Abstract

Indonesia is a mega-diversity country with numerous endemic plants distributed throughout its regions. An Indonesias’ island with the unique and the highest endemic plant species due to being located in the Wallace area is Sulawesi Island. Hopea celebica, an endemic species to Sulawesi Island, is currently categorized as endangered by IUCN. Here, we selected the ISSR primers suitable for the genetic study of H. celebica from Luwu and Konawe provenances and investigated their genetic diversity. Ten ISSR primers were employed in primer screening, and fifty H. celebicaindividuals were genetically analyzed for their genetic diversity. The selected ISSR primers for genetic diversity analysis were UBC 810, UBC 813, UBC 814, UBC 820, UBC 822, UBC 823, and UBC 827. The evaluated H. celebica individuals have high genetic diversity, and this information will be beneficial for designing H. celebica breeding and conservation strategies

Abstract

Cashew is a promising plant in Southeast Sulawesi due to its reasonably high economic value. This species has been cultivated for years in Southeast Sulawesi, particularly in South Konawe, Konawe, and East Kolaka, with 20 to 30-years-old average plant age. This research aimed to identify morphological variations at the vegetative stage of the cashew in three evaluated Southeast Sulawesi populations (South Konawe, Konawe, and East Kolaka). The descriptive observations were carried out on the leaf apex shape, leaf shape, leaf margin, leaf venation, the color of young and mature leaves, branching pattern, tree habit, ease of peeling bark from the twig, leaf texture, as well as leaf aroma. We indicated the morphological variations in the evaluated cashew. The variations were observed in leaf shape (obovate, oblong, and circular), leaf apex shape (pointed, rounded, and indented), leaf venation (curved up, curved down, and flat), branching pattern (intensive and extensive), tree habit (upright and open, upright and compact, spreading), ease of peeling bark from twigs (difficult and easy), leaf texture (smooth and rough), and aroma (mango-like and guava-like).

Abstract

Peningkatan konsumsi tomat semakin meningkat, namun produksi tomat masih lebih rendah dari potensi produksinya. Salah satu upaya dalam meningkatkan produksi tomat dapat dilakukan dengan penggunaan varietas unggul yang dapat beradaptasi baik dengan lingkungannya dan berproduksi tinggi. Penelitian ini bertujuan untuk melakukan pengujian lanjutan beberapa galur hasil persilangan Karina x Mawar terhadap karakter pertumbuhan dan produksi. Penelitian dilaksanakan di Kebun Percobaan Fakultas Pertanian Universitas Hasanuddin sejak bulan April hingga Juli 2023. Hasil yang didapatkan yaitu, galur hasil persilangan Karina x Mawar yang memberikan produksi terbaik adalah KM 69.6.8 (g8) dengan rata-rata produksi 1858.83 g dan berbeda nyata dengan kedua tetuanya, karakter yang memiliki nilai heritabilitas tinggi, yaitu karakter tinggi tanaman, tinggi dikotomus, jumlah cabang, umur berbunga, umur panen, jumlah tandan berbuah, jumlah buah total, panjang buah, diameter buah, bobot buah, jumlah biji perbuah dan produksi, serta terdapat 4 karakter yang berkorelasi positif nyata terhadap produksi, yaitu karakter tinggi tanaman, jumlah cabang, jumlah bunga pertandan dan jumlah tandan berbuah. The increase in tomato consumption is increasing, but tomato production is still lower than its production potential. One effort to increase tomato production can be done by using superior varieties that can adapt well to their environment and produce high yields. This research aims to carry out further testing of several lines resulting from the Karina x Mawar cross on growth and production characteristics. The research was carried out at the Experimental Garden of the Faculty of Agriculture, Hasanuddin University from April to July 2023. The results obtained were, the line resulting from the Karina x Mawar cross that gave the best production was KM 69.6.8 (g8) with an average production of 1858.83 g and was significantly different from the two parents, characters that have high heritability values, namely plant height, dichotomous height, number of branches, flowering age, harvest age, number of fruit bunches, total number of fruit, fruit length, fruit diameter, fruit weight, number of seeds per fruit and production, and there are 4 characters that have a real positive correlation with production, namely plant height, number of branches, number of flower bunches and number of fruit bunches.

