Abstract

 Penelitian Penelitian ini bertujuan untuk mendeteksi antibodi spesifik terhadap virus Avian Influenza subtipe H5N1 pada burung merpati. Penelitian ini dilaksanakan di Laboratorium Mikrobiologi Fakultas Kedokteran Hewan Universitas Syiah Kuala, Darussalam Banda Aceh. Penelitian ini menggunakan sampel serum darah burung merpati sebanyak 25 sampel. Pengambilan sampel darah merpati menggunakan spuit 3cc melalui vena brachialis di kiri ataupun kanan bawah sayap merpati, kemudian didiamkan selama 6-10 jam dengan posisi miring. Darah dibiarkan  hingga terpisah dari serum, kemudian serum dikoleksi dan disimpan di suhu -20°C. Serum sebelum digunakan  terlebih dahulu di simpan pada suhu 56°C selama 30 menit untuk menginaktifasi komplemen. Pemeriksaan sampel tersebut menggunakan uji Hemaglutinasi Inhibisi (HI). Hasil dari penelitian ini menunjukkan bahwa dari 25 sampel serum yang diperiksa, 22 sampel (88%) positif mengandung antibodi terhadap virus Avian Influenza subtipe H5N1 namun 3 sampel (12%) tidak mengandung titer antibodi terhadap virus Avian Influenza subtipe H5N1. Dapat disimpulkan bahwa merpati tersebut pernah terpapar oleh virus Avian influenza subtipe H5N1. Kata kunci: merpati, avian influenza, antibodi, uji hiABSTRACT This study aims to detect specific antibodies of  Avian Influenza virus H5N1 subtype on pigeons. This study was carried out at the Microbiology Laboratory, Faculty of Veterinary Medicine, Syiah Kuala University, Banda Aceh. This study used 25 pigeon blood serum samples. Pigeons blood sampling using 3cc syringe through the brachial vein on the left or right under the pigeons wings, then allowed to stand for 6-10 hours with a sloping position. The blood is left frozen so that the serum is separated, then the serum is collected and stored at -20 ° C. Serum samples of pigeons before being used inactivated first at 56 ° C temperature for 30 minutes. Examination of these samples uses the Hemaglutination Inhibition (HI) test. The results of this study showed that the 25 examined serum samples, 22 positive samples contained antibody titers of the H5N1 subtype Avian Influenza virus and only 3 negative samples contained antibody titers against the Avian Influenza virus subtype H5N1. The presence of antibodies formed in the pigeon's serum showed that the pigeon had been exposed to the H5N1 subtype Avian Influenza virus. Keywords: pigeon, avian influenza, antibody, hi test

