Abstract

In the modern era, electrical energy has become a significant need that drives the large consumption of fossil fuels and their environmental impacts. Supercapacitors can reduce this large consumption of natural polymers such as cellulose acetate (CA), which can be synthesized from Gracilaria sp. Composites with CA can be an environmentally friendly alternative electrode with low toxicity. The results show that the cellulose has been successfully synthesized from the algae Gracilaria sp., which was proven by FTIR spectra analysis. The results also show that supercapacitor electrodes have been successfully made where the manufactured electrodes form a composite between CA and nickel oxide (NiO), with the highest specific capacitance and specific energy values of 15.5 × 102 F/g and 13.3 × 102 Wh/kg, respectively, on the CA-NiO2 electrode, but when the NiO concentration is higher than the CA concentration the specific capacitance and specific energy decrease as shown on the CA-NiO electrode with NiO mass of 0.6 g. Thus, the materials results of this study can be applied to electric vehicles and technology that requires electricity storage in the future.

Abstract

Sengon (Paraserianthes falcataria (L) Nielsen) is a forestry plant often developed for industrial purposes in Indonesia. Sengon is a fast-growing species and has a relatively short-cutting period. Sengon wood is widely used in the production of carpentry wood, plywood raw materials, and pulp. The purpose of this study was to optimize the transformation of the entC and pmsB genes into sengon plants using the embryo injury method through Agrobacterium tumefaciens as an intermediary and to determine the efficiency of the transformation using the injury to sengon embryos. This research was conducted from September 2022 to March 2023. The research was conducted at the Genetically Modified Organism (GMO) Tissue Culture Laboratory, Recombinant Protein (Microbe) Laboratory, and Greenhouse of the Genetic Engineering Center, National Research and Innovation Agency (BRIN) Cibinong, Bogor, Java West. The research procedure consisted of several steps, namely Preparation of MS Media, Preparation of LB Media, Preparation of Sengon Seed Infection Using Agrobacterium tumefaciens, Sterilization and Planting of Sengon Seeds, Acclimatization, and Molecular Analysis (including Sengon DNA isolation, PCR Amplification, and Electrophoresis). Transformation efficiency was calculated by the positive strain of hpt PCR bands compared to the number of transformed embryos and multiplied by 100%. In the results of the PCR analysis, it was found that one plant sample carried the hpt marker, which indicates the integration of the entC and pmsB genes. which when analyzed using transformation efficiency calculations, the data obtained for the entire transformation stage was 0.8%, where the success of the transformation obtained was still classified as low. Factors that cause the low success of transformation include the method of transformation, the growth phase of bacteria, genetic material, and culture aseptic conditions.

Abstract

Stingless bees have different local names but are more commonly known as trigona bees. One of the efforts to increase the productivity of trigona bees from their cultivation is colony propagation. The purpose of this community service is to provide training to multiply trigona bee colonies with colony splitting techniques and the introduction of several forms hive of bee. The activity was carried out in the form of a workshop which was attended by two groups of trigona beekeepers. Based on the results of the evaluation through a questionnaire, the training succeeded in increasing the knowledge of participants about the propagation and introduction of the hive form of the trigona bee colony. As a follow-up to this activity, they conducted mentoring activities for trigona beekeeper groups in Rompegading Village, Cenrana District, Maros Regency. Continuous training and mentoring programs will be able to better understand and accelerate independent trigona cultivation groups in managing their meliponiary. Increased income for breeders as direct beneficiaries of meliponiculture.

Abstract

Land-use change significantly affects carbon emissions, particularly in rapidly growing cities such as Makassar. This study uses geospatial technology to analyze the impact of land cover changes on carbon emissions and sequestration in Makassar City from 2022 to 2041. The Rapid Carbon Stock Appraisal (RaCSA) method and the QUES-C feature of the LUMENS software were utilized to model land cover changes based on the Makassar City spatial planning scenario (RTRW). The results indicate that expanding residential areas and reducing green spaces, such as mangrove forests and agricultural land, have increased carbon emissions. However, carbon sequestration in certain areas, especially in the Tallo District and reclamation areas, has increased significantly due to the establishment of local protection zones. This study highlights the importance of spatial planning that supports the protection and restoration of green spaces to enhance carbon sequestration capacity and support sustainable development strategies in Makassar City.

