Abstract

Mangrove Sonneratia alba memiliki potensi sebagai sumber antioksidan yang telah dimanfaatkan oleh berbagai masyarakat berpesisir di Indonesia. Tujuan dilaksanakannya penelitian ini adalah menentukan dan menganalisis potensi aktivitas antioksidan pada kulit batang dan daun mangrove Sonneratia alba di tanjung carat, Kabupaten Banyuasin, Sumatera Selatan. Metode ekstraksi menggunakan pelarut tunggal yaitu etanol dengan melakukan maserasi 48 jam. Analisis antioksidan menggunakan metode DPPH di mana nilainya didapatkan dari persentase inhibisi yang dikonversi melalui tabel probit dan log konsentrasi. Berdasarkan hasil penelitian menunjukkan bahwa kandungan Potensi aktivitas antioksidan pada ekstrak daun S. alba maserasi 1 dan 2 menunjukkan nilai yang lebih kuat dengan didapatkan nilai 20,27 dan 18,62 ppm, sedangkan ekstrak kulit batang S. alba didapatkan nilai sebesar 38,24 dan 22,96 ppm. Baik ekstrak daun dan kulit batang S. alba sama-sama memiliki potensi antioksidan yang sangat kuat.Kata kunci : antioksidan, DPPH, Mangrove, radikal bebas, Sonneratia alba

Abstract

Mangrove is one of the plants that has the potential to be developed into medicinal plants. However, further research is needed to prove scientifically the content of secondary metabolites in it. This study aims to identify the secondary metabolites contained from leaves and roots in Avicennia alba, Rhizopora apiculata and Sonneratia alba. The leaves and roots of mangroves extracted by maceration using ethyl acetate. The secondary metabolites contained in mangrove samples obtained by doing phytochemical tests through color tests. Phytochemical test results showed that secondary metabolites contained in mangrove Avicennia alba are flavonoids, steroids/triterpenoids, saponins, and tannins/phenols in leaf samples, whereas in its root samples it contains flavonoids and steroids/triterponoid compounds. Sonneratia alba contains flavonoids, steroids/teriterpenoids, saponins and tannins/phenols in its leaf samples, while the root samples contain flavonoids, steroids/teriterpenoids and tannins/phenols compounds. The leaf samples of mangrove Rhizopora apiculatacontains flavonoid and steroid/triterpenoid compounds whereas in its root samples contains flavonoids, steroids/triterpenoids, saponins as well as tannins/phenols.

Abstract

S. alba is a mangrove that is commonly found on the east coast of Banyuasin. Endophytic fungus can produce a functional compound e.g antibacterial. The purpose of this study is to determine the type, the increase in diameter of colonies and the antibacterial potential in endophytic fungus of S. alba. The research method used were by the isolation of endophytic fungus from leaf, stem and root, the identification of endophytic fungus, the measurement of colony diameter in endophytic fungus and antibacterial activity test using paper disc method. The results of this study found the two genera of endophytic fungus with four species of Aspergillus sp1., Aspergillus sp2., Paecilomyces sp1., and Paecilomyces sp2. The increase in diameter for each colony of endophytic fungus shows the difference every day. Aspergillus sp1. The exponential phase has been visible on the third day with an average diameter increase of 33.70 mm / day. The exponential phase of Aspergillus sp2 was visible on the third day with an diameter increase of 37.74 mm/day. In the endothelium fungi Paecilomyces sp1, the exponential phase was visible on the third day to the seventh day with diameter increase of 13.57 mm/day to 38.07 mm/day. Exponential phase of Paecilomyces sp2. was visible on the third day to the sixth day with diameter increase of 25.25 mm/day to 75.21 mm/day. These four types of endophytic fungus are resistant to bacteria E. coli and S. aureus. Aspergillus sp2. endophytic fungus has the highest inhibitor potency of 14.49 mm in E. coli bacteria. The endophytic fungus Paecilomyces sp2. inhibits bacterial growth of S. aureus with the highest inhibitory potency of 11.61 mm.