Abstract

ABSTRAK          Telur itik merupakan salah satu produk hasil pengawetan telur dengan cara pengasinan. Penelitian ini bertujuan mengetahui pengaruh penambahan air perasan jeruk purut (Citrus hystrix) pada proses pembuatan telur asin terhadap jumlah total cemaran bakteri. Metode penelitian yang digunakan adalah metode rancangan acak lengkap (RAL) yang terdiri atas 4 perlakuan dan 3 kali ulangan. Perlakuan yang diberikan adalah penambahan larutan perasan jeruk purut dengan konsentrasi 0%, 10%, 20% dan 30% ke dalam media adonan pengasinan telur itik. Pengamatan jumlah total bakteri dilakukan setelah proses pengasinan telur dilakukan yaitu setelah 14 hari masa penyimpanan telur. Pengamatan jumlah total bakteri dilakukan dengan  metode Total Plate Count (TPC) menggunakan media Plate count agar (PCA).  Data jumlah bakteri yang diperoleh di analisis secara statistik menggunakan Analysis Of Variance (ANOVA). Hasil penelitian menunjukkan bahwa penambahan larutan perasan jeruk purut dengan konsentrasi 0%, 10%, 20% dan 30% ke dalam adonan pengasin telur selama proses pembuatan telur asin dapat menurunkan jumlah total bakteri secara signifikan. Hasil uji lanjut menggunakan uji Duncan diketahui bahwa penambahan larutan perasan jeruk purut dengan konsentrasi 30% secara nyata dapat menurunkan jumlah total bakteri pada telur asin yang dihasilkan dibandingkan dengan konsentrasi 0%, 10%, 20%. Sedangkan pembahan larutan perasan jeruk purut dengan konsentrasi 10% sampai dengan 20% tidak dapat menurunkan jumlah total bakteri pada telur asin yang dihasilkan. Berdasarkan penelitian ini dapat disimpulkan bahwa penambahan larutan perasan jeruk purut dengan konsentrasi 30% ke dalam adonan pengasin telur dapat menurunkan jumlah total bakteri pada telur asin yang dihasilkan. ABSTRACT            Duck egg is a product of egg preservation by salting.This study aims to determine the effect of adding citrus hystrix to the process of making salted eggs to the total amount of bacterial contamination. The research method used is a complete randomized design method (RAL) consisting of 4 treatments and 3 replays. The treatment given is the addition of a solution of orange juice purut with a concentration of 0%, 10%, 20% and 30% into the medium of duck egg marinating dough. Observation of the total number of microbes is done after the process of salting eggs is done after 14 days of egg acidification. Observation of the total number of microbes is done by the Method of Total Plate Count (TPC) using media Plate count agar (PCA) Data on the number of microbes obtained in statistical Analysis Of Variance (ANOVA). The results showed that the addition of a solution of orange juice purut with concentrations of 0%, 10%, 20% and 30% into the egg marinating dough during the process of making salted eggs can significantly decrease the total number of microbes. The results of further tests using Duncan's test found that the addition of a solution of orange juice purut with a concentration of 30% can significantly decrease the total number of microbes in the salted eggs produced compared to the concentration of 0%, 10%, 20%. While the deity of orange juice solution purut with a concentration of 10% to 20% can not decrease the total number of microbes in salted eggs produced. Based on this research, it can be concluded that the addition of a solution of orange juice purut with a concentration of 30% into the egg marinating dough can decrease the total number of microbes in the resulting salted eggs.

Abstract

Abstract              The crop is the part of the chicken's digestive tract that is more likely to break during processing. This research aimed to isolate Salmonella sp. from crop in local chickens and broiler from the Peunayong Chicken Market in Banda Aceh. Samples were swab contents of crop from 15 crops of local chickens and 15 crops of broiler. This research is a Cross sectional study with Carter method, starts with a swab of crop contents and then is inserted into the Selenite Cystine Broth (SCB) media, if the color of the media becomes orange, then followed by streaking bacteria on Salmonella Shigella Agar (SSA) media. Then observed the morphology that grew on SSA media including size, shape, surface, elevation, edges, aspects and colony color, followed by Gram stain to observe bacterial morphology microscopically. The research data were analyzed descriptively, displayed in figures and tables. The results of the isolation showed that 7 from 15 samples (46%) of local chicken cropping and 15 from 15 samples (100%) of broiler were positive for Salmonella sp. Therefore, it can be concluded that Salmonella sp. can be isolated from local chicken and broiler, Salmonella sp. prevalence is higher in broiler than in local chicken.Keywords : Eviserasi; Food borne disease; Gram negatif; Zoonotic disease Abstrak              Tembolok merupakan bagian saluran pencernaan ayam yang lebih mungkin pecah selama pemrosesan. Penelitian ini bertujuan untuk mengisolasi Salmonella sp. pada tembolok ayam buras dan ayam ras yang berasal dari Pasar Ayam Peunayong Kota Banda Aceh. Sampel berupa swab isi tembolok dari 15 tembolok ayam buras dan 15 tembolok ayam ras. Penelitian ini merupakan Cross sectional dengan metode Carter dimulai dari swab isi tembolok kemudian dimasukkan kedalam media Selenite Cystine Broth (SCB), apabila warna media menjadi orange dilanjutkan dengan penanaman bakteri pada media Salmonella Shigella Agar (SSA). Kemudian diamati morfologi yang tumbuh pada media SSA meliputi ukuran, bentuk, permukaan, elevasi, tepi, aspek dan warna koloni, dilanjutkan dengan pewarnaan Gram untuk mengamati morfologi bakteri secara mikroskopis. Data hasil penelitian dianalisis secara deskriptif, ditampilkan dalam bentuk gambar dan tabel. Hasil isolasi didapatkan bahwa pada 7 dari 15 (46%) sampel tembolok ayam buras dan 15 dari 15 sampel (100%) sampel tembolok ayam ras positif terdapat Salmonella sp. Oleh sebab itu, dapat disimpulkan bahwa Salmonella sp. dapat disolasi dari tembolok ayam buras dan ayam ras, prevalensi Salmonella sp. lebih tinggi pada ayam ras dibandingkan ayam buras.Kata kunci : Eviserasi; Food borne Disease; Gram negative; Zoonotic disease