Abstract

Kegiatan pembesaran ikan kakap putih di beberapa Kabupaten di Sulawesi Selatan yang menggunakan benih yang diproduksi dari balai perikanan di Sulawesi Selatan dan Ambon telah berkembang selama beberapa tahun terakhir. Akan tetapi, benih yang dihasilkan dari kegiatan pembenihan tersebut memiliki kualitas yang masih rendah yang diduga karena keragaman genetik yang rendah. Penelitian ini menggunakan metode Random Amplified Polymorphic DNA, RAPD untuk menganalisis variasi genetik ikan kakap putih tipe liar dari Bulungan Kalimantan Utara dan ikan hasil domestikasi dari panti pembenihan di Ambon yang dibesarkan di tambak rakyat di Desa Borongkalukua Kabupaten Maros Sulawesi Selatan. Hasil penelitian menunjukkan bahwa ikan kakap putih tipe liar memiliki nilai heterozigositas yang lebih tinggi dibandingkan dengan ikan kakap putih hasil domestikasi. Jarak genetik antar individu-individu di dalam populasi tipe liar lebih tinggi dibandingkan dengan jarak genetik individu-individu hasil domestikasi. Dendogram dan pohon filogenetik yang dibentuk berdasarkan evolutionary dissimilarity menunjukkan bahwa pada tingkat ketidakmiripan 10% ikan kakap putih tipe liar dari Bulungan Kalimantan Timur membentuk klaster yang terpisah dengan ikan kakap putih hasil domestikasi dari Ambon yang dipelihara di tambak Dusun Borongkalukua Kabupaten Maros. Keragaman genetik ikan hasil domestikasi lebih rendah dibandingkan ikan tipe liar dan menunjukkan pola evolusi yang pholyphyly Hasil yang diperoleh tersebut menunjukkan bahwa marka RAPD dapat digunakan untuk menganalisis perubahan keragaman genetik antara populasi ikan kakap putih tipe liar dan hasil domestikasi serta bermanfaat sebagai informasi dalam menyusun strategi pemanfaatan wild population sebagai sumber induk dan konservasi plasma nutfah ikan kakap putih.

Abstract

Desa Oro Gading, Kabupaten Bulukumba, Sulawesi Selatan, Indonesia, merupakan sentra produksi kopi dengan luas 533,0 Ha dan populasi sekitar 1.403 penduduk. Meskipun kaya akan sumber daya alam, petani kopi di Desa Oro Gading menghadapi tantangan serius seperti keterbatasan pengetahuan dan fasilitas pengolahan kopi yang memadai, pendapatan stagnan, dan cacat produksi. Program pemberdayaan petani kopi dirancang untuk meningkatkan kualitas dan daya saing produk kopi melalui fermentasi anaerob kopi. Dengan meningkatkan pengetahuan petani, diharapkan masyarakat dapat menghasilkan biji kopi berkualitas tinggi, meningkatkan pendapatan ekonomi, dan meningkatkan kesejahteraan di Desa Oro Gading. Metode pelaksanaan kegiatan ini dimulai dari penyuluhan interaktif, demonstrasi langsung, praktik langsung, dan dilakukan evaluasi hasil. Hasil implementasi program diikuti dengan jumlah peserta 20 orang terdiri 63,64% Perempuan dan 36,36% laki-laki. Pada program ini tingkat pengetahuan masyarakat mengalami peningkatan dengan adanya materi dan praktik langsung sehingga mendukung dan merangsang motivasi masyarakat untuk mengembangkan kopi bercita rasa tinggi hasil fermentasi anaerob di desa Orogading. Hal ini sejalan dengan beberapa tujuan pembangunan berkelanjutan (SDGs) yang tertuang pada poin 1 dan 12 dalam peningkatan komoditi pertanian berbasis varietas lokal.
