Abstract

Hypercholesterolemia condition was reported can decrease activities of antioxidant superoxidedismutase, catalase, and glutathione peroxide. This study was conducted to observe the role of seaweed(Eucheuma cottonii) powder in increasing the level of antioxidant cooper, zinc-superoxide dismutase (Cu,Zn-SOD) in kidney tissues of hypercholesterolemic rats. Twenty male Wistar rats were used in this study.Those rats were divided into four groups ; (1) Negative control group (A), (2) and (3) werehypercholesterolemic groups that were treated with 5% (B), and 10% (C) of seaweed powder, and (4)Hypercholesterolemic group as positive control (D). The treatment was carried out for 35 days.Hypercholesterolemia condition (>130 mg/dl), except for group A, was achieved by feeding the rat withcommercial diet containing 1% cholesterol and drinking water ad libitum for 40 days. Serum total cholesterolwas analyzed both before and after treatment. Rat kidneys were taken at the end of treatment, andprocessed by using paraffin embedding standard method. The tissues were then stained usingimmunohistochemical technique to Cu,Zn-SOD. The results showed the seaweed powder decreased serumtotal cholesterol, increased caecum cholesterol, repaired alteration of kidney tissues, and increased thelevel of antioxidant Cu,Zn-SOD in the kidney of hypercholesterolemic rats. Dietary fibers and antioxidantactivity of 10% seaweed powder was better than the of 5%.

Abstract

The objective of this study was to observe the effect of functional synbiotics yoghurt (made ofindigenous probiotic + oligofructosaccharides) in rats that were infected with Enteropathogenic Escherichiacoli (EPEC), and to detect its effect on hematological status. A total of 25 male Sprague Dawley rats wereused in this study and divided into five treatment groups: (i) negative-control; (ii) positive-control; (iii)synbiotic yoghurt; (iv) synbiotic yoghurt + EPEC; and (v) conventional prebiotic yoghurt. Yoghurt (109 cfu/mL lactic acid bacteria) was given orally using feeding tube at day one until day 4. At day 8 – day 14animals were infected with EPEC (107 cfu/mL) orally using feeding tube; at day 22 blood samples werecollected for hematology analysis (erythrocytes, hematocrit (Hct), hemoglobulin (Hb), platelets, andleucocytes). Post infection with the EPEC diarrhea was developed in rats both in the control positivegroups and in the synbiotic yoghurt + EPEC group. Animals in the positive-control group had a significantlyhigher thrombocytes and leucocytes counts and Hct compared to that in animals in the other groups(P<0.01). Whereas there were no significant effect on the erythrocytes counts and Hb (P>0.05). The treatmentwith synbiotic yoghurt, in both synbiotic yoghurt only and synbiotic yoghurt + EPEC groups significantlyreduced the platelets and leucocytes counts and Hct of rats

Abstract

The consumption need of soybean that reached up to 2,5-3 million tons per year has made Indonesiaimports this commodity 1,95 million tons from countries that adopt Genetically Modified (GM) soybean.Soybean utilization for tempe producers is about 50% from total its supply. The aim of this research wasto evaluate the safety of tempe made from GM soybean, by a subchronic test approach using male Sprague-Dawley rats. Toxicity evaluation of tempe from soybean GM was done based on EFSA (2011) principalusing animal laboratory. For 90 days, three groups of male rats (each group contain seven rats) werefeeding with tempe flour from GM soybean, tempe flour from non-GM soybean and control group wasfeeding with casein as a protein source. All rats were terminated on day 90,where the blood, liver andkidney were collected. There were no significant differences on liver and kidney weight between treatment.Hematology analysis showed the level of Hb, erythrocyte, leucocyte, trombocyte and hematocryte were notsignificantly different in between group. Serum analysis on SGOT, SGPT, total protein, albumin, globulin,blood glucose, triglyceride, ureum and creatinin level were also showed no difference between groups. As aconclusion, the consumption of tempe derived from GM soybean was safe, and cause no adverse effect onhealth.

Abstract

High level of blood glucose is an indicator for diabetes mellitus (DM) condition. The condition iscaused by low level of insulin secretion or impairement of insulin receptor. The number of DM patientincreases every year. The World Health Organization reported that the number of DM patient in Indonesiawas the 4th highest in the world, after following China, India, and the United States of America, respectively.This study was conducted to analyze the effect of sea cucumber (Holothuria scabra J) on the profile ofantioxidant copper, zinc-superoxide dismutase (Cu, Zn-SOD) in the pancreatic tissues of diabetic rats. Atotal of 25 male white rats (Sprague Dawley) were used in this study. They were divided into five groups;(1) negative control (KN), (2) positive control, diabetic rats (KP), (3) diabetic rats treated with hydrolyzatedprotein of sea cucumber (HDL), (4) diabetic rats treated with concentrated protein of sea cucumber (KST),and (5) diabetic rats treated with isolated protein of sea cucumber (ISL), respectively. Diabetic conditionwas obtained by alloxan injection 110 mg/kg bw. The treatments were done for 28 days. At the end oftreatment period, the rats were sacrificed and pancreatic tissues were collected and fixed in Bouin solution and then processed to paraffin embedding standard method. The tissues were then stained withimmunohistochemical staining techniques using monoclonal antibody of Cu, Zn-SOD. The results showedthat treatment of HDL, KST, and ISL of sea cucumber (Holothuria scabra J) increased the content ofantioxidant Cu, Zn-SOD either in Langerhans islets and acinar cells of pancreatic tissues-diabetic rats.The HDL of sea cucumber treatment gave the best effect in increasing the antioxidant content of Cu, Zn-SOD in pancreatic tissue of diabetic rats.

Abstract

Hyperglycemia in diabetes mellitus results in stress oxidative condition. This study aims to analyze the antioxidant defence in pancreatic experimental-diabetic rats treated with a combined extract of Cajanus cajan leaf and ginger, using an immunohistochemical technique for copper, zinc – superoxide dismutase (Cu,Zn-SOD). A total of 25 male Sprague Dawley rats were used in this study. The rats were randomly divided into five groups; normal control group (NC), diabetic group (DM), two groups DM treated with a different dose combination of Cajanus cajan leaf extract and ginger extract (DME and DMF), and DM treated with glibenclamide (DMG). This study used 110 mg/kg BW alloxan induction to obtain diabetic conditions, and the treatments of the extract were conducted for 28 days. The pancreatic tissues were processed using the paraffin standard method and an immunohistochemical technique using monoclonal antibody Cu,Zn-SOD. The results showed that a combination of Cajanus cajan leaf extract and ginger extract lowered the level of blood glucose, increased body weight, and increased the level of Cu,Zn-SOD content in pancreatic tissues of experimental-diabetic rats. This study concluded that combining Cajanus cajan leaf extract and ginger extract increased antioxidant defence in pancreatic organs of experimental-diabetic rats.
