Abstract

The aim of this study was to investigate the differences in histopathological changes of Malignant Catarrhal Fever in bali cattle based on sheep population in West Nusa Tenggara Province. This studied used 69 samples consist of the brains, lungs, livers hearts and kidneys, respectively from the bali cattles that affected the Malignant Catarrhal Fever disease. Those organs was processed by paraffin embedded blocking and were stained by Harris Haematoxylin-eosin method. The  histopathological preparations were examined based on the pathognomonical lesion that is vasculitis or inflammation of the blood vessel and other lesions. Result of the histopathological examination of the brains, lungs, livers, myocards and kidneys of the samples showed a varied of the level lymphoid cells infiltration in each of the organs. From the 69 samples showed the lymphoid cells infiltration lesions consist of  67 samples (97,10%) on the brain, 60 samples (86,95%) on the lungs, 63 samples (91,30%) on the liver, 45 samples (65,21%) on the myocard, and 47 samples  (68,11%) on the kidney. The conclusion were not difference variation histopathological lesions on the bali cattles that suffered Malignant Catarrhal Fever among the villages in West Nusa Tenggara with the varied sheep population. The  next researchs are needed to study the relationships of other animals as a reservoir of Malignant Catarrhal Fever virus.

Abstract

This study aims to determine the differences dosage and duration of butyric acid supplements on the layer thickness of colon broiler chicken. The sample consisted of 24 broiler chickens. This study used a Completely Randomized Design 3x4 factorial pattern that consisted of 3 factors of supplement dosage (0; 0.5; and 1 g supplement/ kg of feed) and duration of administration (1,2,3 and 4 weeks). Samples were divided into 3 groups: control group (P1), the group with butyric acid supplementation 0.5 (P2), and group with 1 g supplement/kg of feed (P3). Every week, two broiler chickens from each group were necropsied and the colonic tissue was taken for histologically preparations using HE staining. The changes observed were on the thickness of mucosal, submucosal, muscular, and serous’s layer and then examined under a microscope and then calculated using ZEN applications. Data were analyzed using Analysis of Variance and followed by Duncan test if there is a significant difference. The results showed the difference in the dose of butyric acid did not affect the thickness of the colon layer but it affects the colon histological changes. The difference in the duration of butyric acid administration affects the thickness of the tunica mucosa, submucosa, muscular, and serosa. It is concluded that the administration of butyric acid between doses 0; 0,5; and 1 g/kg chicken food did not affect for the thickness of the mucosal, submucosal, muscular, and serous’s layer, but it affected the presence of the colon histological changes. Differences in the duration of butyric acid’s administration at week 1,2,3,4 affects the thickness of the mucosal, submucosal, muscular, and serosal’s layer of broiler chicken.

Abstract

Propolis is a substance produced by bees that has some benefits, such as: antioxidants and antiulkus. Paracetamol is a relatively safe drug for gastric, but in some studies claimed that high doses of paracetamol increase the risk of gastric mucosal disorders, such as: ulceration, desquamation, and erosion. The purpose of this study is to determine the effects of propolis in histopathological gastric rats that were given high doses of paracetamol. This study used a completely randomized design. A sample of 25 male rats were divided into 5 groups i.e.: a negative control without treatment paracetamol (P0), a positive control (P) that was given paracetamol 250 mg/kgbw, and three other groups given treatment with paracetamol 250 mg/kgbw with a variation of propolis, P1 (0.05 ml/rat), P2 (0.10 ml/rat), and P3 (0.15 ml/rat). The treatment was given in 10 days, then necropsy was done and gastric organs were taken. Histologic gastric specimen for histopathological examination was made by using the Kiernan method (1990) and stained with hematoxylin eosin (HE). Variables examined include histopathological lesions of erosion, desquamation, and ulceration of the gastric epithelium. Results indicate that in treatment P severe damage to the gastric (desquamation) has occurred, while the entire treatment P1, P2, and P3 show the repair from the damage caused by the paracetamol. Treatment P3 gives the best results in reducing the effects of the paracetamol. This study concluded that the provision of propolis may reduce the histopathological side effects on white rat’s gastric givenhigh dose of paracetamol.

