Abstract

Hypercholesterolemia condition was reported can decrease activities of antioxidant superoxidedismutase, catalase, and glutathione peroxide. This study was conducted to observe the role of seaweed(Eucheuma cottonii) powder in increasing the level of antioxidant cooper, zinc-superoxide dismutase (Cu,Zn-SOD) in kidney tissues of hypercholesterolemic rats. Twenty male Wistar rats were used in this study.Those rats were divided into four groups ; (1) Negative control group (A), (2) and (3) werehypercholesterolemic groups that were treated with 5% (B), and 10% (C) of seaweed powder, and (4)Hypercholesterolemic group as positive control (D). The treatment was carried out for 35 days.Hypercholesterolemia condition (>130 mg/dl), except for group A, was achieved by feeding the rat withcommercial diet containing 1% cholesterol and drinking water ad libitum for 40 days. Serum total cholesterolwas analyzed both before and after treatment. Rat kidneys were taken at the end of treatment, andprocessed by using paraffin embedding standard method. The tissues were then stained usingimmunohistochemical technique to Cu,Zn-SOD. The results showed the seaweed powder decreased serumtotal cholesterol, increased caecum cholesterol, repaired alteration of kidney tissues, and increased thelevel of antioxidant Cu,Zn-SOD in the kidney of hypercholesterolemic rats. Dietary fibers and antioxidantactivity of 10% seaweed powder was better than the of 5%.

Abstract

Antioxidant activity of clove leaf was investigated. The clove leaves (Eugenia aromatica) wereprepared with reflux extraction using methanol, water, and ethanol. Total activities of the extractsand reducing power were measured with thiocyanate method and reducing potential method. Resultshowed that the highest total antioxidant activity was observed in methanol extract. It appearsthat the ability of this extract for partitioning at the interface of emulsion in tested oxidationsystem was the highest among the other extracts, therefore it had the best activity to inhibitoxidation. The type of phenolic compounds of this extract appeared to be responsible for the highestradical scavenging capacity. The same phenomenon occurred for reducing power, methanol extracthad the highest reducing power, thereby suggesting that each extracts comprised different type ofphenol based on different polarity of reflux used for extraction. Total antioxidant activity and thehighest reducing power obtained from methanol extract. Both are closely related to total phenolcontent of the clove leaves.

Abstract

The objective of this study was to observe the effect of functional synbiotics yoghurt (made ofindigenous probiotic + oligofructosaccharides) in rats that were infected with Enteropathogenic Escherichiacoli (EPEC), and to detect its effect on hematological status. A total of 25 male Sprague Dawley rats wereused in this study and divided into five treatment groups: (i) negative-control; (ii) positive-control; (iii)synbiotic yoghurt; (iv) synbiotic yoghurt + EPEC; and (v) conventional prebiotic yoghurt. Yoghurt (109 cfu/mL lactic acid bacteria) was given orally using feeding tube at day one until day 4. At day 8 – day 14animals were infected with EPEC (107 cfu/mL) orally using feeding tube; at day 22 blood samples werecollected for hematology analysis (erythrocytes, hematocrit (Hct), hemoglobulin (Hb), platelets, andleucocytes). Post infection with the EPEC diarrhea was developed in rats both in the control positivegroups and in the synbiotic yoghurt + EPEC group. Animals in the positive-control group had a significantlyhigher thrombocytes and leucocytes counts and Hct compared to that in animals in the other groups(P<0.01). Whereas there were no significant effect on the erythrocytes counts and Hb (P>0.05). The treatmentwith synbiotic yoghurt, in both synbiotic yoghurt only and synbiotic yoghurt + EPEC groups significantlyreduced the platelets and leucocytes counts and Hct of rats

Abstract

The consumption need of soybean that reached up to 2,5-3 million tons per year has made Indonesiaimports this commodity 1,95 million tons from countries that adopt Genetically Modified (GM) soybean.Soybean utilization for tempe producers is about 50% from total its supply. The aim of this research wasto evaluate the safety of tempe made from GM soybean, by a subchronic test approach using male Sprague-Dawley rats. Toxicity evaluation of tempe from soybean GM was done based on EFSA (2011) principalusing animal laboratory. For 90 days, three groups of male rats (each group contain seven rats) werefeeding with tempe flour from GM soybean, tempe flour from non-GM soybean and control group wasfeeding with casein as a protein source. All rats were terminated on day 90,where the blood, liver andkidney were collected. There were no significant differences on liver and kidney weight between treatment.Hematology analysis showed the level of Hb, erythrocyte, leucocyte, trombocyte and hematocryte were notsignificantly different in between group. Serum analysis on SGOT, SGPT, total protein, albumin, globulin,blood glucose, triglyceride, ureum and creatinin level were also showed no difference between groups. As aconclusion, the consumption of tempe derived from GM soybean was safe, and cause no adverse effect onhealth.

