized using different base combinations namely Na-CMC:PVA, HPMC: PVA and PVP:PVA until the ideal MN base was obtained. The MN preparations obtained were tested for physico-chemical properties which included macroscopic tests, weight uniformity, drying shrinkage, pH, folding power and moisture absorption. MN optimization results showed that the combination of PVP and PVA formed a flexible MN base and needle. Formulation was then continued by varying the concentration of extracts in the MN. The results of the physical and chemical properties test of the MN preparation formula showed flexible characteristics, homogeneous and no bubbles in MN, yellow to blackish green in color, weights ranging from 0,063-0,163 g, drying shrinkage of 3.349-4.513%, pH in the range of 5.61-7.15, folding power >300x, and moisture absorption of 6.472%-8.33%. The conclusion of the study showed that the MN preparation formulas of A. bilimbi extract, namely F1, F2, F3 and F4 meet the requirements of good physico-chemical MN preparations.

Abstract

Free radicals damage cellular structures and accelerate the degradation of important elements in the skin, thereby accelerating premature aging. Antioxidants in skin care products can shield cells from harm brought on by oxidative stress. Pegagan leaves (Centella asiatica (L.) Urban) are a natural source of antioxidants. Pegagan leaf ethanolic extract has antioxidant properties and can perhaps be applied in cosmetic products like peel-off mask gel. The objective of this research is to measure the antioxidant behavior of a mask that peels off gel composed of ethanolic extract of Pegagan leaves (Centella asiatica (L.) Urban). This research applies a one-shot case study laboratory experimental method. Pegagan ethanolic extract was utilized in concentrations of F1 (0.5% ) F2 (1%) and F3 (1.5%). The level of antioxidant activity was measured. for three formulae using the DPPH (2,2-diphenyl-1-picrylhydrazyl) technique. Organoleptic, pH measurement, viscosity, homogeneity, spreadability, and drying time are all considered in the physical evaluation of the preparation. Based on evaluation outcomes of the preparations, three formulas met the test requirements with organoleptic test parameters, a drying time of 15-30 minutes, and a pH value between 4.5 and 6.5. For the spreadability test, only F2 meets the test requirements with 5-7 cm, while F1 and F3 do not follow the criteria because the A preparation's spreadability is influenced by viscosity. The thicker the mask gel preparation, the smaller the resulting spreading area. The result examination for antioxidant activity on the ethanolic extract Pegagan leaf showed an IC50 value for F1 of 28.68 ppm, F2 of 38.51 ppm, and F3 of 48.68 ppm. Study shows making the peel-off mask prepared gel Pegagan ethanol extract meets the criteria for a strong antioxidant. The results of this research indicate the quantity of antioxidant function in addition to the outcomes of the physical assessment have an impact on the concentration of pegagan leaf extract in the gel mask that peels off.

Abstract

Lemon and starfruit are fruit plants used in traditional medicineThe purpose of this study was to determine how differences in sugar content affected the way gummy candy was made. This study's methodology included the creation of five different gummy sweets using starfruit juice and lemon. Physical quality assessments, such as organoleptic testing, pH, weight uniformity, elasticity tests, water content tests, and hedonic tests, were also conducted. The study's findings demonstrated that the gummy sweets' flavor became sweeter with increasing sugar content. As can be seen, formula 5 has the maximum sucrose content at 1.5 grams. The conclusion reached is that changes in the amount of sucrose used as a sweetener may affect the preparation's physical quality evaluation test results as well as the respondent's hedonic test evaluation. It was discovered that formula 2 and 4, which included 0.5 grams and 0,2 grams of sucrose, was the one that responders liked the best.
