Abstract

Liver fluke infections is an important parasitic disease that common attack cattle, buffalo and others ruminant. The disease in the tropics is caused by Fasciola gigantica trematodes. Fasciolosis found in Bali cattle besides causing high economic loss also has the opportunity infect humans. This study aims to determine the pathology changes of Bali cattle hepatobiliary system infected by F. gigantica. In January2019 were examined a hundred of liver tissue Bali cattle. Based on the sex as many as 75 bali cattle are cows and 25 are bulls. On macroscopic examination was found fifteen bali cattles in gallbladder containing of F. gigantica. In positive Fasciolosis the surface of liver look uneven with enlargement of lymph nodes. The tissue of Bali cattle liver infected with F. gigantica is cut into 1x1x1 cm and put in a pot that has been filled with 10% neutral buffered formaline. The fixed tissue then processed in a tissue processor and stained with hematoxylin-eosin (HE). Microscopic examination was found bile ducts proliferation and blocked of the lumen accompanied by cholangio cirrhosis. Found longitudinal pieces of Fasciola giganticaliver fluke surrounded by fibrous connective tissue and multifocal coagulative necrosis with fibroblast proliferation. In some location the infiltration of neutrophil cells around the bile ducts can also be seen. Congestion, bleeding, and neutrophil infiltration are also seen in areas of necrosis. In the bile duct wall bleeding, adenomatus hyperplasia, infiltration of mononuclear cells with mild intensity and foci ofcalcification are found. It can be concluded that there was a change with severe intensity accompanied by chronic inflammation in the hepatobiliary system of Bali cattle with Fasciolosis.

Abstract

Bali cattle are known for having high adaptability to any environmental condition and have proven tosurvive in critical environments including landfills. The aim of this study was to determine the relationshipbetween Serum Glutamic Pyruvic Transaminase /Serum Glutamic Oxaloacetic Transaminase (SGPT/SGOT) levels and lead (Pb) level in blood of bali cattle reared in the landfill Denpasar. A total of 22Balicattle were used in this study was. The blood was drawn from the jugular vein and 10 ml blood werecollected using tubes containing EDTA 0.5% and 10 mL without EDTA tubes. Levels of SGPT and SGOTwere measured by using Auto analyzer (Refloton (R) plus) method, while the measurement of Pb in bloodplasma was conducted by using atomic absorption spectrophotometry (AAS) method. Result of the SGPTand SGOT measurement obtained were relatively varied with an average of SGPT = 29.96±5.67 U/L andSGOT = 78.82±17.62.U/L. Similarly, Pb measurement results vary widely between 4.004 to 10.216 withan average of 6.595±1.85 ppm. Correlation analysis showed that there was an association between SGOTwith Pb levels. However, no correlation was observed between the SGPT with Pblevels. These resultsindicate that the levels of lead may cause pathological tissue cells in organs other than liver. Its concludedthat heavy metalPb is found in the blood of bali cattle reared in Denpasar city landfill, and it may becorrelated with the level of the SGOT and may cause pathological tissue cells organs.

Abstract

This research was aimed to observe serum turbidity and blood sedimentation rate (ESR) as a predictorof hepatic damage in bali cattle. Two hundred whole blood and sera from 80 male and 120 female bullswere sampled from Mambal abbatoir, Badung, Bali. All blood were examined for their ESR and sera werefor their turbidity using ZnSO4 solution, besides we observed some hepatic damages pathology anatomicallywithout incision such as : internal bleeding, formation of connective tissue, swelling, and bile-ductenlargement. all of those damages were scaled graded from 0 (without abnormality) to 4 (more than 75%liver surface has abnormalities). Results showed all 86% bulls (male 32% and female 54%) have their seraturbidity ranged from 1,01-2,00, besides, 89% bulls (45% male and 44% female) have their ESR rangedfrom 3-8 mm in 24 hours. Most of liver abnormalities were : swelling (58%), bile-duct enlargement (50,5%),connective tissue formation (73%), and bleeding (59%) most of them were falling in scale 1 (less than 25%of liver surface area). Also, ESR has positive correlation (P<0,05) with connective tissue formation andserum turbidity value could be used to predict internal bleeding and connective tissue formation. It couldbe concluded ESR value could be used as a predictor of connective tissue formation and serum turbidityvalue could be used as a predictor for internal bleeding and connective tissue formation in bali cattle.

