Abstract

Penelitian ini bertujuan untuk mengetahui dosis optimal Inhibitor Aromatase (IA) pada perkembangan oosit ikan mas koki. Dosis yang digunakan adalah k = kontrol (disuntik NaCl fisiologis), Pl = 2.5 mg/kg berat tubuh (b.t), P2 = 7.5 mg/kg b.t., dan P3 = 12.5 mg/kg b.t. Untuk mengetahui pengaruh IA terhadap perkembangan gonad diukur kandungan hormon estradiol-l7β dalam plasma darah dan level protein gonad mulai dari awal perlakukan kemudian setiap tujuh hari sekali, yaitu hari ke-7, ke-14, dan ke-21. Hasil penelitian menunjukkan bahwa, pada hari ke-7 hasil semua perlakuan P1, P2, P3 menunjukkan penurunan hormon estradiol- 17β diikuti penurunan level protein gonad, hasil analisis pengamatan histologi menunjukkan terjadi atresi pada sel gonad. Pada hari ke-14 kadar hormon estradiol-17β perlakuan menunjukkan terjadi peningkatan diikuti peningkatan level protein gonad. Pada hari ke-21 kadar hormon estradiol-17β perlakuan sudah sama dengan kadar hormon kontrol namun level protein gonad masih dibawah level protein gonad kontrol.Kata kunci: estradiol 17ß, perkembangan oosit, level protein gonad.

Abstract

Artificial insemination (AI) has been widely applied to improve genetic quality in cattle worldwide. One of the most important factors in AI program is the quality of semen.  Many procedures in semen processing have been developed to preserve the sperm quality. The Singosari AI center produces the frozen semen through cryopreservation method. During this process, the number of semen collected will be rejected if the quality is low. Fresh semen with less than 70% of sperm motility, before freezing semen with less than 55%of sperm motility and less than 40% of sperm post-thawing motility evaluation will be rejected, so that it was only the viable sperm will be processed to be frozen semen commercial. Therefore, comparative study was carried out to analyze the freezing capability of sperm.

Abstract

The beef cattle industry in Indonesia hampered by low population and productivity. Apart from genuine adaptability to tropical condition and low input of production, local breeds basically have small body size and low daily weight gain. During the last few years, through artificial insemination (AI) program the government has introduced crossbred of various European breeds with local cattle. Moreover, embryo transfer (ET) program, has also been adopted to support the foundation of elite cattle.    Balai Embrio Ternak (BET) Cipelang, a centre to produce and distribute embryos of high genetic quality cattle is responsible to develop a modern breeding program through embryo technology. The center, since its establishment in 2000, has produced and distributed thousands of bovine embryos of local and exotic breeds [1].  BET Cipelang has mandated to implement a large-scale importation of Belgian Blue (BB) embryos. Nine hundred frozen BB embryos have been imported from Belgium and transferred to recipients at various breeding centres. To study the performance of  the crossbred between local breeds and BB, 1000 frozen semen of BB bulls were imported and inseminated [2}.   The BB cattle has widely known as the breed with double muscling properties. The significance of this project is to study the BB capability on coping up with various local constraints to produce Indonesian BB. The breeding program of the local breed with BB through AI will direct the breeding policy on crossbreeding program. This study was developed to evaluate the success rate of AI and ET, calving ease, calves body weight, daily gains and some other phenotypic performance.

Abstract

Currently, artificial insemination (AI) techniques using cryopreserved semen have been widely used to improve the genetic potential of livestock species. Semen at the Lembang AI Center usually collected by artificial vagina (AV) then the semen should be evaluated to get the good quality of spermatozoa for the freezing process. Several efforts are being made to improve the quality of frozen semen for AI so as to get the most out of this technology. But there are certain obstacles in this path to achieve breeding goals. The trend now is to design procedures or refine methodologies so as to maximize the production of good quality semen without discarding too many poor quality ejaculates. Therefore, to minimize the poor or discarded spermatozoa used in the cryopreservation process, this study might elaborate more about the influencing factors.

Abstract

Livestock Embryo Center/Balai Embrio Ternak (BET) Cipelang is one of the government institution under the supervision of Directorate General of Livestock and Animal Health Services, Ministry of Agriculture of the Republic of Indonesia. BET Cipelang has the main task of carrying out the production, development, and distribution of livestock embryo for all regions of Indonesia. BET Cipelang produced embryos using two methods, there are in vivo and in vitro. The produced embryos will be transferred to the recipient cow using embryo transfer technique.                Embryo transfer biotechnology can be used to enhance genetic improvement and to increase marketing opportunities with purebred cattle. Because of their relatively low reproductive rate and long generation interval, embryo transfer is especially useful with cattle. The success of embryo transfer depends on factors associated with the embryo, the recipient or an interaction among the factors of the embryo and recipient [1].                 Embryo quality is one of the main factors for successful embryo production. In vivo embryo production has better quality than in vitro such as cell number, morphology, growth and development ability and after freezing.  The frequency of occurrence of chromosomal abnormalities in in vivo-derived embryos are lower than in vitro and the consequence is that only 30-40% of oocytes resulting from in vitro maturation develop into blastocysts after in vitro fertilization and the in vitro yield rate of embryos are lower than from in vivo [2]. Recently, there are no studies about the performance of in vivo embryo production at BET Cipelang. The aim of this study is to explore the quality of in vivo embryos produced by BET Cipelang.

