Abstract

Abstract

This study aims to investigate the potential of chalcone derivatives as antimalarial agents. The structure of the  chalcone derivatives was designed by inserting amino substituents on acetophenone and methoxy variants on benzaldehyde to  produce three amino chalcone derivatives (C1, C2, and C3). The synthesis was carried out by carrying out the Claisen-Schmidt  condensation reaction with NaOH 40% as catalyst, resulting in compound yields ranging from 66% - 83%. The structure of the  three compounds was determined by FTIR, MS, and 1H-NMR spectroscopy techniques, which confirmed that the compounds  had structures that were in line with the desired molecular structure. The antimalarial activity test was carried out by inhibiting  the heme polymerization process into hemozoin (β-hematin) using hydroxychloroquine sulfate as a positive control. Absorption  measurements were carried out at two different wavelengths, namely 415 nm and 630 nm. The results of the IC50 antimalarial  activity of the three compounds (C1, C2, C3) were obtained respectively at 227.61; 115.18; 260.01 µg/mL and positive control of  184.98 µg/mL. From these results, it was found that compound C2 showed better antimalarial activity compared to the other two  compounds and positive control.

Abstract

Measuring workload plays a crucial role in ensuring optimal performance, employee welfare, and efficient resource allocation. The Workload Indicator Staffing Need (WISN) method has been widely used to assess workload and workforce needs. This systematic literature review aims to evaluate the applicability, reliability, and validity of the WISN method in the context of health services. A systematic search of relevant databases (PubMed & Google Scholar) was conducted to identify studies published between 2014 and 2024, focusing on the application of the WISN method in healthcare. Adopting the PRISMA methodology, 10 articles were selected from an initial 200 articles based on their relevance and methodological quality. This study shows that the WISN method has been effectively applied in various health service contexts, such as hospitals, primary service centers, and clinics. This method offers a structured approach to determining workforce needs based on workload, thus facilitating the efficient allocation of resources. However, a number of studies have highlighted challenges related to data accuracy and the adaptability of the WISN method to various health service conditions. Therefore, the development and integrated approach to the WISN method is needed to balance with various other workload measurement methods to improve reliability and validity.

Abstract

This study utilized in vitro and in silico methods to assess the antifungal efficacy of synthesized pyrazoline derivatives (4a–e) against Candida species. The compounds were produced by a one-pot process and structurally analyzed using spectroscopic methods. The antifungal efficacy was evaluated against C. albicans, C. glabrata, and C. krusei using minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) assays. Among the derivatives, compound 4e exhibited potent antifungal action, displaying MIC values similar to ketoconazole. Molecular docking and pharmacophore modeling have shown that 4e interacts efficiently with critical residues of lanosterol 14α-demethylase (CYP51). The density functional theory (DFT) study indicated advantageous electrical characteristics, while molecular dynamics simulations validated the structural stability of the 4e–CYP51 complex, evidenced by low RMSD and RMSF values, along with an MM/GBSA binding energy comparable to that of ketoconazole. A robust association between binding energy and MIC substantiates the predictive use of computational data. The results suggest that compound 4e replicates the binding characteristics of ketoconazole and may be a viable candidate for antifungal medication development. This integrative strategy reinforces the justification for additional optimization and preclinical assessment of pyrazoline-based antifungal drugs aimed at CYP51.

Abstract

Because it directly affects the level of viral suppression and the patient's quality of life, adherence to antiretroviral (ART) medication is an important component in HIV/AIDS treatment. According to many studies, high ART adherence is correlated with better physical, psychological, and social quality of life. Many factors can affect adherence, including therapy regimen, drug side effects, patient mental condition, and economic and social issues. This study was conducted based on six studies published in scientific journals to evaluate the relationship between HIV patients' quality of life and their adherence to antiretroviral drugs. This review used publications from the last ten years collected from the Publish or Perish and PubMed databases using the keywords adherence, antiretroviral, quality of life, and HIV. Articles that discussed the relationship between ART adherence and HIV patients' quality of life used quantitative or qualitative methods with valid measurement tools. The results showed that patients with an adherence rate of at least 90% had better viral suppression and better quality of life. The main factors that increased adherence were simpler ART regimens, milder side effects, support from health workers, and patient education. ART side effects, mental health problems (such as depression and anxiety), economic issues, and access are issues that hinder adherence