Abstract

Tanaman kedondong sering dimanfaatkan oleh masyarakat sebagai obat alternatif  untuk mengobati berbagai macam penyakit. Sedangkan penelitian tentang toksisitas daun kedondong pada ginjal belum pernah dilakukan. Dalam penelitian ini tikus putih (Rattus norvegicus) dibagi secara acak menjadi lima kelompok, masing-masing kelompok berjumlah 5 tikus putih. Kelompok A sebagai kontrol (placebo) yang diberi aquades peroral; kelompok B diberikan ekstrak daun kedondong 100 mg/kg bb (0,2 ml/ekor); kelompok C diberikan ekstrak daun kedondong 200 mg/kg bb (0,4 ml/ekor); kelompok D diberikan ekstrak daun kedondong 300 mg/kg bb (0,6 ml/ekor); kelompok E diberikan ekstrak daun kedondong 400 mg/kg bb (0,8 ml/ekor). Pemberian ekstrak daun kedondong dilakukan secara oral menggunakan sonde khusus yang dimasukkan langsung ke lambung dan dilakukan selama 14 hari. Nekropsi untuk pengambilan organ ginjal dilakukan pada hari ke 15. Jaringan ginjal selanjutnya diproses untuk pembuatan preparat histopatologi dengan pewarnaan Hematoxylin Eosin (HE). Pemeriksaan histopatologi pada ginjal tikus putih yang diberikan ekstrak daun kedondong tidak ditemukan adanya peradangan, degenerasi melemak dan nekrosis pada kontrol (placebo). Sedangkan ditemukan adanya nekrosis pada pemberian dengan dosis 100 mg/kg bb (0,2 ml), 200 mg/kg bb (0,4 ml), 300 mg/kg bb (0,6 ml), 400 mg/kg bb (0,8 ml). Pada pemeriksaan yang didasarkan adanya infiltrasi sel-sel radang ditemukan adanya peradangan pada dosis 400 mg/kg bb (0,8 ml). Hasil ini menunjukkan pemberian ekstrak daun kedondong (Spondias dulcis G.Forst) dengan rentang dosis 100 mg/kg bb sampai dengan dosis 400 mg/kg bb selama 14 hari, menyebabkan gangguan histopatologi pada organ ginjal tikus putih (Rattus novegicus).

Abstract

Canine distemper is a contagious disease, which can infect any dog species includingdomestic dog in Bali. This research used ten domestic dogs in Bali, which were naturallyinfected with canine distemper virus and have been positively confirmed by laboratory testusing RT-PCR method. Lung tissue changes have been identified on gross pathologicalobservation, i.e.: on average the lungs were changed in color and size. The lungs mainlychanged into pale or darker; in addition, the lungs tended to become bigger. Mean while,the septa alveoli of the lungs observed thicker and were infiltrated by inflammatory cells.This tissue changes indicated that canine distemper virus which infected domestic dog inBali caused inflammatory reaction in lung, which is known as interstitial pneumoniainterstitial. These changes implied to be related to the disturbance of the respiration tract,which was accompanied by mucus-purulent exudates

Abstract

Penelitian bertujuan untuk mempelajari mencit Balb/c sebagai hewan model penelitian virus penyakit Jembrana. Penelitian menggunakan 32 ekor mencit yang dibagi atas 2 kelompok perlakuan. Kelompok 1 digunakan sebagai kontrol (placebo), sedangkan kelompok 2 divaksin dengan vaksin limpa (JD.vacc.sp.15). Masing-masing mencit disuntikkan 0,2 ml vaksin JD secara intraperitoneal, setiap dua minggu sekali sampai 4 kali. Seminggu pasca vaksinasi ke-4, semua mencit dinekropsi dan limfosit dari limpa diisolasi untuk uji respon kekebalan seluler dengan uji MTT.Hasil penelitian menunjukkan bahwa respon kekebalan seluler akibat divaksin dengan vaksin limpa lebih tinggi (rata-rata = 1,3819) dari pada kontrol (placebo)(rata-rata = 1,2194). Analisis statistik diperoleh bahwa perbedaan tersebut bermakna (p<0,05). Hasil penelitian ini dapat disimpulkan bahwa mencit BALB/c dapat digunakan sebagai hewan model penelitian virus penyakit Jembrana. The presents study axamines the use of Balc/c mice for the animal model to study of Jembrana disease virus. Thirsty two mice were devided in two group (Treated and control group).Mice in the treated group were injected0,2 cc spleen vaccine (JD Vacc. sp.15). The mice were injected by intra peritoneal route, for every two week until 4 time of treatment.The celluler immune response of treated mice was significantly hygest that the control mice. The result of the studyindicated that the Balb/c mice is useful for the animals models to study of Jembrana disease virus.