Abstract

Mangrove memiliki beberapa senyawa yang mampu dijadikan sebagai antibakteri, salah satunya jenis mangrove Rhizophora apiculata. Sampel mangrove jenis R. apiculata diambil dari Tanjung Api-api, Kabupaten Banyuasin, Sumatera Selatan. Tujuan penelitian ini untuk menentukan potensi senyawa bioaktif R. apiculata sebagai antibakteri dan menentukan (KHM) dari ekstrak mangrove terhadap bakteri S. aureus dan E. coli. Metode penelitian mencakup pengambilan sampel mangrove dari bagian akar, batang dan daun, pengeringan dan penghalusan, maserasi dan ekstraksi dengan menggunakan pelarut metanol, uji aktivitas antibakteri, uji (KHM) dan analisis data menggunakan Beda Nyata Jujur (BNJ). Hasil penenlitian menunjukkan mangrove R. apiculata memiliki aktivitas antibakteri paling tinggi 23,9 ± 21,3 mm untuk bakteri E.coli sedangkan bakteri S. aureus memiliki aktivitas antibakteri 14 ± 10,8 mm. Nilai KHM ditunjukkan pada semua ekstrak R. apiculata terhadap bakteri E.coli pada konsentrasi 250 ppm 6,08 ± 6,01 mm dan konsentrasi 2000 ppm batang 6,11 mm.

Abstract

Studi ini dilakukan untuk menentukan kultur spesies dinoflagelata yang menyebabkan red tide di perairan Pantai Lido, Johor Bahru pada Juli tahun 2002. Kultur klon dibuat dalam medium ES-DK pada suhu 26_C dan siklus 14:10 jam terang gelap. Sel-sel yang dikultur berbentuk bulat bujur dengan panjang 13-22 μm dan lebar 12-19 μm. Isolat yang dikultur dapat hidup pada salinitas 10-35 PSU, sekalipun pertumbuhan optimumnya adalah 20-25 PSU. Isolat menggunakan amonia dan nitrat sebagai sumber nitrogennya dengan kepadatan tertinggi dalam kultur adalah sekitar 5.264 sel mL−1

Abstract

Isolation of endophytic fungus is the growth of endophytic fungus on new media to obtain the type of endophytic fungus. One of the mangrove plants containing endophytic fungi is Avicennia marina mangroves taken from Payung Island of South Sumatera. This study aims to isolate and identify the type of endophytic fungus, measure the growth rate of isolate diameter and know the antibacterial activity. Methods of this study include identification, growth rate measurement of isolate diameter for seven days and endophytic fungal activity test as antibacterial using Paper Disc method. The results of this study obtained five isolates with three types of endophytic fungi from each root part, stems and leaves of Aspergillus ochraceus, Aspergillus flavus and Aspergillus niger. The lag phase of these three fungi occurs from day of planting until the first day for one day, the exponential phase from the first day until the sixth day for five days, and the stationary phase on the sixth day to the seventh for one day for Aspergillus ochraceus and Aspergillus niger fungi while for Aspergillus flavus fungi yet occur stationary phase. The highest antibacterial activity with inhibit zone was shown by A. flavus fungal isolates ranging from 13,79 ± 1,26 mm for S. aureus and 11,86 ± 1,48 mm for E. coli bacteria, whereas the lowest inhibition zone was indicated by fungal isolates A. ochraceus 8,01 ± 0,45 mm for S. aureus bacteria and 9,34 ± 1,25 mm for E. coli bacteria.

Abstract

Gastropoda is one of the potentially most common biotas found in mangrove ecosystems and as bioactive compounds that have not been widely studied. Gastropod species Littorina scabra and Terebralia sulcata were found in the Payung Island. This research aimed to determine and compare the potential toxicity between two species of gastropod extract of L. scabra and T. sulcata. This research was conducted in February 2017. The research procedure included sampling and sample preparation, extraction, and toxicity test that Brine Shrimp Lethality Test (BSLT) method. Toxicity test showed LC50 415.58 μg/mL L. scabra while LC50 value from T. sulcata equal to 565.52 μg/mL. From the comparison, the toxicity value of L. scabra gastropod was higher than the T. sulcata type.