Abstract

Lisozim bersifat bakteriolotik dengan cara merusak ikatan glikosida pada dinding bakteri sehingga pelacakan sel penghasil lisozim pada kelenjar ludah sapi perlu dilakukan. Dalam penelitian ini, sel penghasil lisozim diperiksa menggunakan teknik pewarnaan histologi dan imunohistokimia. Hasil penelitian menunjukkan bahwa kelenjar parotis sapi bersifat serous murni. Kelenjar submandibularis bagian asinarnya terdiri atas sel-sel serous dan sel-sel mukous. Sel-sel asinar mukous terlihat lebih dominan jika dibandingkan dengan sel-sel asinar serous. Sel-sel asinar kelenjar parotis sapi bereaksi positif dengan intensitas sedang (++) terhadap pewarnaan imunohistokimia. Sel-sel epitel duktus kelenjar bereaksi sedang (++) sampai dengan kuat (+++). Pola sebaran lisozim yang terdeteksi pada penelitian ini menunjukkan kemiripan dengan hewan mamalia pada umumnya. Sel-sel asinar serous pada kelenjar submandibularis memberikan reaksi positif dengan intensitas rendah (+) sampai dengan sedang (++) dan sel-sel epitel duktus kelenjar dengan intensitas kuat (+++). Lisozim ditemukan pada bagian sel-sel asinar serous dan sel-sel epitel duktus kelenjar parotis dan submandibularis.

Abstract

Glycogen is found at all of body tissue, especially mostly in liver and muscle. The objectives of thisresearch was to evaluate the content of glycogen in liver and muscle of diabetic rats that were treated withextract of tempe. A total of twenty male Spraque Dawley rats of 2 months old were used in this study. Therats were divided into four groups: (1) negative control group (K-), that were not treated extract of tempeand nondiabetic, (2) positif extract of tempe group (ET), that were treated with extract of tempe andnondiabetic (3) positif diabetic group (DM), and (4) diabetic and extract of tempe group (DM+ET). Extractof tempe was orally administered with dose 300 mg/kg bw/day. The treatment was conducted for 28 days.Effect of extract of tempe on body weight of all rats was determined at various time interval at 0, 4, 7, 14,21, and 28 days. At the end of the experiment, all rats were sacrificed by cervical dislocation. Liver andmuscle gastrocnemius were collected for analysis of glycogen level. The result of this study showed thatadministration methanol extract of tempe of 300 mg/kg bw/day can increase body weight, glycogen synthesisin the liver and muscle in normal rats (rat of ET group) and also diabetic rats (rat of DM+ET group). At theend of research, diabetic rats (rat of DM group) were decrease of body weight up to 5.7%. On the rat ofDM+ET group, rat of K(-) group and rat of ET group were increase of body weight of 5.7%, 19.3% and 20.3%,respectively. Glycogen level both liver, and muscle at rat of ET group and rat of DM+ET group wereincrease each of 9.29%, 2.2% in liver and 18.27% and 4.02% in muscle. Glycogen level at rat of DM groupwere decrease up to 42.5% in liver and 31.6% in muscle.

Abstract

High level of blood glucose is an indicator for diabetes mellitus (DM) condition. The condition iscaused by low level of insulin secretion or impairement of insulin receptor. The number of DM patientincreases every year. The World Health Organization reported that the number of DM patient in Indonesiawas the 4th highest in the world, after following China, India, and the United States of America, respectively.This study was conducted to analyze the effect of sea cucumber (Holothuria scabra J) on the profile ofantioxidant copper, zinc-superoxide dismutase (Cu, Zn-SOD) in the pancreatic tissues of diabetic rats. Atotal of 25 male white rats (Sprague Dawley) were used in this study. They were divided into five groups;(1) negative control (KN), (2) positive control, diabetic rats (KP), (3) diabetic rats treated with hydrolyzatedprotein of sea cucumber (HDL), (4) diabetic rats treated with concentrated protein of sea cucumber (KST),and (5) diabetic rats treated with isolated protein of sea cucumber (ISL), respectively. Diabetic conditionwas obtained by alloxan injection 110 mg/kg bw. The treatments were done for 28 days. At the end oftreatment period, the rats were sacrificed and pancreatic tissues were collected and fixed in Bouin solution and then processed to paraffin embedding standard method. The tissues were then stained withimmunohistochemical staining techniques using monoclonal antibody of Cu, Zn-SOD. The results showedthat treatment of HDL, KST, and ISL of sea cucumber (Holothuria scabra J) increased the content ofantioxidant Cu, Zn-SOD either in Langerhans islets and acinar cells of pancreatic tissues-diabetic rats.The HDL of sea cucumber treatment gave the best effect in increasing the antioxidant content of Cu, Zn-SOD in pancreatic tissue of diabetic rats.

