Abstract

A variety of attempts have been made to reduce the dependence on petroleum raw materials based plastics which are limited supply and difficult to degrade, so it can cause the environment problems. Polyvinyl Alcohol/PVA, one of the water-soluble polymer, has good compatibility with fillers, i.e., nanocellulose addition, so it can resulted a composite films that are environmentally friendly and the mechanical properties close to conventional plastic. The objectives of research was to know the effect of pineapple nanocellulose fibers addition on the improvement of the mechanical properties of composite films based on polyvinyl alcohol. The method for manufactured of composite films using solution casting. A simple in a one-pot process was carried out by mixing of PVA solution in various concentration of pineapple nanocellulose fibers (10% - 50%) and glycerol with 2 levels (0% and 1%). Films observations were tensile strength, elongation, crystallinity, and morphology. The results shows that the addition of nanocellulose fibers 10-40% were effective in increasing tensile strength and elongation, but the higher addition (of up to 50%) resulted in decreased elongation. The addition of glycerol on the composite film tends to lower the tensile strength and elongation. This is supported by the XRD data showed that the addition of nanocelullose was also effective to improve the crystallinity properties of the films, but the properties of crystallinity decreased after adding glycerol. The best mechanical properties of composite film produced by the treatment on nanocellulose 40% addition and without glycerol.

Abstract

Bacillus thuringiensis subsp. aizawai is one of well-known bioinsectiside resources, and utilized in organic farming for the replacement of chemical insecticide. The active compounds produced from fermentation process of B. thuringiensis subsp. aizawai are bacterial spores and protein crystal of δ-endotoxins, which are variously toxic to larvae of the Lepidoptera and Coleoptera. The use of bioinsecticides in Indonesia is still rarely because bioinsecticide marketed in Indonesia is still an import product so that price is relatively expensive. This problem can be overcome with producing bioinsecticide contain active B. thuringiensis using agroindustrial by product as a raw materials. This research compared the bioinsecticide production from several agroindustrial by product in solid fermentation of B. thuringiensis subsp. aizawai. This research aimed to find out the ability of B. thuringiensis subsp. aizawai using substrate combination of carbon source (cassava dregs, pulp of coffee, starch fractions of iles-iles, and sago dregs) and nitrogen source (tofu’s waste, peanut meal, palm kernel cake, and corn hominy) in bioinsecticides production using solid media cultivation. The research consisted of three stages, there were substrates characterization, microorganisms characterization, and bioinsecticides production. The main parameters to select the carbon and nitrogen sources are LC50 value and potential bioinsecticides products. The results showed that the best cultivation of B. thuringiensis subsp. aizawai contained in combination cassava dregs and palm kernel cake blends. This cultivation produced total cell counts 11.2 log CFU/g, viable spore counts 8.9 log CFU/g, LC50 0.04 μg/ ml and potential products 20000 IU/mg. Biopesticide produced can be used to kill Crocidolomia binotalis on cabbage. PRODUKSI BIOINSEKTISIDA OLEH Bacillus thuringiensis subs. aizawai PADA KULTIVASI MEDIA PADAT MENGGUNAKAN LIMBAH AGROINDUSTRI Bacillus thuringiensis subsp. aizawai merupakan salah satu bakteri yang digunakan untuk menghasilkan bioinsektisida dan digunakan pada pertanian organik untuk menggantikan pemakaian insektisida kimia. Komponen aktif yang diproduksi dari proses fermentasi B. thuringiensis subsp.aizawai adalah spora dan kristal protein δ-endotoksin, yang bersifat toksin terhadap larva Lepidoptera dan Coleoptera. Penggunaan bioinsektisida di Indonesia masih jarang karena biasanya berupa produk impor yang harganya mahal. Permasalahan ini dapat diselesaikan dengan memproduksi bioinsektisida menggunakan bahan aktif B. thuringiensis dan hasil samping agroindustri sebagai bahan baku. Penelitian ini bertujuan mengkaji kemampuan produksi B. thuringiensis subsp. aizawai pada beberapa jenis substrat sumber C (onggok, kulit kopi, fraksi pati iles-iles, dan ela sagu) dan sumber N (ampas tahu, bungkil kacang tanah, bungkil inti sawit, dan ampok jagung) dalam memproduksi bioinsektisida dari limbah agroindustri. Penelitian ini terdiri dari tiga tahap, yaitu karakterisasi substrat, karakterisasi isolat dan produksi bioinsektisida. Parameter utama untuk menetapkan sumber karbon dan nitrogen yang tepat adalah nilai LC50 dan potensi produk bioinsektisida. Hasil penelitian menunjukkan bahwa produksi bioinsektisida yang terbaik terdapat pada kultivasi B. thuringiensis subsp. aizawai menggunakan kombinasi onggok dan bungkil inti sawit. Pada kultivasi ini dihasilkan jumlah sel hidup 11.2 log CFU/g, jumlah spora 8.9 log CFU/g, nilai LC50 0.04 μg/ ml dan potensi bioinsektisida 20000 IU/mg. Bioinsektisida yang dihasilkan dapat digunakan untuk membasmi larva Croccidolomia binotalis pada tanaman kubis.