Abstract

The objectives of this study were to assess the diversity of Newcastle Disease Virus (NDV) isolates; to detect and isolate NDV from poultry; and to identify and characterize NDV by serological and molecular assays. A total of 84 cloacal-oropharynx isolates of poultry was collected from privately owned poultries and poultry markets from 12 districts in Aceh Besar and Banda Aceh. Screening was performed by real time reverse transcriptation-polymerase chain reaction (rRT-PCR) to 15 isolates of poultry. Selected isolates were inoculated in 9-11 days old embryonated specific pathogen free (SPF) eggs and showed positive hemagglutination (HA). Characterization was performed through hemagglutination inhibition (HI) test using Komarov and Hitchner B1 antisera, elution test, RT-PCR and realtime RT-PCR fusion (F). All isolates had a higher affinity to Komarov antisera (titer up to 4 log), indicating virulent strain. This was supported by elution test which showed that 93.66% isolates were virulent and 6 % non-virulent. In conclusion, RT-PCR can detect Matrix gene from 15 isolates (100%), while Fusion gene only detected from 11 isolates (73.3%). rRT-PCR is more capable of detecting antigenic diversity compared to RT-PCR.

Abstract

Escherichia coli (E. coli) is a normal flora and a facultative anaerobic bacterium that plays an essential role in the humoral nutritional metabolism of broiler intestines. This study aims to determine the effect of salix (Salix tetrasperma roxb) extract 50 and 100 mg/litre in drinking water of E. coli colonies in the broiler intestines experiencing heat stress. The sample is 12 intestines of broiler strain MB-90 aged 28 days. The method used a completely randomized design. The study by user three treatments: P0 was not given salix extract in drinking water; P1 and P2 were given salix extract 50 and 100 mg/litre in drinking water. The data were analyzed using One-way ANOVA and were continued with the Duncan test. We can conclude that the salix extract given to broiler chickens with heat stress could affect many E. coli colonies. This result can be seen from the doses given, indicating that the number of E. coli colonies in chicken intestines that experience heat stress depends on the dose of salix extract. The higher dose is given, the fewer colonies in the chicken intestine

Abstract

The purpose of this study was to detect the existence of the Streptococcus sp bacteria in the horses’ dental caries in Aceh Tengah Regency. The caries samples used in this study were taken from 8 horses in Aceh Tengah Regency. The samples were collected using swab on the teeth and then put into Nutrient Broth (NB) media and further were cultured on selective media TSY20B. The bacteria identification was conducted by observing the bacteria’s morphology, Gram staining and biochemical test. The eight samples of the horses’ dental caries showed that Streptococcus sp bacteria could be isolated from all of the collected samples. From this research it could be concluded that Streptococcus sp bacteria was found in the horses’ dental caries and it was one of the causes of the dental caries.

