Abstract

Cryopreservation of spermatozoa has been used to preserve spermatozoa in very low temperatures. Glycerol is intracelullar cryoprotectant usually used in Tris citrate containing 20% egg yolk (TCEY), while TCEY-1.5% alginate was used as encapsulation media.  The effect of alginate in microencapsulation process and glycerol concentration on viability spermatozoa (motility (%M), live sperm (%L) and intact apical ridge (%IAR) were studied in two steps. In the first step, the effect of alginate and microencapsulation process was studied. Three treatments of this step were: 1) TCEY add with semen (as control), 2) TCEY-Alginate add with semen, 3) TCEY-Alginate add with semen and microencapsulated, followed by conventional sperm cryopreservation using TCEY 7% glicerol. The second steps were done to evaluate the effect of two glycerol concentrations (5 and 7%) and two duration of equilibration time (3 and 4 hours). Result of these experiments showed, that microencapsulation was significantly alter the percentage of post-thawing motility (%M) and intact apical ridge (%IAR). The motility of microencapsulated sperm vs control was 44.7 and 50.83% respectively, whereas IAR 79.33% and 83.50% on microencapsulated sperm and control. There was evidence that alginate act as extracelullar cryoprotectant by protecting sperm during freezing. The mean of decreasing percentage of %M and %IAR in TCEY-Alginate were lower (15.97 and 6.44%) than control (23.80 and 7.37%). The effect of glycerol concentration and equilibration time on the viability of sperm was not significant different. There was no significant interaction of glicerol and equilibration time on the viability of spermatozoa. In conclution, microencapsulation processes altered the viability of sperm, alginate had an important roles as extracelullar cryoprotectant. Moreover the cryopreservation of microencapsulated sperm might be done using 5 or 7% glycerol in 3-4 hours duration of equilibration time. Abstract

In general, local goat productivity in Indonesia is relatively still lower than that from the temperate area. Yearling body weight in local goat about 14-17 kg. However, the local goat have superiority, such as adaptability and a good reproductive performance, which have litter size 1,49-1,61. Increasing local goat productivity may be done by breeding program (cross breeding and selection) and the environmental improvement. Both cross breeding program and selection that involve mating regulation will be efficient by reproductive technology application, such as artificial insemination (AI). Application of AI in goats is limited by four main factors that influence simultaneously, viz. poor quality of frozen semen, the variety of female reproductive performance, improper technical and insemination time, and poor management. To overcome these constraints his obligatory to pay more attention, so that AI will be able to be used as one of tools to increase the population and genetic quality in goats.   Key words: Goat, reproductive efficiency, artificial insemination

Abstract

This study was conducted to determine the earliest day of pregnancy diagnosis in kacang goat using transrectal ultrasonography. The goat were synchronized by using prostaglandin in the luteal phase. Pregnancy was determined by isoechogenic visualization surrounded by hypoechogenic. Early pregnancy was detected on days 20 of embryonic vesicle diameter 1.2±0.1 cm. Fetus was detected on days 22 with a long gestation fetus 0.4±0.1 cm. Average increase until days 30 pregnancy was 0.19 ± 0.1 cm per day. Development of fetus was followed by an increasing the diameter and thickness of uterus. The diameter of uterus increased from days 14 (0.8 ± 0.3 cm) until days 30 (3.6 ± 0.2 cm), and thickness of uterus increased from days 14 (0.4 ± 0, 2 cm) until days 30 (1.8 ± 0.2 cm). It could be concluded that the earliest pregnancy diagnosis showed positive sign on days 20 and fetus was earliest observed on days 22.

Abstract

The development of horse breeding industry in Indonesia was commenced through horse racing events held all over the country. It were accelerated by the development of Thoroughbred-Indonesian local Crossbred horses. There are many broodmares injured during their racing time and retired from the racetracks. They may still has a reproductive vigor to continue on producing offsprings. Very little information has been reported on the monitoring the reproductive capacity of the mares. The objective of this study was to explore ultrasonography imaging of the ovarian dynamics, correlated with the estrus behavior of the Thoroughbred-Indonesian local crossbred mares. Three Thoroughbred-Indonesian local crossbred mares with 6.25-12.5% of local genetics aged 12-20 years old were used in this study. Estrus and ovulation were synchronized by 10 mg PGF2α i.m. at luteal phase and 1,500 IU hCG i.m. injection when the dominant follicle reach ≥30 mm in diameter. Ultrasonography examination was done every morning at approximately at the same time. Estrus behavior was observed by using teaser stallions following a standard method. Results of the experiment indicated that onset of the estrus was reached 1.33±0.58 days after the hCG injection, with the average duration of 4.00±1.00 days. The ovulations were done at 5.33±1.15 days after PGF2α treatment and 66.67±10.07 hours after hCG treatment. Maximum follicle diameter was identified to reach 4.50±0.52 cm at one day before ovulation. The mares performed 25.4±3.38 days length of estrus cycle with 2-3 follicular  waves. It had been identified that the estrus duration  was  6.8±1.92  days  in mares with the age of 12-20 years. The average of maximum diameter of the largest follicle before ovulation was 4.2±1.24 cm. In conclusion, to improve the efficiency of breeding, several information are needed i.e. the optimal time of ovulation, relevan parameters related to follicular development.