Abstract

Sengon (Paraserianthes falcataria (L) Nielsen) is a forestry plant often developed for industrial purposes in Indonesia. Sengon is a fast-growing species and has a relatively short-cutting period. Sengon wood is widely used in the production of carpentry wood, plywood raw materials, and pulp. The purpose of this study was to optimize the transformation of the entC and pmsB genes into sengon plants using the embryo injury method through Agrobacterium tumefaciens as an intermediary and to determine the efficiency of the transformation using the injury to sengon embryos. This research was conducted from September 2022 to March 2023. The research was conducted at the Genetically Modified Organism (GMO) Tissue Culture Laboratory, Recombinant Protein (Microbe) Laboratory, and Greenhouse of the Genetic Engineering Center, National Research and Innovation Agency (BRIN) Cibinong, Bogor, Java West. The research procedure consisted of several steps, namely Preparation of MS Media, Preparation of LB Media, Preparation of Sengon Seed Infection Using Agrobacterium tumefaciens, Sterilization and Planting of Sengon Seeds, Acclimatization, and Molecular Analysis (including Sengon DNA isolation, PCR Amplification, and Electrophoresis). Transformation efficiency was calculated by the positive strain of hpt PCR bands compared to the number of transformed embryos and multiplied by 100%. In the results of the PCR analysis, it was found that one plant sample carried the hpt marker, which indicates the integration of the entC and pmsB genes. which when analyzed using transformation efficiency calculations, the data obtained for the entire transformation stage was 0.8%, where the success of the transformation obtained was still classified as low. Factors that cause the low success of transformation include the method of transformation, the growth phase of bacteria, genetic material, and culture aseptic conditions.

Abstract

Leaf litter or organic matters can decompossed by microorganisms like fungi.This study aims to determine the ability of growth and the rate of decomposition of Suren (Toona sureni Merr.) rhizosphere fungi isolates on various organic media. The testing process was carried out at the Biotechnology and Tree Breeding Laboratory, Faculty of Forestry, Hasanuddin University, Makassar. The research method consisted of making fungi growing media, rejuvenating isolates and making organic media from teak leaf litter (Tectona grandis), Jabon (Anthocephalus cadamba), and Mahogany (Swietenia macrophylla). The highest growth of mycelium was found in Fusarium sp. The growth of the five best fungi isolates in the formulation of mahogany leaf litter and the highest average decomposition rate in the combination treatment of mahogany leaf litter and Fusarium sp.

Abstract

Abstract. POLOBETE Fish Farm is a private hatchery that uses biofloc technology located in Pinrang Regency, South Sulawesi. It has 18 tilapia biofloc ponds and one nursery pond. The problem faced by POLOBETE Fish Farm is that it has not been able to meet market demand for seeds both in quality and quantity. Genetic improvement with selection methods for growth characteristics and fecundity is a solution to increase the quantity and quality of tilapia seed production at POLOBETE Fish Farm. The Fisheries Research Team from the Research and Development Centre for Biotechnology, Institute of Research and Community Service, Universitas Hasanuddin, assisted POLOBETE Fish Farm with truss morphometric methods to assess fish body shape and its association with growth traits and fecundity analysis methods to assess the number of eggs or the number of larvae produced by female tilapia broodstock. Data on the morphological and fecundity characters of selected fish can be used to assess the progress of selection. The result of the community service is that POLOBETE Fish Farm has applied both methods to assess generations/progeny resulting from crosses between Nile tilapia Oreochromis niloticus Kekar (♂) and O. niloticus Kekar (♀) and hybridization between O. niloticus Kekar (♂) and O. niloticus Sultana (♀). Furthermore, both truss morphometric and fecundity methods can be used by POLOBETE Fish Farm on production program of XY neofemale, YY supermale, and YY female. Abstrak. POLOBETE Fish Farm adalah usaha akuakultur yang menerapkan sistem bioflok yang berlokasi di Kabupaten Pinrang Sulawesi Selatan, memiliki 18 kolam terpal bioflok ikan nila dan satu buah kolam pendederan. Permasalahan yang dihadapi oleh POLOBETE Fish Farm adalah belum mampu memenuhi kebutuhan pasar akan benih baik secara kualitas maupun kuantitas. Genetic improvement dengan metode seleksi untuk karakter pertumbuhan dan fekunditas adalah solusi untuk meningkatkan kuantitas dan kualitas produksi benih. Tim Riset Perikanan dari Lembaga Penelitian dan Pengembangan Bioteknologi LPPM Universitas Hasanuddin, memberi pendampingan kepada POLOBETE Fish Farm tentang metode truss morfometrik untuk menilai morfologi ikan dan metode analisis fekunditas untuk menilai jumlah telur atau jumlah larva (fekunditas) yang diproduksi oleh induk ikan nila betina. Data karakter morfologi dan fekunditas ikan-ikan hasil seleksi tersebut dapat digunakan sebagai parameter untuk menilai kemajuan seleksi. Hasil dari pengabdian adalah POLOBETE Fish Farm telah mengaplikasikan kedua metode tersebut untuk menilai progeny hasil persilangan antara Oreochromis niloticus Kekar (♂) dan O. niloticus Kekar (♀) serta hibridisasi antara O. niloticus Kekar (♂) dan O. niloticus Sultana (♀). Metode truss morfometrik dan fekunditas selanjutnya dapat digunakan oleh POLOBETE Fish Farm pada program produksi XY neofemale, YY supermale, dan YY female.