Abstract

This study aims to determine the quality of eggs from Farm, distributors and retail stores. The sample used is 66 eggs with details taken randomly at the farm is 9 eggs, distributors is 27 eggs and retail stores is 30 eggs. The observed egg quality is the value of Haugh unit, Yolk Index and Albumin Index. The data obtained were analyzed descriptively. The results showed that the average Haugh value of egg units in farms, distributors and retail stores were 91, 72 and 7. The Yolk Index values for eggs in farms, distributors and retail stores were 0.45, 0.35. and 0.36. Meanwhile, the albumin index of eggs in farms, distributors and retail stores was 0.074, 0.050 and 0.046. Egg quality in farms, distributors and retail stores is in the category (grade) AA (HU value 72) and A (HU value 60-72). Based on the results of this study, it can be concluded that the quality of eggs traded in Banda Aceh and its surroundings is very good (included in the AA and A grade categories).

Abstract

Candy is a group of snacks high in sugar content, so it has an impact on oral problems such as tooth decay. The use of natural flavors in the form of essential oils in candy which have antimicrobial activity has several obstacles, including not being easily dissolved. Therefore, essential oils need to be formulated to be solvable. One of the ways is by encapsulation using water-soluble coatings such as gum arabic, inulin and maltodextrin using a spray dryer. Product analysis was carried out for solubility tests and growth inhibition tests for Candida albicans and Streptococcus mutans. This research was conducted using two parameters, first parameter was coating materials consisting of 3 levels, namely; gumarabic (A), inulin (B) dan maltodextrin (C) . The second factor is the concentration of patchouli oil based essential oils which consists of 3 levels, namely; 0.01 g (K1), 0,05 g (K2) dan 0,025 g (K3). The results showed that the solubility of essential oils with three coating materials ranged from 44.77% -44.97%. From the analysis of variance, it can be concluded that differences in coating materials, essential oil concentrations and their interactions have a significant effect (P0.05) on inhibiting the growth of Candida albicans and Streptococcus mutans. The best treatment was obtained from the AK1 sample with inhibition against Candida albicans and Streptococcus mutans of 8.6 2.6 and 10 1.6 mm, respectively.

Abstract

This study aims to offer a detailed technical note thorough examination of the pioneering and effectiveness of FMD immediate diagnostic tests in Aceh, Indonesia. We discuss the key components of these tests, including their sensitivity, specificity, and practicality in resource-limited settings. Furthermore, we explore the challenges and opportunities associated with adopting these tests locally and highlight their potential impact on FMD surveillance and control efforts. The findings presented in this technical note contribute to the ongoing efforts to combat FMD in Aceh, providing valuable insights into using rapid diagnostic tests as a vital tool in disease management strategies. The results showed that the FMD rapid diagnostic test kit has limited sensitivity in detecting early clinical signs of FMD in cattle in Aceh. Therefore, Further examination with laboratory molecular detection methods is needed.

Abstract

Pathogenic Escherichia coli is a type of bacteria that has the ability to cause disease in hosts, including humans and animals, although in general E. coli acts as a normal flora in the digestive tract. This study aimed to detect the presence of pathogenic E. coli in the intestines of ducks sold at Lambaro Market, Aceh Besar. E. coli is a normal flora bacteria in the digestive tract of animals and humans, but some strains are pathogenic and can cause zoonotic diseases. The presence of pathogenic E. coli in poultry products has the potential to be a source of infection for humans through consumption of contaminated food. This study used 15 duck intestine samples obtained from cecum, to identify the presence of pathogenic E.coli. The results showed that 13 samples produced clear colonies on Sorbitol MacConky Agar (SMAC), indicating the inability of bacteria to ferment sorbitol, which correlated to the presence of pathogenic E. coli. In contrast, two samples showed pink colonies indicating the presence of nonpathogenic E. coli. The conclusion of this study was the cecum of ducks sold at Lambaro Market, Aceh Besar positive for pathogenic E. coli.
