Abstract

Soft coral is a marine animal that does not have a spine that lives in shallow water. Soft corals are able to interact with microorganisms such as fungi. Symbiotic fungi from the sea are microbes that are rich in bioactive natural products and secondary metabolites. The purpose of this study was to isolate and identify symbiotic fungi with isolated soft corals from the Sinularia polydactyla type and determine the type of media that produced the most symbiotic fungi in the Sinularia polydactyla soft coral. The research method in isolating fungal symbionts was carried out using the Direct Planting method. This study uses 3 media including PDA (Potato Dexktrose Agart), HDA (Host Dekxtrose Agart), and MEA (Malt Extract Agart). The results of this study showed that symbiotic fungi with Sinularia polydactyla soft coral were isolated in 7 isolates, with 3 isolates identified as Aspergillus flavus, 2 isolates identified as Penicillium sp., and 2 isolates identified as Aspergillus niger. The media that produced the most symbiotic mushrooms were PDA media, where 7 isolates were found, there were 3 isolates that grew on PDA media, 2 isolates on MEA media, and 2 other isolates on HDA media.

Abstract

Muara Sungai Musi, Sumatera Selatan merupakan tempat bermuaranya aliran Sungai Musi. Akibat banyaknya kegiatan manusia di perairan Muara Sungai Musi, berpotensi terjadinya pencemaran lingkungan yang sangat berpengaruh pada kelangsungan hidup komunitas moluska. Penelitian ini bertujuan untuk mengidentifikasi jenis dan menganalisis komunitas moluska serta menganalisis hubungan parameter fisika-kimia dengan komunitas moluska. Hasil penelitian menunjukkan terdapat 7 spesies moluska dimana dikelompokkan menjadi 2 kelas yaitu Bivalvia dan Gastropoda. Hasil analisis substrat sedimen rata-rata memiliki substrat lempung. Komunitas moluska menunjukkan nilai keanekaragaman (H’0,56<H<1,24). Tingkat keseragaman (E=0,29-0,64). Dominansi (C=0,33-0,63). Abstract

ABSTRACT Soft corals contain bioactive compounds that can be used as a marine natural product. The puposes of this study was to determine of the soft corals inhibition potential for antibacterial activity. The methodology in this study included field sampling, extraction (in the solvent of n-hexane, ethyl acetate and methanol), antibacterial bioassay, and briefly describe the type of soft coral that possesed the highest inhibition. In sampling treatment, the biomass among wet samples and dry samples varied.  The highest shrinkage due to drying was found in Nephthea, followed by Lobophytum and Sarcophyton, while the lowest was found in the genus of Sinularia. Extracted from 12 samples of soft coral, the lowest extract weight was found in the semi-polar solvent (EtOAc), while the extract weight of n-Hex and MeOH was vary. Inhibition power from all extract samples was found on soft coral extract in the EtOAc and MeOH solvents. The highest value of inhibition power was found in soft coral of Sinularia polydactyla and S. Abstract

Symbiont bacteria on soft coral can produce bioactive compounds that plays an important role in chemical ecology and as a marine natural product. The purpose of this study was to find and characterize the antibacterial activities of active compounds extracted from bacterial symbionts of soft coral S. flexibilis and S. polydactyla. The methods used in this study were culture and isolation of bacterial symbionts, extraction of compounds, antibacterial bioassay, and identification of bioactive compounds using the LC-MS analyses. Four isolates of bacterial symbionts were obtained from two samples of soft corals, 2 isolates of Pseudomonas diminuta (A1) and Edwardsiellla hoshinae (A2) from soft coral S. flexibilis, and 2 isolates of E. hoshinae (B1) and P. acidovorans (B4) from S. polydactila. Antibacterial activity were found only from the extracts of bacterial symbionts P. diminuta (A1) and from  S. flexiblis about 10.16 ± 0.3mm (for B. subtilis), 8.66 ± 0.8 mm (E. coli) and 9.86 ± 1.7mm (S. dysentri). No antibacterial activity found from the extracts of S. polydactyla. The results of LC MS analysis showed that the group of diterpenes sinularin produced by soft corals S. flexibilis and bacterial symbionts isolates of P. diminuta (A1).Keywords:  Bacterial Symbiont, Bioactive Compound, Antibacterial Activity, Soft Coral, Sinularia flexibilis, Sinularia polydactyla

