Abstract

Pestisida merupakan senyawa kimia yang banyak digunakan sebagai pengendali hama pada aktivitas pertanian. Residu penggunaan pestisida ini dapat mengakibatkan polusi pada lingkungan perairan khususnya di sekitar lahan pertanian. Pestisida yang terakumulasi pada lingkungan perairan maupun hasil pertanian memberikan dampak buruk bagi manusia antara lain gangguan pada sistem organ, jaringan, perkembangan, dan pada tingkat sel mengarah ke senesen. Senesen merupakan suatu kondisi saat sel berhenti melakukan proliferasi. Sel yang mengalami senesen secara alami umumnya terjadi pada individu tua, sebagai respons terhadap pemendekan telomer. Senesen dini akibat paparan pestisida pada umumnya melibatkan mekanisme stres oksidatif, kerusakan DNA, dan disfungsi mitokondria. Senesen memicu penurunan fungsi organ yang mengakibatkan berbagai masalah kesehatan seperti kanker, osteoporosis, penyakit kardiovaskuler hingga demensia. Selain itu, senesen juga dapat menyebabkan berhentinya siklus sel punca antara lain pada mesenchymal stem cells (MSCs). Ulasan ini fokus membahas mekanisme senesen akibat paparan pestisida pada sel punca terutama MSCs. Metode yang digunakan yaitu koleksi data dan analisis dari jurnal terindeks Scopus dengan menggunakan VOSviewer. Berdasarkan hasil ulasan diketahui bahwa pestisida menginduksi senesen pada MSCs melalui jaras peningkatan konsentrasi ROS dalam sel dan penurunan aktivitas ALDH. Hal tersebut menyebabkan aktivasi p53, dan p21, yang kemudian akan menyebabkan hambatan pada CDK2 dan  pRB, berakibat pada  inaktivasi E2F serta induksi senesen. Senesen juga akan memberikan respons patofisiologis lain hingga efek tumorigenesis.

Abstract

Hematoxylin-eosin is widely utilized in the field of animal microtechniques. However, the need to develop alternative dyes from natural sources such as plants has gained attention. Several studies have shown that many plants contain secondary metabolites with the potential to be developed as natural dyes. Lonchocarpus cyanescens and Syzygium cumini are promising candidates as alternative dyes for hematoxylin, while Lawsonia inermis and Hibiscus sabdariffa have shown potential as substitute dyes for eosin. These plants contain various secondary metabolites, including anthocyanins, flavonoids, chlorophyll, betalains, alkaloids, saponins, tannins, carbohydrates, proteins, phenolics, terpenoids, quinones, coumarins, xanthones, and resins. L. cyanescens exhibits a strong binding affinity to cells and tissues, particularly testicular tissue. Dyes derived from Syzygium cumini have been shown to provide a good staining result for rat liver cells. In contrast, dyes from Lawsonia inermis can stain cytoplasmic components and muscle fibers. Additionally, the dye from Hibiscus sabdariffa is capable of staining various biological components, including sperm, nerve cells, and blood cells. The dye preparation process involved extraction from different plant organs, such as leaves, flowers, and fruit. These findings suggest that secondary metabolites from these four plants hold significant potential for development as natural dyes to replace hematoxylin-eosin in histological applications.

Abstract

Forensics plays an important role in investigating crimes because it can reveal everything related to a murder case, including the time and cause of the murder. Histopathology is the study of damage to body tissues and organs which can be used to help investigate crimes. The aim of this literature review is to study the role of histopathology applications for forensic practice. The method used is searching journals and books using Google Scholar, ScienceDirect, NCBI, and PubMed with the keywords histopathology, forensics, diagnosis, death, microscopy, disease, staining, and autopsy. The histology approach helps the pathologist make a clear determination on the cause of death. The aim of histopathology is to perfect the macroscopic diagnosis and act as an additional examination in forensic case management. One of the histopathology methods in the forensic field is the paraffin method with hematoxylin eosin staining and special stains, immunohistochemistry, and MITS. The parameters observed with this method are measurements of pulmonary fat embolism; determining the age of myocardial infarction; assessment of liver steatosis, fibrosis, and cirrhosis; determination of the age of thrombus or thromboembolism; and diagnosis of shock with various diseases. The prospect of forensic histopathology is to complement molecular diagnostic methods, so that the combination of molecular and histology methods can play a maximum role in the field of forensic pathology. The conclusion of this literature review is that histopathology can be used to confirm the diagnosis in forensic cases and has good future prospects if combined with other methods such as molecular.

Abstract

BACKGROUND Protein deficiency (PD) can lead to kidney damage. Consuming plant-based proteins may improve this condition. Bambara groundnut (Vigna subterranea)has an essential amino acid score of 80%, which is higher than other legumes; thus, it is potent in overcoming malnutrition. This study aimed to determine the effect of Bambara groundnut supplementation on kidney histology in adult female mice with PD. METHODS The study was conducted for 2 months in randomly selected female mice. These mice were grouped into the control, PD, and PD supplemented with Bambara groundnuts at 100, 200, and 300 g/kg of feed. 1 day after the last treatment, the kidneys of the mice were collected and processed histologically using the paraffin method (stained with hematoxylin and eosin and Masson’s trichrome). Parameters for observation included histopathological scoring (glomerular and interstitial space fibrosis and tubular damage), kidney histomorphometry, and organ index. Semi-quantitative data were analyzed using the Kruskal–Wallis test, while quantitative data were analyzed using one-way ANOVA (followed by Tukey’s test) and nested t-test. Statistical analysis was performed using SPSS software version 20 (IBM Corp., USA) (p≤0.05). RESULTS PD caused cell sloughing (moderate level) and dilatation (severe level) of the kidney tubules. It also reduced glomerular diameter and area by approximately 17.66% and 29%, respectively. PD and Bambara groundnut administration had no significant effects on the glomerular number, cortex and medulla thickness, distal and proximal tubule diameter, and kidney organ index (p>0.05). CONCLUSIONS Bambara groundnut (V. subterranea) administration prevented damage to the kidney’s histological structure of protein-deficient mice.