Abstract

Fround Sagu adalah pucuk batang sagu yang masih dibungkus oleh pelepah dan tidak diamnfaatkan oleh industri pengolahan sagu. Fround sagu memiliki kandungan serat dengan kandungan selulosa yang tinggi serta berpotensi dijadikan bahan baku untuk produksi selulase. Enzim selulase diproduksi melalui kultivasi substrat cair frond sagu oleh Aktinomiset. Subtart berupa tepung sagu dan ampas frond sagu, diinokulasi oleh isolat HJ4 (3b) dan HJ5 (4b). Kedua isolat diremajakan dalam medium ISP-4 selama 5 hari, kemudian diinokulasikan ke dalam media tepung frond sagu dan ampas frond dan diinkubasi dalam shaker pada suhu runag selama 9 hari. Kedua isolat Aktinomiset mampu menghasilkan enzim selulase pada kedua substrat dan metode kultivasi. Isolat HJ4 (3b) dan HJ4 (5b) pada perlakuan kultivasi substrat padat ampas frond sagu menghasilkan aktivitas spesifik yaitu endoglukase (CMCase) tertinggi yaitu 0.314 U mg-1 dan 0.294 U mg-1 dan aktivitas spesifik enzim eksoglukanase (FPase) yaitu 0.269 U mg-1 dan 0.258 U mg-1, sedangkan pada perlakuan kultivasi substat padat menggunakan tepung frond sagu dihasilkan aktivitas spesifik endoglukanase masing-masing sebesar 0.258 U mg-1 dan 0.254 U mg-1 serta aktivitas spesifik eksoglukanase 0.205 U mg-1 dan 0.198 U mg-1. Production of Cellulase Enzyme by Actinomycet Using Sago FrondSago frond is the upper part of sago trunk which is still wrapped by leaflet, and is not used by the sago processing industry. Sago frond contains fiber with high cellulose content that could potentially be used by as raw material for cellulase production. Cellulase enzymes were produced through both solid-state and submerged cultivation of sago frond by Actinomicycetes. Two substrates, sago frond flour and pulp of sago fronds, were inoculated by isolate HJ4 (3b) and HJ4 (5b). Both isolates were rejuvenated in Sp-4 medium for 5 days, then were inoculated into substrate of frond flour and hampas, and were incubated in a shaker at room temperature for 9 days. Both Actinomycetes isolates were able to produce cellulase enzymes by using both substrates and cultivation methods. The isolates of HJ4 (3b) and HJ4 (5b) by using pulp and solid-state cultivation produced the highest endoglucanase (CMCase) specific activity of 0.294 U mg-1 and 0.276 U mg-1 and exoglucanase (FPase) substrate specific activity os 0.252 U mg-1 and 0.241 U mg-1, while in the solid-state cultivation and by using sago fronds flour resulted in specific endoglucanase activities which were 0.242 U mg-1 and 0.238 U mg-1 and exoglucanase specific activities 0.192 U mg-1 and 0.185 U mg-1, respectively.