Abstract

Marine biota has variety of bioactive compound that is potential to be an antibacterial for shrimp ponds diseases. Some of marine biota has potential as antibacterial i.e. soft coral Sarcophyton sp., Sponge Aaptos sp., seaweeds Sargassum sp. and Halimeda sp. and mangroves Avicennia sp. and Rhizophora sp. species. This study is purposed to find the most potential marine biota as antibacterial. The research method was as follows; sampling, sample identification, extraction and antibacterial activity test. The results is soft coral and sponge extract Sarcophyton sp. has bioactivity against of shrimp bacteria pathogen such as Vibrio spp2. bacteria for around 6.3±0.1 mm, while Aaptos sp. has bioactivity against Vibrio spp1. bacteria for around 7.9+0.1 mm, Vibrio spp2. bacteria for around 7.2± 0.1 to 7.9±0.1 mm, Vibrio spp6. bacteria for around 7.5±0.2 mm, Escherichia coli for around 7.2±0.1 mm and Staphylococcus aureus for around 15.9±0.2 mm. Seaweed extract activity found only of Sargassum sp. which has antibacterial activity against for around 7.1±0.0 mm for Vibrio spp6. and mangrove species activity has Rhizophora sp. extract which has bioactivity against Vibrio spp4. were 7.3±0.1 mm and E. coli bacteria were 6.7±0.1 mm. The most potential marine biota as antibacterial is showed on sponge Aaptos sp.. with an inhibitory zone for around 15.9±0.2 mm (for S. aureus bacteria).

Abstract

Ikan Gelodok (Periophthalmus boddarti) merupakan salah satu jenis ikan yang hiduuppada ekosistem mangrove dan diketahui memproduksi toksin. Tujuan penelitian iniadalah untuk mengetahui tingkat toksisitas senyawa bioaktif pada ekstrak ikan Gelodokyang berasal dari Pulau Payung Kabupaten Banyuasin, Sumatera Selatan. Prosedurpenelitian meliputi ; pengambilan dan preparasi sampel, ekstraksi, uji toksisitas denganmetode Brine Shrimp Lethality Test (BSLT) dan analisis data menggunakan Analisa Probit.Hasil dari penelitian ini menunjukkan bahwa ikan Gelodok mempuyai potensi toksisitasyang tinggi pada ekstrak daging (DIGE) maupun organ dalam (ODIGM) denganmenggunakan pelarut etil asetat dan methanol dengan nilai LC50 < 1000 ?g/mL.Kemampuan toksisitas ekstrak dari ikan P boddarti sangat besar, pada daging ikangelodok dengan menggunakan pelarut methanol DIGM mempunyai nilai tinggi 116.013ppm, ODIGM 132.376 ppm, DIGE 164.090 ppm, dan ODIGE 322.606 ppm.

Abstract

This study was carried out to determine the toxicity of the dinoflagellate Prorocentrum minimum species that formed massive blooms in Lido Beach, Johor Bahru in July 2002. Clone cultures were established in ES-DK medium at 26o C under a 14:10 hour light dark cycle. Species toxicity was determined by intra peritoneal (i.p) injection of culture extract into mice. Cultured cell extracts were toxic to mice. The major symptoms were muscular paralysis and diarrhea. However no mouse mortality was observed even after 13 hours.  Extracts of cultured cells were also hemolytic on rabbit red blood cells. Keywords: Dinoflagellate, HAB, Prorocentrum minimum, Red Tide dan Toxicity Kajian dilakukan untuk menentukan ketoksikan dinoflagelata spesies Prorocentrum minimum yang menyebabkan kejadian pasang merah besar-besaran di perairan Pantai Lido, Johor Bahru pada Juli tahun 2002. Kultur klon telah dibuat dalam medium ES-DK pada suhu 26o C dan siklus pencahayaan 14:10 jam terang gelap. Ketoksikan spesies diuji dengan penyuntikan ekstrak kultur secara intra peritoneal (i.p.) terhadap tikus. Hasil pengamatan menunjukan ekstrak sel yang dikultur memberikan pengaruh racun kepada tikus. Gejala utama adalah kejang otot dan menceret/diare. Walau bagaimanapun tidak didapati tikus yang mati setelah dilakukan pengamatan selama 13 jam. Ekstrak toksin spesies ini juga menunjukkan pengaruh hemolitik terhadap eritrosit kelinci. Kata kunci: Dinoflagellata, HAB, Prorocentrum minimum, Red Tide dan Toxicity