Abstract

A relatively high incidence of spongiform lesions (61%) and perivascular cuffing (89%) in brains ofdogs that suffered from rabies in Bali was an interesting background to do further studied. This study aimwas to identify the association between Negri bodies, spongiform and perivascular cuffing in dog’s brainthat infected with rabies in Bali. The research used 28 of dog’s brain samples infected with rabies and havebeen diagnosed using fluorescence antibody technique (FAT) in the Veterinary Disease InvestigationCenter, of Denpasar. Each of the brain samples contained of cerebrum, cerebellum and hippocampus thatwere taken for histological examination using hematoxylin-eosin staining. The presence of negri bodies, spongiform, and perivascular cuffing were analyzed for their association. The results showed thatcombination lesions of Negri bodies and spongiform found in the cerebellum (14%), cerebrum (4%), andhippocampus (32%), while combination lesions of perivascular cuffing and Negri bodies found in cerebellum(18%), cerebrum (7%), and hippocampus (43%). The highest intensity combination lesion of Negri bodiesand spongiform was found in hippocampus compared to the cerebellum and cerebrum. The highest intensityfor combination of spongiform and perivascular cuffing was found in cerebrum 50%, cerebellum 32%, andhippocampus 36%. It can be concluded that there might be association between the existence of Negribodies and perivascular cuffing lesions with the existed area of rabies infected dog’s brain, but not likely asspongiform.

Abstract

Rats are commonly used as animal model in pathological and reproduction research, butunfortunately they are often infected with cysticercosis. The objective of this research was to determinethe pathological changes the of the rats (Rattus novergicus) tissues affected with cysticercus. Thisresearch using 24 of female rats. They were adapted to a new environment for a week and the feeding andwater were provided ad libitum. At the end of adaptation period rats were necropsied and the visceralorgans were examined for pathological changes especially the present of cysticercosis. The liver and kidneyof each rat were soaked in 10% phosphate buffered formalin. Following dehydration process, tissue wereembedded in paraplast, cut at 5 micron and stained with Harris hematoxylin eosin (HE). The resultshowed that 8 of 24 rats were affected by cysticercosis on the liver. The histopathological changes werenecrotic lesions and eosinophylic cells infiltration around the cysticercosis lesion. The results showed that8 of 23 rats were affected by cysticercosis. The presence of necrosis and cells inflammation could interferethe results of the study when such a rats are used. It is therefore necessary to screen rats for cysticercosis.

Abstract

Tukad Badung River is one of the potential contamination of heavy metal sare very highin the city of Denpasar. Tilapia (Oreochromis mossambicus) isa commonspecies of fish found in the river and became the object of fishing by the public. The fish is usually consume das a food ingredient forever yangler. Fish can be used as bio-indicators of chemical contamination in the aquatic environment. Determination of heavy metal bioconcentration and analysis of liver histopathology gills organs and muscles is performed to determine the content of heavy metals Pb, Cd, and Cr+6, and the influence of heavy metal exposure to changes in organ histopathology Tilapia that live in Tukad Badung. In this observational study examined the levels of heavy metal contamination include Pb, Cd and Cr+6 in Tilapia meat with AAS method (Atomic Absorption Spectrofotometric), and observe the histopathological changes in organ preparations gills, liver, and muscle were stained with HE staining (hematoxylin eosin). Low Pb content of the fish that live in Tukad Badung 0.8385 mg/kg and high of 20.2600 mg/kg. The content of heavy metals Pb is above the quality standards specified in ISO 7378 : 2009 in the amount of 0.3 mg / kg. The content of Cr+6 low of 1.1402 mg / kg and the highest Cr+6 is 6.2214 mg / kg. The content of Cr+6 is above the quality standards established in the FAO Fish Circular 764 is equal to 1.0 mg / kg. In fish with Pb bioconcentration of 0.8385 mg / kg and Cr+6 of 1.1402 mg / kg was found that histopathological changes gill hyperplasia and fusion, the liver was found degeneration, necrosis, and fibrosis, and in muscle atrophy found. Histopathologicalchangessuch asedema and necrosis ofthe liveris foundin fishwith Pb bioconcentration of 4.5225mg/kg and Cr+6 amounted to2.5163mg/kg. Bio concentration of heavy metal contamination of lead (Pb) and hexavalent chromium (Cr+6) on Tilapia ( Oreochromis mossambicus ) who lives in Tukad Badung river waters exceed the applicable standard. Histopathological changes occur in organs gills, liver, and muscle as a result of exposure to heavy metals lead and hexavalent chromium. Advised the people not to eat Tilapia that live in Tukad Badung