Abstract

Studi ini dilakukan untuk memastikan identitas dinoflagelata yang menyebabkan ledakan algae di perairan Pantai Lido, Johor Bahru pada Juli tahun 2002. Kultur klon berhasil ditumbuhkan dalam medium ES-DK pada suhu 26_C dengan siklus pencayaan 14:10 jam terang gelap. Pengamatan spesies dilakukan melalui mikroskop cahaya dan mikroskop elektron (SEM). Sel-sel yang dikultur berbentuk bulat bujur dengan ukuran panjang 13-22 m dan lebar 12- 19 μm. Keseluruhan permukaan valva diliputi oleh duri-duri yang pendek dan berpangkal lebar. Duri-duri tersebut tersebar secara merata pada permukaan sel. Satu duri apikal yang pendek juga ditemukan pada sel. Kolar periflagelanya bercabang. Gelang interkaleri berjalur melintang dan sebagian sel terdapat pedunkel. Ciri-ciri morfologi tersebut menunjukkan bahwa spesies ini adalah Prorocentrum minimum atau Prorocentrum balticum. Namun hasil pengamatan tersebut lebih cenderung sebagai sel P. minimum karena bentuk sel yang tidak simetri.

Abstract

Actinomycetes is one type of endophytic bacteria that lives in the mangrove plant tissues. Bioactive compound which form Actinomycetes can be use as potential medicine in medic industry and pharmacy sector. This study aimed to isolate and identify the kinds of the Actinomycetes bacteria at mangrove Rhizophora apiculata from Tanjung Api-api waters, South Sumatera. This research method included measurement of environmental parameters, mangrove sampling, isolation, characterization and identification of bacteria. The result of research found two isolates of Actinomycetes in the lower roots (AB T4) and leaf (Leaf T2). The isolates code AB T4 had characteristics of moderate colony size, circular shape, entire edge, convex elevation, white colony color, had hyphae, rod-shaped cells, non motile, aerobic, gram positive, negative for oxidase, indole and gelatin, positive for catalase, citrate, urea, ornithine, aesculin, galactose, glucose and trehalose, OF oxidative and fermentative. While the isolates code Leaf T2 had characteristics of moderate colony size, irregular shape, undulate edge, raised elevation, creamy white colony color, had hyphae, gram positive, rod-shaped cells, non motile, aerobic, positive for catalase, ornithine, aesculin, celobiosa, glucose, lactose, melibiosa, sorbitol and trehalose, negative for oxidase, citrate, urea, gelatin and indole, OF oxidative and fermentative. Both isolates AB T4 and Leaf  T2 were identified as Nocardia genus in lower root samples (AB T4) and Actinomadura genus in leaf samples (Leaf T2).

Abstract

The Sungsang waters are often used by the surrounding community for water transportation, fishing and wash. The correlation of phytoplankton community with the physical-chemical parameters that can be used to determine the condition of the waters. The purpose of this research was to discuss the types of phytoplankton, analyze the community index and correlation between physical - chemical parameters in Sungsang waters. The research was had been done on November 2018, taking of samples and measurements was performed at 3 station points by tides at the day and night in 3 days. Taking of sampels that is phytoplankton and nutrient water samples, measurements of physical - chemical parameters that is DO, temperature, salinity, pH and current velocity, the correlation of phytoplankton community's with physical-chemical parameters are carried out by principal component analysis (PCA). The phytoplankton composition at night was found 39 genera from 4 classes, while during the day there were 41 genera from 5 classes. Percentage of phytoplankton composition class that is Bacillariophyceae (98.77%), Cyanophyceae (0.87%), Dinophyceae (0.29%), Chlorophyceae (0.055%) and Dictyochophyceae (0.014%). The abundance of phytoplankton is range between 298 - 2,533 cells/l with the diversity index of the dominant is medium category, the uniformity index of the dominant is high category and the dominance index of the dominant is low category. Based on the results of PCA for the correlation between phytoplankton communities with physical-chemical parameters during at day with the identifiers is uniformity index, DO, dominance index, phosphate, salinity and current, the identifiers at night is current, pH, nitrate, temperature, phosphate, DO, diversity index and uniformity index, for combined data day and night the identifiers is diversity index, phosphate, uniformity index, abundance, temperature, pH, dominance index and salinity.