Abstract

Kepadatan limbah plastik menjadi suatu ancaman besar bagi lingkungan dan organisme perairan, pasalnya proses degradasi plastik menjadi mikroplastik membutuhkan waktu yang sangat lama. Salah satu upaya alternatif untuk mengurangi dampaknya dapat menggunakan bakteri.  Penelitian ini bertujuan untuk  mengidentifkasi jenis bakteri pendegradasi limbah mikroplastik dan menentukan kemampuan isolat bakteri dalam mendegradasi limbah mikroplastik. Penelitian ini dilaksanakan pada bulan Juli – September 2019. Pengambilan sampel air menggunakan jaring micro nett berdiameter 90x15 cm dengan mesh size 300 ?m dan dilakukan pengukuran kualitas air, meliputi suhu, salinitas, DO, pH dan kecepatan arus. Identifikasi bakteri menggunakan uji biokimia otomatis (VITEK-2). Hasil penelitian menunjukkan adanya tiga jenis bakteri yang berpotensi mendegradasi mikroplastik dari enam isolat yang berhasil dimurnikan yaitu Vibrio fluvialis, Serratia rubidaea, Serratia marcescens dan Psedomonas putida.Kata Kunci : Bakteri mikroplastik, degradasi, kualitas air, Muara Sungai Musi.

Abstract

Density residential and transportation activities contribute to the damage and declining water quality in the waters of Sungsang Banyuasin. But the community still use river water for their needs / daily consumption is prone to cause disease. The study was conducted in July 2010 in the Village Sungsang III, Banyuasin II South Sumatra. The method used is Cluster Sampling. Sampling unit is the coastal communities that follow the C model of community service activities (KKN-Thematic) to design the pre-test and post-test without a control treatment with t test. The results of t-test with p value <0.05 indicates that the C model of community service activities through counseling and demonstration of a simple water treatment providing community response to increased motivation to process river water into the water ready to use. The results of the feedback showed that this community service activity further increased knowledge and awareness of coastal communities to maintain the river and a healthy lifestyle. Namun masyarakat sekitar tetap memanfaatkan air sungai untuk kebutuhan/konsumsi sehari-hari yang rawan menimbulkan penyakit. Penelitian dilaksanakan pada bulan Juli 2010 di Desa Sungsang III Kecamatan Banyuasin II Sumatera Selatan. Metode yang digunakan adalah Cluster Sampling. Sampling unitnya adalah kelompok masyarakat pesisir yang mengikuti kegiatan pengabdian masyarakat model C (KKN-Tematik) dengan desain pre-test dan post-test tanpa perlakuan kontrol dengan uji t. Hasil uji t-test dengan nilai p < 0,05 menunjukkan bahwa kegiatan pengabdian masyarakat model C melalui penyuluhan dan alat peraga pengolahan air sederhana memberikan respon peningkatan motivasi masyarakat untuk mengolah air sungai menjadi air siap pakai. Hasil umpan balik menunjukkan bahwa kegiatan pengabdian pada masyarakat ini semakin meningkatkan pengetahuan, kepedulian dan kesadaran masyarakat pesisir untuk menjaga sungai dan melakukan pola hidup sehat.   Kata kunci : respon, masyarakat pesisir, pengabdian masyarakat, pengolahan air

Abstract

Bruguiera gymnorrhiza merupakan salah satu jenis mangrove yang memiliki potensisenyawa bioaktif yang dapat digunakan untuk mengawetkan produk perikanan karenabersifat sebagai sumber antimikroba alami. Penelitian ini bertujuan untuk mengetahuipotensi larutan bubuk daun mangrove B. gymnorrhiza sebagai pengawet alami. Penelitianini dilaksanakan pada Bulan Juni – September 2016 dimana sampel daun mangrove B.gymnorrhiza diambil di Tanjung Api-Api. Prosedur penelitian meliputi penanganan sampeldaun B. gymnorrhiza, pembuatan simplisia, pengawetan produk perikanan (udang kupas),uji fitokimia, analisa sensori, menghitung jumlah mikroba dengan Total Plate Count (TPC)dan analisis statistika (Friedman-Conover dan uji Beda Nyata Jujur). Hasil penelitianmenunjukkan bahwa senyawa yang terkandungan pada B. gymnorrhiza meliputi: tannin,saponin, steroid, flavonoid dan fenol hidroquinon. Analisa sensori menunjukkan mutuproduk perikanan (kenampakan, aroma dan tekstur) paling baik pada konsentrasi 60gram/L dengan masa simpan tujuh hari. Hasil TPC menunjukkan bahwa jumlah mikrobaberkisar antara 1,127 x 103 koloni/ml - 3,3 x 103 koloni/ml, dimana konsentrasi terbaikadalah 40 gram/L dengan jumlah mikroba 1,627 x 103 koloni/ml.Kata Kunci: Analisa Sensori, Bruguiera gymnorrhiza, Pengawet Alami, Total PlateCount (TPC), Uji Fitokimi