Abstract

Parentage analysis has been used to evaluate pollen dispersal in Kopyor coconut (Cocos nucifera L.). Investigations were undertaken to elucidate (i) the dispersal of pollen, (ii) the rate of self and out-crossing pollination, and (iii) the distance of pollen travel in Pati kopyor coconut population. The finding of this activities should be beneficial to kopyor coconut farmers to increase their kopyor fruit harvest and to support breeding of this unique coconut mutant. As many as 84 progenies were harvested from 15 female parents. As many as 95 adults coconut provenances surrounding the female parents were analyses as the potential male parents for the progenies. The adult coconut palms were mapped according to their GPS position. All samples were genotyped using six SSR and four SNAP marker loci. Parentage analysis was done using CERVUS version 2.0 software. Results of the analysis indicated that evaluated markers were effective for assigning candidate male parents to all evaluated seedlings. There is no specific direction of donated pollen movement from assigned donor parents to the female ones. The donated pollens could come from assigned male parents in any directions relative to the female parent positions. Cross pollination occured in as many as 82.1% of the progenies analyzed. Outcrossing among tall by tall (TxT), dwarf by dwarf (DxD), hybrid by hybrid (HxH), TxD, DxT, TxH, DxH, and HxD were observed. Self-pollination (TxT and DxD) occurred in as many as 17.9% of the progenies. The dwarf coconut was not always self pollinated. The presence of DxD, TxD, and HxD outcrossing was also observed. The donated pollens could come from pollen donor in a range of at least 0-58 m apart from the evaluated female recipients. Therefore, in addition to the wind, insect pollinators may have played an important role in Kopyor coconut pollination.

Abstract

Ebony is one of slow-growing spe- cies), that of suspected to be the triggering factors to prone to extinction. In addition, the nature of ebony seeds is recalcitrant, so they cannot be stored for a long time. Other factors affecting seed germination are the degree of maturity, size, and weight of the seeds. Seeds harvested before the physiological maturity level would not have high viability and even could not germinate well due to the in-sufficient of food reserves, and un-perfect embryo formatio. Bio-fertilizer is used to help accelerate the growth of ebony. This study aims to analyze the combination of bio fertilizers which would be giving a better effect on the physiological activity and growth of ebony seedlings. Morphological and physiological variables measurement were used in this study. The research design used a Completely Randomized Design at a real level of 5%. If the fingerprint results obtained have a real effect, then a further test of the Duncan Multiple Range Test is carried out. Re- search results showed that seed coating treatment of 45% Penicillium microbial solution + seed coating) gave the best results in each variable of both growth and physiology of ebony seedlings, while in the treatment without seed coating, Sargassum microbial solution 30% indicates good results in each variable of growth and physiology of ebony seedlings. Treatment with the immersion of seeds in microbes can produce a high percentage in each variable.

Abstract

Pengabdian ini bertujuan untuk meningkatkan pengetahuan dan kemampuan pemasaran produk secara online bagi para petani dan peternak di desa Buntu Mondong, Kabupaten Enrekang, dengan menggunakan situs web integrasi WhatsApp. Kegiatan yang dilakukan terdiri dari: 1) Pelatihan penggunaan website e-commerce, 2) Sosialisasi Pengenalan Teknologi, 3) Persiapan dan Observasi, dan 4) Pemantauan dan penilaian program. Alat penilaian yang digunakan adalah observasi langsung terhadap proses pengenalan teknologi dan kuesioner pra dan pasca tes. Untuk mengukur pertumbuhan pengetahuan dan kemampuan, data dianalisis secara deskriptif. Hasil dari layanan ini menunjukkan peningkatan yang signifikan dalam pengetahuan dan kemampuan teknis petani dan peternak berdasarkan situs web integrasi aplikasi WhatsApp. Semua anggota mitra juga mengalami peningkatan dalam hal selain itu Mitra juga mengalami peningkatan  secara mendiri  mengelola websitenya melalui ponsel dalam menjual produknya.

Abstract

Unique and varied bioactive compounds produced by the ocean have drawn attention and served as a focus for creating antibacterial and anti-inflammatory agents. As part of the approach for locating these research sources, databases such as PubMed, Science Direct, MDPI, Google Scholar, Springer Link, Web of Science, Scopus, and Wiley Online Library were used to identify completed studies. Numerous intriguing bioactive compounds have so far been isolated from marine bacteria. A crucial resource in the ongoing search for novel peptides, proteins, lipids, nucleosides, enzymes, alkaloids, polyketides, and terpenoids is the diversity of marine bacterium strains. This review summarizes several bacterial intracellular and extracellular bioactive compounds that have been applied as antibacterial and anti-inflammatory agents in 2016–2024, which we present in the form of structures, species sources, and evaluations of these compounds' antibacterial and anti-inflammatory activities. Therefore, this review article can be used as a recommendation for utilizing biomaterials from marine bacteria that are promising in the future for industrial-scale production of antibacterial and anti-inflammatory agents.