Abstract

The influence of various types of sera i.e. Fetal Calf Serum (FCS), Bovine Post Estrous Serum (SPES) as heterolog sera; and Ewes Fetal Serum (EFS) and Ewes Serum (ES) as homologue sera on in vitro maturation and fertilization of sheep oocytes was investigated using Tissue Culture Medium 199TM (TCM 199. Gibco). Sovine sera were collected from cow at seven days post-estrus, while ewes sera were collected during estrous (ES-HO) and six days post estrous (ES-H6). The result shows that the maturation rate of oocytes cultured in medium supplemented with EFS, ES-HO and ES-H6 at  concentrations of 32.9%, 68.7%, and 67.6% respectively is better than those supplemented with FCS and SPES at concentrations of 22.2% and 28.9%, respectively. In vitro fertilization rate were significantly higher in medium supplemented with ES-HO and ES-H6 at concentration of 65.4% and 65.8% (P < 0.05) than that supplemented with SPES at concentration of 31.7%. In conclusion, ewes sera could be used in culture medium for promoting both in vitro maturation and fertilization rate of sheep oocytes.

Abstract

The research was planned to yield the biologic potential synergism of MoAb-hCG in increasing the embryo number. Twenty five dairy cows were divided into five groups. The Control Group was superovulated with 2,500 IU PMSG intramuscularly (i.m.), the remaining of the Group II, III, IV and V consecutively after being superovulated by 2,500 IU PMSG were treated with MoAb intravenously (i.v.), hCG i.v., MoAb-hCG i.v. and MoAb i.v-hCG i.m. Data evaluation were analyzed using ANOVA and LSD test. The result showed that there were increasing in number of transferable embryo per donor produced (P<0.01) after application with either MoAb i.v. (9.6), hCG i.v. (9.4), MoAb-hCG i.v. (11.2), or MoAb i.v.-hCG i.m. (9.6). The Control Group produced only 2.4 embryo per donor. The combined MoAb and hCG in one treatment were not able to increase the embryo yield (P>0.05).Key words: MoAb, PMSG, hCG, superovulation, donor, embryo

Abstract

The experiment was carried out to study the use of direct transfer method in embryo cryopreservation by using two cryoprotectants and the effectivity of various concentrations of sucrose during cryoprotectant removal. Eighty-fourmorula stage embryos were divided equally into two groups and were treated by using 1.5 M ethylene glycol (EG) and 1.5 M 1,2-propanediol as cryoprotectant. The embryos were frozen using programmable embryo freezing machine on step by step decreasing temperature. Frozen embryos were thawed and cryoprotectants were removed either without sucrose (0 M) or with sucrose in concentration of 0.2 M, 0.4 M and 0,8 M. The results showed that the quality of the thawed embryos cryopreserved using 1.5 M EG was better than that using 1.5 M PROH. The survival rate on the embryos cryopreserved with 1.5 M EG (92.8%) was higher than 1.5 M PROH (78.6%) (P0.05). The viability of embryos exposed to 0 M, 0.2 M, 0.4 M and 0.5 M sucrose were 80.0, 80.8, 90.9 and 81.80% respectively. Abstract

Monitoring of physiological stress in wild and/or endangered animals living in their natural habitat can be generated via measuring faecal glucocorticoid metabolites (FGCMs) through enzyme immunoassay technique. However, a careful validation of each enzyme immunoassays (EIAs) and methodological issues such as samples preservation, and extraction procedures must be a significant concern and validated because  it may influence FGCMs levels. In this present study, the aim was to carry out a biological validation to find reliable EIAs that could be used for non-invasive measurement of glucocorticoid levels in crested macaques. Moreover, we compared three different techniques of preservation: lyophilisation, oven drying and field extraction and three different solvents for extraction: 80% methanol, 90% pure ethanol and 90% commercial alcohol (isopropyl alcohol) to find the simple and practical techniques for sample processing. For hormone analysis, we colIected 189 faecal samples from wild crested macaques living in the Tangkoko-Batuangus Nature Reserve, North Sulawesi, Indonesia. The results show that biological validation can be used to validate measurement of faecal glucocorticoid which 3α,11β-dihydroxy-etiocholanolone was the most reliable EIAsfor this species. Different preservation and solvent extraction were not significantly influenced levels of FGCMs (p0.05). In conclusion, Our study highlights the importance of biological validation of FGCMassays and presents practical techniques for the non-invasive monitoring of physiological stress in crested macaques.
