Abstract

Antibodi imunoglobulin G (IgG) merupakan salah satu immunoglobulin yang dikandung oleh antibodi anti-sperma (ASA) yang terdapat pada saluran reproduksi betina. Imunoglobulin G dapt berikatan dengan protein-protein yang berpotensi sebagai protein antigenik seperti alfa enolase (ENO1). Penelitian ini bertujuan untuk mengidentifikasi dinamika dan potensi ENO1 membran plasma spermatozoa sebagai protein antigenik serta kualitas spermatozoa sapi Bali dengan perlakuan 0,5 mg/ml IgG. Sampel penelitian adalah 16 ejakulat yang diperoleh dari 4 ekor pejantan. Motilitas spermatozoa dievaluasi menggunakan CASA, viabilitas melalui metode pewarnaan diferensial, Keutuhan Membran Plasma (MPU) menggunakan metode Hypo-osmotic Swelling Test (HOS-Test), nilai Mix Anti-globulin Reaction (MAR) diperoleh dari MAR-Test, sedangkan kuantitas ENO1 diukur dengan ELISA. Analisis data menggunakan analisis ragam RAK. Hasil penelitian menunjukkan bahwa perlakuan dengan IgG sampai 0,5 mg/ml menurunkan motilitas, kinematika spermatozoa, viabilitas, MPU dan kuantitas ENO1 membran plasma spermatozoa (P<0,01), serta meningkatkan nilai MAR-test (P<0,05). Penelitian menyimpulkan bahwa IgG dapat menurunkan kualitas spermatozoa dan kuantitas ENO1, sekaligus menunjukkan bahwa ENO1 membran plasma spermatozoa sapi Bali berpotensi sebagai protein antigenik.

Abstract

This research aims to evaluate the protective potency of freeze-dried egg yolk powder on Garut ram sperm during the liquid semen preservation process. Semen with good sperm quality was divided into three groups and diluted using the following Tris diluents: fresh egg yolk-Tris (FEY-Tris), commercial egg yolk powder-Tris (CEY-Tris), and freeze-dried egg yolk powder-Tris (DEY-Tris). Semen that had been diluted was observed every 12 hours until sperm progressive motility was 50%. Sperm quality was tested for progressive motility percentages, viability, intact plasma membrane (IPM), and intact acrosome (IA). Fresh egg yolk had a particle size of 14460.00±330.76 nm, with polydispersity index value (PI) and zeta potential (ZP) being 1.00±0.11 and -9.22±0.30 mV, respectively. The particle size of commercial egg yolk powder was 877.90±168.86 nm with PI value 0.34±0.04 and ZP-28.7±1.24 mV. The particle size of freeze-dried egg yolk powder was 1296.00±86.73 nm, with PI value 0.70±0.04 and ZP -34.5±0.64 mV. Progressive motility percentage showed that DEY-Tris diluent managed to survive to 51.11±6.06% for 168 hours, compared to FEY-Tris (51.25±6.74%) which survived for 156 hours, although no longer than CEY-Tris diluent (53.47±6.33%) which survive for 180 hours (P0.05). Higher sperm motility was also supported by viability percentage, IPM, and IA, which were all higher (P0.05). In conclusion, freeze dried egg yolk powder mixed with Tris buffer was able to maintain sperm quality during the liquid semen preservation process.

Abstract

The objective of this study was to examine the effects of repeated freeze-thaw cycles and short-term storage of fecal extracts at ambient temperature on the stability of fecal glucocorticoid (fGCM) and estrogen metabolite (fEM) levels from crested macaques.In total 100 aliquots of fecal extracts from fecal samples collected from female crested macaques (Macaca nigra) living at the Tangkoko-Batuangus Nature Reserve, North Sulawesi were used. We performed two different experiments: (1) An experiment to investigate if levels of fGCM and fEM measured from fecal extracts that were exposed to two, four, six and eight repeated freeze-thaw cycles (test groups) differ to control samples (i.e. fecal extracts always stored frozen); (2) An experiment to evaluate whether storing fecal extracts at ambient temperature for two, four, six, and eight days (test groups)affects the levels of fGCM and fEM compared to the control group (i.e. fecal extracts frozen immediately).Results showed that hormone levels were significantly increased (P0.05) after four freeze-thaw cycles for fGCM and after eight freeze-thaw cycles for fEM. By contrast, there was no significant difference (P0.05) in levels of fGCM and fEM between the test groups and the control group in fecal extracts stored at ambient temperature. In conclusion, our data show that more than two and six repeated freeze-thaw cycles should be avoided when measuring fGCM and fEM in crested macaque fecal extracts, respectively. We also demonstrate that storing fecal extracts at ambient temperature is possible for at least 8 days without taking a risk of affecting the stability of fGCM and fEM levels.

Abstract

This study aimed to determine the effect of age on bulls’ productivity and fresh semen characteristics of Simmental bull in Indonesia. A total of 1071 data of semen collection and production from four age groups (two years old (yo), four yo, ≥ 10 yo with high semen rejection (≥10 HR), and ≥ 10 yo with low semen rejection (≥10 LR) were used in this study to evaluate the productivity and characteristics of fresh semen. The results showed that the pre-freezing and post-freezing semen rejection rate of ≥10 HR group was higher (P0.05) than the other groups. The four yo group had the percentage of second semen ejaculation each collection was higher (P0.05) than the other groups. Furthermore, semen volume, sperm concentration, and total sperm concentration significantly increased (P0.05) until four yo and then decreased (P0.05) in ≥ 10 yo groups. The ≥10 HR group had the volume and total sperm concentration significantly different (P0.05) with a group of ≥10 LR. Total sperm motility, individual motility, and mass movement were lower (P0.05) in ≥10 HR than the other groups. In conclusion, age differences of bulls can affect the productivity and characteristics of fresh semen.

