Abstract

The effect of mango ginger (Curcuma mangga Val.) ethanolic extract has been experimentally tested to β cell of alloxan-induced mice at the Laboratory Animal Development and Structure. The experiment aimed to identify the effectifity of the ethanolic extract to defend β cell from necrosis caused by the alloxan. The activity of the extract was evaluated by using control (A0 and A1) and treatments. All mice were induced by alloxan (150 mg/kg of body weight). One week later the extract was given orally to the mice at dose 200 (A2) 400 (A3) and 800 (A4) mg/kg of body weight (A4). The extract was given everyday for 21 days. Pancreas were isolated for each week. The observation of pancreas histology showed that extract 200 mg/kg body weight could decrease necrosis at Langerhans higher than the extract of 400 and 800 mg/kg of body weight. This results indicate that giving mango ginger extract at dose 200 mg/kg body weight could decrease β cell necrosis.Key words : mango ginger (Curcuma mangga Val.), β cell, alloxan, necrosis.

Abstract

Tropidolaemus wagleri commonly known as complex species who widely distributed at tropical areas in Southeast Asia. We predicted that widespread distribution may affect the morphological variation among different population geographycally. This study were conducted based on direct survey and sampling in the fieldwork. The study sites was divided into two categories based on Bukit Barisan range which streching from North to South Sumatra island consist of Padang, Padang Panjang in western part of Bukit Barisan, and Payakumbuh, Dharmasraya in eastern part of Bukit Barisan. In total 21 individual T.wagleri were analysed based on 20 mensural characters (morphometric) and 19 multistate characters (meristic). Male and female was analysed separately based on non parametric statistics Kruskall Wallis and Mann Whitney U Test. The relationship between population were analysed based on UPGMA tree and Principal Component Analysis (PCA) plot. The result showed the significant variation between female T. wagleri between western and eastern part of Buki Barisan range. In contrast, the variation has not detected among the population of T. wagleri male. The morphological character which showed the significant value are Head Length (HL) and Neck Scales Row (NSR). The west population of T. wagleri has more shorter Head Length (HL) and Snouth Length (SnL) than east population. Howefer, the west T. wagleri population has more number in Neck Scales Row (NSR) character than east population. The population of female T. wagleri in Padang Panjang has more close relationship to Padang than to female population T. wagleri in Dharmasraya based n UPGMA tree.

Abstract

The Indonesian archipelago is an ideal setting for the study of speciation and biogeography. This archipelago is divided into three island groups based on zoogeography: Sundaland, Wallaceaand the Australian region. In this paper we used frogs in genus Fejervarya (Bolkay) to study biogeography and examine patterns of gene flow across proposed zoogeographic boundaries. Severalmolecular studies on Fejervarya species from Indonesia have been carried out, but comparative studies among members of the genus Fejervarya have yet to be performed. In order to elucidate genetic divergence and geographic distribution of these frogs, we conducted a molecular analysis of the mitochondrial 16S rRNA gene using 179 frogs from five Fejervarya species. In total we collected from 32 localities in Sumatra, Kalimantan (Indonesian part of Borneo), Java, Bali, Sulawesi and Lesser Sunda Islands in Indonesia. Molecular phylogenetic analysis recovered 35 haplotypes and showed that frogs in the genus Fejervarya were divided into two well-supported clades. The first group were of three species, F. limnocharis, F. iskandari and F. cf. verruculosa and the other group clade consisted of Fejervarya cancrivora and Fejervarya sp. (Sulawesi-type). The average sequence divergence among these four species ranged from 1.09 to 16.03% (mean = 11.29±2.83%). The present results clearly show that there are five Fejervarya species in the Indonesian archipelago. Fejervarya limnocharis and F. cancrivora are widely distributed and sympatric in Sumatra, Borneo and Java. Fejervarya iskandari is not endemic to Java and also occurs in the Lesser Sundas. Fejervarya cf. verruculosa and Fejervarya sp. (Sulawesi-type) are endemic to Lesser Sunda and Sulawesi Island, respectively. Abstract

Population of Bilih Fish (Mystacoleucus padangensis) in Singkarak Lake has decreased as the effect of intensive exploitation and changes in water use function. Many appropriate works have been conducted for the fish conservation. One of them is introducing Bilih fish population to a new habitat,  Toba Lake. After nine years of releasing the fish, there was a hypothesis that these fish had undergone some changes on their morphology compared to their original population in Singkarak Lake. The changes could occure in some morphological characters and also the increase or decrease of their growth. This study was aimed at comparing morphology characters between Bilih fish introduced to Toba Lake with the original population in Singkarak Lake. Survey method was used and 400 samples of fish were taken from each Singkarak and Toba Lakes.  Parameters measured were 29 morphological characters. Data were analyzed using Mann Whitney U Test with SPSS 22. The results showed that 27 morphological characters of introduced fish had changed. Characters that had not changed were dorsal fin (anterior end)-anal fin distance (DFAF) and preanal distance (PD). Population of Bilih fish in Toba Lake might keep changing in the process of morphological character differentiation as long as environment factors still support the life of Bilih fish.

Abstract

Abstract

Bufonidae is widely distributed amphibian family around the world, including Indonesia (Sumatra) and Asia. Sumatra is an island which has separated because of sea level increase and create gene flows barrier for amphibian in Sumatra and Asia. The purpose of this research is to analyse Bufonidae phylogenetic which exist in several locations in Sumatra and Asia based on gene cytochrome b analysis. Samples were collected from six localities in West Sumatra. DNA extraction and amplification has been done in Genetics and Cell Biology Laboratory, Andalas University, whereas DNA sequencing were analyzed at MacroGen USA DNA Sequencing Laboratory, South Korea. About 14 tissue samples of Bufonidae from West Sumatra has been analyzed. The relationship of Bufonidae phylogenetics in Sumatra and Asia were divided into three main clusters. Duttaphrynus melanostictus in Sumatra is closer to Duttaphrynus melanostictus from India with 0,3-0,5% sequence divergence and it is a group of paraphyletic with Duttaphrynus melanostictus from Vietnam, Taiwan, and China.