Abstract

Abstract: In recent years, the pharmaceutical and healthcare industries have undergone significant changes, marked by increasing competition among service providers, including pharmacies. Pharmacies must adapt their marketing plans in response to regulatory changes, service digitization, and increasing public health awareness. The purpose of this study is to conduct a systematic review of the existing literature on the relationship between pharmacy turnover success and marketing mix tactics. Databases including Google Scholar, PubMed, and Sciencedirect were searched, analyzing English and Indonesian-language publications published between 2015 and 2025 using the PRISMA method for systematic literature review. Of the 745 articles found, five were eligible for inclusion and further analysis. The results indicate that the key elements of the 7Ps of marketing—competitive pricing, product availability, and high-quality service involving both human resources and processes—have a significant impact on purchasing decisions and consumer loyalty. The study suggests that digital-based marketing initiatives and consumer education are important complementary elements in improving pharmacy performance. Overall, this study concludes that to maximize pharmacy turnover and competitiveness in the ever-changing healthcare environment, the 7P marketing mix must be implemented in an integrated and customer-oriented manner.

Abstract

Abstract. Bacterial keratitis is a corneal infection that may lead to pain, visual impairment, and even permanent blindness if not promptly and appropriately treated. In outpatient settings, empirical antibiotic therapy is often chosen as the initial management because culture and sensitivity results require more time, whereas delays in therapy may worsen the clinical condition. This study aims to evaluate the effectiveness of empirical antibiotic therapy on clinical improvement and follow-up visits in patients with bacterial keratitis. A systematic literature review was conducted by searching PubMed and Google Scholar for articles published between 2015 and 2025. From 242 articles identified, 12 met the inclusion criteria and were analyzed further. The findings revealed that moxifloxacin and ciprofloxacin were the most frequently prescribed topical antibiotics, with treatment durations ranging from 7 to 21 days. Overall, empirical therapy was shown to accelerate clinical improvement, particularly within the first week of treatment. However, evidence supporting a reduction in follow-up visit frequency remains limited and presents varied results across studies, potentially due to differences in study design and patient conditions. Factors such as treatment regimen, duration of therapy, disease severity, and patient characteristics were also reported to influence the variability in outcomes. In conclusion, empirical antibiotic therapy appears to be clinically effective for bacterial keratitis. Nevertheless, to obtain a more comprehensive understanding of its impact on follow-up visits and implications for outpatient services in Indonesia, further research with larger sample sizes and prospective study designs is required.

Abstract

Peroxisome Proliferator-activated Receptor (PPAR-γ) protein is one of the target proteins for insulin sensitivity therapy in Type 2 DM. PPAR-γ has a key role as a nuclear receptor that regulates the expression of several metabolism-related genes. This research aims to   synthesize a novel   benzenesulfonylurea-substituted   pyridazinone     derivative, namely (E)-N'-(1-(4-(3-(4-methoxyphenyl)-6-oxopyridazin-1(6H)- yl)phenyl)ethylidene)-4-methylbenzenesulfonohydrazide (8) and predicted it activity as the PPAR-γ agonist using a molecular docking approach and ADMET profiles. The compound 8 was obtained through a Schiff base condensation reaction between compound 6, p-tosyl hydrazine, and a glacial acetic acid catalyst using monowave. The purity of the compound was determined by TLC test, and melting point measurement. The structure was confirmed through FTIR, 1H-NMR, C-NMR and HRMS analysis. Molecular docking studies were carried out on the crystal structure of the human PPAR-γ Ligand Binding Domain target protein in complex with the α-aryloxyphenyl acetic acid agonist (PDB ID 1ZEO). The results of the docking show that compound 8 has a lower binding free energy than rosiglitazone (positive control) with a free energy value (S score) = -13.513 kcal/mol and -8.3089 kcal/mol, respectively. Compound 8 can form two hydrogen bonds with residues His323 and Ser289, π-π interactions with Phe363 and π-H interactions with Cys285.  The interactions are similar to the interaction between the native ligand agonists α-aryloxyphenyl acetic acid and rosiglitazone with the target protein. Furthermore, compound 8 is predicted to have a moderate ADME profile. The results support that compound 8 can be developed as a PPAR-γ agonist candidate for the antidiabetic therapeutic agent.