Abstract

Acehnese people uses Cissus quadrangula (CQ) Salisb stem traditionally for treatment of variousbone disease. can differentiate into many different cell types such as osteoblast, adipocyteand chondrocytes.A study was conducted to determine the potential uses and the optimal dosage of C. quadrangula(CQ)extractin increasing the proliferation and differentiation of Mesenchymal stem cells derived from bone marrowof rat osteocytes. Mesenchymal stem cells were isolated from femoral and tibial rat bone marrow. Thecells were cultured according to the experimental group consisting of five the treatment groups each ofwhich has 4 replication. The cells were cultured in modified Dulbecco’s modified eagles’s medium (mDMEM).Control were cultured in medium without Cissus quadrangula Salisb stem extract whereas the treatmentgroup were cultured in medium with k 0,1 mg/mL, 0,3 mg/mL, 0,6 mg/mL, dan 0,9 mg/mLCissus quadrangulaSalisb stem extract. The level of the cell proliferation was determined by populationdoubling time (PDT) method. Cell differentiation was determined by counting cells and determining the diameter of osteoblastdan osteocytes.The result showed that CQ stem extract reduced PDT valuesignificantly (P<0,01) ascompared to those of control group. This showed that CQ stem extract increased the rat bone marrow stemcells. The number of h osteoblast in control group were significantly lower than those in CQ stem extracttreatment groups. The highest osteocyte population was observed in 0,3 mg/mL CQ extract treatmentgroup. The CQ stem extract can increase differentiation and proliferation of rat bone marrow masenchymalstem cells into osteoblastand osteocytes with the optimal dose of 0,3 mg/mL.

Abstract

Hyperglycemia in diabetes mellitus results in stress oxidative condition. This study aims to analyze the antioxidant defence in pancreatic experimental-diabetic rats treated with a combined extract of Cajanus cajan leaf and ginger, using an immunohistochemical technique for copper, zinc – superoxide dismutase (Cu,Zn-SOD). A total of 25 male Sprague Dawley rats were used in this study. The rats were randomly divided into five groups; normal control group (NC), diabetic group (DM), two groups DM treated with a different dose combination of Cajanus cajan leaf extract and ginger extract (DME and DMF), and DM treated with glibenclamide (DMG). This study used 110 mg/kg BW alloxan induction to obtain diabetic conditions, and the treatments of the extract were conducted for 28 days. The pancreatic tissues were processed using the paraffin standard method and an immunohistochemical technique using monoclonal antibody Cu,Zn-SOD. The results showed that a combination of Cajanus cajan leaf extract and ginger extract lowered the level of blood glucose, increased body weight, and increased the level of Cu,Zn-SOD content in pancreatic tissues of experimental-diabetic rats. This study concluded that combining Cajanus cajan leaf extract and ginger extract increased antioxidant defence in pancreatic organs of experimental-diabetic rats.

Abstract

Hypercholesterolemia is characterized by levels of total cholesterol and low-density lipoprotein (LDL) above normal. Hypercholesterolemia is a major risk factor for cardiovascular disease. This study was aimed to explore and analyze various bioactive compounds to manage hypercholesterolemia. Efforts to overcome the condition of hypercholesterolemia are to reduce the consumption of a diet rich in cholesterol and calories, and consuming contemporary hypercholesterolemic drugs such as statins, as well as by regulating the body’s cholesterol metabolism with various antihypercholesterolemic bioactive compounds. Various findings of antihypercholesterolemic bioactive compounds have various sources and effects on cholesterol profiles. Bioactive compounds can be classified according to their mechanism of hypocholesterolemic action, which are inhibition of cholesterol absorption, inhibition of cholesterol biosynthesis, increased cholesterol excretion, increased cholesterol reverse transport, and increased cholesterol catabolism. Several bioactive compounds are involved in a dual mechanism. The development of these various bioactive compounds can be used as an alternative treatment for hypercholesterolemia since contemporary medicines have proven to have many side effects.
