Abstract

This study evaluates the effects of NaOCl concentration and immersion time on contamination levels and viability of in vitro mangosteen seed explants, aiming to identify the optimal sterilization protocol. Conducted at YAHDI Tissue Culture Laboratory, Medan, Indonesia, a Randomized Complete Block Design tested three treatment combinations: (1) 15% and 10% NaOCl for 5 minutes, (2) 20% and 15% NaOCl for 15 minutes, and (3) 30% and 20% NaOCl for 25 minutes. After immersion, explants were rinsed three times with sterile distilled water. Observations spanned 12 MST, with data analyzed using ANOVA and Games-Howell post hoc tests; contamination sources were qualitatively described. Results showed the first combination had the lowest effectiveness, the second was better but suboptimal, while the third significantly delayed contamination and yielded the highest explant survival rate of 86.1%. The third treatment also prolonged the contamination-free period compared to others. Fungal and bacterial contaminants were predominant, highlighting the need for effective disinfectants against both. These findings indicate that the third combination is a promising sterilization protocol for mangosteen seed explants, especially for hard endocarp tissues prone to contamination.

Abstract

 Penelitian ini bertujuan untuk Untuk mendapatkan teknik sterilisasi yang terbaik untuk eksplan batang anggrek Cattleya sp. muda. Penelitian ini dilakukan di Laboratorium Kultur Jaringan YAHDI Perum Pelabuhan Jl. Lambung No. 8 Tanah 600 Medan Marelan. Rancangan penelitian yang digunakan dalam penelitian ini adalah metode eksperimental laboratorium dalam mencobakan beberapa teknik sterilisasi. Untuk mengetahui teknik sterilisasi yang tepat, penelitian ini menggunakan eksplan lapang anggrek Cattleya Sp. dengan jenis pendekatan deskriptif kuantitatif. Data dianalisis dan ditabulasi menggunakan parameter penelitian hari terkontaminasi, persentase eksplan yang terkontaminasi, persentase eksplan browning, persentase eksplan yang membengkak, persentase eksplan yang membentuk bakal tunas dan persentase eksplan hidup. Hasil penelitian ini menunjukkan bahwa terdapat 2 teknik sterilisasi yang optimal dalam sterilisasi eksplan lapang Anggrek Cattleya Sp. yaitu teknik sterilisasi 2 dan 4. Teknik sterilisasi 2 menggunakan zat sterilan deterjen, akuades steril, Tween-20, benomil 50%, agrimicin, NaOCl 15% 10% dan alcohol 70% kemudian teknik sterilisasi 4 dengan zat sterilan Clorox 1,05%, HgCl 0,02% dan akuades steril merupakan teknik sterilisasi yang tersebut. Hal ini ditunjukkan dengan rendahnya tingkat kontaminasi eksplan dan tampaknya spot-spot hijau pada eksplan dimana eksplan akan mengalami pertumbuhan tunas. Faktor-faktor yang menyebabkan kontaminasi pada eksplan Anggrek Cattleya Sp. adalah kurangnya kehati-hatian praktikan pada saat melakukan sterilisasi eksplan lapang, kontaminasi endogenus yang tinggi dan penggunaan zat steril yang kurang tepat

Abstract

This research aims to determine the effect of from kepok banana peel extract (Musa acuminata balbisiana Colla) and BAP and their interaction on the growth of red potato plantlets (Solanum tuberosum L.) in vitro. This research was conducted in September-October 2023 at the G10 Agrotech Medan Plant Tissue Culture Laboratory Jl. Sei Bahorok No. 47 F Medan Baru. This research used a factorial Completely Randomized Design (CRD) with two factors. The first factor is the concentration of Kepok banana peel extract in 3 levels, namely (0, 25 and 50 g/L) and the second factor is BAP which consists of 4 levels, namely (0, 0.5, 1 and 1.5 mg/L). 12 treatment combinations were obtained and each treatment was repeated 3 times to obtain 36 experimental units. The research parameters were plantlet height, number of shoots, number of leaves and number of roots analyzed using Analysis of Variance (ANOVA) and if they were significantly different, further tests would be carried out using the Duncan Multiple Range Test (DMRT) at the 5% level. The results showed that the treatment of kepok banana peel extract and BAP had a significant effect on plantlet height, number of shoots, number of leaves and number of roots. The highest average plantlet height was produced by banana peel extract 50gr/L (7.60 cm), BAP 1mg/L (6.70 cm). The highest number of shoots was produced by banana peel extract 50gr/L (6.33 shoots), BAP 1.5 mg/L (6.33 shoots). The highest number of leaves was produced by sweet corn extract 25 gr/L, namely (10.00 leaves), BAP 0.5 mg/L as much as (8.00 leaves). The highest number of roots was produced by the 50gr/L sweet corn extract treatment, namely (12.00 roots), the highest number of roots was produced by BAP 1 mg/L, namely (7.33 roots). The highest average plantlet height, number of shoots and leaves were produced by the A2B1 treatment interaction, namely 7.40 cm plantlet height, 6.33 shoots and 16.33 leaves. Abstract

This research aims to determine the relationship between students' visual, auditory and kinesthetic learning styles with students' biology learning outcomes. The type of research used is descriptive research with a quantitative approach. In this study there were 71 people using a sampling technique, namely random sampling. The instruments used in data collection were questionnaires and tests. The research results showed that students in class XI Science at SMA Negeri 1 Beringin consisting of classes XI IPA 1 and IPA 2 tend to have a visual learning style with 41 students (57.75%), while students who have an auditory learning style are 16 people (22.54%), and students who have a kinesthetic learning style are 14 people (19.71%). This research shows that the visual learning style variable (X1) has rcount = 0.601 > rtable = 0.308, the auditory learning style variable (X2) having rcount = 0.536 > rtable = 0.497 and the kinesthetic learning style variable (X3) has rcount = 0.547 > rtable = 0.533. Based on the research results, it was concluded that student learning styles greatly influence students' biology learning outcomes.

Abstract

Biotechnology is a discipline of biology that examines the use of methods or how to utilize living things and its components in terms of increasing the welfare of human existence. Biotechnology courses currently require instructional materials, particularly those pertaining to the application of DNA isolation and amplification. The aim of this study was to determine the level of validity of research based book on isolation, amplification, and visualization material for biotechnology course in higher education. The  parameters  measured were the validity of the research based book from the aspect of material, language, context, and design. This study uses developmental stage that followed Borg and Gall (1989) development model. The results of the validation by experts were obtained using a questionnaire instrument. The research based book on isolation, amplification, and visualization topic passed validation by materials experts, linguists experts, and design experts with scores of 84% (very good), 77% (very good), and 85%. (very good). It can be concluded that research based book on isolation, amplification, and visualization material product is feasible to use for learning in biotechnology course in higher education based on its validity.