Abstract

Parity gives an indication of physical maturity of cows. Physical maturity state of primiparous was 82 – 90% while that for pluriparous was 100%. In postpartum period, cows have a negative energy balance. Cows experiencing this condition will use the deposit glycogen in their liver and lipids in adipose tissue to be metabolized by liver become glucose. This experiment aimed at studying blood glucose at different parities of post-partum Fries Holland dairy cows. One-way classification design was use to observe 90 heads of post-partum dairy cows (30 heads of cows each in the first, the second and the third parity state. Data were analyze with General Linear Model (GLM) and the significant difference data among blood glucoses of the cows further analyzed with Duncan Multiple Range Test (DMRT). The results of the study showed that there was a significant (P<0.05) difference between blood glucose of the first parity state of cows (51. 57±5. 56 mg/dL) compared to that of the second (45. 57±8.01 mg/dL ) and the third (46.7±8.62 mg/dL ) cows. This was due to cows in the first parity is in growing state than in producing milk and reproduction function. Cows in the second and the third parities are in opposite function. Blood glucose profile at different parities of post-partum Fries Holland dairy cows was in the normal range (35-55 mg/dL). Blood glucose of the first parity of cows was higher (51.57±5.56 mg/dL) compared to that in the second (45.57±8.01 mg/dL) and the third cows (46.7±8.62 mg/dL). Abstract

Profile Intensity And  Estrus Time In Various Parity Fries Holland Dairy Cattle Postpartum. Parity is a period in the reproductive cycle of cattle with an indication of the amount of carrier confinement livestock.  Feliciano, et al, (2003) states that the parity is classified into three parts, namely: (1) nuliparous , (2) primiparous and (3) pluriparous / multiparous.This study used one-way classification design with the observed 90 cows  post-partum  is composed of 30 first parity, 30 second parity  and 30 third parity. Time of estrus observation data were analyzed with General Linear Model (GLM) followed by Duncan Multiple Range Test (DMRT).This study used one-way classification design with the observed 90 cows   post-partum  was composed of 30 first parity, 30 second parity  and 30 third parity. Data observation of  estrus time  were analyzed with General Linear Model (GLM) followed by Duncan Multiple Range Test (DMRT).It could be concluded that the parity  I, II and III with two estrus intensity (+ +), each for 63.3%, 46.7% and 73.4% and third estrus intensity (+ + +) respectively amounted to 36.6%, 53.3% and 26.6%. While the second estrus postpartum in various parity indicates a difference in means that the parity gives effect to the second estrus postpartum. Abstract

Kajian terhadap profil hormon-hormon steroid merupakan kunci penting dalam upaya memahami aspek fisiologis satwa. Beberapa dekade terakhir telah dikembangkan metode alternatif untuk mengetahui profil hormon steroid, yaitu melalui pengukuran metabolit hormon steroid yang diekskresikan melalui ekskreta tubuh seperti feses. Metode tersebut dikenal sebagai metode non-invasif. Metode ini memungkinkan pengumpulan sampel secara terus menerus dalam jangka panjang dengan meminimalisasi gangguan terutama pada satwa liar. Kajian terhadap profil metabolit hormon steroid yang terukur dapat diaplikasikan antara lain untuk mengetahui status reproduksi, penentuan jenis kelamin, studi perilaku hingga monitoring tingkat stres satwa. Berbagai kajian dengan memanfaatkan metabolit hormon steroid telah berhasil diaplikasikan pada berbagai taksa vertebrata. Tulisan ini memberikan gambaran terkini mengenai aplikasi metode non-invasif untuk kajian profil metabolit hormon steroid dari sampel feses.

Abstract

Anoa is an endemic ungulate in Sulawesi and its status now is endangered because the population continues to decline. Conservation genetics is one of the crucial issues in the anoa conservation strategy and action plan 2013-2022 document, but this genetic data is not yet available. To investigate and provide valuable information for conservation genetics measures, thirteen polymorphic microsatellites were used to analyze 20 adult anoa. Anoa has relatively low genetic diversity within populations (HO = 0.58), and high genetic differentiation among populations (FST = 0157). Although the anoa population has a bottleneck signal (T.P.M: 0.019; P0.05), the bottleneck simulation results show that the loss of genetic diversity is being slow over the next 100 years (9.5%). We provide some recommendations for conservation genetics based on the findings in this paper, including monitoring and genetically mapping for other anoa populations due to bottleneck signals, establishing the founder of the ex-situ population by examining their genetic diversity status, maintaining and increasing the number of individuals in the ex-situ population to genetically safe population size, and managing anoa populations by avoiding inbreeding. In-situ and ex-situ conservation programs should be combined to maintain the genetic diversity of anoa.