Abstract

Penelitian ini bertujuan untuk merancang dan mengevaluasi bioavailabilitas implan intravaginal yang terbuat dari poli-ymer kitosan-polietilen glikol (PEG)-polikaprolakton (PCL) yang dapat terbiodegradasi dalam cairan simulasi vagina (SVF) dan vagina sapi. Implan dibuat menggunakan teknik peleburan dan pencetakan dengan 26 formulasi berbeda. Selanjutnya implan direndam dalam 40 mL SVF pada suhu 37,8℃ dan diamati perubahan morfologi implan setiap hari hingga hari ke 5. Implan yang bertahan hingga hari kelima diuji ketersediaannya lebih lanjut pada vagina sapi dan diamati setiap hari selama 2 minggu. Uji perendaman pada hasil SVF menunjukkan bahwa implan dengan kandungan PEG 75-85% lebih cepat meleleh dibandingkan implan dengan kandungan PEG 20-72%. Implan yang mengandung 1-10% PCL terkikis lebih cepat di SVF dibandingkan implan yang mengandung 22-80% PCL. Implan yang mengandung kitosan dapat tetap stabil di SVF lebih lama dibandingkan implan tanpa kitosan. Uji bioavailabilitas pada vagina sapi menunjukkan bahwa implan 21-23 bertahan hingga hari ke 10, sedangkan implan 24-26 dikeluarkan dari vagina sapi selama pengujian. Implan 21-23 merupakan formulasi yang menjanjikan untuk pengembangan implan intravaginal yang dapat terbiodegradasi. Dengan menyesuaikan komposisi kitosan (antara 14-18 berat%), PEG (maksimum 72 berat%), dan PCL (minimal 20 berat%), implan intravaginal mampu meningkatkan ketersediaan hayati cairan vagina dan vagina yang disimulasikan. ternak

Abstract

Standar kualitas semen yang berperan penting dalam keberhasilan inseminasi buatan (IB) sebagai upaya peningkatan populasi dan mutu ternak sapi di Indonesia terbukti tidak seutuhnya mampu menentukan tingkat fertilitas sapi pejantan di Indonesia. Studi terbaru membuktikan bahwa meskipun sudah memenuhi standar kualitas semen yang sudah ditetapkan, namun tiga dari enam ekor sapi pejantan di Indonesia masih tergolong fertilitas rendah (<60%). Potamine-1 (PRM1) merupakan molekul pada inti spermatozoa yang telah terbukti memiliki hubungan erat dengan fungsi fertilitas dan kualitas spermatozoa pada berbagai sapi pejantan di Indonesia. Hal tersebut mendasari usulan pengembangan uji berbasis molekuler PRM1 sebagai penentu tingkat fertilitas yang diharapkan dapat digunakan untuk proses seleksi dan kebijakan pengafkiran sapi pejantan unggul di Indonesia. Melalui policy brief ini diharapkan usulan pengembangan uji tersebut dapat lebih tersampaikan dengan segala solusi yang diberikan, termasuk terkait pendekatan uji berbasis molekuler yang paling tepat digunakan, kajian-kajian lanjutan, dan berbagai harapan dukungan dari segala pihak demi terwujudnya pengembangan uji PRM1.

Abstract

Bali cattle is one of Indonesia's local cattle that has high economic potential. The quality of the semen used plays an important role as a factor in the success of artificial insemination (AI) programs. Climate change, including temperature variations, rainfall, and length of solar exposure, can affect the quality of fresh semen produced by bulls. Differences in environmental conditions can affect semen characteristics, such as spermatozoa concentration, motility, and morphology, which in turn have an impact on the field fertility rate. The aim of this study is to explore the differences in male fertility based on environmental conditions that change throughout the seasons. The research method used in this study is non-experimental, using secondary data on the quality of fresh semen and frozen semen production collected from the Regional AI Center, Bali Province. In addition, this study also used data from the big data of the National Animal Health Information System (iSIKHNAS) to calculate field fertility represented by conception rate. Climate data was obtained from the Indonesian Agency for Meteorological, Climatological and Geophysics (BMKG) Bali Province, the data collected was climate data in Tabanan Regency. The differences in the appearance of fresh semen quality, frozen semen production, and field fertility were analyzed using the Independent T-Test. In this study, there was no significant difference in fresh semen quality, frozen semen production, and field fertility (P>0.05) in all parameters, but the average reproductive performance of bali bulls was higher in the rainy season than in the dry season.

Progressive motility percentage showed that DEY-Tris diluent managed to survive to 51.116.06% for 168 hours, compared to FEY-Tris (51.256.74%) which survived for 156 hours, although no longer than CEY-Tris diluent (53.476.33%) which survive for 180 hours (P0.05). Higher sperm motility was also supported by viability percentage, IPM, and IA, which were all higher (P0.05). In conclusion, freeze dried egg yolk powder mixed with Tris buffer was able to maintain sperm quality during the liquid semen preservation process.