Abstract

Bali cattle eats organic and inorganic waste in Suwung Denpasar landfill as source of feed. The waste was suspected contaminated by heavy metals Pb. The aim of this study is to determine levels of heavy metals Pb and spleen histopathologic changes of bali cattle reared in Suwung Denpasar landfill. Spleen samples were collected from 5 cattle for examination of heavy metals Pb by Atomic Absorption Spectrofotometri method and histopathology preparations made by Hematoxilin-Eosin staining. The results showed the presence of heavy metal Pb contamination which varied of 1.5024 ppm, 1.5002 ppm, 2.0267 ppm, 2.0268 ppm and 0.6818 ppm. Microscopically, showed histopathological changes the form of depletion and proliferation of lymphoid cells.

Abstract

This study aims to evaluate that the administration of ant nest reduces the side effects of toxic doses of gentamicin seen from the histopathological changes in hepar of white rats (Rattus norvegicus). This study used 24 male white rats, divided into four treatment groups, namely negative controls (P0) there were only given food and drink, positive controls (P1) administered gentamicin at a dose of 100mg/kgBB intraperitoneally for 10 days. The group (P2) was given gentamicin 100mg/kgBB intraperitoneally and ant nest extract with a dose of 250mg/kgBB orally for 10 days. The group (P3) was given 250mg/kgBB of ant nest extract for 7 days, after which ant nest extract was given at a dose of 250mg/kgBB and gentamicin with a dose of 100mg/kgBB intraperitoneally for 10 days.White rat hepar was taken and processed for making histopathological preparations, with hematoxylin eosin (HE) staining. The results showed that there was a significant difference between negative control and positive control groups. The mean score of improvement in hepar damage of white rats was significantly different between treatment P1, P2 and P3. The P2 and P3 groups that showed the most change near the mean score of the negative control group. Giving ant nest extracts to white rats for 17 days at a dose of 250mg/kgBB can improve the side effects of the administration of gentamicin 100mg/kgBB in the hepar of white rats.

Abstract

The purpose of this study is to determine the effect of propolis on kidney histopathological representation of white rats were given the toxic dose paracetamol. 25 male rats males were used in this study who were divided into 5 groups, control group (P0) without treatment, group 1 (P1) that was given paracetamol doses 250mg/kg, group 2 (P2), group 3 (P3), and group 4 (P4), given paracetamol dose of 250mg / kg and propolis added 0.25 ml (P2), 0.50 ml (P3), and 0.75 ml (P4). Paracetamol and propolis were given orally by mixing it into the water provided ad libitum for 10 days. Then performed a necropsy and kidney were taken aseptically for making of histopathology preparations with hematoxylin eosin (HE) staining. The examined variables were fatty degeneration and necrosis in the proximal tubules of the kidney. Results indicate that fatty degeneration and necrosis were showed in treatment P1, while the entire treatment P2, P3, and P4 show the repair from the damage caused by the paracetamol. Treatment P4 gives the best results in reducing the effects of the paracetamol. Kruskal-Wallis showed a significant difference from fatty degeneration and necrosis of the tested group. From this study it can be concluded that the propolis with dose 0.75 ml giving better protection than 0.25 ml and 0.50 ml dose.

Abstract

Provision of dexamethasone for the long term and large doses may cause disturbances in the brain. To prevent disruption to the brain, it needs antioxidant such as vitamin E. The aims of this study is to prove the administration of dexamethasone cause interference on white rat brain and determine the effects of vitamin E supplementation in preventing the side effects of dexamethasone in the brains of rats. This study used 25 male rats were divided into 5 groups, namely negative control was only given food and drink, positive control is given dexamethasone at a dose of 0.13 mg/kg bw, P1 given dexamethasone 0.13 mg/kg of body weight and vitamin E at a dose of 100 mg/kg bw, P2 given dexamethasone 0.13 mg/kg bw and vitamin E at a dose of 150 mg/kg bw, P3 given dexamethasone 0.13 mg/kg bw and vitamin E at a dose of 200 mg/kgbw. Treatment was given for 14 days, then the experimental animals necropsied. Brain tissue was taken and processed for making histopathology preparations using hematoxylin-eosin staining. Variables examined include perivascular edema lesions and congestion. The result found a significant difference between negative and positive control. The mean score improvement of brain damage was not significantly different between the treatment P1, P2, and P3. P1 treatment groups showed mean score approached the negative control. The conclusions of this research was the dexamethasone caused histopathological changes to rat brains. Supplementation of vitamin E up to doses of 200 mg /kg of body weight have not been able to reduce the side effects of dexamethasone on the brain.
